Growth-plate cartilage in chronic renal failure.

Bone growth occurs in the growth-plate cartilage located at the ends of long bones. Changes in the architecture, abnormalities in matrix organization, reduction in protein staining and RNA expression of factors involved in cell signaling have been described in the growth-plate cartilage of nephrectomized animals. These changes can lead to a smaller growth plate associated with decrease in chondrocyte proliferation, delayed hypertrophy, and prolonged initiation of mineralization and vascular invasion. As a result, chronic renal failure can result in stunted body growth and skeletal deformities.

Partial growth plate fusion caused by burn.

Growth plate fusion is a rare but important complication of burn injury in childhood. Reported cases are all of complete fusion. We present a case of partial fusion of the growth plates secondary to a burn injury, which to our knowledge has not been reported in the literature before. Partial fusion of the growth plate behaves differently as compared with complete fusion, and if treated at an early stage by appropriate surgery limb deformity and long-term disability may be prevented. The importance of early recognition and treatment cannot be overemphasized.

The Biomechanical Basis of Growth Plate Injuries.

In brief: Children are at high risk of growth plate injury, contrary to previous research data. Of 134 consecutive growth plate injuries seen at the University of Arizona Hospital in Tucson over a six-year period, 22% occurred in team sports and 20% in activities such as using playground equipment, horseback riding, roller and ice skating, skateboarding, and bicycling. The authors review clinically useful anatomical and biomechanical aspects of the growth plate and its injuries, reflecting their own research as well as other studies. They also discuss prevention and diagnosis of this common so

Recent research on the growth plate: Mechanisms for growth plate injury repair and potential cell-based therapies for regeneration.

Injuries to the growth plate cartilage often lead to bony repair, resulting in bone growth defects such as limb length discrepancy and angulation deformity in children. Currently utilised corrective surgeries are highly invasive and limited in their effectiveness, and there are no known biological therapies to induce cartilage regeneration and prevent the undesirable bony repair. In the last 2 decades, studies have investigated the cellular and molecular events that lead to bony repair at the injured growth plate including the identification of the four phases of injury repair responses (infla

Growth hormone regulates the stem cell population in the growth plate.

Growth hormone (GH) is a key systemic regulator of longitudinal bone growth and is widely used in pediatric endocrinology, including in patients without GH deficiency. Its primary target is the growth plate-a cartilaginous structure driving bone elongation-yet the cellular mechanisms underlying GH action remain incompletely understood. Here, we identify a direct role for GH in regulating a recently defined population of cartilaginous stem cells within the growth plate. Using multiple transgenic mouse models, we show that GH reduces the pool of slow-cycling, label-retaining stem cells by promot

Cyp26b1 within the growth plate regulates bone growth in juvenile mice.

Retinoic acid (RA) is an active metabolite of vitamin A and plays important roles in embryonic development. CYP26 enzymes degrade RA and have specific expression patterns that produce a RA gradient, which regulates the patterning of various structures in the embryo. However, it has not been addressed whether a RA gradient also exists and functions in organs after birth. We found localized RA activities in the diaphyseal portion of the growth plate cartilage were associated with the specific expression of Cyp26b1 in the epiphyseal portion in juvenile mice. To disturb the distribution of RA, we

Effects of exogenous growth hormone on growth plate cartilage in rats.

Slipped capital femoral epiphysis (SCFE) occurs with greater frequency in children with growth hormone deficiency than in children in the general population. This epidemiologic observation suggests that SCFE may be a complication of growth hormone deficiency, associated pituitary hormone deficiencies, growth hormone therapy, or a combination of these factors. To clarify the relationship of growth hormone treatment to SCFE, homologous growth hormone was administered to rats that were without growth hormone deficiency. This treatment was found to alter the material properties of the growth plate

Growth plate injuries of the wrist and hand.

Growth plate or physeal injuries are classified according to the zones of the physis that are affected and the disruption of bony structure. All physeal injuries should be reduced anatomically, particularly when an articular surface is involved. Inability to restore normal configuration can result in malunion, incongruous articular surfaces and premature cessation of growth plate activity in the region of injury. Innocuous-appearing injuries that do not involve articular surfaces may result in severe abnormalities.

The effects of delayed puberty on the growth plate.

BACKGROUND: Many athletes are beginning intense training before puberty, a time of increased bone accrual when up to 25% of total bone mineral accrual occurs. Female athletes experiencing late or delayed pubertal onset may have open epiphyseal plates that are vulnerable to injury. This investigation's purpose was to determine whether a delay in puberty (primary amenorrhea) affects the growth plate immediately postpuberty and at maturity. METHODS: Forty-eight female Sprague-Dawley rats (23 d old) were randomly assigned to 4 groups (n=12); short-term control (C-ST), long-term control (C-LT), sh

Sleeper Plate for Guided Growth: Does the Plate Material Change the Risk of Tethering?

BACKGROUND: Pediatric coronal plane deformities are commonly managed by guiding growth by placing an extraperiosteal 2-hole plate across the growth plate with 1 epiphyseal and 1 metaphyseal screw. Once the correction is achieved, removing the metaphyseal screw only (sleeper plate) has been described to facilitate easier future corrections if deformity rebounds. A complication commonly described with this approach is tethering, where the patient continues to overcorrect despite the elimination of tension across the physis. This study aims to evaluate the safety and efficacy of sleeper plates an

The growth plate: Zonal architecture, plasticity, and endocrine control of linear growth.

The growth plate (physis) is a highly specialized cartilaginous organ that drives longitudinal bone growth and ultimately determines adult stature. Its zonal architecture, including the resting, proliferative, hypertrophic, and calcification zones, integrates stem-like progenitor activity, clonal chondrocyte expansion, matrix remodeling, vascular invasion, and replacement by bone. Here, we review how the structural organization of the growth plate emerges from the interplay among extracellular matrix composition, sulfation pathways, and canonical paracrine signaling pathways, including Ihh-PTH

Impact of growth plate senescence on catch-up growth and epiphyseal fusion.

In mammals, longitudinal bone growth occurs rapidly in prenatal and early postnatal life, but then slows and eventually ceases. This deceleration, which reflects a decline in chondrocyte proliferation, was previously attributed to a hormonal or other systemic mechanism. However, new evidence suggests that it is due to a local mechanism within the growth plate. In particular, recent findings suggest that growth plate chondrocytes have a finite proliferative capacity that is gradually exhausted, causing growth to slow and finally stop. This concept has provided insight into clinical phenomena in

Rheb1 is required for limb growth through regulating chondrogenesis in growth plate.

Ras homology enriched in the brain (Rheb) is well established as a critical regulator of cell proliferation and differentiation in response to growth factors and nutrients. However, the role of Rheb1 in limb development remains unknown. Here, we found that Rheb1 was dynamically expressed during the proliferation and differentiation of chondrocytes in the growth plate. Given that Prrx1+ limb-bud-like mesenchymal cells are the source of limb chondrocytes and are essential for endochondral ossification, we conditionally deleted Rheb1 using Prrx1-Cre and found a limb dwarfism in Prrx1-Cre; Rheb1fl

Periosteal stem cells control growth plate stem cells during postnatal skeletal growth.

The ontogeny and fate of stem cells have been extensively investigated by lineage-tracing approaches. At distinct anatomical sites, bone tissue harbors multiple types of skeletal stem cells, which may independently supply osteogenic cells in a site-specific manner. Periosteal stem cells (PSCs) and growth plate resting zone stem cells (RZSCs) critically contribute to intramembranous and endochondral bone formation, respectively. However, it remains unclear whether there is functional crosstalk between these two types of skeletal stem cells. Here we show PSCs are not only required for intramembr

Leptin stimulates aromatase in the growth plate: limiting catch-up growth efficiency.

Catch-up growth (CUG) in childhood is defined as periods of growth acceleration, after the resolution of growth attenuation causes, bringing the children back to their original growth trajectory. Sometimes, however, CUG is incomplete, leading to permanent growth deficit and short stature. The aim of this study was to investigate the mechanisms that limit nutritional-CUG. Specifically, we focused on the crosstalk between leptin, increased by re-feeding, and sex hormones, which increase with age. In vivo studies were performed in young male Sprague Dawley rats fed ad libitum or subjected to 10/3

Histopathology of growth plate changes in induced abnormal bone growth in lambs.

Unilateral hip excision arthroplasty in lambs resulted in osteochondrosis developing in distal tibiae and metatarsi of the contralateral pelvic limb. The physeal cartilage of distal tibiae and metatarsi developed clefts which probably represent shear failure. The histopathology of the meataphyseal defects differed between the medial and lateral growth plate sectors. Medially, the lesions were typical of physeal osteochondrosis in other mammals, with disorganized chondrocyte columns, cell death and matrix necrosis. Laterally, metaphyseal defects were predominantly fibrous, and islands of bone f

New Biomaterials and Technologies in Growth Plate Regeneration.

Growth plate (GP) injuries present a significant clinical challenge due to their potential to induce limb deformities and growth disturbances. Traditional treatments, such as surgical resection and filler implantation, have low success rates and often result in bony bar recurrence. This review systematically summarizes recent advances in biomaterials and technologies for GP regeneration, including scaffolds, hydrogels, and microspheres, and emerging cell-free approaches. Pro-chondrogenic growth factors, extracellular vesicles (EVs), and stem cells are integrated into these biomaterials to enha

Transcriptional, epigenetic and microRNA regulation of growth plate.

Endochondral ossification is a critical event in bone formation, particularly in long shaft bones. Many cellular differentiation processes work in concert to facilitate the generation of cartilage primordium to formation of trabecular structures, all of which occur within the growth plate. Previous studies have revealed that the growth plate is tightly regulated by various transcription factors, epigenetic systems, and microRNAs. Hence, understanding these mechanisms that regulate the growth plate is crucial to furthering the current understanding on skeletal diseases, and in formulating effec
