Longitudinal and spatial declines in equine Hendra virus vaccination despite continued risk.

Hendra virus is an emerging zoonosis endemic to Australia with high fatality of infection in horses and humans. The role of infected domestic horses as amplifying bridge hosts for onward transmission to humans led to the creation of an equine vaccine as a One Health solution against spillover. Despite the risk this disease represents, previously reported equine vaccination rates were low. This study explored vaccination trends between 2012 and 2023 at fine spatial scales and identified demographic and ecological predictors of vaccination against emerging zoonoses. Over 1 million doses of Equiv

Cohorts of immature Pteropus bats show interannual variation in Hendra virus serology.

Understanding the drivers of seasonal disease outbreaks remains a fundamental challenge in disease ecology. Periodic outbreaks can be driven by several seasonally varying factors, including pulses of susceptible individuals through births, changes in host behaviour and social aggregation and variation in host immunity. However, when these potential drivers overlap temporally, isolating their relative contributions to outbreak patterns becomes challenging. We studied Hendra virus, a zoonotic pathogen with seasonal spillovers from bats to horses and humans. Multiple seasonal factors have been hy

Microclimates Might Limit Indirect Spillover of the Bat Borne Zoonotic Hendra Virus.

Infectious diseases are transmitted when susceptible hosts are exposed to pathogen particles that can replicate within them. Among factors that limit transmission, the environment is particularly important for indirectly transmitted parasites. To try and assess a pathogens' ability to be transmitted through the environment and mitigate risk, we need to quantify its decay where transmission occurs in space such as the microclimate harbouring the pathogen. Hendra virus, a Henipavirus from Australian Pteropid bats, spills-over to horses and humans, causing high mortality. While a vaccine is avail

Natural Hendra Virus Infection in Flying-Foxes - Tissue Tropism and Risk Factors.

Hendra virus (HeV) is a lethal zoonotic agent that emerged in 1994 in Australia. Pteropid bats (flying-foxes) are the natural reservoir. To date, HeV has spilled over from flying-foxes to horses on 51 known occasions, and from infected horses to close-contact humans on seven occasions. We undertook screening of archived bat tissues for HeV by reverse transcription quantitative polymerase chain reaction (RT-qPCR). Tissues were tested from 310 bats including 295 Pteropodiformes and 15 Vespertilioniformes. HeV was detected in 20 individual flying-foxes (6.4%) from various tissues including spleen

A neutralizing human monoclonal antibody protects african green monkeys from hendra virus challenge.

Hendra virus (HeV) is a recently emerged zoonotic paramyxovirus that can cause a severe and often fatal disease in horses and humans. HeV is categorized as a biosafety level 4 agent, which has made the development of animal models and testing of potential therapeutics and vaccines challenging. Infection of African green monkeys (AGMs) with HeV was recently demonstrated, and disease mirrored fatal HeV infection in humans, manifesting as a multisystemic vasculitis with widespread virus replication in vascular tissues and severe pathologic manifestations in the lung, spleen, and brain. Here, we d

Foals of mares vaccinated for Hendra virus have a suboptimal response to HeV vaccination.

Hendra virus (HeV) is lethal to horses and a zoonotic threat to humans in Australia, causing severe neurological and/or respiratory disease with high mortality. An equine vaccine has been available since 2012. Foals acquire antibodies from their dams by ingesting colostrum after parturition, therefore it is assumed that foals of mares vaccinated against HeV will have passive HeV antibodies circulating during the first several months of life until they are actively vaccinated. However, no studies have yet examined passive or active immunity against HeV in foals. Here, we investigated anti-HeV a

No Evidence of Hendra Virus Infection in the Australian Flying-fox Ectoparasite Genus Cyclopodia.

Hendra virus (HeV) causes potentially fatal respiratory and/or neurological disease in both horses and humans. Although Australian flying-foxes of the genus Pteropus have been identified as reservoir hosts, the precise mechanism of HeV transmission has yet to be elucidated. To date, there has been limited investigation into the role of haematophagous insects as vectors of HeV. This mode of transmission is particularly relevant because Australian flying-foxes host the bat-specific blood-feeding ectoparasites of the genus Cyclopodia (Diptera: Nycteribiidae), also known as bat flies. Using molecu

Periodic shifts in viral load increase risk of Hendra virus spillover from Pteropus bats.

Prediction and management of zoonotic spillover requires an understanding of infection dynamics within reservoir host populations. Spillover risk is commonly inferred from infection prevalence based on detection of viral genomic material, yet detection alone does not indicate the presence of infectious virus or a sufficient dose for transmission. We undertook a comprehensive investigation of Hendra virus shedding in its primary reservoir, Pteropus bats, analyzing quantitative PCR with reverse transcription (RT-qPCR) data from 6151 pooled urine samples collected across five sites over 3 years.

C-terminal tyrosine residues modulate the fusion activity of the Hendra virus fusion protein.

The paramyxovirus family includes important human pathogens such as measles, mumps, respiratory syncytial virus, and the recently emerged, highly pathogenic Hendra and Nipah viruses. The viral fusion (F) protein plays critical roles in infection, promoting both the virus-cell membrane fusion events needed for viral entry as well as cell-cell fusion events leading to syncytia formation. We describe the surprising finding that addition of the short epitope HA tag to the cytoplasmic tail (CT) of the Hendra virus F protein leads to a significant increase in the extent of cell-cell membrane fusion.

Hendra virus: research progress on a bat-borne threat lurking at the human-animal interface.

Hendra virus (HeV) is a highly lethal zoonotic henipavirus that has caused recurrent cross-species spillover events in Australia since 1994. Its capacity for cross-species spillover, together with its tropism for vascular endothelial and neural tissues, makes HeV important to veterinary medicine and public health. Flying foxes are the natural reservoir hosts, and horses act as intermediate and amplifying hosts through which all confirmed human infections have occurred. Detection of antibodies and viral RNA from related henipaviruses in Southeast Asian bats indicates regional henipavirus divers

Experimental challenge of African green monkeys with contemporary Hendra virus isolates produces divergent clinical disease.

Hendra virus (HeV) is a medically important, zoonotic paramyxovirus that emerged over thirty years ago which causes severe, often fatal disease in humans and animals. There are presently no approved medical countermeasures to prevent or treat human HeV disease, although many are in various stages of development. Critical to the stringent evaluation of these experimental countermeasures are nonhuman primate models of HeV disease which accurately recapitulate the pathogenesis of human infection. The continued emergence of HeV since its initial discovery in 1994 has recently expanded to include a

A single-cycle, recombinant VSV platform Nipah vaccine cross-protects against Hendra virus in nonhuman primates.

Nipah virus (NiV) and Hendra virus (HeV) are highly pathogenic paramyxoviruses that produce severe, often fatal disease in humans and animals. Zoonotic spillover of these henipaviruses from the Pteropid bat natural reservoir occurs near-annually in Southeast Asia and Oceania. Outbreaks of NiV disease frequently exceed case fatality rates of 75%, and person-to-person transmission makes controlling outbreaks in low-resource environments challenging. HeV is less transmissible between humans; however, the overall mortality rate is 57%. Approaches to human vaccine development have largely focused o

Evaluation of two point-of-care molecular diagnostic platforms for rapid detection of equine Hendra virus.

Hendra virus (HeV) is a lethal zoonotic pathogen endemic to eastern Australia, posing significant risks to equine and human health. Rapid field detection of HeV enables timely intervention and outbreak management. This study evaluated candidate point-of-care (POC) molecular diagnostic platforms for HeV detection in equine samples: including a loop-mediated isothermal amplification (DARQ RT-LAMP) assay and real-time reverse transcription quantitative polymerase chain reaction (RT-qPCR). Comparative analytical evaluation demonstrated that RT-qPCR exhibited superior analytical sensitivity relativ

Evaluation of a Single-Dose Nucleoside-Modified Messenger RNA Vaccine Encoding Hendra Virus-Soluble Glycoprotein Against Lethal Nipah virus Challenge in Syrian Hamsters.

In the absence of approved vaccines and therapeutics for use in humans, Nipah virus (NiV) continues to cause fatal outbreaks of encephalitis and respiratory disease in Bangladesh and India on a near-annual basis. We determined that a single dose of a lipid nanoparticle nucleoside-modified messenger RNA vaccine encoding the soluble Hendra virus glycoprotein protected up to 70% of Syrian hamsters from lethal NiV challenge, despite animals having suboptimally primed immune responses before challenge. These data provide a foundation from which to optimize future messenger RNA vaccination studies a

Development and validation of a chemiluminescent immunodetection assay amenable to high throughput screening of antiviral drugs for Nipah and Hendra virus.

There are currently no antiviral drugs approved for the highly lethal Biosafety Level 4 pathogens Nipah and Hendra virus. A number of researchers are developing surrogate assays amenable to Biosafety Level 2 biocontainment but ultimately, the development of a high throughput screening method for directly quantifying these viruses in a Biosafety Level 4 environment will be critical for final evaluation of antiviral drugs identified in surrogate assays, in addition to reducing the time required for effective antiviral drug development. By adapting an existing immunoplaque assay and using enzyme

Designing novel multiepitope mRNA vaccine targeting Hendra virus (HeV): An integrative approach utilizing immunoinformatics, reverse vaccinology, and molecular dynamics simulation.

Human and animal health is threatened by Hendra virus (HeV), which has few treatments. This in-silico vaccine design study focuses on HeV G (glycoprotein), F (fusion protein), and M (matrix protein). These proteins were computationally assessed for B and T-cell epitopes after considering HeV strain conservation, immunogenicity, and antigenicity. To improve vaccination immunogenicity, these epitopes were selectively ligated into a multiepitope construct. To improve vaccination longevity and immunological response, adjuvants and linkers were ligated. G, F, and M epitopes were used to create an m

Vaccination against Hendra virus of previously unvaccinated mares and their foals demonstrates the effectiveness of the current prime and boost vaccination schedule for foals.

Hendra virus (HeV) is a high consequence pathogen that causes severe respiratory and/or neurological disease in both horses and humans with high mortality. The equine vaccine, Equivac® HeV, is a recombinant soluble version of the G glycoprotein of HeV (HeV-sG) that elicits a potent HeV-neutralising antibody response in adult horses and also provides indirect protection for humans. Maternally derived HeV-G specific antibodies could interfere with subsequent foal vaccination, although other factors such as foal age and the suitability of vaccination protocols that have been optimised for adult h
