Aspergillosis is a disease caused by the opportunistic pathogen Aspergillus fumigatus and other related fungi. It occurs mainly in immunosuppressed people and causes very high mortality rates. A fumigatus and other pathogenic fungi have been shown to produce a metabolite, gliotoxin, which has immunosuppressive properties in vitro, but little is known about its in vivo activity. Here we report that gliotoxin has increased toxicity in mice after irradiation. A single injection of gliotoxin delayed the recovery of immune cells after immunosuppression by sublethal irradiation by 2 weeks. Study of

This study aimed to develop gliotoxin derivatives with high yield and low toxicity through fermentation optimisation and structural modification. First, the culture conditions were optimised, resulting in a yield of 208 mg L-1. Subsequently, 17 new salt-type derivatives were synthesised by introducing amino acid esters at the 6-position. These compounds retained the essential disulphide pharmacophore while exhibiting significantly reduced toxicity towards normal GES-1 cells. Representative compounds 3b and 3e exhibited good inhibitory activity against breast cancer MCF-7 and oesophageal cancer

Anoikis is a form of apoptosis induced by cell detachment. Integrin inactivation plays a major role in the process but the exact signalling pathway is ill-defined. Here we identify an anoikis pathway using gliotoxin (GT), a virulence factor of the fungus Aspergillus fumigatus, which causes invasive aspergillosis in humans. GT prevents integrin binding to RGD-containing extracellular matrix components by covalently modifying cysteines in the binding pocket. As a consequence, focal adhesion kinase (FAK) is inhibited resulting in dephosphorylation of p190RhoGAP, allowing activation of RhoA. Seque

Gliotoxin is an immunosuppressive apoptogenic mycotoxin produced by a number of fungi including important human pathogens as Aspergillus fumigatus. In order to elucidate the potential role of gliotoxin as immunoevasive fungal virulence factor we studied the effects of gliotoxin on the innate and adaptive T cell-mediated immune response against the facultatively intracellular bacterium Listeria monocytogenes. Gliotoxin induced apoptosis of bone marrow-derived macrophages, dendritic cells and CD8 T cells in a dose- and cell type-dependent manner. In vitro the apoptogenic effect of gliotoxin corr

BACKGROUND: Disseminated invasive aspergillosis is an exceptional finding in immunocompetent hosts. As in immunocompromised patients, it has high mortality rates. Early diagnostic methods are required in order to properly manage the patient. Bis(methylthio)gliotoxin (bmGT) is a novel biomarker, useful in onco-hematological patients.
CASE REPORT: A 70-year-old male, with non-insulin dependent type II diabetes mellitus and a past surgery history of aortic valve replacement with coronary by-pass five years ago, was seen in the emergency department with blurred vision. Three days later, endogen e

Gliotoxin is a member of the epipolythiodioxopiperazine class of toxins and is both the major and the most potent toxin produced by Aspergillus fumigatus. Since the discovery of the putative gliotoxin biosynthetic 12-gene cluster in the genome of A. fumigatus, five different laboratories have attempted to determine the role of this toxin in the virulence of A. fumigatus. The genes in the cluster that have been disrupted to study the pathobiological importance of gliotoxin include gliZ that encodes a transcription factor and gliP that encodes a nonribosomal peptide synthase. Two of the five lab

Fibrosis is generally characterized by an excessive accumulation and deposition of extracellular matrix (ECM) that progressively leads to the destruction of functional nephrons of the kidney. Current study is aimed to propose a probable antifibrotic mechanism of gliotoxin, isolated from marine Aspergillus fumigatus strain, in proximal tubule renal epithelial cells by inhibiting HIF-1α, Ang-II and NF-κB in a dose dependent manner. GTX at a concentration of 1.6 ng/ml also showed more than 80% cell viability. Scanning Electron Microscopy, fluorescence microscopy, FACS, mRNA and protein profiles f

In susceptible hosts, angioinvasion by Aspergillus fumigatus triggers thrombosis, hypoxia, and proinflammatory cytokine release, all of which are stimuli for angiogenesis. We sought to determine whether A fumigatus directly modulates angiogenesis. A fumigatus culture filtrates profoundly inhibited the differentiation, migration, and capillary tube formation of human umbilical vein endothelial cells in vitro. To measure angiogenesis at the site of infection, we devised an in vivo Matrigel assay in cyclophosphamide-treated BALB/c mice with cutaneous invasive aspergillosis. Angiogenesis was signi

Antimicrobial resistance (AMR) is a major global problem and threat to humanity. The search for new antibiotics is directed towards targeting of novel microbial systems and enzymes, as well as augmenting the activity of pre-existing antimicrobials. Sulphur-containing metabolites (e.g., auranofin and bacterial dithiolopyrrolones [e.g., holomycin]) and Zn2+-chelating ionophores (PBT2) have emerged as important antimicrobial classes. The sulphur-containing, non-ribosomal peptide gliotoxin, biosynthesised by Aspergillus fumigatus and other fungi exhibits potent antimicrobial activity, especially i

Sclerotium rolfsii causes destructive soilborne disease in numerous plant species, and biological control may be a promising and sustainable approach for suppressing this widespread pathogen. In this study, the antagonistic effect against S. rolfsii of 10 Trichoderma strains was tested by the dual culture method, and a gliotoxin-producing strain, T. virens T23, was shown to be the most effective, inhibiting growth of S. rolfsii in vitro by 70.2%. To clarify the antagonistic mechanism and gliotoxin biosynthesis regulation of T23, a gliotoxin-deficient mutant was constructed via Agrobacterium tu

Gliotoxin, one of the most toxic metabolites produced during the growth of Aspergillus fumigatus, can cause direct damage to the immune system and results in infection and spread of Aspergillus, or even leads to invasive aspergillosis. Accurate, rapid, and sensitive detection of the disease-specific marker gliotoxin, particularly in serum, urine, or other body fluids, is therefore an important approach to achieving early and rapid diagnosis of Invasive Aspergillus Fumigatus Infection (IAFI). In this study, aptamers that specifically bind to gliotoxin were successfully obtained using immobiliza

Demyelinating disorders such as multiple sclerosis (MS) or transverse myelitis are devastating neurological conditions with no effective cure. Prevention of myelin loss or restoration of myelin are key for successful therapy. To investigate the disease and develop cures animal models with good clinical relevance are essential. The goal of the current study was to establish a model of focal demyelination in the brain of domestic pig using MRI-guided gliotoxin delivery. The rationale for developing a new myelin disease model in the domestic pig was based on the fact that the brain in pigs is ana

The opportunistic human pathogenic fungus Aspergillus fumigatus primarily reproduces by forming a large number of asexual spores (conidia). Sequential activation of the central regulators BrlA, AbaA and WetA is necessary for the fungus to undergo asexual development. In this study, to address the presumed roles of these key developmental regulators during proliferation of the fungus, we analyzed and compared the proteomes of vegetative cells of wild type (WT) and individual mutant strains. Approximately 1300 protein spots were detectable from 2-D electrophoresis gels. Among these, 13 proteins

As a pivotal secondary metabolite of Aspergillus fumigatus, gliotoxin (GT) has many toxicological effects on mammalian cells; however, its function on LC3-associated phagocytosis (LAP) induced by A. fumigatus in macrophages is poorly understood. Here, it was found that pretreatment of macrophages with GT can significantly attenuate the conversion of LC3-II. In parallel, the expression of Rubicon, the putative indicator of LAP in macrophages, was dampened in a similar trend. Loss of ability to produce GT made the conidia of gliPΔ mutant of A. fumigatus induce more LC3-II conversion in THP1 cell

Pyruvate kinase M2 (PKM2) functions as an important rate-limiting enzyme of aerobic glycolysis that is involved in tumor initiation and progression. However, there are few studies on effective PKM2 inhibitors. Gliotoxin is a marine-derived fungal secondary metabolite with multiple biological activities, including immunosuppression, cytotoxicity, and et al. In this study, we found that Gliotoxin directly bound to PKM2 and inhibited its glycolytic activity in a dose-dependent manner accompanied by the decreases in glucose consumption and lactate production in the human glioma cell line U87. The epidithiodioxopiperazine gliotoxin is a virulence factor of Aspergillus fumigatus, the most important airborne fungal pathogen of humans. Gliotoxin suppresses innate immunity in invasive aspergillosis, particularly by compromising neutrophils, but the underlying molecular mechanisms remain elusive. Neutrophils are the first responders among innate immune cells recruited to sites of infection by the chemoattractant leukotriene (LT)B4 that is biosynthesized by 5-lipoxygenase and LTA4 hydrolase (LTA4H). Here, we identified gliotoxin as inhibitor of LTA4H that selectively abrogates LTB4 format

A linear ubiquitin chain, which consists of ubiquitin molecules linked via their N- and C-termini, is formed by a linear ubiquitin chain assembly complex (LUBAC) composed of HOIP, HOIL-1L, and SHARPIN, and conjugation of a linear ubiquitin chain on the NF-κB essential modulator (NEMO) is deeply involved in NF-κB activation induced by various signals. Since abnormal activation of NF-κB is associated with inflammatory disease and malignancy, we searched for an inhibitor of LUBAC by high-throughput screening (HTS) with a Tb(3+)-fluorescein FRET system. As a result, we found that the fungal metabo

Lung cancer and breast cancer are among the most common malignant tumours worldwide. Gliotoxin (GTX) is an epipolythiodioxopiperazine (ETP)-class fungal toxin with broad biological activities; however, its low yield and the difficulty to synthesise have limited its application. To thoroughly explore the anticancer potential of GTX, this study systematically evaluated its inhibitory effects on non-small cell lung cancer (NSCLC) H1975 cells and breast cancer MCF-7 cells. For this purpose, we isolated and identified a high-yielding GTX-producing strain of Aspergillus fumigatus ZSS02 from local so

Mycotoxins are low molecular weight compounds present in food and feed. Although their effects on human health have been widely described, their mechanisms of action are still undefined. Gliotoxin (GTX) and ochratoxin A (OTA) are among the most dangerous mycotoxins produced by Aspergillus spp. Therefore, their toxicity was studied in the Daphnia magna model, which has high capacity to predict cytotoxicity and assess ecotoxicity, comparable to mammalian models. The study consisted of a series of tests to evaluate the effects of mycotoxins GTX, OTA and their combinations at different dilutions o

Multidrug-resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA) is one of the major causes of hospital-acquired and community infections and pose a challenge to the human health care system. Therefore, it is important to find new drugs that show activity against these bacteria, both in monotherapy and in combination with other antimicrobial drugs. Gliotoxin (GT) is a mycotoxin produced by Aspergillus fumigatus and other fungi of the Aspergillus genus. Some evidence suggests that GT shows antimicrobial activity against S. aureus in vitro, albeit its efficacy against mult
