Prevention of organophosphate intoxication with presynaptic cholinergic blockers.

In Vivo cardiovascular neuromuscular preparation of rats was used to study the prophylaxis and mechanism of diisopropyl fluorophosphate (DFP) intoxication by presynaptic cholinolytic agents (ganglionic blocking agents), including hexamethonium, trimethaphan and mecamylamine. The lethal action of DFP was partially or completely prevented by pretreatment of rats with hexamethonium (10 mg/kg), trimethaphan (80 mg/kg) and mecamylamine (30 mg/kg). Combined use of these drugs with 2-PAM (100 mg/kg) improved further the prophylaxis of DFP lethality. The mechanism of prophylaxis could be due to the re

Aging increases venous compliance in awake instrumented rats.

To determine the effects of aging on total venous compliance, mean circulatory filling pressure (MCFP) was determined at several different blood volumes in 10 young (10-mo-old) and 10 older (30-mo-old) awake instrumented male Fischer 344/Brown Norway hybrid rats. Baseline weight and mean arterial pressure were similar in the two groups; heart rate was higher in the young (426 +/- 9 beats/min) than in the older rats (376 +/- 8 beats/min). Although MCFP was similar in the two groups at baseline blood volume, MCFP rose less with transient volume expansion and fell less with transient volume deple

Evidence for a respiration-modulated cholinergic action on the activity of medullary respiration-related neurons in the rabbit. An iontophoretic study.

Effects of the iontophoretically administered cholinergic agonists acetylcholine, bethanechol and DMPP on the activity of medullary respiration-related neurons were examined in urethane-anaesthetized rabbits. Inhibitory effects prevailed over excitatory effects. Analysis of cholinergic effects by cycle-triggered averaging revealed three major types of neuronal responses: (i) constant alterations of spike-density throughout the whole period of activity ("constant effects"), (ii) effects increasing during the progression of the burst of discharge or effects restricted to a particular fraction of

Local effect of substance P on colonic motor activity in different experimental states.

We studied the effects of close intra-arterial injections of substance P on colonic motor activity in the conscious state, during anesthesia, and during acute laparotomy. In the conscious state, with enteric nerves intact, substance P stimulated postsynaptic cholinergic neurons to induce a large amplitude and long duration contraction. This response was blocked by prior close intra-arterial injection of atropine and tetrodotoxin (TTX) but not hexamethonium. Hexamethonium and TTX given alone, close intra-arterially, induced a series of short-duration contractions. Prior close intra-arterial adm

Basal and spontaneous exocrine pancreatic secretion in conscious and anaesthetized chickens, Gallus domesticus.

Basal exocrine pancreatic secretion was studied in anaesthetized and conscious chickens and spontaneous secretion was studied in anaesthetized chickens. Results are compared with those of other vertebrates to estimate the specific pattern of this secretion in birds. The flow of pancreatic juice was greater in conscious chickens than when anaesthetized (Table 1). Amylase activity was greater than that of the other species and was of the same order in anaesthetized and conscious birds (Table 1). A spontaneous exocrine pancreatic secretion was seen to remain after eliminating a large part of the

The rabbit cornea lacks cholinergic receptors.

Cholinergic receptors were studied in membranes prepared from rabbit cornea, iris-ciliary body, and retina, using 3H-quinuclidinyl benzilate (3H-QNB) to identify muscarinic receptors and 125I-alpha-bungarotoxin (125I-BGT) to identify nicotinic receptors. Muscarinic cholinergic receptors were not found in the cornea. As a positive control, muscarinic cholinergic receptors were characterized in preparations of the iris-ciliary body. Specific binding of 3H-QNB to iris-ciliary body membrane preparations was saturable, with a Kd of 1.3 nM QNB. Specificity of the assay for muscarinic receptors was c

Sodium and volume depletion activates neurogenic mechanisms in renal hypertensive dogs.

The acute response to ganglionic blockade (hexamethonium bromide, 30 mg/kg, i.v.) was used to evaluate the neurogenic contributions to mean arterial pressure maintenance in the conscious one-kidney, one clip hypertensive dog. Approximately 2 hours (112 minutes) after ganglionic blockade, captopril (10 mg/kg, i.v.) was given to block the renin-angiotensin system. Hypertensive animals were studied 3 days after clipping (group 2) or 2 to 4 weeks after clipping (groups 3 and 4). Groups 2 and 3 were fed a regular sodium diet, but group 4 animals were sodium and volume depleted. Normotensive control

Amnesia induced by stimulation of the amygdala is attenuated by hexamethonium.

Bilateral subseizure stimulation of the amygdala given immediately following training in an inhibitory avoidance task produced retrograde amnesia. Hexamethonium (3.0 and 10.0 mg/kg), a peripherally acting nicotinic cholinergic antagonist, attenuated the retention deficits induced by amygdala stimulation if the drug was given 30 min prior to, but not immediately following training. Hexamethonium had no effect in normal unoperated animals, but did produce a retention deficit in operated control (nonstimulated) animals if it was given immediately following training (3.0 and 10.0 mg/kg). The resul

[Pharmacoprophylaxis of an experimental neurogenic pancreatic injury].

Experimental neurogenous injury to the rabbit pancreas was used to show that mechanical traumatization of the pyloroduodenal region leads to a considerable decrease in the creatine phosphate content and to the rise in the level of lactic acid in the gland tissue. Pharmacological analysis of the changes obtained demonstrated the reflex nature of the injury under consideration. High doses of noradrenaline were also found to cause pronounced metabolic alterations in the pancreatic tissue.

Autonomic nervous system activity in Macaca fascicularis.

This study was performed to determine the contributions of the sympathetic and parasympathetic nervous systems to cardiovascular control. Hexamethonium was administered to block the autonomic ganglia, propranolol to block beta adrenergic receptors of the sympathetic nervous system, and methylatropine to block the parasympathetic nervous system. The results of this study indicate high sympathetic tone and low parasympathetic tone in resting Macaca fascicularis. These findings are different from those in man but are similar to other nonhuman primates.

On the character of the electrical activity of the stomach, pyloric sphincter and duodenum of cats.

In cats with chronically implanted electrodes in the muscle wall of the gastro-intestinal tract, the electrical activity of the stomach (S) pyloric sphincter (PS) and duodenum (D) is characterized by cycles consisting of two phases: active--whereby 2 to 3 slow potential changes (SPC) are accompanied by spike potentials (SP) and a pause during which for 60-90 s only SPC are recorded. After bilateral transthoracic vagotomy there is a disruption of the cyclic character of the electrical activity in the investigated regions of the gastro-intestinal tract--SP from S and PS are recorded with each SP

Efficacy and toxicity of drug combinations in treatment of physostigmine toxicosis.

Atropine, in combination with 1 of 6 other drugs, was tested in mice for the ability to prevent death by an otherwise lethal dose of the cholinesterase inhibitor, physostigmine. The atropine dose (4 mg/kg, i.p.) was kept constant, while the dose of the other drug in the pair was tested in 5 geometrically spaced doses, ranging down to 1/16 of the maximum dose (which caused no gross behavioral signs). Atropine alone saved 20% of the mice. The combination of atropine and benactyzine saved 100% of the mice at all 5 doses of benactyzine; similar complete protection was afforded by the combination o

Interaction between histamine and dichloroisoproterenol, hexamethonium, pempidine, and diphenhydramine, in normal and reserpine-treated heart preparations.

Histamine stimulated the isolated auricles and heart of the guinea-pig. The effect was best seen in auricles which had been previously depressed by treatment with reserpine. Ganglionic blocking drugs (hexamethonium and pempidine), applied to auricles which had been previously treated with reserpine, abolished the diphasic effect of nicotine, but did not alter the response to histamine. Dichloroisoproterenol did not modify the stimulant action of histamine in isolated auricles, either before or after treatment with reserpine; nor did it alter the response of the isolated heart. Diphenhydramine

Effect of sodium azide on catecholamine release from isolated adrenal gland and on guanylate cyclase.

Sodium azide and other compounds which activate guanylate cyclase could stimulate catecholamine (CA) release from perfused dog adrenals. Verapamil reduced the secretory effect of sodium azide, but atropine and hexamethonium did not affect it while isobutyl methylxanthine potentiated it. Ca2+ deprivation abolished the stimulating effect of sodium azide on CA release and cyclic AMP output but the increased output of cyclic GMP remained. These results suggest the involvement of the Ca2+ influx mechanism in the secretory action of sodium azide.

Cholinergic interneurons in the feeding system of the pond snail Lymnaea stagnalis. II. N1 interneurons make cholinergic synapses with feeding motoneurons.

The N1 neurons are a population of interneurons active during the protraction phase of the feeding rhythm. All the N1 neurons are coupled by electrical synapses which persist in a high Mg/low Ca saline which blocks chemical synapses. Individual N1 spikes produce discrete electrotonic postsynaptic potentials (PSPS) in other N1 cells, but the coupling is not strong enough to ensure 1:1 firing. Bursts of N1 spikes generate compound PSPS in the feeding motoneurons. The sign (excitation or inhibition) of the N1 input corresponds with the synaptic barrage recorded during the protraction phase. Role of angiotensin and vasopressin on blood pressure of ganglionic blocked dogs.

This study was designed to investigate the possible role of angiotensin and vasopressin in the maintenance of arterial blood pressure during acute blockade of the autonomic nervous system. Two groups of eight dogs each were anesthetized with pentobarbital sodium, and autonomic ganglia were blocked with hexamethonium (20 mg/kg). Thirty minutes later group 1 received the vasopressin antagonist 1-(beta-mercapto-beta, beta-cyclopentamethylene propionic acid),2-(O-methyl)tyrosine arginine vasopressin (10 micrograms/kg) followed after a 30-min interval by captopril (1 mg/kg).
