Insecticidal Effects of Triterpene Glycosides Extracted From Holothuria atra (Echinodermata: Holothuroidea) Against Spodoptera litura (Lepidoptera: Noctuidae).

Spodoptera litura (Fabricius) is economically important in many countries. Many biopesticides have been developed and studied to control this pest species; however, few natural products from marine organisms have been studied for their pesticidal efficiency. This study compared the acute contact toxicity of solvent extracted from the viscera of Holothuria atra and isolated triterpene glycoside compounds on third-instar S. litura. Our results showed that crude methanolic extracts were the most toxic to the larvae (LC50 ~ 10.37 mg/ml) compared with other crude extracts and contained the bioactiv

Sex steroids and steroidogenesis-related genes in the sea cucumber, Holothuria scabra and their potential role in gonad maturation.

The sea cucumber Holothuria scabra is an economically valuable marine species which is distributed throughout the Asia-Pacific region. With the natural population declining due to over fishing, aquaculture of this species is deemed necessary. Hence, it is essential to understand the mechanisms regulating the reproduction in order to increase their populations. Sex steroids, including estrogens, androgens and progestogens, play an important role in reproduction in most vertebrates and several invertebrates. It has been proposed that sea cucumbers have the same sex steroids as vertebrates but th

Epidemiology of a SKin Ulceration Disease (SKUD) in the sea cucumber Holothuria scabra with a review on the SKUDs in Holothuroidea (Echinodermata).

Aquacultivated sea cucumbers often suffer from SKin Ulceration Diseases (SKUDs). SKUDs have been observed in six holothuroid species from nine countries. All SKUDs present a similar symptom-the skin ulceration-and can be induced by bacteria, viruses, or abiotic factors. We here provide an update on SKUDs in holothuroids and analyse the case of the SKUD observed in Holothuria scabra in Madagascar. Field observations revealed a seasonality of the disease (i.e. wintertime maximum peak). Morphological analyses of integument ulcers showed that sea cucumbers react by forming a collagen fibre plug. Tensilin-like stiffening protein from Holothuria leucospilota does not induce the stiffest state of catch connective tissue.

The dermis of sea cucumbers is a catch connective tissue or mutable connective tissue that exhibits large changes in mechanical properties. A stiffening protein, tensilin, has been isolated from the sea cucumber Cucumaria frondosa. We purified a similar protein, H-tensilin, from Holothuria leucospilota, which belongs to a different family to C. frondosa. H-tensilin appeared as a single band with an apparent molecular mass of 34 kDa on SDS-PAGE. No sugar chain was detected. Tryptic fragments of the protein had homology to known tensilin. H-tensilin aggregated isolated collagen fibrils in vitro

Genome-wide analysis unveils the differentiated "external" and "internal" lectin system in the tropical sea cucumber Holothuria leucospilota.

To date, the understanding of lectin-related immunity in sea cucumbers remains limited. Through a comprehensive genome-wide analysis, 133 lectin genes were identified from Holothuria leucospilota, a sea cucumber species with high ecological and economic significance in the Pacific and Indian Oceans. These lectins were classified into five subtypes, with C-type lectin being the predominant form, and the presence of SUEL-like lectin and Malectin reported here for the first time in sea cucumbers. Transcriptomic analysis across twelve tissues and sixteen developmental stages revealed ubiquitous ex

Assessment of the therapeutic efficacy of Holothuria polii extract during the muscular phase of Trichinella spiralis infection in albino mice.

Marine invertebrates are an important source of bioactive natural products with potential therapeutic applications, owing to their chemically diverse defense mechanisms. Trichinellosis is a widespread food-borne zoonosis caused by Trichinella spiralis (T. spiralis). Albendazole (ABZ) is the main treatment, but its efficacy against encysted and newborn larvae is limited. This limitation, together with emerging resistance, has prompted the search for safer alternative antiparasitic agents, particularly natural compounds. Holothuria polii (H. polii) is a bioactive-rich sea cucumber with promising

HOLOTHURIA SCABRA METHANOL EXTRACT INHIBITS CANCER GROWTH THROUGH TGF-β/PI3K/PTEN SIGNALING PATHWAY IN BREAST CANCER MICE MODEL.

BACKGROUND: Molecules and cytokines can be targeted in cancer therapy. Transforming growth factor-beta (TGF-β) is a cytokine that acts on protein kinase receptors in the plasma membrane. The signaling pathway of TGF-β can trigger the phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K) pathway, a signal transduction pathway important in cancer growth and development. However, this PI3K/AKT cascade can be inhibited by phosphatase and tensin homolog (PTEN) tumor suppressor genes. AIM: To determine the inhibitory effect of Holothuria scabra methanol extract (HSE) on breast cancer growth through

Fucosylated chondroitin sulfates from the body wall of the sea cucumber Holothuria forskali: conformation, selectin binding, and biological activity.

Fucosylated chondroitin sulfate (fCS) extracted from the sea cucumber Holothuria forskali is composed of the following repeating trisaccharide unit: → 3)GalNAcβ4,6S(1 → 4) [FucαX(1 → 3)]GlcAβ(1 →, where X stands for different sulfation patterns of fucose (X = 3,4S (46%), 2,4S (39%), and 4S (15%)). As revealed by NMR and molecular dynamics simulations, the fCS repeating unit adopts a conformation similar to that of the Le(x) blood group determinant, bringing several sulfate groups into close proximity and creating large negative patches distributed along the helical skeleton of the CS backbone.

A Synthetic Derivative of Antimicrobial Peptide Holothuroidin 2 from Mediterranean Sea Cucumber (Holothuria tubulosa) in the Control of Listeria monocytogenes.

Due to the limited number of available antibiotics, antimicrobial peptides (AMPs) are considered antimicrobial candidates to fight difficult-to-treat infections such as those associated with biofilms. Marine environments are precious sources of AMPs, as shown by the recent discovery of antibiofilm properties of Holothuroidin 2 (H2), an AMP produced by the Mediterranean sea cucumber Holothuria tubulosa. In this study, we considered the properties of a new H2 derivative, named H2d, and we tested it against seven strains of the dangerous foodborne pathogen Listeria monocytogenes. This peptide was
