Radiation-induced transformation of peptides into nanoparticles for nano-theranostics of pancreatic cancer with controllable drug delivery and sustained release.

Nano-theranostics using biodegradable polymers are effective for the diagnosis and treatment of intractable cancers; however, there is a need for technology that controls bioavailability and biodegradability without compromising biocompatibility. In this study, peptide nanoparticles (NPs) with controlled particle size and biodegradability were prepared by ionizing radiation and loaded with imaging agents and anticancer drugs to develop novel nano-theranostics for pancreatic cancer diagnosis and treatment. Peptides composed of histidine, glycine, glutamic acid, and an arginine-glycine-aspartic

The glycine deportation system and its pharmacological consequences.

The glycine deportation system is an essential component of glycine catabolism in man whereby 400 to 800mg glycine per day are deported into urine as hippuric acid. The molecular escort for this deportation is benzoic acid, which derives from the diet and from gut microbiota metabolism of dietary precursors. Three components of this system, involving hepatic and renal metabolism, and renal active tubular secretion help regulate systemic and central nervous system levels of glycine. When glycine levels are pathologically high, as in congenital nonketotic hyperglycinemia, the glycine deportation

Protection by glycine of proximal tubules from injury due to inhibitors of mitochondrial ATP production.

We have determined whether glycine or glutathione can protect rabbit proximal tubules damaged by chemical inhibitors of oxidative phosphorylation: antimycin A, rotenone, cyanide, oligomycin, or carbonyl cyanide m-chlorophenylhdrazone (CCCP). All the agents severely depleted cell ATP levels within 15 min and caused lethal cell injury, as quantified by lactate dehydrogenase (LDH) release. Glycine and glutathione largely prevented this injury without altering the primary effects of the inhibitors on tubule respiration or the depletion of ATP. Glycine and gamma-aminobutyric acid(A) receptor function is enhanced by inhaled drugs of abuse.

Inhalable solvents possess significant abuse liability and produce many of the neurobehavioral effects typically associated with central nervous system-depressant agents, including motor incoordination, anxiolysis, and the elicitation of signs of physical dependence on withdrawal. We tested the hypothesis that the commonly abused solvents toluene, 1,1,1-trichloroethane (TCE), and trichloroethylene (TCY) affect ligand-gated ion channel activity, as do other classes of central nervous system-depressive agents. TCE and toluene, like ethanol, reversibly enhanced gamma-aminobutyric acid (GABA)(A) r

Renal ischemia/reperfusion injury in rats is attenuated by a synthetic glycine derivative.

Renal ischemia/reperfusion is a common cause of acute renal failure. Glycine is an effective anti-inflammatory, cytoprotective agent and is reported to have a beneficial effect against ischemia/reperfusion injury in various organs. Previous research notes that free radicals and inflammatory leukocytes both play important roles in the pathogenesis of renal ischemia/reperfusion injury. To develop new therapeutic agents against renal ischemia/reperfusion injury, we sought to link an antioxidant moiety (nitronyl nitroxide) to glycine in the hope that the resulting glycine-nitronyl nitroxide conjug

[SUBUNIT SPECIFIC MODULATION OF GLYCINE RECEPTORS BY GINKGOLIC ACID.].

Ginkgo biloba extract is a multicomponent pharmacological agent widely used in neurological disorders therapy. It was shown that ginkgolic acid, a constituent of lipophylic Ginkgo biloba extract, has numerous biological activities. In the present study we have focused on the features of ginkgolic acid action on αl and α2 glycine receptors that make part of the inhibitory system of the brain. Using whole-cell configuration of patch-clamp recording we analysed effects of ginkgolic acid on different subunits of glycine receptors. Experiments were performed on cultured Chinese hamster ovary cells

Priming agents combat copper stress in wheat (Triticum aestivum L.) under hydroponic conditions: Insights in impacts on morpho-physio-biochemical traits and health risk assessment.

In recent years, the use of priming agents, such as silicon, melatonin, salicylic acid, glycine betaine, and ascorbic acid has gained significant attention for their role in mitigating abiotic stresses across various plant species. While previous research has been conducted on the individual impact of silicon, melatonin, salicylic acid, glycine betaine, and ascorbic acid in metal stress resistance among various crop species, their combined effects in the context of heavy metal stressed conditions remain underexplored. Wheat (Triticum aestivum L.) seedlings was grown under the toxic concentrati

Glycine protects against hepatocyte killing by KCN or hypoxia by preventing intracellular Na+ overload in the rat.

Glycine has been shown to prevent hepatocyte death induced by anoxia and by several toxic agents. However, the mechanisms responsible for such a cytoprotective effect have not yet been entirely clarified. We have previously shown that an uncontrolled increase in intracellular Na+ is critical for hepatocyte killing induced by adenosine triphosphate (ATP) depletion. We herein report that protection by glycine (2 mmol/L) against cytotoxicity induced in isolated rat hepatocyte by potassium cyanide (KCN) or hypoxia was associated with the prevention of cytosolic Na+ accumulation. The addition of th

Glycine and Sodium Glycinate in Water: Experiment and Molecular Dynamics with Different Model Parameters.

Amino acids play a fundamental role in biological processes and have various practical applications. Specifically, they have been considered as promising agents for the preparation of environmentally friendly deep eutectic solvents. To understand the physicochemical behavior of amino acids and their salts on a molecular level, atomistic molecular dynamics (MD) is quite helpful. In this work, glycine and sodium glycinate aqueous solutions were simulated with short-range interactions described by the OPLS-AA force field, using different atomic charge sets for the glycine zwitterion (GLZ) and the

Lead (Pb) is a highly toxic heavy metal widely dispersed in the environment because of human industrial activities. Many studies revealed that Pb could adversely affect several organs, including the male reproductive system. Pb-induced reproductive toxicity could lead to infertility. Thus, finding safe and clinically applicable protective agents against this complication is important. It has been found that oxidative stress plays a fundamental role in the pathogenesis of Pb-induced reprotoxicity. Glycine is the simplest amino acid with a wide range of pharmacological activities. It has been fo

Enhancement of homomeric glycine receptor function by long-chain alcohols and anaesthetics.

1. The effects of n-alcohols (ethanol to dodecanol) and anaesthetics on strychnine-sensitive glycine receptors were studied in Xenopus oocytes expressing homomeric alpha 1 or alpha 2 glycine receptor subunits, with the two electrode voltage-clamp recording technique. 2. The glycine-induced chloride conductance of homomeric alpha glycine receptors was potentiated by all the alcohols tested when an EC2 concentration of glycine was used. Homomeric alpha 1 and alpha 2 receptors were potentiated similarly by the n-alcohols, except that low concentrations of ethanol produced greater potentiation wit

Palliative Care Approach in a Case of Severe Nonketotic Hyperglycinemia With Long Survival: A Case Report.

Nonketotic hyperglycinemia (NKH) is a rare autosomal recessive metabolic disorder characterized by defective glycine cleavage, resulting in glycine accumulation, particularly in the central nervous system. The severe neonatal form typically presents with early-onset encephalopathy, refractory epilepsy, and poor neurodevelopment, often leading to early mortality. We report a male patient with genetically confirmed severe NKH who is currently alive at seven years of age - one of the longest reported survivals for this form. He presented within the first day of life with hypotonia, myoclonic seiz

Cytoprotection of kidney epithelial cells by compounds that target amino acid gated chloride channels.

Glycine, strychnine and certain chloride channel blockers were reported to protect cells against lethal cell injury. These effects have been attributed to interactions with membrane proteins related to CNS glycine gated chloride channel receptors. We have investigated the pharmacology of these actions. Madin-Darby canine kidney (MDCK) epithelial cells were depleted of adenosine triphosphate (ATP) by incubation in glucose free medium containing a mitochondrial uncoupler. Medium Ca2+ was adjusted to 100 nM in the presence of an ionophore such that intracellular Ca2+ did not increase, and Ca(2+)-

Action of renal vasodilators in dogs following acute biliary obstruction.

The renal vasodilator properties of six endogenous substances were tested before and 4 hr after ligation of the common bile duct. Two substances, acetylcholine and a glucocorticoid, retained their vasodilator properties at a fixed dose following acute biliary obstruction. Dopamine was still able to increase glomerular filtration rate and renal blood flow, but demonstrated an attenuated response. Several other agents, glucagon, glycine, and bradykinin, lost their renal vasodilator actions at the dosages employed. For these latter three compounds, control studies using sham-obstructed dogs and i

Use of trifluoroperazine isolates a [(3)H]Ifenprodil binding site in rat brain membranes with the pharmacology of the voltage-independent ifenprodil site on N-methyl-D-aspartate receptors containing NR2B subunits.

The use of trifluoroperazine in a well washed rat brain membrane preparation revealed [(3)H]ifenprodil binding to a single high affinity state with the pharmacology of N-methyl-D-aspartate (NMDA) receptors containing NR2B subunits. Inhibition of [(3)H]ifenprodil binding in the presence of trifluoroperazine by 10 NR1a/NR2B selective agents was highly correlated with their inhibition at rat NR1a/NR2B receptors expressed in Xenopus ooctyes and [(3)H]TCP binding to rat brain NR2B subunit containing NMDA receptors but not with their inhibition of [(3)H]DTG binding. Antimicrobial activity of novel symmetrical antimicrobial peptides centered on a hydrophilic motif against resistant clinical isolates: in vitro and in vivo analyses.

UNLABELLED: Antibiotic resistance poses a significant public health threat worldwide. The rise in antibiotic resistance and the sharp decline in effective antibiotics necessitate the development of innovative antibacterial agents. Based on the central symmetric structure of glycine-serine-glycine, combined with tryptophan and arginine, we designed a range of antimicrobial peptides (AMPs) that exhibited broad-spectrum antibacterial activity. Notably, AMP W5 demonstrated a rapid and effective sterilization against methicillin-resistant Staphylococcus aureus (MRSA), displaying both a minimum inhi

A copper/O2-mediated direct sp3 C-H/N-H cross-dehydrogen coupling reaction of acylated amines and N-aryl glycine esters.

A copper salt-catalyzed cross-dehydrogenative coupling reaction between N-aryl glycine esters and acylated amines has been developed. The reaction proceeded effectively under an oxygen atmosphere without the use of peroxide agents. This simple protocol allows for the preparation of a series of new compounds in a moderate to excellent yield via the CDC reaction of a wide range of N-aryl glycine derivatives with acylated amines, which are of great interest in the field of medicinal chemistry. A plausible mechanism involving the formation of an iminium ion intermediate, followed by coupling with

Improvement of glycine biosynthesis from one-carbon compounds and ammonia catalyzed by the glycine cleavage system in vitro.

Glycine cleavage system (GCS) plays a central role in one-carbon (C1) metabolism and receives increasing interest as a core part of the recently proposed reductive glycine pathway (rGlyP) for assimilation of CO2 and formate. Despite decades of research, GCS has not yet been well understood and kinetic data are barely available. This is to a large degree because of the complexity of GCS, which is composed of four proteins (H, T, P, and L) and catalyzes reactions involving different substrates and cofactors. In vitro kinetics of reconstructed microbial multi-enzyme glycine cleavage/synthase syst

Glycine administration attenuates skeletal muscle wasting in a mouse model of cancer cachexia.

BACKGROUND AND AIMS: The non-essential amino acid, glycine, is often considered biologically neutral, but some studies indicate that it could be an effective anti-inflammatory agent. Since inflammation is central to the development of cancer cachexia, glycine supplementation represents a simple, safe and promising treatment. We tested the hypothesis that glycine supplementation reduces skeletal muscle inflammation and preserves muscle mass in tumor-bearing mice. METHODS: To induce cachexia, CD2F1 mice received a subcutaneous injection of PBS (control, n = 12) or C26 tumor cells (n = 32) in ac
