Continuous fixation of dissolved arsenite from flooded soil by cooperating ferrihydrite with Geobacter sulfurreducens.

The fixed arsenic in soil is easy to be released into the aquatic environment in the form of arsenite (As(III)) with high toxicity and mobility due to the eutrophication of environment under anaerobic conditions. However, As(III) is difficult to be fixed in situ continuously by traditional methods, especially for the most efficient fixation method by iron ores. Based on that Fe(II) could promote the fixation of As(III), this study investigated the possibility that Geobacter sulfurreducens (G. sulfurreducens) cooperates with ferrihydrite to fix released As(III) from flooded soil in a glass colu

Geobacter Autogenically Secretes Fulvic Acid to Facilitate the Dissimilated Iron Reduction and Vivianite Recovery.

Biosynthetic organic matters, such as humus, play important roles in iron and phosphorus cycling in soil and aquatic systems. As an important member of humus, fulvic acid (FA) is ubiquitous in different environmental media, such as water, soil, and sediments. In this study, we fabricated the network among phosphate supply, metabolism pathway of FA, iron reduction, and vivianite recovery at the batch scale. Both the vivianite recovery performance and the content of biosynthetic FA were positively related to the phosphorus dosage. The highest vivianite formation efficiency of 53% was obtained in

Cation-driven envelope dynamics modulate outer membrane vesiculation and extracellular electron transfer in Geobacter.

Geobacter bacteria use conductive pili and redox-active outer membrane vesicles to mediate metal transformations critical to the effectiveness of bioremediation and energy technologies. Mechanistic knowledge into these processes primarily comes from studies with Geobacter sulfurreducens grown in media closely formulated to mirror the mineral chemistry of contaminated sites. Here we show that, although subtle differences in the media's cationic strength did not measurably change permeability, they reprogrammed outer membrane-peptidoglycan crosslinks to modulate vesiculation and envelope functio

Metabolic turnover of cysteine-related thiol compounds at environmentally relevant concentrations by Geobacter sulfurreducens.

Low-molecular-mass (LMM) thiol compounds are known to be important for many biological processes in various organisms but LMM thiols are understudied in anaerobic bacteria. In this work, we examined the production and turnover of nanomolar concentrations of LMM thiols with a chemical structure related to cysteine by the model iron-reducing bacterium Geobacter sulfurreducens. Our results show that G. sulfurreducens tightly controls the production, excretion and intracellular concentration of thiols depending on cellular growth state and external conditions. The production and cellular export of

Coupled mercury-cell sorption, reduction, and oxidation on methylmercury production by Geobacter sulfurreducens PCA.

G. sulfurreducens PCA cells have been shown to reduce, sorb, and methylate Hg(II) species, but it is unclear whether this organism can oxidize and methylate dissolved elemental Hg(0) as shown for Desulfovibrio desulfuricans ND132. Using Hg(II) and Hg(0) separately as Hg sources in washed cell assays in phosphate buffered saline (pH 7.4), we report how cell-mediated Hg reduction and oxidation compete or synergize with sorption, thus affecting the production of toxic methylmercury by PCA cells. Methylation is found to be positively correlated to Hg sorption (r = 0.73) but negatively correlated t

Geobacter lovleyi sp. nov. strain SZ, a novel metal-reducing and tetrachloroethene-dechlorinating bacterium.

A bacterial isolate, designated strain SZ, was obtained from noncontaminated creek sediment microcosms based on its ability to derive energy from acetate oxidation coupled to tetrachloroethene (PCE)-to-cis-1,2-dichloroethene (cis-DCE) dechlorination (i.e., chlororespiration). Hydrogen and pyruvate served as alternate electron donors for strain SZ, and the range of electron acceptors included (reduced products are given in brackets) PCE and trichloroethene [cis-DCE], nitrate [ammonium], fumarate [succinate], Fe(III) [Fe(II)], malate [succinate], Mn(IV) [Mn(II)], U(VI) [U(IV)], and elemental sul

Transformation of carbon tetrachloride by biogenic iron species in the presence of Geobacter sulfurreducens and electron shuttles.

The transformation of carbon tetrachloride (CT) by biogenic iron species produced from the bioreduction of various Fe(III) oxides in the presence of Geobacter sulfurreducens and electron shuttles were investigated. Cysteine and anthraquinone-2,6-disulfonate (AQDS) at concentrations of 0.5mM and 10microM, respectively, were added as the electron shuttles. Addition of electron shuttles enhanced the extent of reduction and rate of ferric oxide reduction. The bioreduction extents of ferric oxides by G. sulfurreducens in the presence of electron shuttles were 22.8-48.3% for ferrihydrite, 6.5-17.2%

Enhanced dechlorination of carbon tetrachloride by Geobacter sulfurreducens in the presence of naturally occurring quinones and ferrihydrite.

The effect of naturally occurring quinones including lawsone (LQ), ubiquinone (UQ), juglone (JQ), and 1,4-naphthoquinone (NQ) on the biotransformation of carbon tetrachloride (CT) in the presence of Geobacter sulfurreducens and ferrihydrite was investigated. AQDS was used as the model compound for comparison. The reductive dissolution of ferrihydrite by G. sulfurreducens was enhanced by AQDS, NQ, and LQ. However, addition of UQ and JQ had little enhancement effect on Fe(II) production. The bioreduction efficiency and rate of ferrihydrite was highly dependent on the natural property and concent

Fast removal of toxic hexavalent chromium from an aqueous solution by high-density Geobacter sulfurreducens.

Hexavalent chromium (Cr(VI)) is a carcinogenic compound that can be removed from contaminated sites by the activity of metal-reducing bacteria. The model bacterium Geobacter sulfurreducens reduces Cr(VI) to less toxic Cr(III) and accumulates Cr ions intracellularly. However, this process is usually slow with small concentrations of Cr(VI) removed in a matter of days. Here, high-density G. sulfurreducens cultures were tested for the capacity to remove Cr(VI) readily. With an initial G. sulfurreducens density of 5.8 × 108 cells ml-1, 99.0 ± 0.8% of 100 mg l-1 Cr(VI) was removed after 20 min. Wit

Geobacter metallireducens-evaluation of its sustainability in nitrate-reducing Fe(II) oxidation under autotrophic conditions.

Bacterial nitrate (NO3-) -reducing Fe(II) oxidation is often regarded as an autotrophic process, although its long-term sustainability is rarely demonstrated. Here, we assessed whether Geobacter metallireducens (ATCC 53774) can sustain chemolithoautotrophic nitrate-reducing Fe(II) oxidation. Cultures were incubated with Fe(II) and NO3- as electron donor and acceptor, respectively, and CO2 as the sole carbon source. Negative controls contained either (1) electron acceptor, (2) donor or (3) no substrates. We found that first-generation cultures transferred from a washed NO3-/acetate-grown precul

Spatial and temporal distributions of Geobacter lovleyi and Dehalococcoides spp. during bioenhanced PCE-NAPL dissolution.

The spatial and temporal distributions of multiple reductively dechlorinating bacteria were simultaneously assessed in a one-dimensional sand column containing a tetrachloroethene (PCE) nonaqueous phase liquid (NAPL) source and associated plume zones. The column was uniformly inoculated with a PCE-to-ethene dechlorinating microbial consortium that contained Dehalococcoides spp., Dehalobacter spp., and Geobacter lovleyi strain SZ. Geobacter and Dehalococcoides populations grew and colonized the column material, including the mixed-NAPL (0.25 mol/mol PCE in hexadecane) source zone. In contrast,

Effects of redox-modified biochar on mercury reduction and methylation on electron transfer in Geobacter sulfurreducens PCA.

Geobacterplays a key role in mercury (Hg) methylation and reduction in rice fields.Biochar boosts microbial electron transfer, but its impact on Hg reduction and methylation remains unclear. This study investigates how oxidative (OBC) and reductive (RBC) biochar influence Hg reduction and methylation by Geobacter sulfurreducens PCA. OBC increased electron donating capacity (EDC) but reduced electron accepting capacity (EAC), while RBC decreased electric resistance. Correlation analysis revealed biochar's electron exchange capacity (EEC) positively correlated with -OH (r = 0.73*), O-CO (r = 0.8

Influence of fumarate on interspecies electron transfer and the metabolic shift induced in Clostridium pasteurianum by Geobacter sulfurreducens.

AIMS: In previous studies, it was demonstrated that co-culturing Clostridium pasteurianum and Geobacter sulfurreducens triggers a metabolic shift in the former during glycerol fermentation. This shift, attributed to interspecies electron transfer and the exchange of other molecules, enhances the production of 1,3-propanediol at the expense of the butanol pathway. The aim of this investigation is to examine the impact of fumarate, a soluble compound usually used as an electron acceptor for G. sulfurreducens, in the metabolic shift previously described in C. pasteurianum. METHODS AND RESULTS: E

Relationship of electrochemical performance and biofilm development of Desulfuromonas acetexigens and Geobacter sulfurreducens in microbial electrolysis cells.

Desulfuromonas acetexigens has gained attention as a biocatalyst in microbial electrolysis cells (MECs) due to its inability to utilize hydrogen as an electron donor, which favors beneficial Coulombic efficiencies (CE). In this study, the electrochemical performance and biofilm morphology of D. acetexigens were compared with the model organism Geobacter sulfurreducens in flow cell MECs. Biofilm morphology was assessed non-invasively via optical coherence tomography (OCT), providing insight into quantitative parameters, including spatially resolved thickness, biovolume and anode surface coverag

A Simple and Low-Cost Procedure for Growing Geobacter sulfurreducens Cell Cultures and Biofilms in Bioelectrochemical Systems.

Anaerobic microorganisms play a central role in several environmental processes and regulate global biogeochemical cycling of nutrients and minerals. Many anaerobic microorganisms are important for the production of bioenergy and biofuels. However, the major hurdle in studying anaerobic microorganisms in the laboratory is the requirement for sophisticated and expensive gassing stations and glove boxes to create and maintain the anaerobic environment. This appendix presents a simple design for a gassing station that can be used readily by an inexperienced investigator for cultivation of anaerob

Detection and quantification of Geobacter lovleyi strain SZ: implications for bioremediation at tetrachloroethene- and uranium-impacted sites.

Geobacter lovleyi strain SZ reduces hexavalent uranium, U(VI), to U(IV) and is the first member of the metal-reducing Geobacter group capable of using tetrachloroethene (PCE) as a growth-supporting electron acceptor. Direct and nested PCR with specific 16S rRNA gene-targeted primer pairs distinguished strain SZ from other known chlorinated ethene-dechlorinating bacteria and closely related Geobacter isolates, including its closest cultured relative, G. thiogenes. Detection limits for direct and nested PCR were approximately 1 x 10(6) and 1 x 10(4) 16S rRNA gene copies per mul of template DNA,

Complexation by cysteine and iron mineral adsorption limit cadmium mobility during metabolic activity of Geobacter sulfurreducens.

Cadmium (Cd) adversely affects human health by entering the food chain via anthropogenic activity. In order to mitigate risk, a better understanding of the biogeochemical mechanisms limiting Cd mobility in the environment is needed. While Cd is not redox-active, Cd speciation varies (i.e., aqueous, complexed, adsorbed), and influences mobility. Here, the cycling of Cd in relation to initial speciation during the growth of Geobacter sulfurreducens was studied. Either fumarate or ferrihydrite (Fh) was provided as an electron acceptor and Cd was present as: (1) an aqueous cation, (2) an aqueous c

Integrated analysis of transcriptomic and protein-protein interaction data reveals cadmium stress response in Geobacter sulfurreducens.

Bacteria have evolved several mechanisms to resist Cd toxicity, which are crucial for Cd detoxication and have the potential to be used for bioremediation of Cd. Geobacter species are widely found in anaerobic environments and play important roles in natural biogeochemical cycles. However, the transcriptomic response of Geobacter sulfurreducens under Cd stress have not been fully elucidated. Through integrated analysis of transcriptomic and protein-protein interaction (PPI) data, we uncovered a global view of mRNA changes in Cd-induced cellular processes in this study. We identified 182 differ

Bioremediation of Cr (VI) contaminated groundwater by Geobacter sulfurreducens: Environmental factors and electron transfer flow studies.

In this study, the removal of Cr (VI) was examined in the presence of bio-produced Fe (II) from hematite, sulfate and dissolved organic matter by Geobacter sulfurreducens. The adaptation results of G. sulfurreducens showed that cells growth was stimulated up to 576 μM of Cr (VI) concentration. The first-order rate and electron transfer rate in each step during Cr (VI) reduction by G. sulfurreducens in the presence of hematite was clearly modeled and calculated. For Cr (VI) reduction rate, both separately dissolved and adsorbed bio-produced Fe (II) were faster than G. sulfurreducens although bi

Contrasting Effects of Dissolved Organic Matter on Mercury Methylation by Geobacter sulfurreducens PCA and Desulfovibrio desulfuricans ND132.

Natural dissolved organic matter (DOM) affects mercury (Hg) redox reactions and anaerobic microbial methylation in the environment. Several studies have shown that DOM can enhance Hg methylation, especially under sulfidic conditions, whereas others show that DOM inhibits Hg methylation due to strong Hg-DOM complexation. In this study, we investigated and compared the effects of DOM on Hg methylation by an iron-reducing bacterium Geobacter sulfurreducens PCA and a sulfate-reducing bacterium Desulfovibrio desulfuricans ND132 under nonsulfidic conditions.
