Genotyping, pathogenicity, and immunogenicity of Avibacterium paragallinarum serovar B-1 isolates from the Americas.

The bacterium Avibacterium paragallinarum is the etiologic agent of infectious coryza of chickens. Among the nine Kume serovars currently recognized in this bacterium, serovar B-1 is a common serovar in the Americas. In the current study, serovar B-1 isolates from Ecuador (seven isolates), Mexico (seven isolates) and Panama (two isolates) were genotyped. In addition one Panamanian, one Ecuadorian, and two Mexican isolates were used in a vaccination-challenge trial in which the vaccine was based on the 2671 serovar B-1 reference strain. Genotyping by enterobacterial repetitive intergenic consen

Characterization of an Outbreak of Infectious Coryza (Avibacterium paragallinarum) in Commercial Chickens in Central California.

In 2017, the Turlock branch of the California Animal Health & Food Safety laboratory system received a significant increase in infectious coryza (IC) necropsy cases, with a total of 54 submissions originating from commercial broilers (n = 40), commercial layers (n = 11), and backyard chickens (n = 3). Layer flocks positive for IC were distributed within the adjacent counties of Merced and Stanislaus, while broiler flocks were concentrated within Merced County. The backyard flocks were located in Alameda and Sacramento counties. The clinical and pathologic presentation was consistent with IC, a

Identification of candidate antigens for a vaccine against Avibacterium paragallinarum using reverse vaccinology.

Infectious coryza, caused by Avibacterium paragallinarum, is a respiratory disease that results in significant economic losses for the global poultry industry. Current inactivated whole-cell vaccines often fail to provide broad protection, largely due to the extensive antigenic diversity among circulating strains. To address this limitation, we applied a reverse vaccinology strategy to identify candidate antigens for rational subunit vaccine development. Using genomic and proteomic data from 95 Av. paragallinarum strains and a subtractive genomics-immunoinformatics pipeline, we prioritized pro

Prevalence and genomic-based antimicrobial resistance analysis of Avibacterium paragallinarum isolates in Guangdong Province, China.

Infectious coryza (IC) is an acute infectious respiratory disease in chickens that is caused by Avibacterium paragallinarum (A. paragallinarum). A. paragallinarum poses a significant threat to poultry health due to its virulence and multidrug resistance. This study isolated and identified 21 A. paragallinarum isolates from Guangdong between 2022 and 2023. Biochemical tests showed that 100% of A. paragallinarum isolates fermented glucose but did not ferment alginate and galactose, and only YZ18 was nicotinamide adenine dinucleotide independent. To determine the genetic relatedness between these

The current epidemiological status of infectious coryza and efficacy of PoulShot Coryza in specific pathogen-free chickens.

Infectious coryza (IC) is an infectious disease caused by Avibacterium (Av.) paragallinarum. IC is known to cause economic losses in the poultry industry via decreased egg production in layers. Between 2012 and 2013, Av. paragallinarum was isolated from seven chicken farms by Chungbuk National University. We identified Av. paragallinarum, the causative pathogen of IC by polymerase chain reaction (PCR) and serovar serotype A, by multiplex PCR. Antibiotic sensitivity tests indicated that a few field-isolated strains showed susceptibility to erythromycin, gentamicin, lincomycin, neomycin, oxytetr

Identifcation of the genes involved in biofilm formation of Avibacterium paragallinarum using random transposon mutagenesis.

Infectious coryza (IC) is a respiratory disease in poultry caused by Avibacterium paragallinarum (Av. paragallinarum). The disease caused growth retardation in broilers and reduced egg production in laying hens, resulting in significant economic losses to the global chicken industry. The biofilm is an important virulence factor for many bacterial pathogens, yet there is a paucity of research on the biofilm of Av. paragallinarum. This study aimed to construct a random mutant library of Av. paragallinarum using the Tn5-Kan transposon to identify genes involved in biofilm formation. A total of ap

Development of a counterselection system for efficient marker-free genetic manipulation in Avibacterium paragallinarum.

UNLABELLED: Avibacterium paragallinarum causes infectious coryza in chickens, a condition that is prevalent worldwide and causes considerable losses for the poultry sector. However, the scarcity of information concerning the virulence factors of the bacterium and the underlying pathogenic mechanisms is largely due to the lack of effective and practical tools for genetic manipulation. Herein, the point mutation A294G was introduced in the gene pheS of Av. paragallinarum to establish a counterselectable marker AppheSm. The host strain harboring the gene AppheSm exhibited sensitivity to para-chlo

Validation of a real-time PCR assay for high-throughput detection of Avibacterium paragallinarum in chicken respiratory sites.

Avibacterium paragallinarum is the causative agent of infectious coryza, a highly contagious respiratory disease in chickens. Given its fastidious nature, this bacterium is difficult to recover and identify, particularly from locations colonized by normal bacterial flora. Standard PCR methods have been utilized for detection but are labor-intensive and not feasible for high-throughput testing. We evaluated a real-time PCR (rtPCR) method targeting the HPG-2 region of A. paragallinarum, and validated a high-throughput extraction for this assay. Using single-tube extraction, the rtPCR detected 4

Phylogenetic relationship of serovar C-1 isolates of Avibacterium paragallinarum.

The bacterium Avibacterium paragallinarum is the etiologic agent of infectious coryza of chickens. Serovar C-1 has emerged in infectious coryza outbreaks in layer hens of Ecuador and Mexico. In the current study, genotyping and phylogenetic analyses of five Ecuadorian and 10 Mexican isolates of Av. paragallinarum serovar C-1 were performed. All 15 isolates share a unique enterobacterial repetitive intergenic consensus-based-PCR fingerprint and have identical 16S ribosomal RNA and hemagglutinin antigen gene sequences. Results indicate that Ecuadorian and Mexican isolates of serovar C-1 of Av. p

Antimicrobial resistance patterns and genomic characterization of Avibacterium paragallinarum isolates collected in China from 2013 to 2021.

BACKGROUND: Avibacterium paragallinarum is the causative agent of infectious coryza in chickens and contributes to significant economic losses in poultry production. Antimicrobial therapy is widely used for disease control; however, increasing antimicrobial resistance (AMR) may compromise treatment efficacy. Long-term surveillance data on AMR in Av. paragallinarum in China remain limited. RESULTS: A total of 117 Av. paragallinarum isolates were analyzed, including 113 clinical isolates collected from 13 provinces in China between 2013 and 2021 and 4 historical international reference strains.

Evaluation of the ERIC-PCR as a probable method to differentiate Avibacterium paragallinarum serovars.

Infectious coryza, an upper respiratory tract disease in chickens, caused by Avibacterium paragallinarum, leads to huge economic losses. The disease is controlled through vaccination; but vaccination efficacy is dependent on correct identification of the infecting serovar, as limited cross-protection is reported amongst some serovars. Current identification methods include the heamagglutination inhibition test, which is demanding and could be subjective. To overcome this, molecular typing methods proposed are the Multiplex polymerase chain reaction (PCR) and Restriction Fragment Length Polymor
