Dystrophin deficiency markedly increases enterovirus-induced cardiomyopathy: a genetic predisposition to viral heart disease.

Both enteroviral infection of the heart and mutations in the dystrophin gene can cause cardiomyopathy. Little is known, however, about the interaction between genetic and acquired forms of cardiomyopathy. We previously demonstrated that the enteroviral protease 2A cleaves dystrophin; therefore, we hypothesized that dystrophin deficiency would predispose to enterovirus-induced cardiomyopathy. We observed more severe cardiomyopathy, worsening over time, and greater viral replication in dystrophin-deficient mice infected with enterovirus than in infected wild-type mice. This difference appears to

Insulin Resistance Exacerbates Genetic Predisposition to Nonalcoholic Fatty Liver Disease in Individuals Without Diabetes.

The accumulation of excess fat in the liver (hepatic steatosis) in the absence of heavy alcohol consumption causes nonalcoholic fatty liver disease (NAFLD), which has become a global epidemic. Identifying metabolic risk factors that interact with the genetic risk of NAFLD is important for reducing disease burden. We tested whether serum glucose, insulin, insulin resistance, triglyceride (TG), low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, body mass index (BMI), and waist-to-hip ratio adjusted for BMI interact with genetic variants in or near the patatin-like phospho

Frailty, genetic predisposition, and incident chronic kidney disease.

Frailty is common among individuals with chronic kidney disease (CKD), whereas its impact on incident CKD risk remains unknown. This study aimed to prospectively evaluate the association between frailty and incident CKD risk, exploring the potential modification role of genetic risk factors (GRS). A cohort of 275,442 UK Biobank participants (mean age 55.3 ± 8.1 years, 43.4% male) without CKD were included. Physical frailty was defined by Fried Frailty phenotypes (FP) and Rockwood Frailty Index (FI). New-onset CKD was identified through hospital inpatient records and death register. GRS for CKD

[Molecular diagnostics of renal diseases with underlying genetic predisposition].

The genetic basis for dysplasia and tumors of the kidney has increasingly become the subject of cytogenetic and molecular genetic investigations over the last decade. For that reason, it is now possible to define the risk of disease recurrence more precisely in families with kidney diseases caused by genetic alterations. The relevant genes and the mutations have been identified for most of these diseases and genetic diagnostics are possible. However, it is necessary to evaluate in each individual case whether genetic diagnostics are reasonable. This will be discussed for polycystic renal disea

Genetic predisposition to smoking in relation to 14 cardiovascular diseases.

AIMS: The aim of this study was to use Mendelian randomization (MR) to determine the causality of the association between smoking and 14 different cardiovascular diseases (CVDs). METHODS AND RESULTS: Our primary genetic instrument comprised 361 single-nucleotide polymorphisms (SNPs) associated with smoking initiation (ever smoked regularly) at genome-wide significance. Data on the associations between the SNPs and 14 CVDs were obtained from the UK Biobank study (N = 367 643 individuals), CARDIoGRAMplusC4D consortium (N = 184 305 individuals), Atrial Fibrillation Consortium (2017 dataset; N =

Preimplantation diagnosis for diseases with genetic predisposition and nondisease testing.

Preimplantation genetic diagnosis has become a method of choice in genetic practices. The present experience includes thousands of clinical cases of preimplantation genetic diagnosis, demonstrating the safety, accuracy and reliability of the technique and its clinical relevance for those at-risk couples who cannot accept traditional methods for prenatal diagnosis and termination of pregnancy. Most recently, preimplantation genetic diagnosis has been performed for late-onset disorders, such as genetic predisposition to cancers, Alzhelmer's disease and nondisease testing involving human leukocyt

[Hereditary neoplastic diseases (genetic predisposition and cancer syndromes)].

After one decade of molecular genetic studies, little doubt remains that cancer is a genetic disease. While mutations in somatic cells may cause cancer, they don't carry forward to the next generation. In fact, most cancers arise in this way. In some individuals mutations in cancer related genes may be present at the constitutional level, inherited from one of the parents. An individual who carries a mutant allele of an inherited cancer gene has a variable risk of cancer that is influenced by other genetic and non-genetic factors (such as other cellular genes, dietary, lifestyle, and environme

Genetic predisposition to Kawasaki disease and the risk of coronary atherosclerosis: a Mendelian randomisation study.

BACKGROUND: Kawasaki disease is linked to coronary artery abnormalities, yet whether this reflects a direct causal relationship with atherosclerosis remains unclear due to potential confounding factors. This study aimed to investigate, using two-sample Mendelian randomisation, whether a genetic predisposition to Kawasaki disease is associated with subsequent atherosclerosis. METHODS: Instrumental variables were single-nucleotide polymorphisms significantly associated with Kawasaki disease from a large-scale genome-wide association study (119 cases, 6071 controls). Summary data for coronary at

Human papillomavirus disease in GATA2 deficiency: a genetic predisposition to HPV-associated female anogenital malignancy.

OBJECTIVE: Patients with pathogenic variants in the GATA Binding Protein 2 (GATA2), a hematopoietic transcription factor, are at risk for human papillomavirus-related (HPV) anogenital cancer at younger than expected ages. A female cohort with GATA2 haploinsufficiency was systematically assessed by two gynecologists to characterize the extent and severity of anogenital HPV disease, which was also compared with affected males. METHODS: A 17-year retrospective review of medical records, including laboratory, histopathology and cytopathology records was performed for patients diagnosed with GATA2

ADAMTS7 and ZC3HC1 Share Genetic Predisposition to Coronary Artery Disease and Large Artery Ischemic Stroke.

Coronary artery disease (CAD) and ischemic stroke (IS) are commonly considered distinct disease phenotypes. However, there is some evidence in favor of a degree of overlap between genetic susceptibility to CAD and genetic risk factors for IS. In the present study, we aimed to examine the role of two single-nucleotide polymorphisms (SNPs), rs3825807 and rs11556924, located in the ADAMTS7 and ZC3HC1 genes, respectively, associated with CAD in published GWASs in European populations and their possible contribution to the development of coronary atherosclerosis and cerebral LA atherosclerosis in a

Genetic predispositions and nutritional strategies: a review of nutrigenomics in Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disease that manifests motor and non-motor symptoms due to the progressive loss of dopaminergic neurons in the substantia nigra. While its pathogenesis is multifactorial, with genetic as well as environmental components, more and more evidence suggests that nutrition and diet play an important role in regulating both the development and progression of the disease through crosstalk with genetics, a term known as nutrigenomics. This review discusses genetics and nutrition in PD, emphasizing putative mechanisms underlying neurodegeneration, includin

Association of a Healthy Lifestyle with Risk of Parkinson's Disease and Genetic Predisposition.

INTRODUCTION: Parkinson's disease (PD) is the second most common neurodegenerative disease with largely unknown etiology. Evaluating the association between a healthy lifestyle with PD and genetic risk is necessary. METHODS: The study included 438,241 participants from the UK Biobank, with lifestyle information collected via baseline questionnaires. Polygenic risk scores (PRS) were divided into quartiles. The healthy lifestyle, including alcohol consumption, body mass index, physical activity, sleep duration, sedentary time, social connections, and diet, was categorized into favorable (scorin

Genetic predisposition to celiac disease in Kazakhstan: Potential impact on the clinical practice in Central Asia.

BACKGROUND: Celiac disease (CD) is a systemic immune-mediated disorder developing in HLA genetically predisposed individuals carrying HLA-DQ2 and/or HLA-DQ8 molecules. Recent evidences supported a predominant importance of HLA-DQB1 locus and, in particular, HLA-DQB1*02 alleles. This diagnosis is poorly considered in Kazakhstan, because of the assumption that CD is not prevalent in this population. OBJECTIVE: To demonstrate that the genetic predisposition to CD in Kazakhstan is not negligible and is actually comparable to Western populations. METHODS: Through the analysis of HLA-DQ genotypes

The CREM gene is involved in genetic predisposition to inflammatory bowel disease in the Tunisian population.

The identification of susceptibility genes for inflammatory bowel disease (IBD) is key to understanding pathogenic mechanisms. Recently, the results of genetic association studies have highlighted many loci that are shared among several autoimmune diseases. We aimed to study the genetic epidemiology of polymorphisms in specific genes previously associated with other autoimmune diseases, namely the CREM, STAT4, STAT5a, Stat5b, and IRF5 genes. Twelve polymorphisms in the CREM, STAT4, STAT5a, Stat5b, and IRF5 genes were genotyped in a cohort of 107 IBD patients (39 Crohn's disease [CD] and 68 ulc
