Construction of an infectious cDNA clone for Omsk hemorrhagic fever virus, and characterization of mutations in NS2A and NS5.

Omsk hemorrhagic fever virus (OHFV) is a member of the tick-borne encephalitis serocomplex of flaviviruses, and causes hemorrhagic disease in humans. In this study, an infectious cDNA of OHFV was constructed to investigate the molecular mechanisms involved in OHFV pathogenesis for the first time. Our cDNA clone was capable of producing infectious virus which is genetically identical to the parental Guriev strain, and the recombinant virus showed similar biological properties to the parental virus including growth kinetics and virulence characteristics. While characterizing the cDNAs, fortuitou

Omsk hemorrhagic fever virus: review of the biology, ecology, and disease associated with a historic tick-borne pathogen.

Omsk hemorrhagic fever virus (OHFV) is a tick-borne flavivirus associated with acute febrile illness and hemorrhagic disease in humans. OHFV was first identified in 1947 and since its identification has largely remained endemic to the Omsk region of the Russian Federation, where it continues to result in illnesses each year. However, the 2 primary tick vectors of OHFV exhibit ranges that extend far beyond the Omsk region, and the recent positive confirmation of OHFV in samples outside of the historic endemic range, in both the Russian Federation and neighboring Kazakhstan, to the south raises

Molecular variability and genetic structure of Omsk hemorrhagic fever virus, based on analysis of the complete genome sequences.

Omsk hemorrhagic fever virus (OHFV) is the etiological agent of Omsk hemorrhagic fever, a disease described in the 1940s in Western Siberia. However, until now, it has been represented in GenBank by just four complete genome sequences, which do not reflect the real genetic diversity of the virus in nature. In this study, we analyzed the molecular variability and genetic structure of OHFV based on 20 complete genome sequences, fifteen of which were obtained for the first time. All these sequences belong to virus strains isolated at different times from three regions of Western Siberia. The resu

Comparative analysis of immune responses to Russian spring-summer encephalitis and Omsk hemorrhagic fever viruses in mouse models.

Omsk hemorrhagic fever virus (OHFV) and Russian spring-summer encephalitis virus (RSSEV) are tick-borne flaviviruses that have close homology but different pathology and disease outcomes. Previously, we reported that C57BL/6 and BALB/c mice were excellent models to study the pathology and clinical signs of human RSSEV and OHFV infection. In the study described here, we found that RSSEV infection induced robust release of proinflammatory cytokines (IL-1α, IL-1β, IL-6 and TNF-α) and chemokines (MCP-1, MIP-1β, RANTES and KC) in the brain at 9 and 11dpi, together with moderate to low Th1 and Th2 c

Omsk Hemorrhagic Fever Virus and Powassan Virus Infections at Different Times After Immunization with the TBE Vaccine in In Vivo Experiments.

Ranges of distribution of some orthoflaviviruses, including the Omsk hemorrhagic fever virus (OHFV) and the Powassan virus (POWV), have been identified in the range endemic for the tick-borne encephalitis virus (TBEV). In contrast to TBEV, no registered vaccine against OHFV and POWV is currently available. Nevertheless, recent studies have indicated that the anti-TBEV vaccine may offer partial protection against other orthoflaviviruses. The present study assesses OHFV and POWV infection in an experimental model on BALB/c mice one, three- and twelve-months post-immunization with inactivated TBE

Cross-Reactive Antibodies to the NS1 Protein of Omsk Hemorrhagic Fever Virus Are Absent in the Sera of Patients with Tick-Borne Encephalitis.

Omsk hemorrhagic fever virus (OHFV) is a member of the tick-borne encephalitis virus (TBEV) complex of the Flaviviridae family. Currently, there are no data on the cross-reactivity of antibodies to the NS1 proteins of OHFV and TBEV. Such data are of major interest for monitoring viral encephalitis of unknown etiology due to the increasing geographical distribution of OHFV. In this study, a recombinant OHFV NS1 protein was produced using the Escherichia coli expression system and purified. The recombinant OHFV NS1 protein was recognized by specific mice immune ascetic fluids to the native OHFV

The First Record of Omsk Hemorrhagic Fever Virus and Tick-Borne Encephalitis Virus of Baltic Lineage from the Kemerovo Region of Russia.

Objectives: Tick-borne encephalitis virus Siberian subtype (TBEV-Sib) and Omsk hemorrhagic fever virus (OHFV) are causative agents of natural focal infections in Western Siberia, Russia. The distribution of TBEV phylogenetic lineages and OHFV in the Kemerovo Region of Western Siberia remains poorly investigated. Methods: The phylogenetic analyses of fragment genome sequences 26 flaviviruses identified in 2019 were performed, and the amino acid variation was determined to reveal to which clusteron they belong. The age of Baltic and Asian lineages of the TBEV-Sib was calculated for Kemerovo Dist

Orthoflavivirus omskense NS1 Protein Induces Microvascular Endothelial Permeability In Vitro.

Orthoflavivirus omskense (Omsk hemorrhagic fever virus, OHFV) is a tick-borne flavivirus that causes Omsk hemorrhagic fever (OHF), a severe zoonotic disease endemic to Western Siberia. Despite the fact that the role of NS1 proteins of various mosquito-borne flaviviruses in pathogenesis was investigated and their ability to affect human endothelial permeability was shown, the role of the NS1 protein of OHFV in pathogenesis is unstudied. In this work, the ability of OHFV NS1 to induce human endothelial permeability was investigated for the first time. It was shown that recombinant OHFV NS1 produ

[Experience with using the indirect hemagglutination reaction in studying natural foci of tick-borne encephalitis].

Using the indirect hemagglutination test, tick-borne encephalitis (TBE) virus could be detected in the nutrient medium of the infected cell cultures, in brain suspensions of the infected suckling mice and in most specimens of suspensions of Ixodid ticks containing active virus. Positive results were obtained only with erythrocyte immunoglobulin diagnostic preparation to TBE virus but not with the diagnostic preparation for Omsk hemorrhagic fever. In the indirect hemagglutination inhibition test sera from human patients similarly inhibited antigens of tick-borne encephalitis and Omsk hemorrhagi

[Differences in the electrophoretic mobility of the low-molecular virus-specific proteins of viruses of the tick-borne encephalitis complex].

Analysis of low-molecular virus-specific proteins of all the known members of the tick-borne encephalitis (TBE) complex viruses: TBE (2 strains), Langat, louping-ill, Negishi, Kyasanur Forest disease, Omsk hemorrhagic fever (2 strains), and Powassan, was performed. The cells infected with the above viruses were found to contain low-molecular virus-specific NVX (with the exception of Powassan, Omsk hemorrhagic fever, and Kyasanur Forest disease viruses), NV21/2, NV2, NV11/2, and NV1 proteins. These proteins (except NV1) differ in the electrophoretic mobility in the viruses under study and in TB

In vitro antiviral activity of adenosine analog NITD008 against tick-borne flaviviruses.

There are currently no antiviral therapies available for the tick-borne flaviviruses associated with hemorrhagic fevers: Kyasanur Forest disease virus (KFDV), both classical and the Alkhurma hemorrhagic fever virus (AHFV) subtype, and Omsk hemorrhagic fever virus (OHFV). In this brief study, we describe the in vitro antiviral activity of adenosine analog NITD008 against KFDV, AHFV, OHFV, as well as Tick-borne Encephalitis virus (TBEV). Alongside the well-established activity of NITD008 against mosquito-borne flaviviruses, our results have demonstrated the feasibility of identifying nucleoside
