Effects of hypergravity environment on the parathyroid gland of the isoproterenol-treated hamster.

The ultrastructure of the parathyroid glands of hamsters subjected to 5 g environment after an intraperitoneal injection of isoproterenol was studied. In the isoproterenol-treated hamsters exposed to hypergravity environment, the Golgi complexes, cisternae of the granular endoplasmic reticulum and lipid droplets were significantly increased and secretory granules were significantly decreased compared with those of the control group. In addition, many chief cells contained numerous prosecretory granules in the Golgi areas and several secretory granules were situated close to the plasma membrane

Hypergravity reduces F-actin accumulation in osteoclasts, with attenuated bone resorption.

Bone loss occurs in astronauts during prolonged spaceflight, thus indicating the sensitivity of skeletal homeostasis to altered gravitational environments. Previous studies have shown that microgravity affects osteoclast differentiation and bone resorption, which suggests that osteoclasts possess mechanisms to sense and respond to gravity-generated mechanical forces. For testing of the related mechanisms, hypergravity can be experimentally reproduced with use of a centrifuge. In the present study, osteoclasts derived from mouse bone marrow were subjected to hypergravity under three conditions:

Enhanced nasal drug delivery efficiency by increasing mechanical loading using hypergravity.

Nasal route drug administration for local and systemic delivery of many therapeutics has received attention because the nasal cavity is highly vascularized and provides a large surface area for drug absorption. However, nasal mucosa exhibits limited permeability to polar molecules. In this study, we developed a novel method for improving absorption efficiency of polar drugs by applying hypergravity. RPMI 2650 cells and primary human nasal epithelial cells were exposed three times to a 20 min hypergravitational condition (10 × g) with a 20 min rest period after each exposure. The applied hyperg

Morphometry of fish inner ear otoliths after development at 3g hypergravity.

Size and asymmetry (size difference between the left and right sides) of inner ear otoliths of larval cichlid fish were determined after a long-term stay in moderate hypergravity conditions (3g; centrifuge), in the course of which the animals completed their ontogenetic development from hatch to freely swimming. Neither the normal morphogenetic development nor the timely onset and gain of performance of swimming behaviour were impaired by the experimental conditions. However, both utricular and saccular otoliths (lapilli and sagittae, respectively) were significantly smaller after hyper-g expo

Modulation of vestibular function and receptor expression by experimental hypergravity in a rat model.

Motion sickness develops because of a mismatch or conflict between the information gathered from the visual input, somatosensory input, and the vestibular system. Altered gravity can cause motion sickness while traveling in space. In this study, we aimed to establish a rat model of hypergravity-stimulated motion sickness using hypergravity equipment and investigate changes in serotonin and histamine receptors. Overall, 21 rats were allocated to three experimental groups (4G-24 h, 4G-2 weeks, and 4G-4weeks) and one control group, with seven rats in each group. We performed western blotting and

Effects of hypergravity environment on the parathyroid gland of the propranolol-treated golden hamster.

The fine structure of the parathyroid glands of propranolol-treated hamsters subjected to 5 x gravity environment was studied. In the parathyroid glands of the propranolol-treated hamsters exposed to hypergravity environment, the volume density occupied by the Golgi complexes and cisternae of the granular endoplasmic reticulum was increased as compared to that of propranolol-treated hamsters and was decreased as compared to that of hamsters exposed to a hypergravity environment but was almost similar to that of control hamsters. In addition, many chief cells contained rich free ribosomes, abun

Effects of amygdala or hippocampus lesion on hypergravity-induced motion sickness in rats.

We examined the effects of amygdala lesion (AL) or hippocampal lesion (HL) on hypergravity-induced motion sickness in rats. Rats do not vomit, but the behavior known as pica, the eating of non-nutritive substances such as kaolin, can be used as an index of motion sickness. In the present study, hypergravity-induced kaolin intake and apomorphine-induced kaolin intake were measured before and after brain lesions. After AL, hypergravity-induced kaolin intake and the ratio of the hypergravity- to apomorphine-induced kaolin intakes were decreased. These results indicate that AL suppressed motion si

Repeated hypergravity training facilitates vestibular compensation associated with dorsal root ganglion remodeling.

Vestibular compensation relies on sensory reweighting, but whether repeated hypergravity training (HT) facilitates recovery after bilateral vestibular lesion (BVL) remains unclear. We investigated the effects of HT on motor function and dorsal root ganglion (DRG) transcriptomes in mice following BVL. Mice underwent sham surgery or BVL and were assigned to either HT (2 G, 60 min/day for 10 days) or no HT (NHT). Body weight, righting reflex, and rotarod performance were evaluated, and RNA sequencing of L5 DRGs was performed on day 7. HT did not improve body weight loss or vestibular-dependent ri

Suppression of essential oil biosynthesis in sweet basil cotyledons under hypergravity conditions.

The mechanism through which gravity influences the biosynthesis of essential oils in herbs is an important issue for plant and space biology. Sweet basil (Ocimum basilicum L.) seedlings were cultivated under centrifugal hypergravity conditions at 100 g in the light, and the growth of cotyledons, development of glandular hairs, and biosynthesis of essential oils were analyzed. The area and fresh weight of the cotyledons increased by similar amounts irrespective of the gravitational conditions. On the abaxial surface of the cotyledons, glandular hairs, where essential oils are synthesized and st

Medial gastrocnemius activation during graded axial hypergravity induced by short-arm human centrifugation.

BACKGROUND: Gravitational loading is a key determinant of antigravity muscle activation, yet neuromuscular responses to graded + Gz loading in humans remain incompletely characterized. Reduced mechanical loading, whether due to microgravity or physical inactivity, leads to musculoskeletal deconditioning across both spaceflight and clinical contexts. METHODS: Twenty-five adults with stable neuromuscular or neurodegenerative disorders and twenty-one age-matched healthy controls completed a short-arm human centrifugation protocol under a no-rotation baseline condition (ω = 0) and at 1.5, 1.7, an

Counteracting inactivity deconditioning with graded hypergravity loading: From clinical neurorehabilitation to space medicine.

BACKGROUND: Neuromuscular and neurodegenerative diseases such as Parkinson's disease result in progressive mobility restrictions, accelerating cardiovascular, muscular, and functional deconditioning. Such changes resemble the multisystem deconditioning that occurs during microgravity exposure, highlighting shared physiological consequences of reduced mechanical loading, even though the underlying mechanisms differ. OBJECTIVE: This study evaluated short-arm human centrifugation (SAHC) training as a whole-body rehabilitation intervention for patients with multiple sclerosis and neurodegenerativ

Hypergravity susceptibility of ventral root activity during fictive swimming in tadpoles (Xenopus laevis).

1. Fictive swimming is an experimental model to study early motor development. As vestibular activity also affects the development of spinal motor projections, the present study focused on the question whether in Xenopus laevis tadpoles, the rhythmic activity of spinal ventral roots (VR) during fictive swimming revealed age-dependent modifications after hypergravity exposure. In addition, developmental characteristics for various features of fictive swimming between stages 37/38 and 47 were determined. Parameters of interest were duration of fictive swimming episodes, burst duration, burst fre

The importance of gravity vector on adult mammalian organisms: Effects of hypergravity on mouse testis.

In the age of space exploration, the effect of hypergravity on human physiology is a relatively neglected topic. However, astronauts have several experiences of hypergravity during their missions. The main disturbance of altered gravity can be imputed to cell cytoskeleton alteration and physiologic homeostasis of the body. Testis has proved to be a particularly sensible organ, subject to environmental alteration and physiological disturbance. This makes testis an organ eligible for investigating the alteration following exposure to altered gravity. In our study, mice were exposed to hypergravi

Space cycle: a human-powered centrifuge that can be used for hypergravity resistance training.

INTRODUCTION: This study represents the first step toward testing the hypothesis that hypergravity can be used as a unique resistance training modality for maintaining the health and function of skeletal muscle in microgravity. The primary objectives of this study were to use a human-powered short-arm centrifuge for the following: 1) to determine whether subjects could perform squats under hypergravity conditions without developing motion sickness or illusory motion; 2) to measure foot forces while performing squats under hypergravity conditions; and 3) to determine the mechanical power requir

Stimulation of cyclic GMP efflux in human melanocytes by hypergravity generated by centrifugal acceleration.

Gravity alteration (micro- and hypergravity) is known to influence cell functions. As guanosine 3',5'-cyclic monophosphate (cGMP) plays an important role in human melanocyte functions and different guanylyl cyclase isoforms are responsible for cGMP synthesis in human non-metastatic and metastatic melanoma cells, we investigated the effects of hypergravity on the regulation of cGMP levels in cultured human melanocytes and in melanoma cell lines with different metastatic potentials. Hypergravity was produced by horizontal centrifugal acceleration. Here we report that long-term application of hyp

Hormonal modulation of food intake in response to low leptin levels induced by hypergravity.

A loss in fat mass is a common response to centrifugation and it results in low circulating leptin concentrations. However, rats adapted to hypergravity are euphagic. The focus of this study was to examine leptin and other peripheral signals of energy balance in the presence of a hypergravity-induced loss of fat mass and euphagia. Male Sprague-Dawley rats were centrifuged for 14 days at gravity levels of 1.25, 1.5, or 2 G, or they remained stationary at 1 G. Urinary catecholamines, urinary corticosterone, food intake, and body mass were measured on Days 11 to 14. Plasma hormones and epididymal

Blood-spinal cord barrier alterations in a mouse model of centrifugation-induced hypergravity.

Astronauts endure repetitive episodes of hypergravity (HG) during takeoff and landing of spaceflights, and also in space due to microgravity. Blood-brain barrier alterations and disruptions were recently reported in mice subjected to a short period of HG induced by centrifugation. In this study, we have evaluated if the blood-spinal cord barrier (BSCB) was also damaged by centrifugation-induced HG in mice. For that purpose, adult C57Bl/6J male mice were subjected to HG in a centrifuge at 2 g for 24 h, while control mice stayed in normogravity (n = 7-8 per group). Mice were sacrificed after cen

Impact of peripheral skin cooling on neuroendocrine leukocytic and hematological reactions during Hypergravity.

Optimal neuroendocrine responses are essential during hypergravity (+Gz) exposure. Peripheral skin cooling (PSC) may enhance neuroendocrine function, potentially improving +Gz resiliency and influencing leukocyte and hematologic factors. This study investigated whether PSC augments the cumulative +Gz stress index (CGSI) and shifts it toward noradrenergic dependency. Eighteen men underwent a graded +Gz profile in a crossover design, with PSC applied using Arctic Sun cooling pads. Neuroendocrine and blood profiles were assessed pre- and post-+Gz. CGSI did not differ between groups, but serum osm

Stability Studies of UV Laser Irradiated Promethazine and Thioridazine after Exposure to Hypergravity Conditions.

Pharmaceuticals carried into space are subjected to different gravitational conditions. Hypergravity is encountered in the first stage, during spacecraft launching. The stability of medicines represents a critical element of space missions, especially long-duration ones. Therefore, stability studies should be envisaged before the implementation of drugs for future deep space travel, where the available pharmaceuticals would be limited and restocking from Earth would be impossible. Multipurpose drugs should be proposed for this reason, such as phenothiazine derivatives that can be transformed b
