Sustainable Gossypium arboreum biodiesel production using an industrial waste heterogeneous catalyst for RCCI engine applications.

Industrial waste-based catalysts provide a sustainable and cost-efficient solution for biodiesel production, improving yield, quality, and environmental impact. When this biodiesel is used in advanced reactivity-controlled compression ignition (RCCI) mode, it enhances the combustion process within direct injection (DI) diesel engines. These strategies effectively reduce nitrogen oxide (NOx) emissions and smoke without compromising engine performance. This study used cottonseed (Gossypium arboreum) methyl ester (CSME) as the pilot injection fuel. It was produced under optimal conditions of 2 wt

Salicylic acid-related cotton (Gossypium arboreum) ribosomal protein GaRPL18 contributes to resistance to Verticillium dahliae.

BACKGROUND: Verticillium dahliae is a phytopathogenic fungal pathogen that causes vascular wilt diseases responsible for considerable decreases in cotton yields. The complex mechanism underlying cotton resistance to Verticillium wilt remains uncharacterized. Identifying an endogenous resistance gene may be useful for controlling this disease. RESULTS: We cloned the ribosomal protein L18 (GaRPL18) gene, which mediates resistance to Verticillium wilt, from a wilt-resistant cotton species (Gossypium arboreum). We then characterized the function of this gene in cotton and Arabidopsis thaliana pla

A Ve homologous gene from Gossypium barbadense, Gbvdr3, enhances the defense response against Verticillium dahliae.

The tomato Ve1 gene and several Ve1 homologues are involved in the resistance to Verticillium dahliae. Here, we report on another Ve homologous gene, Gbvdr3, from a Verticillium wilt-resistant cotton cultivar, Gossypium barbadense Hai7124, which has a 3207-bp region that encodes a predicted receptor-like protein. Transient expression analyses indicated that Gbvdr3 is localized in the plasma membrane, and virus-induced gene silencing of Gbvdr3 compromised the resistance of Hai7124 cotton to a defoliating strain of V. dahliae, V991, but not to a non-defoliating strain, BP2. This resistance patte

Genetic Dynamics of Allotetraploid Cotton Species (Gossypium spp.) and Their Utilization in Breeding for Superior Fiber Quality.

The restricted genetic diversity of current cotton varieties (Gossypium hirsutum L.) considerably limits breeding opportunities aimed at enhancing fiber quality and resistance to environmental factors. Wild types of allotetraploid cottons are valuable sources of genetic variation, and their breeding potential has not yet been fully utilized. In this study, the inheritance and variability of important agronomic traits were analyzed in F1 and F2 hybrid generations obtained from intra- and interspecific crosses involving G. hirsutum, G. barbadense, and G. darwinii. The phylogenetic relationships

Identification of GASA Protein Family Expression Levels in Cotton (Gossypium hirsutum L.) Affected by Verticillium wilt.

Verticillium wilt, caused by Verticillium dahliae, is a major constraint on cotton production. The GASA (Gibberellic Acid-Stimulated in Arabidopsis) proteins are cysteine-rich peptides induced by gibberellin signaling; however, their systemic role in upland cotton (Gossypium hirsutum) resistance to Verticillium wilt remains elusive. Here, we identified 40 GhGASA members from the G. hirsutum genome, which were clustered into three subfamilies. All members possessed a typical gibberellin-regulated domain, and segmental duplication was the primary driver of family expansion. Transcriptomic analys

Two temporally synthesized charge subunits interact to form the five isoforms of cottonseed (Gossypium hirsutum) catalase.

Five charge isoforms of tetrameric catalase were isolated from cotyledons of germinated cotton (Gossypium hirsutum L.) seedlings. Denaturing isoelectric focusing of the individual isoforms in polyacrylamide gels indicated that isoforms A (most anodic) and E (most cathodic) consisted of one subunit of different charge, whereas isoforms B, C and D each consisted of a mixture of these two subunits. Thus the five isoforms apparently were formed through combinations of two subunits in different ratios. Labelling cotyledons in vivo with [35S]methionine at three daily intervals in the dark, and trans

Pan-Genome Analysis of the Tubulin Gene Family Reveals Candidates for Fiber Strength in Gossypium barbadense.

Background/Objectives: Tubulins (Tub) are central components of microtubules, but intraspecific variation and developmental expression of the Tub family in Gossypium barbadense remain poorly characterized. This study aimed to characterize the GbTub family using a pan-genome framework and identify candidates associated with fiber development and strength. Methods: A total of 50 GbTub genes were identified in the G. barbadense 3-79 reference genome, and their orthologous presence-absence patterns were subsequently assessed across 12 additional G. barbadense accessions. Gh4CL20/20A involved in flavonoid biosynthesis is essential for male fertility in cotton (Gossypium hirsutum L.).

Flavonoids have been shown to play an essential role in plant growth and fertility. 4-Coumarate CoA ligase (4CL) is one of the indispensable enzymes involved in the biosynthesis of flavonoids. However, the role of 4CL and flavonoids in impact on cotton fertility is still unknown. In this study, on the basis of identification of an additional Gh4CL gene, Gh4CL20A, by using an updated G. hirsutum genome, we found that Gh4CL20A and its homologous Gh4CL20 were preferentially expressed in petals and stamens. The petals of the loss-of-function Gh4CL20/Gh4CL20A mutant generated by CRISPR/Cas9 gene ed

Dynamic transcriptome analysis and volatile profiling of Gossypium hirsutum in response to the cotton bollworm Helicoverpa armigera.

In response to insect herbivory, plants emit elevated levels of volatile organic compounds for direct and indirect resistance. However, little is known about the molecular and genomic basis of defense response that insect herbivory trigger in cotton plants and how defense mechanisms are orchestrated in the context of other biological processes. Here we monitored the transcriptome changes and volatile characteristics of cotton plants in response to cotton bollworm (CBW; Helicoverpa armigera) larvae infestation. Analysis of samples revealed that 1,969 transcripts were differentially expressed (l

Identification and Functional Characterization of a Microtubule-Associated Protein, GhCLASP2, From Upland Cotton (Gossypium hirsutum L.).

Cytoplasmic linker-associated proteins (CLASPs) are microtubule-associated proteins (MAPs) involved in regulation of dynamics of microtubules (MTs) that play an important role in plant growth and development. In this study, we identified cotton CLASP genes and investigated the function of GhCLASP2. GhCLASP2 was mainly expressed in stem and developing fibers, especially in fibers of the secondary cell wall deposition stage. Ectopic expression of GhCLASP2 in Arabidopsis increased the branching number of leaf trichomes and rescued the defective phenotypes of clasp-1. In cotton, overexpression of

Detection of Stress in Cotton (Gossypium hirsutum L.) Caused by Aphids Using Leaf Level Hyperspectral Measurements.

: Remote sensing can be a rapid, accurate, and simple method for assessing pest damage on plants. The objectives of this study were to identify spectral wavelengths sensitive to cotton aphid infestation. Then, the normalized difference spectral indices (NDSI) and ratio spectral indices (RSI) based on the leaf spectrum were obtained within 350⁻2500 nm, and their correlation with infestation were qualified. The results showed that leaf spectral reflectance decreased in the visible range (350⁻700 nm) and the near-infrared range (NIR, 700⁻1300 nm) as aphid damage severity increased, and significan

Fusarium wilt (Fusarium oxysporum f. sp. vasinfectum) genes expressed during infection of cotton (Gossypium hirsutum)dagger.

SUMMARY We sought to identify Fusarium oxysporum f. sp. vasinfectum (Fov) genes that may be associated with pathogenicity. Initially we utilized microarray and Q-PCR technology to identify Fov genes expressed in root and hypocotyl tissues during a compatible infection of cotton. We identified 218 fungal clones representing 174 Fov non-redundant genes as expressed in planta. The majority of the expressed sequences were expressed in infected roots, with only six genes detected in hypocotyl tissue. The Fov genes identified were predominately of unknown function or associated with fungal growth an

Gossypium hirsutum gene of unknown function Gohir.A03G0737001 encodes a potential Chaperone-like Protein of protochlorophyllide oxidoreductase (CPP1).

A gene of unknown function identified in Gossypium hirsutum , Gohir.A03G0737001.1, was studied using sequence and bioinformatic tools. The encoded protein (referred to here as GhCPP1-A0A1U8HKT6) was predicted to function as a Chaperone-like protein of protochlorophyllide oxidoreductase (CPP1), which is involved with initiation of photochemical reactions of chlorophyll biosynthesis. Sequence analysis indicates it is embedded in the chloroplast envelope membrane through four transmembrane regions and contains a J-like domain that is structurally similar to the J domain of DnaJ/Hsp40 "holdase" ch

Azotobacter chroococcum as a potentially useful bacterial biofertilizer for cotton (Gossypium hirsutum): Effect in reducing N fertilization.

The aim of this research was to evaluate whether the application of two plant growth-promoting (rhizo)bacteria might reduce nitrogen fertilization doses in cotton. We used strains Azotobacter chroococcum AC1 and AC10 for their proven ability to promote seed germination and cotton growth. These microorganisms were characterized by their plant growth-promoting activities. Then, we conducted a glasshouse study to evaluate the plant growth promoting ability of these strains with reduced doses of urea fertilization in cotton. Results revealed that both strains are capable of fixing nitrogen, solubi

Identification and functional characterization of GhCSLD3, a negative regulator of fruit branch elongation in cotton (Gossypium hirsutum L.).

The fruit branch is a critical determinant of plant architecture in cotton, and its length optimization is pivotal for high-density planting and mechanical harvesting. However, the genetic mechanisms governing fruit branch length (FBL) are not fully elucidated. In this study, an F2 population was generated from two contrasting upland cotton cultivars, Blanco3363 (BL, compact) and Sanjiang (SJ, loose). Using two different methodologies of bulked-segregant analysis (BSA)-seq, we identified three stable overlapping QTLs on chromosomes A08 (119.8-122.46 Mb), D01 (7.51-10.10 Mb), and D09 (16.33-18.

Dose-Dependent Effects and Metabolomic Analysis of Foliar-Applied Carbon Dots on Cotton (Gossypium hirsutum L.) Growth.

While carbon dots (CDs) are promising green nanomaterials for sustainable agriculture, how they regulate cotton growth via physiological and metabolic reprogramming remains poorly understood. This study evaluated foliar CD application (0-200 mg·L-1) to uncover dose-response patterns and metabolic drivers. CDs exerted a concentration-dependent impact, with 100 mg·L-1 yielding optimal results. This application markedly boosted shoot and root biomass (fresh weight: 103.43% and 44.28%; dry weight: 56.03% and 40.39%) and enhanced specific leaf area by 52.51% over the control. Unveiling the role of heat shock protein 83 (HSP83) in Gossypium hirsutum in enhancing whitefly stress tolerance.

Heat shock proteins (HSPs) are key molecular chaperones that control protein homeostasis and stress signalling in plants. They have been widely studied for abiotic stresses, such as drought and heat; however, their role in insect-mediated biotic stress is little known, especially in polyploid crops like cotton, which is an important economic crop. Whitefly, Bemisia tabaci (Gennadius), infestation is a serious agronomic constraint in cotton (Gossypium spp.); however, efforts to completely investigate the genetic and molecular components driving host resistance against this pest have not been en

Identification of candidate genes associated with major QTL controlling Vertcillium wilt resistance in cotton (Gossypium hirsutum L.).

Vertcillium wilt (VW), a soil-borne disease causing cotton yield loss and fiber quality reduction, is a major stumbling block in cotton production. It is a challenging job to detect key genes associated with major quantitative trait loci (QTL) that can be used to develop VW-resistant varieties. In this study, several genome regions associated with major QTL controlling VW resistance were fine mapped by constructing the high-density genetic map of major QTL regions using Kompetitive Allele-Specific PCR (KASP) markers. Candidate genes were subsequently identified by combining gene annotation, ge
