The Clot Thickens: Differential Coagulotoxic and Cardiotoxic Activities of Anguimorpha Lizard Venoms.

Despite their evolutionary novelty, lizard venoms are much less studied in comparison to the intense research on snake venoms. While the venoms of helodermatid lizards have long been assumed to be for defensive purposes, there is increasing evidence of toxic activities more useful for predation than defence (such as paralytic neurotoxicity). This study aimed to ascertain the effects of Heloderma, Lanthanotus, and Varanus lizard venoms on the coagulation and cardiovascular systems. Anticoagulant toxicity was demonstrated for the Varanus species studied, with the venoms prolonging clotting times

Purification and inflammatory edema induced by two PLA2 (Anch TX-I and Anch TX-II) from sea anemone Anthothoe chilensis (Actiniaria: Sagartiidae).

The Anch TX-I and II PLA(2) were purified from Anthothoe chilensis (Lesson, 1830) from the extract of the anemone after only two chromatographic step using molecular exclusion chromatography (Sephadex G-75) and reverse phase HPLC on μ-Bondapak C18 column. Both PLA(2) showed a molecular mass of ~14kDa determined by MALDI-TOF mass spectrometry and showed a high catalytic activity (data not showed). Although homologous with mammalian or snake venom group I PLA(2)s, Anch TX-I and II is sufficiently structurally different for the question of its placement into the existing PLA(2) classification sch

Amino acid sequences of helospectins, new members of the glucagon superfamily, found in Gila monster venom.

The amino acid sequences of two closely related peptides from Gila monster (Heloderma suspectum) venom are reported. Helospectin I is a 38-residue peptide, His-Ser-Asp-Ala-Thr-Phe-Thr-Ala-Glu-Tyr-Ser-Lys-Leu-Leu-Ala-Lys-Leu-Ala- Leu-Gln - Lys-Tyr-Leu-Glu-Ser-Ile-Leu-Gly-Ser-Ser-Thr-Ser-Pro-Arg-Pro-Pro-Ser-Ser, and helospectin II is a 37-residue peptide identical to helospectin I except that it lacks serine 38. Helospectins are pancreatic secretagogues with structures and bioactivities similar to vasoactive intestinal peptide and other members of the glucagon superfamily. The relative significa

The effect of helodermin in rat dispersed pancreatic acini.

Helodermin is a 35 amino acid-residue peptide of the vasoactive intestinal polypeptide (VIP) family, which was originally isolated from the venom of Heloderma suspectum on the basis of its capacity to stimulate adenylate cyclase in the rat pancreas. In the present study, using rat dispersed pancreatic acini, we examined the binding characteristics of helodermin, its action on amylase secretion, and the production of intracellular cyclic AMP (cAMP). Helodermin stimulated intracellular cAMP production dose dependently in a manner that was nearly identical to that of VIP. Helodermin stimulated am

CROSS-SPECIES TRANSMISSION OF A GENOGROUP C FERLAVIRUS IN A ZOOLOGICAL COLLECTION IN THE UNITED STATES.

Ferlaviruses have been associated with significant mortality events in squamates. Over a 5-month period in 2012, a mortality event at the Phoenix Zoo involved seven rattlesnakes (Crotalus spp.), a Sonoran gopher snake (Pituophis catenifer affinis), and a Gila monster (Heloderma suspectum). All individuals had been managed within the same group of enclosures at the zoo, and many of the affected individuals had previously been housed together. Clinical signs during the outbreak included sudden death, agonal behavior, anorexia, regurgitation, and loss of body condition. Histologic findings were s

Varanoid Tooth Eruption and Implantation Modes in a Late Cretaceous Mosasaur.

Erupting teeth are some of the oldest witnesses of developmental processes in the vertebrate fossil record and provide an important resource for vertebrate cladistics. Here, we have examined a mosasaur jaw fragment from central Texas using ultrathin ground section histology and 3D tomographic imaging to assess features critical for the cladistic placement of mosasaurs among varanoids vs. snakes: (i) the orientation of replacement teeth compared to the major tooth axis, (ii) the occurrence of resorption pits, and (iii) the mode of tooth implantation/attachment to the tooth bearing element (TBE)

From Gila Monster Venom to Dermatology: The History of GLP-1 Receptor Agonists.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have evolved from novel therapies for type 2 diabetes mellitus into widely used treatments for obesity and metabolic disease. Their origins can be traced to the Gila monster (Heloderma suspectum), a venomous lizard native to the deserts of the southwestern United States and northwestern Mexico. The discovery of exendin-4, a peptide isolated from the venom of the Gila monster, led to the subsequent development of longer acting GLP-1RAs. Long before its contribution to modern medicine would be recognized, the Gila monster was revered in Navajo

Exendin-4 is a high potency agonist and truncated exendin-(9-39)-amide an antagonist at the glucagon-like peptide 1-(7-36)-amide receptor of insulin-secreting beta-cells.

Exendin-4 purified from Heloderma suspectum venom shows structural relationship to the important incretin hormone glucagon-like peptide 1-(7-36)-amide (GLP-1). We demonstrate that exendin-4 and truncated exendin-(9-39)-amide specifically interact with the GLP-1 receptor on insulinoma-derived cells and on lung membranes. Exendin-4 displaced 125I-GLP-1, and unlabeled GLP-1 displaced 125I-exendin-4 from the binding site at rat insulinoma-derived RINm5F cells. Exendin-4 had, like GLP-1, a pronounced effect on intracellular cAMP generation, which was reduced by exendin-(9-39)-amide. When combined,

Thirst Responders: Effects of Hydration State on Body Temperature and Immune Responses to an Antigen Challenge.

Physiological performance is highly temperature sensitive, and enhanced physiological activity (e.g., digestion, reproductive, immune activation) often leads to a thermophilic response that elevates body temperature. Previous studies have shown that the hydration state of the organism may influence the extent of the thermophily, and other studies have shown that hydration state can alter physiological performance. However, it is rare for studies to comprehensively examine the interaction among hydration state, physiological performance, and the thermophilic response. We simultaneously examined

Amelioration of D-galactose-induced hyposalivation in aging rats by the GLP-1 receptor agonist Exendin-4.

Xerostomia is commonly associated with aging, diabetes, and other systemic conditions. It disrupts oral homeostasis and consequently elevates the likelihood of dental caries, periodontal disorders, and oral infections. Accordingly, mitigating xerostomia is expected to help prevent these oral pathologies and improve patients' quality of life. Because rodents cannot report subjective sensations of oral dryness, experimentally induced hyposalivation is used as an appropriate surrogate to study underlying mechanisms and potential interventions. Exendin-4 is a GLP-1 receptor agonist initially disco

A novel peptide isolated from Aphonopelma chalcodes tarantula venom with benefits on pancreatic islet function and appetite control.

Proof-of-concept for therapeutic application of venom-derived compounds in diabetes is exemplified by the incretin mimetic, exenatide, originally extracted from the saliva of the venomous Heloderma suspectum lizard. In this regard, we have isolated and sequenced a novel 28 amino acid peptide named Δ-theraphotoxin-Ac1 (Δ-TRTX-AC1) from venom of the Mexican Blond tarantula spider Aphonopelma chalcodes, with potential therapeutic benefits for diabetes. Following confirmation of the structure and safety profile of the synthetic peptide, assessment of enzymatic stability and effects of Δ-TRTX-AC1 o

Black widow spider alpha-latrotoxin: a presynaptic neurotoxin that shares structural homology with the glucagon-like peptide-1 family of insulin secretagogic hormones.

alpha-Latrotoxin is a presynaptic neurotoxin isolated from the venom of the black widow spider Latrodectus tredecimguttatus. It exerts toxic effects in the vertebrate central nervous system by depolarizing neurons, by increasing [Ca2+]i and by stimulating uncontrolled exocytosis of neurotransmitters from nerve terminals. The actions of alpha-latrotoxin are mediated, in part, by a GTP-binding protein-coupled receptor referred to as CIRL or latrophilin. Exendin-4 is also a venom toxin, and it is derived from the salivary gland of the Gila monster Heloderma suspectum. It acts as an agonist at the

Diet-dependent production of calcium- and phosphorus-rich 'spheroids' along the intestine of Burmese pythons: identification of a new cell type?

Burmese pythons, Python molurus bivittatus, digest the skeleton of their prey; this must result in a high amount of calcium and phosphorus passing through the intestinal lining. To determine how Burmese pythons can process this ion influx, the effects of different nutritional diets were examined in juveniles reared in captivity using three different diets: a normal diet with calcium and phosphorus provided from entire rodents; a low-calcium and phosphorus diet using rodents with no bones ('boneless prey'); and a calcium-rich diet composed of boneless rodents supplemented with calcium carbonate

Connectivity maps for biosimilar drug discovery in venoms: the case of Gila monster venom and the anti-diabetes drug Byetta®.

Like most natural product libraries animal venoms have long been recognized as potentially rich source of biologically active molecules with the potential to be mined for the discovery of drugs, drug leads and/or biosimilars. In this work we demonstrate as a proof of concept a novel approach to explore venoms for potential biosimilarity to other drugs based on their ability to alter the transcriptomes of test cell lines followed by informatic searches and Connectivity Mapping to match the action of the venom on the cell gene expression to that of other drugs in the Connectivity Map (C-Map) dat
