Diazepam (DZ), a common benzodiazepine, is increasingly detected in aquatic environments, yet its immunological effects in fish remain poorly understood. We investigated the impact of DZ at concentrations ranging from 1 to 10 mg/L on head kidney-derived macrophages of Amur catfish (Silurus asotus). Cell viability was unaffected, but DZ induced dose-dependent macrophage aggregation and significantly reduced phagocytic activity. Transcriptomic profiling revealed 1053 differentially expressed genes, with suppression of immune-related pathways, including PI3K-Akt, MAPK, phagosome, and toll-like re

The A2b adenosine receptor (A2bR) is a critical modulator of inflammatory responses. Despite its high expression in Japanese flounder (Paralichthys olivaceus) macrophages, the expression characteristics and immunomodulatory functions of A2bR in this species remain poorly understood. Here, we molecularly characterized flounder A2bR and established its function as a potent negative regulator of teleost inflammation. The flounder A2bR gene displays an exon-intron architecture analogous to that of its mammalian orthologs and is broadly expressed across tissues, with maximal abundance in the skin.

BACKGROUND: The incidence of acute kidney injury (AKI) is high among head and neck cancer (HNC) patients following Platinum-based concurrent chemo-radiotherapy (CCRT). However, the effect of renin-angiotensin-aldosterone system (RAAS) blockade on the risk of AKI in HNC patients undergoing CCRT is controversial. This study aimed to investigate the association between RAAS blockade, AKI and survival in HNC patients undergoing CCRT.
METHOD: This retrospective cohort study included 989 HNC patients treated between January 2016 and July 2022, with follow-up extending to July 2022. Among them, 65 (

Two continuous macrophage cell lines (LCM07 and LCM10) were established for the first time from the head kidney of the marine fish large yellow croaker (Larimichthys crocea). To date, both cell lines have been subcultured for more than 100 passages in 12 months. Notably, the LCM07 and LCM10 cells have distinct morphology and immune function. LCM07 cells showed strong contact inhibition in crowded conditions, while this was not observed in the LCM10 cells because they could grow in an overlapping manner. Correspondingly, LCM10 cells were slenderer than LCM07 cells. LCM07 cells had stronger phag

Proteins of the suppressor of cytokine signaling (SOCS) family function as inducible feedback inhibitors of cytokine signaling via the JAK/STAT pathway. Although several SOCS isoforms have been identified in teleosts, their immunological functions remain largely unknown. In this study, we identified in turbot Scophthalmus maximus a SOCS homologue (named SmSOCS3) of the mammalian SOCS3 type. The deduced amino acid sequence of SmSOCS3 contains 205 residues and shares extensive overall identities (60-82%) with those of known fish SOCS3. In silico analyses revealed that, like typical SOCS3, SmSOCS

Copper is used in treatment mixtures to control fungal diseases in vineyards plants. High concentrations of copper are inducing antioxidant stress in some aquatic ecosystems, and potential bioaccumulation in aquatic organisms has prompted the demand for alternative use of low toxic molecules in culture treatments. Chitosan is a biomolecule with antifungal and heavy metal ion chelating properties that may be used as a biopesticide. In this study, we investigate the potential toxicity of chitosan for aquatic animal health, alone or associated with copper. Carp (Cyprinus carpio L.) were exposed t

OBJECTIVE: Pacific salmon Oncorhynchus spp. reared in production hatcheries are typically fed high-lipid, energy-dense diets to achieve large size and high body condition prior to release. In contrast, juveniles in natural environments tend to consume low-lipid, high-protein diets, and fish reared for research or conservation purposes are sometimes fed diets that are formulated to mimic natural diets and promote wild-like phenotypes. Understanding how these alternative diets affect fish health beyond growth and body condition could ultimately contribute to improving hatchery fish fitness.

Trained immunity represents an evolutionarily conserved form of innate immune memory established through epigenetic and metabolic reprogramming. This study has demonstrated for the first time that butyrate, a key gut microbiota-derived metabolite, induces trained immunity in teleost macrophages. Our results revealed that priming head kidney macrophages (HKMs) from turbot (Scophthalmus maximus L.) with 1 μM of sodium butyrate (NaB) for 24 h followed by a 5-day training period, established a durable memory phenotype of HKMs. Importantly, the bactericidal capacity against Edwardsiella tarda (E.ta

As a kind of environmental pollutant, heavy metal Cadmium (Cd) exists widely in the environment. It is well known that Cd can accumulate and cause damage in liver, kidney and other organs. However, there are few studies on the immune cytotoxicity of Cd to fish. In particular, there are few studies on the toxicity of Cd to the head kidney lymphocytes of common carp. In order to further explore these mechanisms, we established an Cd exposure model in vitro. At the same time, we used the natural antioxidant astilbin (AST) to treat the cells to study its antagonistic effect on the toxicity of Cd.

Infection of carp with Listeria monocytogenes 4b resulted in decreased liver, spleen, and head kidney enzyme activities, involved in the metabolism of xenobiotics. After infection, cytochrome P-450 levels and ethoxyresorufin O-deethylase (EROD) activity were decreased while conjugation enzymes remained unaffected. The maximum decrease for phase I enzymes occurred on d 3. This loss of monooxygenase levels and activity could not be directly correlated with an increase in the number of organisms, as consistently high bacterial counts were observed in all three organs during infection. The effect

Triploid fish are considered a promising candidate for aquaculture; however, their immunological properties remain incompletely characterized. In this study, we investigated the immunological differences between 10-month-old triploid and diploid grass carp. Triploids exhibited a higher head kidney index and obvious macrophage aggregation in the head kidney compared to diploids. Among the immune and antioxidant indices, serum IgM content, head kidney lysozyme and acid phosphatase activities, and catalase and total superoxide dismutase activities, were significantly elevated in the triploids. Vaccination is an important strategy in the protection of aquaculture species from major diseases. However, we still do not have a good understanding of the mechanisms underlying vaccine-induced disease resistance. This is further complicated by the presence of several lymphoid organs that play different roles when mounting an immune response. In this study, we attempt to elucidate some of these mechanisms using a microarray-based approach. Asian seabass (Lates calcarifer) were vaccinated against Streptococcus iniae and the transcriptomic changes within the spleen and head kidney at one and se

BACKGROUND: High-dose cisplatin (≥200 mg/m2 cumulative) remains the standard of care in concurrent chemoradiotherapy (CRT) for locally advanced head and neck squamous cell carcinoma (LA-HNSCC). However, its use is frequently limited by nephrotoxicity, including acute kidney disease (AKD). This recently described clinical renal syndrome encompasses functional alterations of the kidney lasting fewer than 3 months post-exposure. Although hydration protocols and antiemetic strategies are routinely applied to avoid reduction in oral liquid intake and to prevent dehydration that could worsen renal f

Sichuan taimen (Hucho bleekeri) is a glacial relict and critically endangered fish species restricted to the Yangtze River drainage in China. The lack of knowledge regarding the morphology of immune organs may limit the timely detection of health threats, thereby hindering effective conservation efforts. To address this gap, the pilot study provides the first histological and ultrastructural characterisation of the main immune organs (spleen, head kidney, and trunk kidney) of H. bleekeri. The spleen comprises red and white pulp; the red pulp is abundant in erythrocytes, while the white pulp co

Lactococcus garvieae is a significant pathogen in aquaculture with a potential zoonotic risk. To begin to characterize the late immune response of trout to lactococcosis, we selected infected individuals showing clinical signs of lactococcosis. At the time lactococcosis clinical signs appeared, infection by L. garvieae induced a robust inflammatory response in the spleen of rainbow trout, which correlated with abundant granulomatous lesions. The response in kidney goes in parallel with that of spleen, and most of the gene regulations are similar in both organs. A correlation existed between th

This study focused on the effects of dietary selenium deficiency on structural integrity of the head kidney, spleen and skin in young grass carp (Ctenopharyngodon idella). A total of 540 healthy grass carp (mean weight 226.48 ± 0.68 g) were randomly divided into six groups and fed six separate diets with graded dietary levels of selenium (0.025-1.049 mg/kg diet) for 80 days. Results showed that selenium deficiency (1) caused oxidative damage in part by reducing the activities of antioxidant enzymes (such as SOD, CAT, GPx, GST and GR) and glutathione (GSH) content, down-regulating the transcrip

Lead (Pb) is a toxic heavy metal and an aquatic pollutant. Various amounts of heavy metals are released into the environment through industrial discharge, causing excessive contamination of aquatic ecosystems. The head kidney is a unique immune organ of the bony fish and plays an important role in the metabolism of heavy metals. Studies of toxic Pb exposure that have investigated the head kidney of carp are limited. This study was carried out to explore the potential immunotoxicity effects of Pb and the specific related mechanisms in the carp head kidney. Pb poisoning was shown to induce the p

Melatonin, the ubiquitous hormone marks its presence from plants to animals with remarkable antioxidant and anti-inflammatory properties. In the teleost, melatonin is secreted from the pineal gland, which functions as a modulator of circadian rhythm, being homologous to the suprachiasmatic nuclei of mammals. The current study revolves around evaluating the role of in vitro melatonin in the immune mechanism of the snakehead fish, Channa punctatus. Reactive oxygen and nitrogen species production was assayed by respiratory burst and nitrite assay. Mitogen-stimulated lymphocyte proliferation was d
