The comparison of melt technologies based on mesoporous carriers for improved carvedilol dissolution.

High-shear (HS) melt granulation and hot melt extrusion (HME) were compared as perspective melt-based technologies for preparation of amorphous solid dispersions (ASDs). ASDs were prepared using mesoporous carriers (SyloidⓇ 244FP or NeusilinⓇ US2), which were loaded with carvedilol dispersed in polymeric matrix (polyethylene glycol 6000 or SoluplusⓇ). Formulations with high carvedilol content were obtained either by HME (11 extrudates with polymer:carrier ratio 1:1) or HS granulation (6 granulates with polymer:carrier ratio 3:1). DSC and XRD analysis confirmed the absence of crystalline carved

High-throughput eutectic screening and hot-melt extrusion enable rapid development of nifedipine amorphous solid dispersions.

The development of eutectic-based amorphous solid dispersions (ASDs) is often constrained by the time and material required to identify suitable drug-coformer combinations. Here, we present a miniaturised high-throughput workflow that integrates picolitre-volume 2D inkjet printing, predictive thermodynamic modelling, and scalable processing to accelerate eutectic formulation design for the model active pharmaceutical ingredient (API) nifedipine. Binary systems were printed as microarrays and screened microscopically, enabling rapid identification of promising eutectic pairs using minimal mater

Process optimization in pharmaceutical hot-melt extrusion: real-time volatile detection via SIFT-MS combined with multivariate analysis.

Establishing robust processing windows for pharmaceutical polymers during hot-melt extrusion (HME) remains challenging, as conventional thermal analyses reveal little about early chemical change. Here, selected-ion-flow-tube-mass-spectrometry (SIFT-MS) combined with principal component analysis (PCA) was used to characterise real-time volatile evolution under both thermogravimetric (TGA) and extrusion conditions. Centroid-distance mapping and PCA loadings revealed distinct transitions, providing a data-driven means of defining the onset of significant chemical change. Across four representativ

A solid-state-dependent correlation between drug-polymer hot melt extrudate mechanical behavior and milled extrudate tablet tensile strength.

The mechanical behavior of drug-polymer hot melt extrudates, whether amorphous or partially crystalline, was found to predict the tensile strength of tablets directly compressed from the milled extrudates, with the most predictive test depending on the extrudate solid state. Griseofulvin (GF), a poorly water-soluble BCS class II drug prone to crystallization, was selected as the model drug. Two polymers were compared, Hydroxypropyl Cellulose SL (HPC) and Kollidon VA 64 (PVPVA), to uncover the influence of drug-polymer interactions and identify (anti)plasticization phenomena. Extrudates were pr

Hot Melt Extruded High-Dose Amorphous Solid Dispersions Containing Lumefantrine and Soluplus.

Over the last 15 years, a small number of paediatric artemisinin-based combination therapy products have been marketed. These included Riamet® and Coartem® dispersible tablets, a combination of artemether and lumefantrine, co-developed by the Medicines for Malaria Venture and Novartis. Disappointingly, patient compliance, requirement for high-fat meal, and sporadic drug dissolution behaviours following administration still result in considerable challenges for these products. The first and foremost barrier that needs addressed for successful delivery of the artemether/lumefantrine combination

Development of amorphous solid dispersion of delamanid, an extremely water-insoluble drug, by hot melt extrusion applying acid-base supersolubilization (ABS) principle.

Delamanid, an antituberculosis drug used for multidrug-resistant tuberculosis, has extremely poor water solubility and a high melting point of ∼195 °C. The current commercial product, Deltyba®, shows incomplete dissolution and food-dependent bioavailability. This study is the first to report the preparation of amorphous solid dispersions (ASD) of delamanid by hot-melt extrusion (HME), made possible by applying the acid-base supersolubilization (ABS) principle using malic acid. Malic acid increased the aqueous solubility of delamanid to 46 mg/mL at pH 1.8, approximately 660 times higher than it

System-agnostic prediction of pharmaceutical excipient miscibility via computing-as-a-service and experimental validation.

We applied computing-as-a-service to the unattended system-agnostic miscibility prediction of the pharmaceutical surfactants, Vitamin E TPGS and Tween 80, with Copovidone VA64 polymer at temperature relevant for the pharmaceutical hot melt extrusion process. The computations were performed in lieu of running exhaustive hot melt extrusion experiments to identify surfactant-polymer miscibility limits. The computing scheme involved a massively parallelized architecture for molecular dynamics and free energy perturbation from which binodal, spinodal, and mechanical mixture critical points were det

Enhancing drug solubilization using a surface-modified edible biopolymer through hot melt extrusion: A design space methodology.

This study investigates the use of an octenylsuccinate-modified dendrimer-like biopolymer (OS-DLB) as a carrier matrix in the formulation of biodendrimeric solid dispersions (BDSDs) using hot melt extrusion (HME). Ibuprofen (IBU) and griseofulvin (GSF) were selected as model compounds due to their poor aqueous solubility - one limited by its hydrophobicity and the other by its strong crystal lattice, respectively. This study demonstrates that the BDSD formulation can significantly enhance the dissolution rates of the model compounds through a parallel liquid phase equilibrium, while retaining

Hot melt extrusion-driven cocrystallization of aceclofenac with nicotinamide: A strategy to enhance solubility, bioavailability, and therapeutic effect.

Aceclofenac (ACF) is a commonly used non-steroidal anti-inflammatory drugs (NSAID) for numerous inflammatory conditions; however, its therapeutic potential is limited due to suboptimal solubility. Pharmaceutical cocrystals have demonstrated favorable and sustainable results in improving the physicochemical and biopharmaceutical characteristics of poorly soluble drugs. To enhance the solubility, dissolution rate, and bioavailability of ACF, we aimed to prepare cocrystals of this biopharmaceutics classification system (BCS) class II drug with a coformer including nicotinamide (NCT), using hot me

Leveraging solid solubility and miscibility of etoricoxib in Soluplus® towards manufacturing of 3D printed etoricoxib tablets by additive manufacturing.

This research focuses on exploring the solid solubility and miscibility of Etoricoxib, a poorly water-soluble anti-inflammatory drug, within Soluplus®, a polymer used as a matrix for 3D-printed tablets. By utilizing hot-melt extrusion (HME), the drug was dispersed within Soluplus® to enhance its solubility. The extrudates were then employed in 3D printing to create customized solid oral dosage form. This study's novelty lies in combining HME and 3D printing, aiming to improve drug incorporation, stability, and effectiveness in the final formulation. Comprehensive characterization techniques, i

Multi-dose oral abuse deterrent formulation of loperamide using hot melt extrusion.

Loperamide, an over the counter anti-diarrheal drug, also infamously referred to as "poor man's methadone". Due to the ease of availability and low price, people/patients abuse it by consuming more than 30 tablets to achieve euphoric effect and to combat opioid withdrawal. But supratherapeutic doses of loperamide result in severe respiratory depression, cardiac dysrhythmia and mortality. To address this issue, we developed a unique and innovative technology to deter multi-dose oral abuse. The concept is to design a tablet which can immediate release loperamide in diarrheic patients (single tab

Advanced Pharmaceutical Applications of Hot-Melt Extrusion Coupled with Fused Deposition Modelling (FDM) 3D Printing for Personalised Drug Delivery.

Three-dimensional printing, also known as additive manufacturing, is a fabrication process whereby a 3D object is created layer-by-layer by depositing a feedstock material such as thermoplastic polymer. The 3D printing technology has been widely used for rapid prototyping and its interest as a fabrication method has grown significantly across many disciplines. The most common 3D printing technology is called the Fused Deposition Modelling (FDM) which utilises thermoplastic filaments as a starting material, then extrudes the material in sequential layers above its melting temperature to create

Preparation of self-dispersible eutectic mixtures for poorly soluble drugs: Hot-melt extrusion vs. melt casting.

Solid eutectic mixtures of self-dispersible polyethylene glycol-40-stearate with fenofibrate or ibuprofen - prepared by casting or by twin-screw extrusion - were compared in terms of processability and dissolution rate at, below, and above the eutectic concentration. Furthermore, the effects of extruding with two distinct screw configurations - a shear screw and a conveyor screw - were investigated. The eutectic mixtures containing ibuprofen had a poor recrystallization tendency and therefore did not yield the desired solid state in the casting process. In contrast, they could be successfully

Low-Temperature Hot Melt Extrusion: Solid-State Organization and Mechanical Performance of Lipid Filaments.

The bioavailability of highly lipophilic, poorly soluble drugs can be improved effectively, when formulated in lipid-based systems. Solid lipids generally offer greater physical robustness than liquid lipids but slower release. Hot-melt extrusion (HME) and additive manufacturing (AM) enable personalized release by tailoring composition and geometry. However, the limited availability of suitable solid lipid-based materials, particularly for processing thermolabile and lipophilic drugs, restricts their application. Polyglycerol esters of fatty acids (PGFAs) represent a promising class of lipids

Development of indomethacin amorphous solid dispersion by applying acid-base supersolubilization (ABS) principle to enhance solubility and enable low-temperature hot melt extrusion.

This study explores a novel acid-base supersolubilization (ABS) principle for enhancing the solubility and dissolution rate of indomethacin, a poorly water-soluble and weakly acidic drug, by interaction with the weak base tromethamine. The reported aqueous solubilities of indomethacin are 1.5 and 105.2 µg/mL at pH 1.2 and 7.4, respectively. However, in the present investigation, they were increased to 2.5 mg/mL at pH 7.3 and greater than 240 mg/mL at pH ∼ 8.0 by interaction with tromethamine. Such an acid-base interaction also occurred in the solid state, which reduced the melting temperature

Insight into manufacturing of bespoke combination drug products containing carvedilol and simvastatin by fused deposition modeling.

The goal of this study was to explore the fabrication of a combination drug product containing two poorly soluble active pharmaceutical ingredients (APIs), carvedilol (CAR) and simvastatin (SIM), in therapeutically relevant doses (25 mg of each API) with a distinct, easily distinguishable shape. Fused deposition modeling, combined with hot-melt extrusion (HME), was used to produce hollow heart-shaped dual-loaded tablets in which the two APIs were spatially separated with an intermediate API-free layer. Water-soluble hydroxypropyl methylcellulose of varying molecular weights was used as the pri
