Myelination and remyelination in the regenerating visual system of the goldfish.

The remyelination of regenerated optic axons was investigated in goldfish following either optic nerve crush or ouabain retinal intoxication. Axons grown after nerve crushing acquire thinner myelin sheaths than axons originating from reconstituted ganglion cells. If axons of reconstituted ganglion cells are crushed and allowed to regenerate, the subsequent myelination is weaker than that of control axons not interrupted by crushing, but stronger than that of axons of preexisting retinal ganglion cells. The present results suggest that a neuron is capable of inducing a normally developed myelin

Hippocampal Pallium Lesion Impairs Transitive Inference in Goldfish.

Transitive inference, a process that involves drawing logical conclusions based on preliminary information, is considered a cornerstone of human deductive reasoning. Furthermore, transitive inference is a clear instance of representational flexibility as it implies the novel expression of learned information. In mammals and birds, both episodic memory and transitive inference critically depend on the integrity of the hippocampus. Comparative neurobiological evidence indicates that a hippocampus homologue can also be found in the telencephalic pallium of teleost fish. Here, we investigated whet

Fish Lice (Argulus japonicus) in Goldfish (Carassius auratus).

Fish lice (Argulus spp; family Argulidae) are branchiuran crustaceans that parasitize both marine and freshwater fishes. Argulus spp can be a major threat to fish health, because heavy infestations can cause significant morbidity and mortality. In addition, fish lice are known to be the vehicle for other fish diseases. During rounds at our facility, Argulus japonicus was collected from the caudal and anal fins of 3 goldfish (Carassius auratus). These goldfish were asymptomatic, and no additional cases were noted after manual removal of the lice. As soon as any Argulus organisms are identified,

Trivalent chromium induces oxidative stress in goldfish brain.

Although information on the effects of Cr(6+) in biological systems is abundant, Cr(3+) has received less attention. Toxic effects of chromium compounds are partially associated with activation of redox processes. Recently we found that Cr(6+) induced oxidative stress in goldfish tissues and the glutathione system was shown to play a protective role. The present study aimed to investigate free radical processes in brain of goldfish exposed to CrCl(3). Trivalent chromium at a concentration of 50 mg L(-1) was lethal and therefore we chose to examine sublethal dosages of 1.0-10.0 mg L(-1) in aqua

Intraocular tetrodotoxin in goldfish hinders optic nerve regeneration.

Repeated intraocular injection of tetrodotoxin (TTX) was used to produce a maintained block of neural activity in goldfish optic axons which were regenerating following a crush of the optic nerve. The recovery of visual function was delayed in the TTX-treated fish, as demonstrated by delays in the return of the startle reaction to sudden illumination, the dorsal light reflex and food pellet localization. A single injection of TTX at the time of optic nerve crush delayed recovery of the startle reaction but not of food localization. There was no loss of ganglion cells in the TTX-treated animals

Importation of CyHV-2-infected goldfish into the Netherlands.

Cyprinid herpesvirus 2 (CyHV-2) is known as the causative agent of herpesviral haematopoietic necrosis in goldfish Carassius auratus auratus. However, the virus has also been detected in Prussian carp C. gibelio and crucian carp C. carassius from European and Asian countries. To prevent spread of the causative virus to other areas, investigation of the risk factors of spread of this virus is important. In this study, 8 batches of goldfish imported into the Netherlands by airfreight from Asia and the Middle East were investigated for the presence of the virus. CyHV-2 DNA was detected by PCR in

A model of MPTP-induced Parkinson's disease in the goldfish.

Parkinson's disease (PD) has been modeled in humans, lower primates, and to a lesser extent in some other vertebrates by the administration of the potent neurotoxin 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine (MPTP). The MPTP model has thus drawn considerable attention during the past 15 years, as a system to search for anti-PD drugs. It has been previously reported that a Parkinsonian syndrome can be elicited in the common goldfish (Carassius auratus) by a single dose of MPTP. This protocol describes the relatively simple and inexpensive MPTP model of PD in goldfish. The procedure takes 14-3

Melatonin signal transduction in the goldfish, Carassius auratus.

Generation and reception of melatonin signals in the goldfish, Carassius auratus, are reviewed. The photoreceptive pineal gland of the goldfish generates circulating melatonin rhythms according to a given photoperiod under light-dark cycles and in a circadian manner under continuous dark conditions. Melatonin is also produced in the retina in a similar fashion. Melatonin produced in the pineal gland and retina is considered to act as internal zeitgeber in the brain and retina, respectively, controlling various physiological events via specific melatonin binding sites that are coupled with G pr

Retinal changes in a mutant form of goldfish with megalophthalmia.

The retinal changes of a mutant strain of goldfish with megalophthalmia were studied by histology, electron microscopy and biochemistry. Changes in the morphology of the pigment epithelium, decrease in number of photoreceptors, thinning out of retinal layers and the existence of spaces in the retina were all features as the eyes grew in size. Invasion of macrophages was also evident in the retina. A decrease in leucine uptake per milligram of retina was also detected as the eye grew beyond 0.8 millilitre in volume. These changes, although related to volume changes (i.e., growth) of the eyes, w

Degenerative and regenerative changes in the trochlear nerve of goldfish.

The features of unlesioned and lesioned trochlear nerves of goldfish have been examined electron microscopically. Lesioned nerves were studied between 1 and 107 days after cutting or crushing the nerve. Unlesioned nerves contained, on average, 77 myelinated axons and 19 unmyelinated axons. The latter were found in 1-2 fascicles per nerve. A basal lamina surrounded each myelinated axon and fascicle of unmyelinated axons. The numbers of myelinated axons, fascicles of unmyelinated axons and basal laminae varied by less than 5% over the intraorbital extramuscular segment of the nerve. Following in

Medulloepithelioma of the ciliary body in the goldfish (Carassius auratus).

A medulloepithelioma of the ciliary body in the right eye of a 1-year-old goldfish (Carassius auratus) had spindle and epithelial cells arranged either in columns or around a central lumen. Vitreous-like material was found within the tumor. A medulloepithelioma is a most unusual tumor in fish in which ocular tumors are rare.

Melatonin reduces locomotor activity and circulating cortisol in goldfish.

The present study focused on the effects of a subchronic melatonin treatment on locomotor activity and cortisol plasma levels in goldfish. We compared two different administration routes: peripheral (10 microg/g body weight) versus central (1 microg/microl) injections of melatonin for 7 or 4 days, respectively. Daily locomotor activity, including both diurnal and nocturnal activities, food anticipatory activity and circulating cortisol at 11:00 (under 24 h of food deprivation and 17 h postinjection) were significantly reduced after repeated intraperitoneal injections with melatonin for 7 days,

Bioassay of thyroid stimulating hormone using the f goldfish.

A new method for the bioassay of thyroid stimulating hormone using goldfish is described. The technique can detect thyroid stimulating substances in euthyroid unconcentrated plasma. Early results show that the method compares well with other techniques. Some evidence has been obtained which suggests that in primary myxoedema there may be some impairment of the thyropituitary "servo-mechanism" in older patients.

Dissociation of place and cue learning by telencephalic ablation in goldfish.

This study examined the spatial strategies used by goldfish (Carassius auratus) to find a goal in a 4-arm maze and the involvement of the telencephalon in this spatial learning. Intact and telencephalon-ablated goldfish were trained to find food in an arm placed in a constant room location and signaled by a local visual cue (mixed place-cue procedure). Both groups learned the task, but they used different learning strategies. Telencephalon-ablated goldfish learned the task more quickly and made fewer errors to criterion than controls. Probe trials revealed that intact goldfish could use either

Fine structure of the olfactory bulb in the goldfish, Carassius auratus.

Ultrastructural studies have aided in identifying the principal cell types and the layers of the olfactory bulb in goldfish. The concentrically arranged four layers are: the primary olfactory nerve layer (most peripheral), the glomerular layer, the mitral cell layer, and the granule cell layer (deepest). Three types of cell bodies were differentiated by their size and cytoplasmic organelles: mitral cells, granule cells and short axon cells. The axons of the primary olfactory nerve formed synaptic contacts with mitral cell dendrites in the glomeruli. Reciprocal synapses were observed between th

Conditioned place preference and conditioned place aversion in goldfish.

Some drugs possess reinforcing properties, which are commonly studied using the conditioned place preference (CPP) paradigm. CPP has been applied across a wide range of taxa, including nonmammalian vertebrates and invertebrates. In the present study, goldfish were used as an experimental model to examine CPP induced by nicotine, a substance that remains a major public health concern. Nicotine produced CPP at lower doses and conditioned place aversion (CPA) at higher doses. One advantage of fish models is the feasibility of drug administration by immersion, an animal-friendly alternative to inj

Causal gene identification and desirable trait recreation in goldfish.

Goldfish (Carassius auratus) have long fascinated evolutionary biologists and geneticists because of their diverse morphological and color variations. Recent genome-wide association studies have provided a clue to uncover genomic basis underlying these phenotypic variations, but the causality between phenotypic and genotypic variations have not yet been confirmed. Here, we edited proposed candidate genes to recreate phenotypic traits and developed a rapid biotechnology approach which combines gene editing with high-efficiency breeding, artificial gynogenesis, and temperature-induced sex revers
