Research progress in the genetics of hyperuricaemia and gout.

Gout is one of the most common inflammatory arthritis caused by hyperuricaemia, which is affected by both genetic factors and environmental factors. Early researches show that a few of rare monogenic mutations, such as PRPS1 and HPRT1 mutations, lead to abnormal purine anabolism and then cause hyperuricaemia and gout. In recent years, genome-wide association studies (GWAS) have identified dozens of susceptibility loci and/or candidate genes associated with hyperuricemia and gout. Loss-of-function mutations in SLC2A9, SLC22A11, and SLC22A12 cause hereditary hypouricaemia, while their overexpres

[Advances of genetic research on the SPG4 gene].

The hereditary spastic paraplegia (HSP), also known as Strumpell-Lorrain familial spasmodic paraplegia, is a highlighted clinical and genetic heterogeneity disorder with the prevalence of (2-9.6)/100,000. This disorder is characterized by progressive, usually severe spasticity and pyramidal weakness, predominantly in the lower limbs. Inheritance of this disease has been reported to be autosomal dominant (AD), autosomal recessive (AR), or X-linked recessive (XR), with the AD forms of HSP (ADHSP) being the most common type. At least 40 HSP gene loci have been localized and 19 genes have been ide

A forward genetics platform for papillomavirus research.

The inability to perform forward genetics has been a major barrier to human papillomavirus (HPV) research. Here, we report a platform for phenotypic selection of HPV mutants. Our method relies on large "concordant" virus libraries, where each mutant virus particle encapsidates the mutant plasmid that encodes it, thereby directly linking viral genotype to phenotype. This platform can achieve a ∼2500-fold enrichment of an L1 capsid protein mutant from an excess of wild-type virus. To demonstrate that this system could select rare mutants from complex libraries, we selected and validated antibody

Modern research criteria and the genetics of schizophrenia.

The authors assessed the relevance of narrowly defined diagnostic criteria to genetic research in schizophrenia in the nuclear families of 84 chronic schizophrenic probands compared with families of 90 normal control probands. The morbidity risk for narrowly defined schizophrenia in first-degree relatives of patients with the narrow diagnosis was significantly higher than the control rate (3.8% versus 0.3%). The rate of chronic schizophrenia in the relatives of all schizophrenic patients was also significantly higher than the control rate (7.1% versus 0.6%), as was the rate of "spectrum" disor

[Advances in molecular genetic research on Myelodysplastic syndrome].

Myelodysplastic syndrome (MDS) is a chronic hematologic disorder characterized by ineffective hematopoiesis, dysplasia of one or more cell lines with or without definite genetic changes. Its diagnosis requires a comprehensive analysis combining morphology, immunology, cytogenetics, and molecular biology findings. In recent years, the development of second-generation sequencing (NGS) has provided great assistance in exploring the molecular pathogenesis of hematological malignancies and guidance for clinical practice. Mutations of a series of gene involved in RNA splicing, DNA methylation, trans

[Progress in genetic research on primary pulmonary hypertension].

Primary pulmonary hypertension (PPH) consisting of hereditary pulmonary arterial hypertension and idiopathic pulmonary arterial hypertension is an obstructive pulmonary hypertension caused by primary pulmonary artery hyperplasia. Both environmental and genetic factors are involved in the pathogenesis of PPH. Genes associated with TGF-β signaling pathway including BMPR2, ALK1, ENG, SMAD8 and other genes including NFU1, CAV1, KCNK3 and TopBPl have been associated with PPH. In this review, the function and mechanism of the above genes in the pathogenesis PPH are summarized.

[Clinical implications of molecular genetic research in otorhinolaryngology].

Molecular-genetic research in Otolaryngology has seen a rapid advancement during the last ten years, especially in the fields of otology and head and neck tumors. The results of this basic research have now started to be implemented in the clinic. In otology the understanding of auditive function has dramatically improved. The syndromic and non-syndromic forms of hereditary hearing impairment can be subdivided into their underlying genetic defects, as more and more genes are identified. Diagnostic of syndromic hearing loss has been improved and can be done earlier. But the molecular-genetic an

Filippi syndrome is a rare autosomal recessive genetic disorder characterized primarily by syndactyly, microcephaly, distinctive facial features, and intellectual disability. It may also involve multiple systemic manifestations including growth retardation, language disorders, visual impairment, cryptorchidism, abnormal dental and skeletal development, and so forth. The pathogenic gene associated with Filippi syndrome has been identified as CKAP2L, which encodes a mitotic spindle protein localized in the spindle, centrosome, and neural progenitor cells. Deficiency of CKAP2L may disrupt the div

Genetic eye research in Tasmania: a historical overview.

Although considerable recent work on hereditary eye diseases in Tasmanian families has been published, much of this depended on a century of meticulous pedigree collection by earlier clinical researchers. This article reviews some of the historical papers and the importance they have played in gene discovery and understanding of ophthalmic genetics. Tasmanian families have contributed to the identification of genes for X-linked megalocornea, Leber's hereditary optic neuropathy, retinitis pigmentosa, congenital cataract, ptosis, keratoconus, glaucoma and myopia. The true value of the Tasmanian

Current status of genetic research in Rett syndrome.

Rett syndrome is a progressive neurological development syndrome. Evidences for a genetic transmission, but no conclusive data which entirely support the X chromosome involvement, exist. The chromosomal region 11p14-pte has been investigated with polymorphic markers and genes in the region have been sequenced. No sequence divergencies were detected.

Multiracial individuals' perspectives on participating in genetics research.

The vast majority of genetics research is confined to a relatively narrow subset of the global population, limiting the benefits of this research. Recommendations to ameliorate this issue frequently include calls to recruit diverse populations through community engagement, an approach that has been effective in genetics, but has not explicitly included Multiracial individuals, despite this being the fastest-growing population in the United States. As such, this study explored Multiracial individuals' perspectives on: (1) what "Multiracial community" means; (2) concerns regarding ancestry-based

[Advances in genetic research of congenital hypothyroidism in China].

Congenital hypothyroidism (CH), which results from insufficient thyroid hormone biosynthesis, is one of the most common neonatal endocrine disorders. Thyroid dysgenesis and thyroid dyshormonogenesis are the two causes of CH and either one will lead to deficiencies of enzymes during thyroid hormone biosynthesis and insufficient thyroid hormone biosynthesis. Recently, researchers have performed extensive studies on genetics of CH. This paper reviews genes reported to be associated with CH in China.

Factors associated with enrollment in cancer genetics research.

Previous studies have identified low patient accrual in large-scale cancer clinical trials, particularly for underrepresented groups, such as ethnic minorities, females, and patients >65 years. As there have been few studies examining participation in cancer genetics epidemiologic research, our objective was to identify clinical and demographic factors predicting enrollment in these studies. A total of 1,111 patients diagnosed with colorectal cancer presenting to a gastrointestinal oncology clinic were approached to enroll in a study investigating the role of the MSH6 gene in familial colorect
