Thermostable homologues of the periplasmic siderophore-binding protein CeuE from Geobacillus stearothermophilus and Parageobacillus thermoglucosidasius.

Siderophore-binding proteins from two thermophilic bacteria, Geobacillus stearothermophilus and Parageobacillus thermoglucosidasius, were identified from a search of sequence databases, cloned and overexpressed. They are homologues of the well characterized protein CjCeuE from Campylobacter jejuni. The iron-binding histidine and tyrosine residues are conserved in both thermophiles. Crystal structures were determined of the apo proteins and of their complexes with iron(III)-azotochelin and its analogue iron(III)-5-LICAM. The thermostability of both homologues was shown to be about 20°C higher t

Refolding, characterization, and dye decolorization ability of a highly thermostable laccase from Geobacillus sp. JS12.

A putative laccase gene (lacG) from Geobacillus sp. JS12 was cloned and expressed as a fusion protein with six histidine residues in Escherichia coli BL21 (DE3) cells, and the protein was primarily found in inclusion bodies. The resulting insoluble proteins were solubilized with 6 M guanidine HCl and refolded using an on-column refolding procedure. Ni-chelation affinity chromatography found the laccase to be a 30 kDa monomeric protein. Spectrophotometry and electron paramagnetic resonance (EPR) analysis indicated LacG as a multi-copper oxidase, with the usual laccase copper sites, Type 1, 2, a

Structural basis for substrate recognition in l-lysine 6-dehydrogenase from Geobacillus stearothermophilus by Cryo-EM.

l-lysine 6-dehydrogenase (LysDH; EC 1.4.1.18) oxidatively deaminates the ε-amino group of l-lysine. Due to its high substrate specificity, LysDH serves as a valuable tool for l-lysine quantification. However, the molecular basis of this specificity has remained unclear because of the lack of substrate-bound structures. In this study, we determined the cryo-electron microscopy (cryo-EM) structures of LysDH from the thermophilic bacterium Geobacillus stearothermophilus (GstLysDH) in the apo form at 2.9 Å resolution and in complex with NAD+ and l-lysine at 2.5 Å resolution. GstLysDH assembles as

Methanol response of V171S lipase from Geobacillus sp. 42 explored via experimental and in silico approaches.

The stability of lipase in organic solvents is crucial for biocatalytic processes in industrial biotechnology. Previously, we described a thermostable and solvent-tolerant Lip 42 from Geobacillus sp. Despite these features, the wild-type Lip 42 activity deteriorated and became unstable in methanol at high temperatures, limiting its effectiveness in solvent-driven catalysis. This study aims to integrate experimental data with molecular dynamics simulation, free energy landscape (FEL), and principal component analysis (PCA) to explore the structural and dynamic properties of V171S mutant lipase,

Bioinformatics-guided mutagenesis and biochemical characterization of the GDSL lipase GstLipC from Geobacillus stearothermophilus ATCC 7953.

GDSL lipases, members of the SGNH hydrolase superfamily, exhibit broad substrate specificity and catalytic adaptability, making them valuable model enzymes for investigating structure-function relationships under high-temperature conditions. Despite these features, residue-level determinants influencing the catalytic performance of thermophilic GDSL lipases remain insufficiently characterized. In this study, we investigated the effects of rationally selected amino acid substitutions on the catalytic properties of a thermophilic GDSL lipase using an in silico-guided strategy combined with exper

Novel sterilization method of Bacillus atrophaeus and Geobacillus stearothermophilus spores by low concentration chlorine dioxide gas.

Chlorine dioxide (ClO2) is a powerful disinfectant widely regarded as a safe and effective hygienic agent in pharmaceutical plants and other manufacturing facilities that require sterility. However, the efficacy of low concentrations of ClO2 gas on sterilizing spore-forming bacteria remains uncertain. In this study, we investigated the optimal conditions for disinfection of spore-forming bacteria with low concentrations of ClO2 gas using biological indicators. The results showed that 0.05 ppmv (0.13 mg/m3) ClO2 gas at 37±1℃, 86±2%RH sterilized Bacillus atrophaeus in 14 d and Geobacillus stearo

Self-assembly of surface layer protein SbsC from Geobacillus stearothermophilus and common principles of Bacilli S-layers.

S-layers are (glyco)protein coats displayed on the external surface of many bacterial and archaeal species. By combining X-ray crystallography, electron cryo-tomography and SymProFold predictions, we propose the structure of the self-assembled S-layer protein (SLP) SbsC from the Gram-positive bacterium Geobacillus stearothermophilus ATCC 12980. SbsC consists of nine domains arranged like beads on a string. Domains 2 through 7 form a closed ring-like conformation, while the secondary cell wall polymer binding domain 1 and the auxiliary domains 8 and 9 protrude from the ring. Within the assemble

Engineering of Geobacillus kaustophilus lipase for enhanced catalytic efficiency and methanol tolerance in biodiesel production from sunflower oil.

Lipase-mediated biodiesel production offers a sustainable and environmentally friendly alternative to conventional chemical methods. However, enzyme limitations such as low activity, poor thermal stability, and limited solvent tolerance remain challenges. In this study, a lipase from Geobacillus kaustophilus (Gklip) was engineered for improved biodiesel production using molecular docking, molecular dynamics (MD) simulations, and molecular mechanics/generalized born surface area (MM/GBSA) free energy calculations. Five mutants (Y29S, Q114T, F289D, Q184M, and Q114F) were generated via site-direc

Submerged Bioreactor Production of Geobacillus stearothermophilus ATCC 7953 Spores for Use as Bioindicators to Validate Hydrogen Peroxide Inactivation Processes.

In the food and pharmaceutical industries, evaluating the sterilization performance preceding aseptic production processes is of central importance. In the case of hydrogen peroxide sterilization of solid surfaces, bioindicators (BI) consisting of spores of Bacillus atrophaeus or Geobacillus stearothermophilus are used to validate the effectiveness and efficiency of the inactivation procedure. Commercial production of G. stearothermophilus is commonly performed on agar plates, where cultivation and sporulation conditions are not well-defined. Therefore, the produced BI can vary in their resist

Gene replacement of adenylate kinase in the gram-positive thermophile Geobacillus stearothermophilus disrupts adenine nucleotide homeostasis and reduces cell viability.

Thermophilic bacteria are of great value for industry and research communities. Unfortunately, the cellular processes and mechanisms of these organisms remain largely understudied. In the present study, we investigate how the inactivation of adenylate kinase (AK) affects the adenine nucleotide homeostasis of a gram-positive moderate thermophile, Geobacillus stearothermophilus strain NUB3621-R. AK plays a major role in the adenine nucleotide homeostasis of living cells and has been shown to be essential for the gram-negative mesophile Escherichia coli. To study the role of AK in the maintenance
