Cysteine import contributes to hypochlorous acid resistance in Staphylococcus aureus.

Hypochlorous acid (HOCl) is a potent antimicrobial oxidant produced by innate immune cells to eliminate invading bacterial pathogens, including Staphylococcus aureus. HOCl readily oxidizes thiols, amines, and macromolecules in S. aureus. Yet, some bacterial cells within an HOCl-challenged population withstand this oxidative stress and proceed to establish infection. The mechanisms that enable S. aureus to resist the effects of HOCl remain incompletely understood. Here, we show that cysteine uptake is a major determinant of HOCl resistance in S. aureus. Inactivation of the cysteine transporters

Linking Inflammation and Parkinson Disease: Hypochlorous Acid Generates Parkinsonian Poisons.

Inflammation is a common feature of Parkinson Disease and other neurodegenerative disorders. Hypochlorous acid (HOCl) is a reactive oxygen species formed by neutrophils and other myeloperoxidase-containing cells during inflammation. HOCl chlorinates the amine and catechol moieties of dopamine to produce chlorinated derivatives collectively termed chlorodopamine. Here, we report that chlorodopamine is toxic to dopaminergic neurons both in vivo and in vitro Intrastriatal administration of 90 nmol chlorodopamine to mice resulted in loss of dopaminergic neurons from the substantia nigra and decrea

Hypochlorous Acid: Considerations for Use in Skin Care and Skin Preparation.

Hypochlorous acid (HOCl) is endogenously produced by neutrophils, macrophages, and other cells of the innate immune system and acts as a natural antiseptic. The active components are chlorine and reactive oxygen species in the form of a weak acid. It can be manufactured by electrolysis or chemical acidification of sodium or calcium hypochlorite, producing a mixed solution of HOCl and hypochlorite ions (OCl-) in equilibrium. The ratios of HOCl to OCl- within the solutions vary depending upon the manufacturing process and method used for stabilization. These ratios can change over time, and main

Melatonin prevents hypochlorous acid-mediated cyanocobalamin destruction and cyanogen chloride generation.

Hypochlorous acid (HOCl) is a potent cytotoxic oxidant generated by the enzyme myeloperoxidase (MPO) in the presence of hydrogen peroxide (H2 O2 ) and chloride (Cl- ). Elevated levels of HOCl play an important role in various pathological conditions through oxidative modification of several biomolecules. Recently, we have highlighted the ability of HOCl to mediate the destruction of the metal-ion derivatives of tetrapyrrole macrocyclic rings such as hemoproteins and vitamin B12 (VB12 ) derivatives. Destruction of cyanocobalamin, a common pharmacological form of VB12 mediated by HOCl, results i

Germicide wound pad with active, in situ, electrolytically produced hypochlorous acid.

We describe a new wound dressing technology that can actively generate an inorganic germicide agent, in situ, within the wound pad. The technology provides real time control over the quantitative, spatial and temporal delivery of the germicide. The identity of the germicide is hypochlorous acid (HClO). The HClO is produced in a flexible wound pad, made of a composite of thin (micrometer scale) layers of various materials, with different electrochemical properties that enhance HClO production. Active control over the production of HClO is achieved by control of the pH and of the electric potent

Structural characteristics of histamine H2 receptor antagonists that scavenge hypochlorous acid.

Gastric and duodenal ulcers are characterized by hypersecretion of gastric acid and pepsin, inflammation of the mucosa and an influx of neutrophils. These cells can produce reactive oxygen species after stimulation. Particularly hydroxyl radicals and hypochlorous acid (HOCl) can be cytotoxic and damage the gastroduodenal mucosa. It has been shown that histamine H2 receptor antagonists such as cimetidine, ranitidine and famotidine are good hydroxyl radical scavengers. This study was undertaken to investigate whether these agents were able to scavenge the cytotoxic HOCl, and if so, which part of

Reaction of cyclamate with hypochlorous acid in vitro and formation of chlorcyclohexylamine and cyclohexylamine.

In this work, we investigated the reaction of cyclamate with hypochlorous acid (HOCl) in simulated gastric juice. The reaction products were detected by high-performance liquid chromatography diode array detection (HPLC-DAD) and ultra-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry (UPLC-ESI-MS/MS). We also explored the changes in product concentration as a function of reaction time, cyclamate and HOCl concentrations. Cyclamate reacted with hypochlorous acid instantly in the simulated gastric fluid. N, N-dichlorcyclohexylamine and cyclohexylamine

The role of hypochlorous acid in the management of eye infections: a case series.

Experiences from clinical practice suggest that hypochlorous acid (HOCl) used as adjuvant treatment can shorten the time to clinical resolution of blepharitis, particularly in relation to the ophthalmic complications that often occur with a blepharitis diagnosis. In addition, in clinical practice, HOCl adjuvant treatment also resulted in a viable option for the management of ophthalmic disorders other than blepharitis. This case series presents and discusses real-life experiences with using an HOCl ophthalmic spray to manage eye infections, further characterizing the use of HOCl for ophthalmic

Flow-through Instillation of Hypochlorous Acid in the Treatment of Necrotizing Fasciitis.

INTRODUCTION: Necrotizing fasciitis (NF) is a rare and rapidly progressing bacterial infection of soft tissues. Bacterial toxins cause local tissue damage and necrosis, as well as blunt immune system responses. A self-propagating cycle of bacterial invasion, toxin release and tissue destruction can continue until substantial amounts of tissue become necrotic. Neutralization of bacterial toxins should improve the results. MATERIALS AND METHODS: Pure hypochlorous acid (HOCl) (0.01% w/v) with no sodium hypochlorite impurity in saline pH 4-5, which was recently shown to both eradicate bacteria an

Reactions of 1-methyl-2-mercaptoimidazole with hypochlorous acid and superoxide.

Reactions of thioureylene antithyroid drugs (1-methyl-2-mercaptoimidazole and carbimazole) with hypochlorous acid (HOCl) and superoxide were followed optically and products were analyzed by mass spectrometry. 1-methyl-2-mercaptoimidazole (MMI) and carbimazole reacted rapidly with HOCl with a rate constant of 1 x 10(7) and 7 x 10(6) M(-1)s(-1), respectively. The characteristic spectrum assigned to MMI disulfide appeared immediately after addition of HOCl, followed by a slow conversion to a final spectrum. The conversion was dependent upon the ratio of HOCl to MMI and both antithyroid drugs upta
