Presence of Protozoan Viruses in Vaginal Samples from Pregnant Women and Their Association with Trichomoniasis.

This study was conducted in Armenia and included 32 pregnant women with TV infection and 30 healthy controls. The vaginal virome includes viruses that infect human cells and unicellular eukaryotes such as Trichomonas vaginalis (TV). Among these are Trichomonas vaginalis viruses (TVVs), double-stranded RNA viruses from the Totiviridae family, and giant DNA viruses that replicate in protozoa. This study investigated the presence of TVVs and giant protozoan viruses in pregnant women with trichomoniasis in Armenia and explored their potential associations with adverse pregnancy outcomes. Reassessing viral origins and evolutionary placement in the tree of life.

The quest to fit all cellular beings in one picture frame as the universal Tree of Life (ToL) has always been a daunting task for evolutionary biologists. Over the decades, ToL has emerged from a dichotomous topology to its present form with three domains; bacteria, archaea and eukarya. But this phylogenetic placement is also questionable due to the miscellaneous nature of certain housekeeping genes, horizontal gene transfers (HGT), and also due to incomplete pathways of pathogenic organisms. Furthermore, the ambiguous nature of viruses has always puzzled researchers about their placement in T

Looking at protists as a source of pathogenic viruses.

In the environment, protozoa are predators of bacteria and feed on them. The possibility that some protozoa could be a source of human pathogens is consistent with the discovery that free-living amoebae were the reservoir of Legionella pneumophila, the agent of Legionnaires' disease. Later, while searching for Legionella in the environment using amoeba co-culture, the first giant virus, Acanthamoeba polyphaga mimivirus, was discovered. Since then, many other giant viruses have been isolated, including Marseilleviridae, Pithovirus sibericum, Cafeteria roenbergensis virus and Pandoravirus spp. T

Integrative genomic analyses support the division of the extended Asfarviridae clade into multiple viral families.

Giant viruses continue to challenge our understanding of virology, blurring boundaries of what a virus can be. The so-called "extended Asfarviridae"- such as African swine fever virus, faustovirus, kaumoebavirus, pacmanvirus, and AbALV-has long puzzled taxonomists. By integrating comparative genomics, pangenomics, and phylogenomics, we show these lineages are deeply divergent, forming multiple families rather than one. This work underscores the huge unexplored diversity of giant viruses and demonstrates the value of integrative genomic analyses for proper taxonomic delineation.

Diving into the hidden viral world of marine protists.

As the most abundant biological entities in the ocean, viruses of microbes play important roles in regulating host population dynamics and influencing biogeochemical cycles. Metagenomic surveys have revealed an astounding reservoir of viral genetic diversity in single-celled marine eukaryotes known as protists, but the vast majority of these viruses have not been directly observed, and information about their protist hosts remains fragmentary. The 2023 discovery of mirusviruses provides a striking example, whereby metagenomic surveys of samples collected by the Tara Oceans expedition led to th

Megamimivirus double-stranded DNA linear genomes flanked by highly diverse terminal inverted repeats.

UNLABELLED: Giant viruses have fundamentally expanded our understanding of virology by challenging the conventional boundaries of both virion size and genome complexity. However, the scarcity of isolates has left many of their unique biological features unexplored. Here, we report the isolation and characterization of four new giant virus species belonging to the subfamily Megamimivirinae, sampled from distinct environments across China. Among these, Megavirus daqingense is the first giant virus isolated from an oil reservoir; it exhibits virion stability under high salinity, chloroform exposu

Single-cell genomics reveals complex microbial and viral associations in ciliates and testate amoebae.

Protists play important roles in nutrient cycling across ecosystems, yet the composition and function of their associated microbiomes remain poorly studied. Here, we use cultivation-independent single-cell isolation and genome-resolved metagenomics to investigate the microbiomes and viromes of more than 100 uncultivated ciliates and amoebae from diverse environments. Our findings reveal unique microbiome structures and complex associations with bacterial symbionts and viruses, with stark differences between ciliates and amoebae. We recover 117 microbial genomes affiliated with known eukaryotic

Taxonomic update for the giant algal chloroviruses of the family Phycodnaviridae.

Chloroviruses were first described in the late 1970s, and this opened a new area in virology. These giant DNA viruses, capable of infecting unicellular eukaryotic algae, are ubiquitous in inland waters and have been isolated from various locations worldwide, revealing extensive and complex genetic diversity. These viruses belong to the genus Chlorovirus, family Phycodnaviridae, order Algavirales. Currently, only six species are officially recognized by the International Committee on Taxonomy of Viruses (ICTV), but recent data from our group suggest the existence of many others. In this work, w

Amoebal Tubulin Cleavage Late during Infection Is a Characteristic Feature of Mimivirus but Not of Marseillevirus.

Mimivirus and Marseillevirus infections of Acanthamoeba castellanii, like most other viral infections, induce cytopathic effects (CPE). The details of how they bring about CPE and to what extent and how they modify the host cytoskeletal network are unclear. In this study, we compared the rearrangement of the host cytoskeletal network induced by Mimivirus and Marseillevirus upon infection. We show that while both Mimivirus and Marseillevirus infections of A. castellanii cells cause retraction of acanthopodia and depolymerization of the host actin filament network, the Mimivirus infection also r

Facing the Unknown: Idiopathic Giant Cell Hepatitis.

Giant cell hepatitis is a rare infiltrative disease associated with several viruses, drugs, malignancies, and autoimmune conditions. To date, treatment aims at controlling the underlying etiology, and there are limited data on the clinical course and treatment of idiopathic cases. We present a case of idiopathic giant cell hepatitis in an otherwise healthy adult man and review the literature regarding treatment and outcomes in this population.

Questions on unusual Mimivirus-like structures observed in human cells.

Background: Mimiviruses or giant viruses that infect amoebas have the ability to retain the Gram stain, which is usually used to colour bacteria. There is some evidence suggesting that Mimiviruses can also infect human cells. Guided by these premises, we performed a routine Gram stain on a variety of human specimens to see if we could detect the same Gram positive blue granules that identify Mimiviruses in the amoebas.  Methods: We analysed 24 different human specimens (liver, brain, kidney, lymph node and ovary) using Gram stain histochemistry, electron microscopy immunogold, high resolution

The polinton-like supergroup of viruses: evolution, molecular biology, and taxonomy.

SUMMARYPolintons are 15-20 kb-long self-synthesizing transposons that are widespread in eukaryotic, and in particular protist, genomes. Apart from a transposase and a protein-primed DNA polymerase, polintons encode homologs of major and minor jelly-roll capsid proteins, DNA-packaging ATPases, and proteases involved in capsid maturation of diverse eukaryotic viruses of kingdom Bamfordvirae. Given the conservation of these structural and morphogenetic proteins among polintons, these elements are predicted to alternate between transposon and viral lifestyles and, although virions have thus far no

Comparison of the ultrastructure of a malignant tumor of the mandible containing giant cells with Paget's disease of bone.

Distinction between lesions of the jaws containing giant cells can be difficult. This study reports the results of the study of the ultrastructure of 30 patients with Paget's disease in comparison with a patient diagnosed as having malignant giant cell tumor of the mandible. The ultrastructure of the malignant giant cell tumor of the mandible was found to contain nuclear inclusions morphologically identical with the 12-15 nm tubules characteristic of the nuclei of osteoclasts in Paget's disease of bone. No clinical or laboratory signs of Paget's disease was found in this patient. A search of t

Elucidating the protein interaction network of one of the largest icosahedral capsids in the virosphere.

Giant viruses challenge traditional boundaries of virology with their large particle sizes, complex genomes, and unique replication strategies. Yet, despite its 750 nm diameter and incorporation of dozens of proteins, the mimivirus virion retains an icosahedral symmetry, a trait often associated with smaller viruses. The functional roles and interactions of most proteins composing such complex icosahedral particles remain elusive. Here, we dissect the spatial and functional organization of mimivirus morphogenesis by integrating bioinformatics, genetics, and interactomics. We performed protein

Rapid Diversification of a Natural Heterosigma akashiwo Virus Population during a Host Bloom.

Despite the ecological importance of viruses, our understanding of their evolutionary dynamics in natural environments remains limited. This gap is particularly pronounced for giant dsDNA viruses of the phyla Nucleocytoviricota and Mirusviricota. Knowledge on their population genetic dynamics is mostly derived from a small number of laboratory-based experiments, while patterns in nature are rarely observed. To overcome this limitation, we traced the genetic structure and transcription status of Heterosigma akashiwo virus (HaV) using high-frequency, time-resolved sampling during a host bloom in

Polykaryocytosis induced by certain arboviruses in monolayers of BHK-21-528 cells.

Multinucleated giant cell formation in a clone of BHK-21 cells, BHK-21-528, was tested with certain arboviruses. Eleven out of 19 viruses tested, Chikungunya, Getah, Sagiyama, Sindbis, Western equine encephalitis, St. Louis encephalitis, Japanese encephalitis, West Nile, Bunyamwera, Germiston and California encephalitis virus induced cell fusion after infection. All cases where giant cells were observed during the experiments, involved the B type cell fusion (fusion from within).

Pantanal virophage: a new virophage species associated with a moumouvirus and a transpoviron, expanding the genus Sputnikvirus.

Amoeba-infecting giant viruses, such as mimiviruses, challenged paradigms of virology since their discovery. Their extensive and complex genomes and particle structures can interact with virophages and transpovirons, revealing unique aspects of the viral ecology and evolution. Here, we describe the isolation of Pantanal virophage, a new virophage obtained from Brazilian pantanal biome. The virophage was found in association with moumouvirus pantanense and a transpoviron sequence. Sequencing and genomic analysis revealed that Pantanal virophage presents a 17,964 bp genome with 21 coding sequenc

Measles pneumonia in a newborn.

We report a 21-day-old preterm infant who had severe respiratory distress of 6 days' duration and whose lungs revealed a giant cell pneumonia at necropsy. Measles antigen was demonstrated in mononuclear and multinucleated epithelial cells of the lung by immunoperoxidase staining. We recommend the immunostaining procedure to differentiate measles from other viruses, such as parainfluenza 2 or 3, and respiratory syncytial virus, all of which may produce giant cell pneumonias.

Usurpativirus massiliensis, a new giant virus related to clandestinovirus.

Giant virus discovery in 2003 revolutionized the virus paradigm. In 2015, we introduced a new host, Vermamoeba vermiformis, that led to the discovery of Faustovirus, Orpheovirus and two giant viruses (Clandestinovirus and a faustovirus) having distinct cytopathic effects despite being co-isolated from the same environmental sample and culture. This raised concerns about the possibility that the most "discreet" virus be overlooked. Here we report on Usurpativirus, a new giant virus closely related to Clandestinovirus and co-isolated with Faustovirus LCD7 in V. vermiformis. Its non-lytic replica
