Conjugates of horseradish peroxidase (HRP) and sheep anti-human immunoglobulin (anti-Ig) were prepared by means of a heterobifunctional reagent, N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP). The molar ratios of the conjugates could be changed by varying the degree of chemical substitution of anti-Ig and HRP. Gel filtration and affinity chromatography were used to separate conjugated anti-Ig from free HRP and unconjugated anti-Ig. The distribution of complex sizes and the presence of free HRP and anti-Ig in the final products were monitored by sodium dodecyl sulphate (SDS) polyacrylamid

The present study utilizes 1H NMR spectroscopy to characterize the binding of substrate to heme active site of three different peroxidases, horseradish peroxidase, lignin peroxidase, and manganese peroxidase. Information has been obtained on the site of p-cresol binding to the active-site cavity of the cyanide derivative of horseradish peroxidase. This information was obtained by relaxation enhancements of the substrate protons and connectivities between the latter and heme 8-CH3 and a Phe residue. Manganese(II) is shown to bind to ferri-manganese peroxidase and its cyanide derivative in a spe

Detection of the immune response to external antigen of the palatine tonsil of the rabbit is an interesting problem. Employing horseradish peroxidase (HRP) as an external antigen, the reactions of a perfusion sensitized group and those of a group sensitized by intravenous injection were examined. The results were as follows: 1. Hypertrophy of the palatine tonsil was recognized only in the perfusion sensitized group. 2. The serum titer level of the antibody to HRP was recognized to be high in the intravenous injection sensitized group, which was confirmed by the production of antibody in the sp

A kinetic study has been carried out over the pH range of 2.63-9.37 for the reaction of horseradish peroxidase with hydrogen peroxide to form compound I of th;e enzyme. Analysis of the results, indicates that there are two kinetic influencing, ionizable groups on the enzyme with pKa values of 3.2 and 3.9. Protonation of these groups results in a decrease in the rate of reaction of the enzyme with H2O2. A previous study of the kinetics of cyanide binding to horseradish peroxidase (Ellis, W.D. & Dunford, H.B.: Biochemistry 7, 2054-2062 (1968)) has been extended to down to pH 2.55, and analysis o

Chitosan beads were prepared with phytic acid to develop a biocompatible matrix for enzyme immobilization. Horseradish peroxidase (HRP) was immobilized on phytic acid-modified chitosan beads, and its catalytic activity was measured using 2-methoxyphenol. The concentration of phytic acid used was varied, with optimal enzyme activity at a concentration of 25 mM phytic acid. The phytic acid/chitosan beads were characterized using FTIR, SEM, and their swelling behavior was investigated at room temperature. The beads exhibited slightly higher levels of water absorption, 20 ± 2 of mass for phytic ac

Protein uptake and transport within the brain stem vasculature of mechanically brain injured cats was studied by means of both light and electron microscopy utilizing intravenously injected horseradish peroxidase as the protein tracer. In animals sustaining low grade head injuries not of sufficient intensity to elicit either microscopic, intraparenchymal hemorrhages or subtle, neuropathological responses, peroxidase extravasation was noted both in the vascular walls and in the surrounding parenchyma of the ventromedial aspect of the brain stem. At the ultrastructural level as early as 3 min af

Enzyme-mediator systems generate radical intermediates that abstract hydrogen atoms under mild conditions. These systems have been employed extensively for alcohol oxidation, primarily in biomass degradation, but they are underexplored for direct activation of C(sp3)-H bonds in alkyl groups. Here, we combine horseradish peroxidase (HRP), H2O2, and redox mediator N-hydroxyphthalimide (NHPI) for C(sp3)-H functionalization of alkylbenzene-type substrates. The HRP-NHPI system is >10-fold more active than existing enzyme-mediator systems in converting alkylbenzenes to ketones and aldehydes under ai

Horseradish peroxidase (HRP) is widely used to visualize blood-brain barrier (BBB) breakdown. In addition to its visual properties, HRP activity can be measured and can, therefore, be used to quantitate barrier leakage rapidly and simply. In addition, the same tissue can be used for electron microscopic studies in which the structural basis for the leakage can be resolved. Here we describe two methods for quantitation of barrier damage using HRP: one in small, multifocal barrier lesions caused by ethanol combined with barbiturates, and a second in large, single-focus, necrotic lesions. We dete

Atomic force microscopy (AFM)-based fishing is a promising method for the detection of low-abundant proteins. This method is based on the capturing of the target proteins from the analyzed solution onto a solid substrate, with subsequent counting of the captured protein molecules on the substrate surface by AFM. Protein adsorption onto the substrate surface represents one of the key factors determining the capturing efficiency. Accordingly, studying the factors influencing the protein adsorbability onto the substrate surface represents an actual direction in biomedical research. Herein, the in

The photodissociation of cyanide from ferric myoglobin (MbCN) and horseradish peroxidase (HRPCN) has definitively been observed. This has implications for the interpretation of ultrafast IR (Helbing et al. Biophys. J. 2004, 87, 1881-1891) and optical (Gruia et al. Biophys. J. 2008, 94, 2252-2268) studies that had previously suggested the Fe-CN bond was photostable in MbCN. The photolysis of ferric MbCN takes place with a quantum yield of ~75%, and the resonance Raman spectrum of the photoproduct observed in steady-state experiments as a function of laser power and sample spinning rate is ident

There has been experimental evidence of the tethered cord syndrome being a gray matter disease resulting from excessive tensile force in the lumbosacral cord. Clinical findings also support this concept. This study was designed to elucidate the possibility of the involvement of the long tract in tethered cord. Long tract continuity was evaluated utilizing the retrograde axonal transport of horseradish peroxidase in fasciculus gracilis. There were no significant differences between the normal and experimental tethered cord in the number of labeled dorsal root ganglion cells of the lumbosacral c

We describe here the morphology of the inferior olive and the localization of labeled cells after HRP injections into various lobules of vermis and hemisphere of the cerebellum of the sheep. The medial part of the caudal half of the medial accessory olive projects to a medial zone in the anterior lobe, the simple lobule, and the lobules VII and VIII. The lateral part of the medial accessory olive projects to more lateral parts of these lobules with the exception of lobule VII. The group beta projects in a differential manner to the lateral parts of the lobules VII and VIII and the medial parts

Previous studies of the reduction of metmyoglobin and adducts by dithionite have been extended to horseradish peroxidase and its complexes. In addition, the reduction of metmyoglobin, horseradish peroxidase and adducts by a much bulkier reactant, cobalt(II) sepulchrate has been studied. Similar patterns of kinetic behavior were observed, namely, direct reduction of cyanide and imidazole adducts of the iron(III) proteins and indirect (via dissociation) reduction of the fluoride adduct. In the reduction of horseradish ferriperoxidase by cobalt(II) sepulchrate, three steps are observed and the sp

Several N-N-and N-O-containing compounds were analysed for their ability to act as substrates for horseradish peroxidase and peroxidases in Mycobacterium tuberculosis extracts. Aminoguanidine, diaminoguanidine, isoniazid, hydroxylamine and hydrazine were found to be weak substrates for horseradish peroxidase in reaction I and to inhibit the reaction of horseradish peroxidase with hydrogen peroxide. The same compounds inhibited the reaction of Mycobacterium tuberculosis peroxidase-catalase with hydrogen peroxide, and hydroxylamine was found to be a weak substrate for this enzyme. In growth inhi

Fighting cancer still relies on chemo- and radiation therapy, which is a trade-off between effective clearance of malignant cells and severe side effects on healthy tissue. Targeted cancer treatment on the other hand is a promising and refined strategy with less systemic interference. The enzyme horseradish peroxidase (HRP) exhibits cytotoxic effects on cancer cells in combination with indole-3-acetic acid (IAA). However, the plant-derived enzyme is out of bounds for medical purposes due to its foreign glycosylation pattern and resulting rapid clearance and immunogenicity. In this study, we ge

A horseradish peroxidase (HRP)-based electrochemical biosensor was developed for melatonin (MLT) detection using a graphene-modified screen-printed electrode (G/SPE). HRP was immobilized at different temperatures (4 °C and 20 °C) and enzyme amounts (10 and 20 µL of 5 mg/mL HRP solution) to optimize the performance of the biosensors. FTIR analysis confirmed successful enzyme immobilization, while differential pulse voltammetry (DPV) showed a clear oxidation peak of MLT at 1.1 V with enhanced current responses for HRP-modified electrodes. The optimal biosensor, prepared at 4 °C with 10 µL enzyme

Horseradish peroxidase (HRP) is a heme-containing oxidoreductase with extensive applications in biotechnology, medical diagnostics, and environmental protection. In this study, Pichia pastoris was utilized to produce HRP. Successfully, expression strains with 1-5 copies of HRP-C were constructed, and the strain with the highest expression level and activity of HRP-C was obtained. Different molecular chaperones (PDI1, HAC1, BIP1) were selected, and co-expression was carried out through co-induction and separate induction methods. The results showed that the yield of HRP increased approximately
