Hormesis Effects of Nano- and Micro-sized Copper Oxide.

The concerns about the possible risk of manufactured nanoparticles (NPs) have been raised recently. Nano- and micro-sized copper oxide (CO and CONP) are widely used in many industries. In this regard, in-vitro studies have demonstrated that CONP is a toxic compound in different cell lines. Despite their unique properties, NPs possess unexpected toxicity profiling relative to the bulk materials. This study was designed to examine and compare the toxic effects of CO and CONPs in-vivo and in isolated rat mitochondria. Male Wistar albino rats received 50 to 1000 mg/kg CO or CONP by gavage and seve

Ionizing Radiation and Translation Control: A Link to Radiation Hormesis?

Protein synthesis, or mRNA translation, is one of the most energy-consuming functions in cells. Translation of mRNA into proteins is thus highly regulated by and integrated with upstream and downstream signaling pathways, dependent on various transacting proteins and cis-acting elements within the substrate mRNAs. Under conditions of stress, such as exposure to ionizing radiation, regulatory mechanisms reprogram protein synthesis to translate mRNAs encoding proteins that ensure proper cellular responses. Interestingly, beneficial responses to low-dose radiation exposure, known as radiation hor

Mitochondrial stress extends lifespan in C. elegans through neuronal hormesis.

Progressive neuronal deterioration accompanied by sensory functions decline is typically observed during aging. On the other hand, structural or functional alterations of specific sensory neurons extend lifespan in the nematode Caenorhabditis elegans. Hormesis is a phenomenon by which the body benefits from moderate stress of various kinds which at high doses are harmful. Several studies indicate that different stressors can hormetically extend lifespan in C. elegans and suggest that hormetic effects could be exploited as a strategy to slow down aging and the development of age-associated (neu

Hormesis, cell death, and regenerative medicine for neurode-generative diseases.

Although the adult human brain has a small number of neural stem cells, they are insufficient to repair the damaged brain to achieve significant functional recovery for neurodegenerative diseases and stroke. Stem cell therapy, by either enhancing endogenous neurogenesis, or transplanting stem cells, has been regarded as a promising solution. However, the harsh environment of the diseased brain posts a severe threat to the survival and correct differentiation of those new stem cells. Hormesis (or preconditioning, stress adaptation) is an adaptation mechanism by which cells or organisms are pote

Mechanisms of hormesis through mild heat stress on human cells.

In a series of experimental studies, it was shown that repetitive mild heat stress has antiaging hormetic effects on growth and various other cellular and biochemical characteristics of human skin fibroblasts undergoing aging in vitro. We have reported the hormetic effects of repeated challenge at the levels of maintenance of stress protein profile; reduction in the accumulation of oxidatively and glycoxidatively damaged proteins; stimulation of the proteasomal activities for the degradation of abnormal proteins; improved cellular resistance to ethanol, hydrogen peroxide, and ultraviolet-B ray

Myocardial Redox Hormesis Protects the Heart of Female Mice in Sepsis.

Mice challenged with lipopolysaccharide develop cardiomyopathy in a sex and redox-dependent fashion. Here we extended these studies to the cecal ligation and puncture (CLP) model. We compared male and female FVB mice (wild type, WT) and transgenic littermates overexpressing myocardial catalase (CAT). CLP induced 100% mortality within 4 days, with similar mortality rates in male and female WT and CAT mice. 24 h after CLP, isolated (Langendorff) perfused hearts showed depressed contractility in WT male mice, but not in male CAT or female WT and CAT mice. In WT male mice, CLP induced a depression

Health effects of intermittent fasting: hormesis or harm? A systematic review.

BACKGROUND: Intermittent fasting, alternate-day fasting, and other forms of periodic caloric desistance are gaining popularity in the lay press and among animal research scientists. Whether clinical evidence exists for or is strong enough to support the use of such dietary regimens as health interventions is unclear. OBJECTIVE: This review sought to identify rigorous, clinically relevant research studies that provide high-quality evidence that therapeutic fasting regimens are clinically beneficial to humans. DESIGN: A systematic review of the published literature through January 2015 was per

Hormesis and vitagenes in aging and longevity: mitochondrial control and hormonal regulation.

Abstract Average life span has increased because of medical and environmental factors, but maximal life span remains unchanged. Understanding the mechanisms of aging will help to reduce age-related morbidity and facilitate healthy aging. Unlike female menopause, which is accompanied by an abrupt and permanent cessation of ovarian function (both folliculogenesis and estradiol production), male aging does not result in either cessation of testosterone production or infertility. Although the circulating serum testosterone concentration does decline with aging, in most men this decrease is small,

Rethinking Bioactivity Screening Through the Lens of Hormesis and Functional Windows.

The interaction between a compound and a biological system changes across the concentration gradient: it is selective and target-mediated at low concentrations and non-specific and stress-dominated at high concentrations. However, bioactivity screening favours concentrations high enough to produce clear, statistically robust effects, which preferentially captures the stress-dominated regime and overlooks the range in which regulated, adaptive responses occur. This review examines how sparse concentration spacing, monotonic analytical models and effect-size-driven selection interact to bias dis

The crucible of resilience: hormesis as the unifying principle of evolution, genetics, and epigenetics.

The traditional linear no-threshold model of toxicity and stress fails to capture the complex, adaptive nature of biological systems. Hormesis-a biphasic dose-response phenomenon where low-dose stress induces beneficial, adaptive responses while high doses cause harm-offers a more comprehensive paradigm. This article asserts that hormesis is a major, unifying explanatory principle of life that efficiently integrates evolutionary theory, genetics, and epigenetics. From an evolutionary perspective, hormesis is an energy-efficient strategy of adaptive homeostasis, permitting organisms to build bi

Integrating Hormesis and Xenohormesis in Plants Through Fitness and Response Variables.

Hormesis is an adaptive response empirically characterized by a biphasic dose-response pattern, in which low or moderate levels of stress induce stimulatory responses whereas higher levels result in inhibitory or toxic effects. Although the biphasic dose-response pattern provides an empirical basis for identifying hormesis, it does not necessarily establish the adaptive significance of the observed response. Individual physiological, biochemical, or molecular variables may exhibit transient or ontogenetically dependent responses; therefore, fitness-related outcomes may provide an integrative c

Sex hormonal regulation and hormesis in aging and longevity: role of vitagenes.

Aging process is accompanied by hormonal changes characterized by an imbalance between catabolic hormones, such as cortisol and thyroid hormones which remain stable and hormones with anabolic effects (testosterone, insulin like growth factor-1 (IGF-1) and dehydroepiandrosterone sulphate (DHEAS), that decrease with age. Deficiencies in multiple anabolic hormones have been shown to predict health status and longevity in older persons.Unlike female menopause, which is accompanied by an abrupt and permanent cessation of ovarian function (both folliculogenesis and estradiol production), male aging

Hormesis associated with a low dose of methylmercury injected into mallard eggs.

We injected mallard (Anas platyrhynchos) eggs with methylmercury chloride at doses of 0, 0.05, 0.1, 0.2, 0.4, 0.8, 1.6, 3.2, and 6.4 μg mercury/g egg contents on a wet-weight basis. A case of hormesis seemed to occur because hatching success of eggs injected with 0.05 μg/g mercury (the lowest dose) was significantly greater (93.3%) than that of controls (72.6%), whereas hatching success decreased at progressively greater doses of mercury. Our finding of hormesis when a low dose of methylmercury was injected into eggs agrees with a similar observation in a study in which a group of female malla

Radiation hormesis and reactive oxygen species-mediated stress priming in plants.

Radiation hormesis refers to the beneficial biological effects of low-dose ionizing radiation (LDIR) that do not conform to linear-no-threshold or threshold models. Ionizing radiation has been extensively used in the bioindustry for mutation induction, food sterilization, and structural modification of natural compounds. Despite its industrial potential, the application of LDIR-induced hormetic effects remains limited owing to insufficient scientific evidence, reproducibility concerns, and policy considerations related to radiation safety management. This review examines representative cases o

The current insights of mitochondrial hormesis in the occurrence and treatment of bone and cartilage degeneration.

It is widely acknowledged that aging, mitochondrial dysfunction, and cellular phenotypic abnormalities are intricately associated with the degeneration of bone and cartilage. Consequently, gaining a comprehensive understanding of the regulatory patterns governing mitochondrial function and its underlying mechanisms holds promise for mitigating the progression of osteoarthritis, intervertebral disc degeneration, and osteoporosis. Mitochondrial hormesis, referred to as mitohormesis, represents a cellular adaptive stress response mechanism wherein mitochondria restore homeostasis and augment resi

Hormesis enables cells to handle accumulating toxic metabolites during increased energy flux.

Energy production is inevitably linked to the generation of toxic metabolites, such as reactive oxygen and carbonyl species, known as major contributors to ageing and degenerative diseases. It remains unclear how cells can adapt to elevated energy flux accompanied by accumulating harmful by-products without taking any damage. Therefore, effects of a sudden rise in glucose concentrations were studied in yeast cells. This revealed a feedback mechanism initiated by the reactive dicarbonyl methylglyoxal, which is formed non-enzymatically during glycolysis. Low levels of methylglyoxal activate a mu

Nitroxide Hormesis in Yeast: 4-Hydroxy-TEMPO Modulates Aging, and Cell Cycle.

4-hydroxy-TEMPO is a water-soluble nitroxide radical with potent antioxidant and redox-modulating properties. Its small molecular weight and membrane permeability enable it to act as a superoxide dismutase mimetic, efficiently scavenging reactive oxygen species and mitigating oxidative damage. In this study, we investigated the physiological and transcriptomic effects of 4-hydroxy-TEMPO in Saccharomyces cerevisiae, using wild-type and mutant strains deficient in key redox and DNA repair pathways (sod1Δ, sod2Δ, yap1Δ, rad52Δ). RNA-Seq analysis revealed widespread transcriptional reprogramming.

Ethanol Hormesis in Honeybees (Apis mellifera L.) Infected with Vairimorpha (Nosema) spp.

This study investigates the phenomenon of ethanol hormesis in honeybees (Apis mellifera) infected with Vairimorpha (Nosema) spp., a widespread parasite that significantly impacts bee health and colony survival. Hormesis refers to a biphasic response where low doses of potentially harmful substances may elicit beneficial effects, contrasting with the detrimental impacts observed at higher concentrations. We hypothesized that low ethanol concentrations could reduce Vairimorpha spp. infection severity and improve bee lifespan. In a controlled experiment, foraging bees were divided into groups of

Nutrient removal by Chlorella vulgaris F1068 under cetyltrimethyl ammonium bromide induced hormesis.

Toxicants are generally harmful to biotechnology in wastewater treatment. However, trace toxicant can induce microbial hormesis, but to date, it is still unknown how this phenomenon affects nutrient removal during municipal wastewater treatment process. Therefore, this study focused on the effects of hormesis induced by cetyltrimethyl ammonium bromide (CTAB), a representative quaternary ammonium cationic surfactant, on nutrient removal by Chlorella vulgaris F1068. Results showed that when the concentration of CTAB was less than 10 ng/L, the cellular components chlorophyll a, proteins, polysacc

Inverse hormesis of cancer growth mediated by narrow ranges of tumor-directed antibodies.

Compelling evidence for naturally occurring immunosurveillance against malignancies informs and justifies some current approaches toward cancer immunotherapy. However, some types of immune reactions have also been shown to facilitate tumor progression. For example, our previous studies showed that although experimental tumor growth is enhanced by low levels of circulating antibodies directed against the nonhuman sialic acid N-glycolyl-neuraminic acid (Neu5Gc), which accumulates in human tumors, growth could be inhibited by anti-Neu5Gc antibodies from a different source, in a different model.
