Inhibition of osteoblastic differentiation by warfarin and 18-α-glycyrrhetinic acid.

Anticoagulation therapy with vitamin K antagonists such as warfarin is widely used to prevent and treat stroke in patients with chronic atrial fibrillation or mechanical heart valves. Because vitamin K is an essential factor for ggg-carboxylation of osteocalcin, vitamin K antagonists might cause bone loss. Although the association between warfarin use and bone metabolism is still controversial, several studies show that bone mineral density is decreased and fracture risk is increased with warfarin therapy. Meanwhile, attenuation of gap junctional communication (GJC) by warfarin is reported in

Antibacterial activity of 18β-glycyrrhetinic acid against Neisseria gonorrhoeae in vitro.

Gonorrhea is the second most common sexually transmitted diseases worldwide. Chronic infection of Neisseria gonorrhoeae (N. gonorrhoeae) can lead to severe complications. Presently, N. gonorrhoeae has developed resistance to almost all antibiotics used for the treatment of gonorrhea. Thus, it's urgent to explore new approaches to treat gonorrhea. Presently, nontraditional treatment method as an alternative to antibiotic use is getting more and more attention. Here we demonstrated that 18β-glycyrrhetinic acid (GRA) exhibited robust antimicrobial activity against N. gonorrhoeae in vitro. GRA led

Glycyrrhetinic acid inhibits contact hypersensitivity induced by trichophytin via dectin-1.

Trichophyton infection is highly prevalent and tends to be recurrent. Therefore, it is important to develop new therapeutic agents. Previously, we established a mouse model of Trichophyton-induced contact hypersensitivity (CHS) and demonstrated that dectin-1 was involved in inflammation induced by trichophytin, the Trichophyton antigen. Here, we used that model to investigate glycyrrhetinic acid (GA) from plants of the genus Glycyrrhiza as a potential anti-inflammatory agent against superficial mycoses. GA suppressed swelling and the expression of inflammatory cytokines, including macrophage i

Antiulcer activities of glycyrrhetinic acid derivatives in experimental gastric lesion models.

Glycyrrhetinic acid (Ia) and eighteen related derivatives were examined for antiulcer activity using stress-induced gastric lesions (restraint plus water immersion at 25 degrees C) in mice and rats as screening tests. Among the compounds tested, dihemiphthalate derivatives of 18 alpha- or 18 beta-olean-12-ene-3 beta,30-diol (IV, IIId), 18 beta-olean-9(11)12-diene-3 beta,30-diol (VIc), and olean-11,13(18)-diene-3 beta,30-diol (VIIc) showed potent inhibition of gastric lesion formation at a dose of 12 or 25 mg/kg (p.o.); carbenoxolone sodium (Ib) significantly suppressed the lesion formation at

Glycyrrhetinic acid attenuates vascular smooth muscle vasodilatory function in healthy humans.

Abnormal glucocorticoid metabolism contributes to vascular dysfunction and cardiovascular disease. Cortisol activation of vascular mineralocorticoid and glucocorticoid receptors is regulated by two types of 11beta-HSD (11-beta hydroxysteroid dehydrogenase), namely 11beta-HSD2 and 11beta-HSD1 (type 2 and type 1 11beta-HSD respectively). We hypothesized that inhibition of 11beta-HSD would attenuate vascular function in healthy humans. A total of 15 healthy subjects were treated with the selective 11beta-HSD inhibitor GA (glycyrrhetinic acid) or matching placebo in a randomized double-blinded cro

18beta-glycyrrhetinic acid alleviates deoxynivalenol-induced hepatotoxicity by inhibiting GPX4-dependent ferroptosis.

Deoxynivalenol (DON), a mycotoxin that severely contaminates agri-food products can cause hepatotoxicity. Ferroptosis is an iron-dependent form of cell death, and the liver is an important organ for iron accumulation. 18beta-glycyrrhetinic acid (GA) has anti-ferroptosis and hepatoprotective effects. This study aimed to investigate the role of ferroptosis in the protective effects of GA against DON-induced hepatotoxicity in HepG2 cells and mice. The in vitro results revealed that DON (0.4 μM) decreased GPX4, SLC7A11, GCLC, NQO1, and Nrf2 expression; promoted TFR-1 expression and MDA, 4-HNE, and

Synthesis and anti-hepaticfibrosis of glycyrrhetinic acid derivatives with inhibiting COX-2.

Many tests have shown cyclooxygenase-2 (COX-2) was closely related to the activation of hepatic stellate cells (HSCs), which further promoting the onset and development of hepatic fibrosis. According to these research findings, a series of glycyrrhetinic acid derivatives were designed and synthesized. Meanwhile, their anti-hepaticfibrotic activities were evaluated in vitro and in vivo. Firstly, in the tests of the cell models, all the compounds displayed anti-proliferative effect on the HSC-T6 activated by (transforming growth factor beta) TGF-β1 (10 ng/mL). Among them, compounds 2 and 16 exhi

Suppressive effects of glycyrrhetinic acid derivatives on tachykinin receptor activation and hyperalgesia.

Glycyrrhetinic acid (GA), an aglycone of glycyrrhizin, isolated from the licorice root (Glycyrrhizia), and its semi-synthetic derivatives have a wide range of pharmacological effects. To investigate whether GA derivatives may be used as a new class of analgesics, we examined the effects of these compounds on human tachykinin receptors expressed in CHO-K1 cells. Among the GA derivatives examined, the disodium salt of olean-11,13(18)-dien-3β,30-O-dihemiphthalate inhibited the mobilization of [Ca(2+)](i) induced by substance P, neurokinin A, and neurokinin B in CHO-K1 cells expressing the human N

Exploring the Pivotal Immunomodulatory and Anti-Inflammatory Potentials of Glycyrrhizic and Glycyrrhetinic Acids.

Licorice extract is a Chinese herbal medication most often used as a demulcent or elixir. The extract usually consists of many components but the key ingredients are glycyrrhizic (GL) and glycyrrhetinic acid (GA). GL and GA function as potent antioxidants, anti-inflammatory, antiviral, antitumor agents, and immuneregulators. GL and GA have potent activities against hepatitis A, B, and C viruses, human immunodeficiency virus type 1, vesicular stomatitis virus, herpes simplex virus, influenza A, severe acute respiratory syndrome-related coronavirus, respiratory syncytial virus, vaccinia virus, a

Recent Advances in Glycyrrhetinic Acid-Functionalized Biomaterials for Liver Cancer-Targeting Therapy.

Liver cancer is one of the most common causes of cancer mortality worldwide. Chemotherapy and radiotherapy are the conventional therapies generally employed in patients with liver tumors. The major issue associated with the administration of chemotherapeutics is their high toxicity and lack of selectivity, leading to systemic toxicity that can be detrimental to the patient's quality of life. An important approach to the development of original liver-targeted therapeutic products takes advantage of the employment of biologically active ligands able to bind specific receptors on the cytoplasmati

Glycyrrhetinic acid food supplementation lowers serum potassium concentration in chronic hemodialysis patients.

Hyperkalemia is a common life-threatening problem in hemodialysis patients. Because glycyrrhetinic acid (GA) inhibits the enzyme 11beta-hydroxy-steroid dehydrogenase II and thereby increases cortisol availability to the colonic mineralocorticoid receptor, it has the potential to lower serum potassium concentrations. To test this, 10 patients in a 6 month prospective, double-blind, placebo-controlled crossover study were given cookies or bread rolls supplemented with glycyrrhetinic acid or placebo. Twenty-four-hour blood pressure measurements were performed at baseline and week 6 and 12 of each

Anti-allergic activity of 18beta-glycyrrhetinic acid-3-O-beta-D-glucuronide.

Glycyrrhizin (18beta-glycyrrhetinic acid-3-O-beta-D-glucuronopyranosyl-(1 --> 2)-beta-D-glucuronide, GL) was transformed to 18beta-glycyrrhetinic acid-3-O-beta-D-glucuronide (GAMG) by Streptococcus LJ-22. The antiallergic activities of GL and GAMG was measured using a RBL cell assay system and contact hypersensitivity model mice. GAMG exhibited anti-allergic activity with IC50 values of 0.28 mM. GAMG, which is sweeter than GL, and 18beta-glycyrrhetinic acid, which is a GAMG metabolite by human intestinal bacteria, also inhibited the passive cutaneous anaphylaxis and skin contact inflammation.

Advances in Drug Delivery Systems Utilizing Glycyrrhizic Acid and Its Aglycone Glycyrrhetinic Acid in Cancer Therapy.

Glycyrrhizic acid (GL) and glycyrrhetinic acid (GA) exhibit antitumor activity, favorable biocompatibility, receptor-targeting capabilities, and amphiphilicity. These properties make them promising multifunctional components for developing antitumor drug delivery systems. Unlike previous reviews that focus on individual applications, this review systematically categorizes GL/GA according to their three functional roles. First, as therapeutic agents, GL/GA can be incorporated into passively targeted, actively targeted, and stimuli-responsive nanocarriers. They can be combined with chemotherapeu

Reengineering glycyrrhetinic acid into a therapeutic oligomer for targeted tumor therapy with cardioprotection.

To overcome the inherent limitations of conventional nanocarriers such as therapeutic inefficacy and suboptimal drug loading capacity, a novel "polymeric nanodrug" strategy was developed. This approach redefines nanodrug design by employing glycyrrhetinic acid (GA) simultaneously as a bioactive agent and a polymerizable targeting monomer to synthesize poly-glycyrrhetinic acid (PGA), achieving near-theoretical drug loading approaching 100%. Subsequent PEGylation yielded hepatocellular carcinoma (HCC)-targeting PGA-PEG-GA nanodrugs, which exhibited a 1.89-fold enhancement in antitumor efficacy c

In Vitro and In Silico Studies of Glycyrrhetinic Acid Derivatives as Anti- Filarial Agents.

BACKGROUND: Lymphatic filariasis is one of the chronic diseases in many parts of the tropics and sub-tropics of the world despite the use of standard drugs diethylcarbamazine and ivermectin because they kill microfilaries and not the adult parasites. Therefore, new leads with activity on adult parasites are highly desirable. OBJECTIVE: Anti-filarial lead optimization by semi-synthetic modification of glycyrrhetinic acid (GA). METHODS: The GA was first converted into 3-O-acyl derivative, which was further converted into 12 amide derivatives. All these derivatives were assessed for their antif

Carrier-as-Prodrug Glycyrrhizic Acid Nanogel Co-Delivers Mannose and Glycyrrhetinic Acid for Metabolic-Ionic Cancer Therapy.

Cancer cells evade conventional therapies by dynamically switching between glycolysis and oxidative phosphorylation (OXPHOS). Simultaneous blockade of both pathways could overcome this plasticity but requires coordinated delivery of two inhibitors to the same cell. Here we design a glycyrrhizic acid (GA) nanogel that functions as both a structural carrier and a prodrug. GA itself serves as the structural carrier. It hydrolyzes in the acidic tumor microenvironment to become glycyrrhetinic acid (GRA), an OXPHOS inhibitor. The nanogel also encapsulates mannose, a glycolysis inhibitor. A crisis-in

3-Monoglucuronyl-glycyrrhetinic acid is a major metabolite that causes licorice-induced pseudoaldosteronism.

18 beta-Glycyrrhetinic acid (GA) has been thought to be one of the major metabolites that causes licorice-induced pseudoaldosteronism. However, we found no difference in the blood level of GA between the patients with and without pseudoaldosteronism. 18β-Glycyrrhetinic acid ameliorates hepatic fibrosis through modulating bile acid profile and inhibiting TGF-β/SMAD3 pathway.

Oxidative stress drives the progression of liver disease from hepatitis to fibrosis, underscoring the need for dietary strategies to protect liver health. 18β-glycyrrhetinic acid (GA), a natural bioactive compound from Glycyrrhiza glabra, exhibits hepatoprotective, antioxidant, and anti-inflammatory properties. This study investigated the protective effects of dietary GA against D-galactose-induced hepatic injury in weaned piglets and explored its impact on hepatic function and gut-liver metabolism. The results showed that GA supplementation markedly reduced liver damage and fibrosis, as evide
