Engineered Halomonas sp. Y3 enables highly efficient upcycling of poly(ethylene terephthalate) to polyhydroxyalkanoates.

The global plastic crisis demands innovative solutions for recycling polyethylene terephthalate (PET), a chemically stable polymer constituting 23 % of annual plastic waste. This study presents a significant advance in PET upcycling using engineered Halomonas sp. Y3, a halophilic bacterium uniquely suited for industrial bioprocessing. We addressed key challenges in PET valorization-inefficient assimilation of its depolymerized monomers, terephthalic acid (TPA) and ethylene glycol (EG)-through systematic metabolic engineering. First, TPA catabolism was enabled by integrating heterologous tph op

Biofilm formation on polystyrene in detached vs. planktonic cells of polyhydroxyalkanoate-accumulating Halomonas venusta.

Biofilm development is characterized by distinct stages of initial attachment, microcolony formation and maturation (sessile cells), and final detachment (dispersal of new, planktonic cells). In this work we examined the influence of polyhydroxyalkanoate (PHA) accumulation on bacterial surface properties and biofilm formation on polystyrene in detached vs. planktonic cells of an environmental strain isolated from microbial mats, Halomonas venusta MAT28. This strain was cultured either in an artificial biofilm in which the cells were immobilized on alginate beads (sessile) or as free-swimming (

Elucidating the salt-tolerant mechanism of Halomonas cupida J9 and unsterile ectoine production from lignocellulosic biomass.

BACKGROUND: Ectoine as an amino acid derivative is widely applied in many fields, such as the food industry, cosmetic manufacturing, biologics, and therapeutic agent. Large-scale production of ectoine is mainly restricted by the cost of fermentation substrates (e.g., carbon sources) and sterilization. RESULTS: In this study, Halomonas cupida J9 was shown to be capable of synthesizing ectoine using xylose as the sole carbon source. A pathway was proposed in H. cupida J9 that synergistically utilizes both WBG xylose metabolism and EMP glucose metabolism for the synthesis of ectoine. Transcripto

The inhibition effects of Cu and Ni alloying elements on corrosion of HSLA steel influenced by Halomonas titanicae.

Halomonas titanicae accelerated steel corrosion by dissimilatory Fe(III) reduction under anaerobic environments, and their adhesion was the key to achieving extracellular electron transfer between cells and Fe(III). This work investigated the inhibition effects of Cu and Ni alloying elements on corrosion of high strength low alloy (HSLA) steel affected by H. titanicae. It was found that both the addition of Cu (1.3%) and high content of Ni (7.2%) brought better corrosion resistance than the steel containing 4.8% Ni via decreasing the amount of sessile bacterial cells. And the inhibition effici

An efficient co-culture of Halomonas mongoliensis and Dunaliella salina for phenol degradation under high salt conditions.

Phenol is one of the major organic pollutants in high salt industrial wastewater. The biological treatment method is considered to be a cost-effective and eco-friendly method, in which the co-culture of microalgae and bacteria shows a number of advantages. In the previous study, a co-culture system featuring Dunaliella salina (D. salina) and Halomonas mongoliensis (H. mongoliensis) was established and could degrade 400 mg L-1 phenol at 3% NaCl concentration. In order to enhance the performance of this system, D. salina strain was subjected to adaptive laboratory evolution (ALE) by gradually in

PHB production from food waste hydrolysates by Halomonas bluephagenesis Harboring PHB operon linked with an essential gene.

Food wastes can be hydrolyzed into soluble microbial substrates, contributing to sustainability. Halomonas spp.-based Next Generation Industrial Biotechnology (NGIB) allows open, unsterile fermentation, eliminating the need for sterilization to avoid the Maillard reaction that negatively affects cell growth. This is especially important for food waste hydrolysates, which have a high nutrient content but are unstable due to batch, sources, or storage conditions. These make them unsuitable for polyhydroxyalkanoate (PHA) production, which usually requires limitation on either nitrogen, phosphorou

Halomonas, a newly recognized human pathogen causing infections and contamination in a dialysis center: three new species.

Our Renal Care Center (RCC) is a separate building, performing almost 2500 outpatient dialysis runs per month. In May 2007, 2 patients developed, days apart, bacteremia with an apparently identical nonfermentative Gram-negative rod. Because of difficulty identifying the organism, testing in the Biolog system identified them as a Halomonas species. Sequencing of approximately 1500 bases of the 16S rRNA gene in both organisms in 3 reference laboratories confirmed, searching against 3 databases, that the organisms were identical and were Halomonas species. There were 54 recognized species of this

Taxogenomic and Physiological Characterization of Halomonas shizuishanensis sp. nov. Reveals Osmotic-Adaptation Traits and an Ammonium-Utilization Phenotype.

Extreme saline-alkaline environments impose simultaneous osmotic and nitrogen stresses, yet the traits that enable microbial persistence and nitrogen utilization under these conditions remain incompletely understood. Here, we isolated and characterized a novel bacterium, Halomonas shizuishanensis DWK9T, from a saline-alkaline soil (salinity 5.75 g/kg, pH 8.35) in Shizuishan City, Ningxia, China, using a polyphasic taxonomic and genomic approach. Strain DWK9T grew at 0-20% (w/v) NaCl and pH 7.0-10.0. Although its 16S ribosomal RNA gene shared 98.4% similarity with H. lutescens Q1UTT, phylogenom

Identifying potential nutrient acquisition mechanisms for long-term survival: adaptive evolution of Halomonas isolated from subseafloor crustal fluids.

In nature, microbes must often survive for long periods of time under conditions of nutrient and carbon limitation while also facing extremes in temperature, pressure, and competition with other microbes. One low-carbon, cold, and high pressure environment is the subseafloor crustal aquifer, where fluids circulate through old ocean crust. While microbial communities are known to be present in these fluids and contribute to biogeochemical cycling, the survival strategies of microbes in these communities is poorly constrained. In this study, multiple Halomonas strains were isolated from subseafl

Effective production of Poly(3-hydroxybutyrate-co-4-hydroxybutyrate) by engineered Halomonas bluephagenesis grown on glucose and 1,4-Butanediol.

Halomonas bluephagenesis has been engineered to produce flexible copolymers P34HB or poly(3-hydroxybutyrate-co-4-hydroxybutyrate) from glucose and petrol-chemical precursor, γ-butyrolactone. Herein, gene cluster aldD-dhaT was constructed in recombinant H. bluephagenesis for catalyzing 1,4-butanediol (BDO) into 4-hydroxybutyrate, which could grow to 86 g L-1 dry cell mass (DCM) containing 77 wt% P(3HB-co-14 mol% 4HB) in 7-L bioreactor fed with glucose and bio-based BDO. Furthermore, 4HB monomer ratio could be increased to 16 mol% by engineered H. bluephagenesis TDH4-WZY254 with defected outer-m

An Efficient CO2-Upcycling Platform Based on Engineered Halomonas TD with Enhanced Acetate-Utilizing Capacity via Adaptive Laboratory Evolution.

Biohybrid conversion of carbon dioxide (CO2) into value-added bioproducts via engineered microbes using CO2-derived electrolytes (CDE) addresses global CO2 emissions, but most recombinants have poor saline CDE tolerance and low carbon conversion rate (CCR). Herein, Halomonas TD (salt-resistant) was adaptively evolved into TD80, which efficiently uses acetate; its aceE gene mutation (encoding pyruvate dehydrogenase) drives acetate utilization. Subsequently, different biosynthesis pathways in TD80 enabled high yields of poly-3-hydroxybutyrate (PHB), poly-3-hydroxybutyrate-co-4-hydroxybutyrate (P

Simultaneous conversion of chromium and malachite green coexists in halophilic bacterium Halomonas xianhensis SUR308 isolated from a solar saltern.

Many industries are known to use heavy metals like chromium (Cr) to fix dyes in the fabrication processes and malachite green (MG) as colorant. Alkalinity, elevated temperature, or salinity of the industrial effluents makes conventional physicochemical removal of MG and hexavalent chromium [Cr(VI)] more difficult to apply and demands to perceive potential cost-effective and environment-friendly treatment methods to eliminate or convert them into less toxic compounds. Here, we report simultaneous removal and bioconversion of MG and Cr(VI) by a halophilic biofilm-forming bacterium Halomonas xian

The Assembly of Alginate-Chitosan-Xanthine Hydrogel and Halomonas Improves Soil Quality of Protected Crop Growth Under Saline-Alkali Stress.

Soil saline-alkali stress has become a great risk, leading to significant decline in crop yields, especially in protected agriculture that is frequently threatened by excessive chemical fertilization. There is an urgent need to develop friendly strategies for improving the quality of saline-alkali soil. Microbial inoculants are promising agents in attenuating soil stress, but their colonization ability in the crop rhizosphere is often limited. To improve the efficiency of microbial inoculants, this study constructed the assembly of a salt-tolerant bacterium Halomonas (Homs) and a hydrogel comp

An exopolysaccharide produced by the novel halophilic bacterium Halomonas stenophila strain B100 selectively induces apoptosis in human T leukaemia cells.

Microbial exopolysaccharides (EPSs) are highly heterogeneous polymers produced by fungi and bacteria and have recently been attracting considerable attention from biotechnologists because of their potential applications in many fields, including biomedicine. We have screened the antitumoural activity of a panel of sulphated EPSs produced by a newly discovered species of halophilic bacteria. We found that the novel halophilic bacterium Halomonas stenophila strain B100 produced a heteropolysaccharide that, when oversulphated, exerted antitumoural activity on T cell lines deriving from acute lymp

Characterization of Halomonas sp. ZM3 isolated from the Zelazny Most post-flotation waste reservoir, with a special focus on its mobile DNA.

BACKGROUND: Halomonas sp. ZM3 was isolated from Zelazny Most post-flotation mineral waste repository (Poland), which is highly contaminated with heavy metals and various organic compounds. Mobile DNA of the strain (i.e. plasmids and transposons) were analyzed in order to identify genetic information enabling adaptation of the bacterium to the harsh environmental conditions. RESULTS: The analysis revealed that ZM3 carries plasmid pZM3H1 (31,370 bp), whose replication system may be considered as an archetype of a novel subgroup of IncU-like replicons. pZM3H1 is a narrow host range, mobilizable

Integrated Analysis of Paired Crude Oil and Produced Water Reveals Stress-Tolerant Hydrocarbon-Degrading Bacteria with Bioremediation Potential.

Oilfield microbiome studies have extensively characterized microbial community composition or isolated individual hydrocarbon degraders, but integrated exploration of cultivable functional bacteria from paired crude-oil and produced-water environments remains limited, particularly for strains combining hydrocarbon utilization with stress-tolerance and biosurfactant-related traits. In this study, paired crude-oil and produced-water samples from the Huoshaoshan Oilfield were investigated by combining 16S rRNA amplicon sequencing with culture-dependent isolation and functional screening.
