Nutrients are essential to the human host and to its invading pathogens. The purpose of this International Nutrition Council symposium on Micronutrient Regulation of Host-Pathogen Interactions held at Experimental Biology 2006 was to examine new knowledge about the mechanisms by which certain limiting micronutrients can mediate the balance of power between the human host and its numerous potential pathogens. In this introductory article, we briefly review how competition for nutrients is critical to the survival of both host and pathogen and describe some of the evolved mechanisms by which eac

As a leading cause of healthcare-associated infections, catheter-associated urinary tract infections (CAUTIs) present a unique challenge due to the high prevalence of multidrug-resistant and polymicrobial pathogens. Distinct from uncomplicated UTIs, the insertion of a urinary catheter provokes tissue damage that triggers inflammation, plasma extravasation, and the deposition of host fibrinogen. This inflammatory response drives the robust recruitment of immune cells and plasma extravasation, leading to the accumulation of fibrinogen─a key coagulation protein─on the device and bladder epitheliu

Periodontitis is an infectious, inflammatory, non-communicable disease characterized by tissue destruction driven by host responses to dysbiotic shifts in oral microbial communities. The subgingival microbiome constitutes a complex ecosystem in which bacteria, fungi, viruses, and archaea interact via interkingdom communication to modulate the inflammatory response through molecular mechanisms that remain largely unknown. This narrative review aims to understand how functional imbalances within the microbiome alter the microenvironment and promote uncontrolled inflammation responsible for perio

BACKGROUND: A strategic way to gain a deeper understanding of how infection occurs is to characterize the mechanisms of pathogen binding and uptake. To accomplish this, we have focused our efforts on developing approaches that enable real-time monitoring of the initial stages of pathogen binding and the dynamics of the corresponding cellular response, including how membrane receptors are recruited to the binding sites. Past efforts have relied largely on widefield fluorescence imaging to track receptor recruitment and pathogen uptake.
METHODS: In this work, we have applied holotomography (HT)

Understanding the virulence mechanisms of pathogenic bacteria is crucial for elucidating their ability to cause disease. Among these, type secretion systems (TSS) are important for mediating host-pathogen interactions. This review focuses on the TSS in Burkholderia species, emphasizing their molecular structures, mechanisms, and their role in virulence. Burkholderia spp. employ a variety of TSS, each contributing distinctly to pathogenicity. Type 1 SS (T1SS) facilitate the export of toxins/enzymes to the extracellular space, promoting host-tissue damage and immune modulation. T2SS secretes hyd

Listeria monocytogenes is primarily a saprophytic bacterium, but it can transition into a pathogenic form when ingested by humans, vertebrates, or invertebrates, thereby initiating infection and proliferating within the host. This bacterium serves as a model for studying the molecular and cellular mechanisms by which environmental microorganisms adapt to mammalian and other host systems, thereby promoting the successful colonization and survival of these microorganisms. Environmental contamination is a significant factor in the transmission of listeriosis and other foodborne diseases. This rev

Cryptosporidium, a protozoan parasite affecting the gastrointestinal system, is primarily known for causing diarrhea, especially in those with weakened immune systems. However, there is increasingly persuasive evidence that it may be directly involved in tumorigenesis. This review examines some of the potential mechanisms through which Cryptosporidium infections can induce cancer, specifically chronic inflammation, manipulation of the immune system, and alteration of cell signaling pathways. Persistent inflammation with immune system changes due to chronic infection, particularly among immunoc

In recent years, substantial progress has been made in the understanding of the intracellular lifestyle of Chlamydia trachomatis and how the bacteria establish themselves in the human host. As an obligate intracellular pathogenic bacterium with a strongly reduced coding capacity, C. trachomatis depends on the provision of nutrients from the host cell. In this Review, we summarize the current understanding of how C. trachomatis establishes its intracellular replication niche, how its metabolism functions in the host cell, how it can defend itself against the cell autonomous and innate immune re

Shigella spp. are Gram-negative enteric pathogens and the leading cause of bacterial dysentery worldwide. Since the discovery more than three decades ago that the large virulence plasmid of Shigella is essential for pathogenesis, our understanding of how the bacterium orchestrates inflammation and tissue destruction at the mucosal surface has been informed by studies employing the rabbit ileal loop model. Here, we outline how Phillippe Sansonetti, together with his co-workers and collaborators, exploited this model to provide a holistic view of how Shigella survives in the intestinal tract, tr

Both tick-borne encephalitis (TBE) virus and Borrelia burgdorferi depend for their survival on a tick-borne transmission cycle involving diverse vertebrate hosts and ixodid (primarily Ixodes) tick species. Increasing evidence strongly suggests that a key factor in successful transmission is the ability of these pathogens to exploit the pharmacological properties of their vector tick's saliva. Proteins and other chemicals, secreted in tick saliva, control the vertebrate hosts' haemostatic, inflammatory and immune responses in order to facilitate blood-feeding. Such bioactive saliva molecules in

Malaria remains a major global health burden despite decades of effort toward its prevention and treatment. Much of this difficulty stems from the complexity of the Plasmodium parasite's life cycle and its interactions with the human host. Here, we review receptor-ligand interactions between the Plasmodium parasite and human host, with a focus on clinically relevant antigens. The parasite uses a variety of proteins at each stage of its life cycle to achieve requisite functions such as motility, cell traversal, and invasion. In turn, the human immune response targets these key parasite antigens

Recent advances in single-cell technologies have revealed the dynamic and heterogeneous nature of host-pathogen interactions at the single-cell level. This review explores how cellular variability-both within clonal bacterial populations and among genetically identical host cells-gives rise to distinct infection outcomes, from pathogen clearance to persistence across multiple biological scales, from single cells to tissues and the whole organism. We highlight the conceptual and technological progress that has enabled the dissection of these interactions at single-cell resolution, including mic

The toxin-producing bacterium Bacillus cereus is an important and neglected human pathogen and a common cause of food poisoning. Several toxins have been implicated in disease, including the pore-forming toxins hemolysin BL (HBL) and nonhemolytic enterotoxin (NHE). Recent work revealed that HBL binds to the mammalian surface receptors LITAF and CDIP1 and that both HBL and NHE induce potassium efflux and activate the NLRP3 inflammasome, leading to pyroptosis. These mammalian receptors, in part, contribute to inflammation and pathology. Other putative virulence factors of B. cereus include cytot

The bacterial genus Salmonella includes a large group of food-borne pathogens that cause a variety of gastrointestinal or systemic diseases in hosts. Salmonella use several secretion devices to inject various effectors targeting eukaryotic hosts, or bacteria. In the past few years, considerable progress has been made towards understanding the structural features and molecular mechanisms of the secretion systems of Salmonella, particularly regarding their roles in host-pathogen interactions. In this review, we summarize the current advances about the main characteristics of the Salmonella secre

Helicobacter pylori (H. pylori), discovered in 1982, is a microaerophilic, spiral-shaped gram-negative bacterium that is able to colonize the human stomach. Nearly half of the world's population is infected by this pathogen. Its ability to induce gastritis, peptic ulcers, gastric cancer and mucosa-associated lymphoid tissue lymphoma has been confirmed. The susceptibility of an individual to these clinical outcomes is multifactorial and depends on H. pylori virulence, environmental factors, the genetic susceptibility of the host and the reactivity of the host immune system. Despite the host imm

Necrotizing fasciitis, a prototypical form of necrotizing soft-tissue infection, is a rapidly progressive and life-threatening soft-tissue infection characterized by extensive fascial necrosis, systemic toxicity, tissue hypoperfusion, septic shock, and multiorgan dysfunction. Although early recognition, urgent surgical source control, appropriate antimicrobial therapy, toxin suppression when indicated, and organ support remain the foundations of management, the dynamic host-response states that link microbial virulence, local tissue injury, impaired pathogen clearance, perfusion failure, and s

The eukaryotic protein CD98hc (also known as 4F2, FRP-1, or SLC3A2) is a membrane glycoprotein and one of the heavy chains of the family of heterodimeric amino acids transporters. It can associate with any of 6 different light chains to form distinct amino acid transporters. CD98hc is also involved in mediation of intracellular integrin signaling. Besides its physiological roles in the development of the placenta and the immune system, CD98hc is important during pathological processes such as tumorigenesis and host-pathogen interaction. Since its first identification as Fusion Regulatory Prote
