Monitoring of insulin-like growth factors during growth hormone treatment: adulthood growth hormone deficiency.

Decisions on growth hormone (GH) dosing in paediatric practice have depended on body weight or surface area calculations. Although this policy was used initially in the treatment of adult patients, it has become apparent both in clinical studies and longitudinal observation that this results in relative overtreatment of male patients, with a high incidence of immediate side effects, and undertreatment of females. Use of individualized GH dosing against serum IGF-I has proven useful in optimizing therapy across the whole adult age range, including the elderly and in patients who are in transiti

Hemibrain growth as a biomarker for whole-brain growth.

OBJECTIVE: Accurate estimation of CSF and brain volume is an important component in evaluating hydrocephalus treatments, including shunt and endoscopic third ventriculostomy procedures. While MRI-based segmentation typically provides precise measurements, metallic artifacts from implanted shunts in patients with hydrocephalus can impede accurate volume determination. This study introduces a method for assessing brain growth in hydrocephalus patients using artifact-affected MR images and presents an efficient, automated AI-based pipeline for hemibrain segmentation and subsequent volume assessme

Growth and Growth Hormone Treatment in Noonan Syndrome.

Short stature is a common characteristic of Noonan Syndrome (NS), a genetic condition caused by mutations affecting the RAS / mitogen-activated protein kinase (MAPK) cascade. Growth hormone (GH) has been used to normalize childhood growth and increase adult height in NS. GH is effective in increasing growth velocity, and significantly improves height SDS at adult height. Studies of GH treatment to adult height have shown height gains of 9.5-13.0 cm for males and 9.0 - 9.8 cm for females. Factors associated with improved height outcomes are earlier initiation of therapy, a greater height SDS at

Growth patterns and the use of growth hormone in the mucopolysaccharidoses.

Short stature is characteristic of patients with mucopolysaccharidosis (MPS) diseases. For children with skeletal dysplasias, such as MPS, it is important to know the natural history of growth. An understanding of the natural growth pattern in each MPS disease provides a measurement to which treatments can be compared, as well as data which can help families and providers make individualized decisions about growth promoting treatments. Multiple advancements have been made in the treatment of MPS with both hematopoietic cell transplantation (HCT) and enzyme replacement therapy (ERT). The long t

Standards for growth and growth velocity in Turner's syndrome.

Turner-specific natural growth and growth velocity curves based on the retrospectively surveyed growth data of 704 Japanese patients in a mixed cross-sectional and longitudinal mode are reported. There was no significant difference in growth between the patients with karyotype 45,X and non-45,X. Twenty-three patients had a history of genital bleeding between the ages of 11 and 14 years (12 years +/- 9 months). The mean height of the patients with genital bleeding did not differ significantly from that without bleeding from birth until 1 year 3 months of age but thereafter the former were alway

[Growth retardation and compensatory growth in Perthes disease].

In 12 boys with a unilateral Morbus Perthes' disease who were not surgically treated, growth and skeletal maturation were prospectively recorded from the time of diagnosis onward. There were no significant deviations from the norm at the time the condition was diagnosed. Most of the boys manifested a distinctly retarded growth rate in the first phase following diagnosis; this was followed by a phase of excessive compensatory growth. Skeletal maturation showed a similar course. The course of growth would appear to be an expression of a transitory systemic disturbance.

Growth throughout childhood of children born growth restricted.

OBJECTIVE: Many studies that examine growth in growth-restricted children at birth do not discriminate between fetal growth restriction (FGR) and small for gestational age (SGA). These terms however are not synonymous. In SGA, stunting and increased weight gain have been reported. We do not know if this holds true for FGR. Our aim was to study postnatal growth until age 12.5 years in a cohort of children born FGR due to early onset placental insufficiency, and its relation to FGR severity. DESIGN: Prospective cohort study, follow-up of an antenatal randomised controlled trial in two tertiary

Growth Hormone and Insulin-Like Growth Factor-1.

Human growth hormone (GH) was first isolated from the human pituitary gland in 1945 and found to promote the growth of children with hypopituitarism. Since the formation of the World Anti-Doping Association, human GH has appeared on the list of forbidden substances. There is a significant amount of anecdotal evidence that human GH is misused by athletes to enhance performance, and there have been a number of high-profile cases of GH use in professional sport. GH secretagogues (GH-Ss), which increase GH secretion, and insulin-like growth factor (IGF-1), which mediates many of the effects of GH,

Growth without growth hormone (GH): a case report.

Growth without growth hormone (GH) has occasionally been described in patients with organic pituitary pathology, and even more rarely in patients with idiopathic pituitary hormone deficiency. The mechanism of growth without GH remains a mystery. We describe a 17-year old male who grew 38.5 cm in height over a 7-year period, despite the fact that he had established panhypopituitarism. The hypopituitarism was initially attributed to a presumptive hypothalamic hamartoma which was not, however, confirmed on subsequent and prolonged follow-up. Regular endocrine evaluation confirmed persistent anter

Growth Hormone, Not Simply Just a Hormone for Growth.

Since its discovery almost 100 years ago, growth hormone (GH) has been extensively studied to elucidate its structural characteristics, receptor interactions and its physiologic and non-physiologic effects. These actions include but are not limited to its effects on somatic growth, substrate metabolism, body composition, bone mineral density, cardiovascular system, and cognitive function. Contextually, recombinant human GH was approved for growth promotion in children and to enhance metabolic health in adult patients with GH deficiency (GHD), along with other clinical indications. Growth and Growth-Promoting Treatments in Turner Syndrome.

Short stature is a common presenting feature and an important concern for families of children with Turner syndrome. In this review, we summarize the data that shaped the updated international consensus guidelines for Turner syndrome published in 2024. The natural history of growth in Turner syndrome, the safety and efficacy of recombinant human growth hormone therapy, and the alternate growth promoting agents under consideration are presented. Timely, adequate growth hormone therapy can counter growth failure in childhood, promote catch-up growth and help many individuals with Turner syndrome

The effect of growth hormone on pituitary tumour growth.

PURPOSE OF REVIEW: In the last decade there has been an increased use of growth hormone replacement in patients with pituitary insufficiency. It has been shown to improve patient's quality of life, bone mass and cardiovascular risk factors but there are concerns about its mitogenic potential. RECENT FINDINGS: The most common aetiology of pituitary insufficiency in adults is hypothalamo-pituitary tumours or as a consequence of hypophysectomy or radiotherapy to treat pituitary tumours. Long-term growth hormone replacement is well established in clinical practice. We explore the available eviden

Enhancement of intestinal growth and repair by growth factors.

Recently, glucagon-like peptide 2 has emerged as a potent stimulator of epithelial growth, joining insulin-like growth factor I, hepatocyte growth factor and keratinocyte growth factor as potential treatment modalities for intestinal disorders associated with loss of mucosal mass, such as short bowel syndrome. Investigations into other members of the expanded epidermal growth factor peptide family, the development of more potent peptide analogues, and advances in the development of enterally administered bioactive growth factor formulations further expands the repertoire of epithelial growth f

Growth of infants with neonatal growth hormone deficiency.

In view of contradictory reports on the growth hormone dependency of early postnatal growth we studied the growth curves of 15 infants with neonatal growth hormone deficiency. In seven infants the growth curve was parallel to the standards of the infancy-childhood-puberty (ICP) model (group 1), but in the remaining infants there was an immediate deviation of the growth curve (group 2). In this group the mean (SD) at 4 and 9 months of age was -3.3 (1.1) and -4.9 (1.4), respectively, when compared with the ICP model. The mean birth length in both groups was decreased and five out of 15 had a bir

The role of the growth plate in longitudinal bone growth.

The epiphyseal growth plate is the main site of longitudinal growth of the long bones. At this site, cartilage is formed by the proliferation and hypertrophy of cells and synthesis of the typical extracellular matrix. The formed cartilage is then calcified, degraded, and replaced by osseous tissue. Proliferation and differentiation of cartilage cells (i.e., chondrocytes) as studied mostly in culture, is regulated by various endocrine, paracrine, and autocrine agents such as growth hormone, insulin-like growth factor-I (IGF-I), transforming growth factor (TGE-beta), and vitamin D metabolites (1

Growth hormone for the treatment of growth failure in children.

The etiology, diagnosis, and clinical features of growth failure in children are presented, with discussion of exogenous growth hormone (GH) replacement, its indications, efficacy, and adverse effects. Causes of growth delay include malnutrition, systemic illness, emotional deprivation, deficiency of endogenous growth hormone, thyroid hormone deficiency, and cortisol excess. Growth hormone (somatotropin) is secreted by the anterior pituitary gland in response to various stimuli, including exercise, hypoglycemia, and arginine. This hormone stimulates growth of skeletal muscle and connective tis

Targeting RACGAP1 suppresses growth hormone pituitary adenoma growth.

PURPOSE: Growth hormone pituitary adenoma (GHPA) is a major subtype of pituitary adenoma (PA), with tumor enlargement and abnormal secretion of growth hormone (GH) often causing complications. Rac GTPase-activating protein 1 (RACGAP1), a member of the guanine triphosphatase-activating protein family, is highly overexpressed in multiple tumors and promotes tumor growth. However, the role of RACGAP1 in GHPA remains unelucidated. Besides, specific inhibitors targeting RACGAP1 have not yet been developed. In this study, we aimed to determine the expression and function of RACGAP1 in GHPA and ident
