Interstitiospinal action on forelimb, hindlimb, and back motoneurons.

Responses of motoneurons supplying muscles of the forelimbs, hindlimbs, and back to stimulation of the interstitial nucleus of Cajal (INC) were recorded intracellularly in cats under chloralose anesthesia. Stimulation of the ipsilateral and contralateral INC evoked predominantly excitatory postsynaptic potentials in these motoneurons. Response latencies and properties of responses to multiple shock stimuli indicated that the responses were evoked by a di- or polysynaptic pathway. Stimulation of the anterior MLF (P2), which should have activated the entire interstitiospinal tract, but few retic

ImmunoPET of CD146 in a Murine Hindlimb Ischemia Model.

Peripheral arterial disease (PAD) consists of a persistent obstruction of lower-extremity arteries further from the aortic bifurcation attributable to atherosclerosis. PAD is correlated with an elevated risk of morbidity and mortality as well as of deterioration of the quality of life with claudication and chronic leg ischemia being the most frequent complications. Therapeutic angiogenesis is a promising therapeutic strategy that aims to restore the blood flow to the ischemic limb. In this context, assessing the efficacy of pro-angiogenic treatment using a reliable noninvasive imaging techniqu

Hindlimb lameness and gait abnormalities in bitches with pyometra.

The objective of this study was to assess the frequency of gait abnormalities and lameness (GAL) in bitches with pyometra, and their association with clinical and laboratory findings. The study included 79 bitches diagnosed with pyometra and 35 negative control intact bitches presented with other soft tissue surgical disorders. Dogs with a history of chronic lameness due to orthopaedic or neurological origin were excluded. A history of GAL was more frequent in the pyometra group (47 per cent) compared with the control group (20 per cent) (P=0.007). In the pyometra group, bitches presenting GAL

Arthrodesis of the proximal interphalangeal joints of a hindlimb in a heifer.

A two-year-old Braunvieh heifer was presented with a traumatic luxation of the second phalanx of the medial digit and concurrent subluxation of the second phalanx of the lateral digit of the right hindlimb. Closed reduction of both luxations was possible. Surgical arthrodesis was achieved using one narrow 4.5 mm three-hole equine locking compression plate for each joint. Placement of the bone plates resulted in stable arthrodesis of both proximal interphalangeal joints of the right hindlimb but there was persistent residual lameness. The heifer delivered a healthy calf but was slaughtered eigh

Murine hindlimb lymphedema model: optimization and evaluation of radiation.

BACKGROUND: Post-surgical lymphedema frequently occurs following lymph node dissection. The murine tail is the most commonly used model to study secondary lymphedema. The murine hindlimb model offers a more clinically translatable approach but results in the literature have been inconsistent. The purpose of this study is to optimize the murine hindlimb lymphedema to achieve consistent results and assess the utility of radiation. METHODS: C57BL/6 mice underwent either 20-Gy irradiation of one hindlimb 7 days prior to surgery (n = 11) or no radiation (n = 9). For all mice, a circumferential ski

Goats decrease hindlimb stiffness when walking over compliant surfaces.

Leg stiffness, commonly estimated as the 'compression' of a defined leg element in response to a load, has long been used to characterize terrestrial locomotion. This study investigated how goats adjust the stiffness of their hindlimbs to accommodate surfaces of different stiffness. Goats provide a compelling animal model for studying leg stiffness modulation, because they skillfully ambulate over a range of substrates that vary in compliance. To investigate the adjustments that goats make when walking over such substrates, ground reaction forces and three-dimensional trajectories of hindlimb

Embryonic blastemic changes in retinoic acid-induced hindlimb deformity.

In this study we analyzed, on an experimental model, blastemic changes occurring during the embryonic period that may later cause fetal hindlimb deformity. Experimental induction of clubfoot-like deformity in rat fetuses was performed by maternal administration of retinoic acid (RA) (120 mg/kg body weight) as a single intragastric dose on day 10 of pregnancy (previous phase of this assay). The caudal and hindlimb blastemic changes were studied by mitosis count and stereological, immunohistochemistry and AgNOR techniques in the hindlimbs of 15-day embryos and in the caudal somites of 11-day emb

Targeted disruption of Hoxd-10 affects mouse hindlimb development.

Targeted disruption of the Hoxd-10 gene, a 5' member of the mouse HoxD linkage group, produces mice with hindlimb-specific defects in gait and adduction. To determine the underlying causes of this locomotor defect, mutant mice were examined for skeletal, muscular and neural abnormalities. Mutant mice exhibit alterations in the vertebral column and in the bones of the hindlimb. Sacral vertebrae beginning at the level of S2 exhibit homeotic transformations to adopt the morphology of the next most anterior vertebra. In the hindlimb, there is an anterior shift in the position of the patella, an oc

Development and validation of a kinematic hindlimb cycling model for rats.

Functional electrical stimulation (FES) bicycle training is a physical rehabilitation technique used to promote muscle recovery and/or cardiorespiratory health in persons with lower extremity impairment due to neurologic injury. FES cycling may also increase bone mineral density (BMD) in such populations, although no consensus exists that supports FES-induced skeletal improvement, in-part due to the extended 9-12 + month duration necessary to detect BMD gain in humans and the multitude of FES parameter permutations that require optimization to improve bone strength and/or reduce fracture risk,

Imaging of a subcutaneous abscessation in the back of a calf with hindlimb paralysis.

BACKGROUND: Ultrasonography is not chosen as the common imaging modality to diagnose spinal cord diseases. The present report indicates good diagnostic efficacy of ultrasonography for identifying spinal cord compressed by subcutaneous mass when scanning through the defected vertebral laminae and spinous process. CASE DESCRIPTION: A five-month-old female Holstein calf presented with progressive hindlimb paralysis following a surgical resection of a back mass conducted at 21 days of age. The mass was subsequently histopathologically diagnosed as a pulmonary choristoma. Alongside hindlimb paraly

Celsr3 is required in motor neurons to steer their axons in the hindlimb.

The cadherin Celsr3 regulates the directional growth and targeting of axons in the CNS, but whether it acts in collaboration with or in parallel to other guidance cues is unknown. Furthermore, the function of Celsr3 in the peripheral nervous system is still largely unexplored. Here we show that Celsr3 mediates pathfinding of motor axons innervating the hindlimb. In mice, Celsr3-deficient axons of the peroneal nerve segregate from those of the tibial nerve but fail to extend dorsally, and they stall near the branch point. Mutant axons respond to repulsive ephrinA-EphA forward signaling and glia

An investigation of the relationship between hindlimb lameness and saddle slip.

REASONS FOR PERFORMING STUDY: We have observed saddle slip consistently to one side because of a crooked rider, an ill-fitting saddle, asymmetry in a horse's thoracolumbar shape and lameness. Currently, there are no objective data to permit assessment of the relative importance of each factor. OBJECTIVES: To document the frequency of occurrence of saddle slip in horses with hindlimb lameness compared with other horses. To describe the effect of lameness characteristics and grade, the abolition of lameness by diagnostic analgesia, breed, size, thoracolumbar shape and symmetry and the rider's w

Myelodysplasia as a cause of hindlimb ataxia in two beef calves.

Myelodysplasia is a general term referring to abnormal development of the spinal cord. Unless associated with vertebral malformations, it can be difficult to distinguish clinically from other causes of spinal cord disease. These case reports describe the clinical and pathological findings in two calves with a distinctive non-progressive pelvic limb ataxia. The syndrome was observed in two calves on a large, extensively managed beef cattle property near Richmond, north Queensland. Both calves had similar clinical signs, including hindlimb ataxia with swaying of the pelvis and a well-coordinated

New experimental model of crush injury of the hindlimbs in rats.

BACKGROUND: Crush injury (CI) remains a life-threatening condition. Because there is a shortage of animal models of CI, we purposed to develop a reproducible model of CI of hindlimbs in rats and to evaluate correlation between the volume of muscles traumatized and the severity of CI. METHODS: The right or both hindlimbs of anesthetized rats were compressed for 6 hours under blocks weighing 3 kg. This was followed by 3 hours of reperfusion. Serum lactate, base excess (BE), and potassium (K) were measured at 10 minutes after cannulaton (baseline), immediately before release (compression), and 3

Papaverine injection into the hindlimb circulation stimulates ventilation in sheep.

To test the hypothesis, previously suggested by Huszczuk et al. (1993), that distention of the peripheral microvascular network could, per se, stimulate ventilation, the ventilatory effects of papaverine-induced muscular vasodilation were studied in ten anaesthetized sheep. Because systemic action of papaverine may involve the arterial baro- and chemoreceptors, the animals were surgically prepared for a reversible isolation of the hindlimb circulation. Papaverine injection (1-2 mg/kg) into the arterial inflow of the isolated limbs provoked a 13 +/- 6 sec-delayed increase in VE by 1.8 +/- 0.2 L

Anoxia and capillary filtration coefficient in the isolated cat hindlimb.

Isolated cat hindlimbs were perfused from a reservoir with an albumin-blood mixture at a constant flow of 20 ml X min-1 X 100 g muscle-1 while alternately bubbling the perfusate with either 95% O2-5% CO2 gas mixture or a 95% N2-5% CO2 mixture for 50- to 60-min periods. Capillary filtration coefficient (CFC), vascular resistance (R), and perfusate O2 content were measured in each period. The arterial O2 content fell from fully equilibrated to 0.25 vol% during the use of N2. R fell from about 6 mmHg X min X 100 g X ml-1 during O2 bubbling to approximately 1.0 with N2. CFC averaged 0.012 +/- 0.00

Effects of enkephalins on perfusion pressure in isolated hindlimb preparations.

A series of studies were conducted to determine the effects of leucine-(leu-) enkephalin and methionine-(met-) enkephalin on perfusion pressure. These experiments utilized isolated perfused femoral arterial preparations in pentobarbital-anesthetized cats. The enkephalins were administered intraarterially into the femoral artery and changes in perfusion pressure recorded. Leu-enkephalin in doses of 1 microgram to 320 micrograms produced significant dose-dependent decreases in perfusion pressure (4.0 +/- 1.3% with 1 microgram to 19.1 +/- 2.1% with 320 microgram). Similar declines in perfusion pr

The functional anatomy of the hindlimb of some African Viverridae (Carnivora).

The functional anatomy of the hindlimb of 12 species of viverrids was studied with relation to locomotion. The animals were allocated to primary locomotor categories on the basis of their anatomy and locomotion. The climbing, arboreal walking category (Nandinia binotata) is characterized by a small sacroiliac articulation, the iliopsoas inserts onto a medially located lesser trochanter and the femoral condyles are not posteriorly placed. The hindfoot is plantigrade and its structure permits considerable movement. The pads are soft and the claws retractile. Representatives of the arboreal and t

Discharges of single hindlimb afferents in the freely moving cat.

1. Implanted semimicroelectrodes were used to record single afferent fiber discharges from L7 dorsal roots during unrestrained walking in the conscious cat. 2. A series of tests were used to identify an afferent during a short period of anesthesia following each recording session. The majority of afferents were from muscle spindle primary endings in hindlimb muscles. 3. Ankle extensor spindle primaries generally showed their highest firing rates during that phase of stepping in which they were passively stretched. During active muscle contraction there was evidence of fusimotor drive, although
