Characterization of the hemolytic activity of Haemophilus ducreyi.

H. ducreyi is the causative agent of chancroid, a genital ulcer disease most prevalent in developing countries. Chancroid enhances the heterosexual transmission of human immunodeficiency virus and is identified in focal outbreaks in the United States, but little is known about its pathogenesis. We studied the hemolysin produced by H. ducreyi because this molecule might be an important virulence factor in the pathogenesis of chancroid. Ten strains of H. ducreyi were tested on newly devised blood agar plates and were found to have hemolytic activity. We examined the hemolytic activity of H. ducr

A novel extracellular protease of Vibrio mimicus that mediates maturation of an endogenous hemolysin.

Vibrio mimicus, a human pathogen that causes gastroenteritis, produces an enterotoxic hemolysin as a virulence factor. The hemolysin is secreted extracellularly as an inactive protoxin and converted to a mature toxin through removal of the N-terminal propeptide, which comprises 151 amino acid residues. In this study, a novel protease having the trypsin-like substrate specificity was purified from the bacterial culture supernatant. The N-terminal amino acid sequence of the purified protein was identical with putative trypsin VMD27150 of V. mimicus strain VM573. The purified protease was found t

Heterogeneity of hemolysin expression during neonatal Streptococcus agalactiae sepsis.

The beta-hemolysin of Streptococcus agalactiae is a major virulence factor; consequently, nonhemolytic strains rarely cause infections. We report on a case of neonatal sepsis caused by a strain displaying heterogeneous hemolysin expression. It was detected by the simultaneous isolation of hemolytic and nonhemolytic colonies from cultures of the infant's blood.

A novel fully human recombinant antibody neutralizing α-hemolysin of Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is resistant to almost all β-lactam antibiotics. Hence, new ways to control MRSA infection, such as antibacterial antibodies, need to be explored. α-hemolysin is the most important virulence factor widely expressed in S. aureus. This study aimed to develop a new fully human antibody against α-hemolysin of S. aureus and research its neutralizing effect. The single-chain antibody fragments (scFvs) against S. aureus were screened from a fully human scFv library using phage display technology. The selected scFvs had good binding affinities to α-he

Diversity of β-hemolysins produced by the human opportunistic streptococci.

The genus Streptococcus infects a broad range of hosts, including humans. Some species, such as S. pyogenes, S. agalactiae, S. pneumoniae, and S. mutans, are recognized as the major human pathogens, and their pathogenicity toward humans has been investigated. However, many of other streptococcal species have been recognized as opportunistic pathogens in humans, and their clinical importance has been underestimated. In our previous study, the Anginosus group streptococci (AGS) and Mitis group streptococci (MGS) showed clear β-hemolysis on blood agar, and the factors responsible for the hemolysi

Streptococcus agalactiae 1a capsule (cpsE) and hemolysin (cylE) deletion mutants display attenuated virulence in Nile tilapia (Oreochromis niloticus).

Streptococcus agalactiae (S. agalactiae), also known as Group B Streptococcus (GBS), is an increasingly concerning pathogen affecting humans and animals, including aquatic species. This bacterium significantly impacts aquaculture, causing disease outbreaks in various freshwater and marine fish, particularly in warm-water environments. This study examined the biological impact of two major virulence factors of S. agalactiae 1a and their roles in host pathogenesis. We attenuated a strain of S. agalactiae 1a (NAM) through targeted deletions of selected virulence genes, cpsE and cylE, involved in

Transposon mutagenesis of group B streptococcus beta-hemolysin biosynthesis.

Beta-hemolysin production by group B streptococci (GBS) is speculated to be a major virulence factor of the organism. A virulent, beta-hemolytic group B streptococcus strain was mutagenized with the self-conjugative transposon Tn916 to derive isogenic strains with mutations only in the gene(s) responsible for beta-hemolysin biosynthesis. There was no significant difference between the virulence of the parent strain and that of the mutant strains in a neonatal rat sepsis model.

Characterization of Aqx and its operon: the hemolytic RTX determinant of Actinobacillus equuli.

Actinobacillus equuli, a member of the family Pasteurellaceae is the etiologic agent of a frequently lethal septicemia in neonatal foals as well as other more chronic diseases like arthritis, pleuritis, pneumonia or peritonitis. It may also be isolated from the oral cavity of healthy horses. Hemolytic isolates of A. equuli are known but so far no virulence determinants have been described for this bacterial species. By screening hemolytic A. equuli strains with specific gene probes, a hemolysin, designated Aqx (A. equuli RTX (repeats in the structural toxin)) was identified. This hemolysin was

[Pathogenicity of Burkholderia pseudomallei: extracellular and surface antigen functions].

The data of literature and the results of investigations carried out by the authors on the analysis of B. pseudomallei pathogenicity factors. They include mucoid, endotoxin, lecithinase, proteases, hemolysins, etc. In addition to fimbriae and pili, the adhesive properties of B. pseudomallei are supposed to be associated with surface capsule-like structures whose composition includes Ag8. The characterization of exoproteases, hemolysins and lethal toxins is given. The virulence and immunogenicity of B. pseudomallei were shown to be linked with the components of surface glycoprotein Ag8. The con

Group B streptococcal beta-hemolysin promotes injury of lung microvascular endothelial cells.

Group B streptococci (GBS) are the leading cause of pneumonia and sepsis in human newborns. Exudative pulmonary edema and alveolar hemorrhage seen in GBS pneumonia indicate vascular damage, and we reported that GBS injure lung microvascular endothelial cells (LMvEC) both in vivo and in vitro. The specific GBS factors causing LMvEC injury are uncertain, but GBS beta-hemolysin activity is associated with lung epithelial cell injury. We hypothesized that GBS beta-hemolysin contributes to LMvEC injury and exudative pulmonary edema. To test this hypothesis we used isogenic nonhemolytic and hyperhem

Identification and enumeration of virulent Listeria strains.

Recent outbreaks and a continuous increase in cases of listeriosis underscore the need for rapid, sensitive and reliable techniques to detect Listeria. Of the species of Listeria, only L. monocytogenes has been found to be associated with human infections. One factor which definitely contributes to its pathogenicity is the presence of hemolysins, although L. ivanovii and L. seeligeri also elaborate hemolysins. Based upon cloned hemolysin genes, we have developed DNA probes specifically for detecting L. monocytogenes. The technique combines growth of bacterial colonies on selective agar plates

[Pathogenic factors of vibrios with special emphasis on Vibrio vulnificus].

Bacteria of the genus Vibrio are normal habitants of the aquatic environment and play roles for biocontrole of aquatic ecosystem, but some species are believed to be human pathogens. These species can be classified into two groups according to the types of diseases they cause: the gastrointestinal infections and the extraintestinal infections. The pathogenic species produce various pathogenic factors including enterotoxin, hemolysin, cytotoxin, protease, siderophore, adhesive factor, and hemagglutinin. We studied various pathogenic factors of vibrios with special emphasis on protease and hemol

Thromboxane-mediated hypertension and vascular leakage evoked by low doses of Escherichia coli hemolysin in rabbit lungs.

Escherichia coli hemolysin has been implicated as a pathogenicity factor in extraintestinal E. coli infections including sepsis. In the present study the effects of intravascular administration of hemolysin were investigated in isolated blood-free perfused rabbit lungs. Low concentrations of the toxin in the perfusate (0.05-5 hemolytic units/ml, corresponding to approximately 5-500 ng/ml), caused a dose- and time-dependent release of potassium, thromboxane A2, and prostaglandin I2, but not of lactate dehydrogenase, into the recirculating medium, as well as a dose-dependent liberation of the pr

Acquired immunity to an intracellular pathogen: immunologic recognition of L. monocytogenes-infected cells.

Listeria monocytogenes (L. monocytogenes) is a pathogenic bacterium, and subclinical infection in mice is utilized as a prototypic model to investigate the development and expression of acquired resistance to facultative intracellular organisms. A key virulence factor of L. monocytogenes is the hemolysin listeriolysin O (LLO), and BALB/c mice immunized with hemolysin-secreting strains of L. monocytogenes develop specific acquired resistance, while mice immunized with hemolysin-negative strains or non-viable preparations of L. monocytogenes do not develop a protective immune response. Glucose and nonmaintained pH decrease expression of the accessory gene regulator (agr) in Staphylococcus aureus.

The effect of glucose on accessory gene regulator (agr) expression in Staphylococcus aureus was examined. agr is a global regulator that affects the expression of numerous genes, including those for some factors implicated in virulence, such as toxic shock syndrome toxin 1, alpha-hemolysin, and protein A. The agr locus determines two divergent transcripts, designated RNAII and RNAIII. RNAII contains four open reading frames (agrABCD), and RNAIII encodes delta-hemolysin. The mechanisms responsible for agr-mediated regulation are not well understood, but it appears that the RNAIII transcript pla

Inhibitory effect of totarol on exotoxin proteins hemolysin and enterotoxins secreted by Staphylococcus aureus.

Staphylococcus aureus (S. aureus) causes a wide variety of infections, which are of major concern worldwide. S. aureus produces multiple virulence factors, resulting in food infection and poisoning. These virulence factors include hyaluronidases, proteases, coagulases, lipases, deoxyribonucleases and enterotoxins. Among the extracellular proteins produced by S. aureus that contribute to pathogenicity, the exotoxins α-hemolysin, staphylococcal enterotoxin A (SEA) and staphylococcal enterotoxin B (SEB) are thought to be of major significance. Totarol, a plant extract, has been revealed to inhibi

Disruption of Functional Membrane Microdomains Enhances Methicillin-Resistant Staphylococcus aureus Pathogenesis via Hyperexpression of Hemolysins.

(1) Background: methicillin‑resistant Staphylococcus aureus (MRSA) is a zoonotic pathogen, and its hemolysins serve as key virulence factors. Functional membrane microdomains (FMMs) are protein-enriching platforms and regulate diverse physiological functions by recruiting and assembling various proteins. However, whether and how FMMs regulate the hemolytic ability of MRSA remains unclear. This study aimed to investigate FMM-mediated regulation of MRSA hemolysins and the underlying mechanisms. (2) Methods: Homologous recombination was employed to generate FMM-disrupted (N315ΔfloA) and complemen
