Three-dimensional super-resolution fluorescence imaging of DNA.

Recent advances in fluorescence super-resolution microscopy are providing important insights into details of cellular structures. To acquire three dimensional (3D) super-resolution images of DNA, we combined binding activated localization microscopy (BALM) using fluorescent double-stranded DNA intercalators and optical astigmatism. We quantitatively establish the advantage of bis- over mono-intercalators before demonstrating the approach by visualizing single DNA molecules stretched between microspheres at various heights. Finally, the approach is applied to the more complex environment of int

CUBIC pathology: three-dimensional imaging for pathological diagnosis.

The examination of hematoxylin and eosin (H&E)-stained tissues on glass slides by conventional light microscopy is the foundation for histopathological diagnosis. However, this conventional method has some limitations in x-y axes due to its relatively narrow range of observation area and in z-axis due to its two-dimensionality. In this study, we applied a CUBIC pipeline, which is the most powerful tissue-clearing and three-dimensional (3D)-imaging technique, to clinical pathology. CUBIC was applicable to 3D imaging of both normal and abnormal patient-derived, human lung and lymph node tissues.

Three-dimensional CT imaging in pediatric calvarial pathologies.

In children with suspected cranial pathologies, three-dimensional (3D) computed tomography (CT) imaging is superior to other modalities. It can help differentiate actual pathology from normal or variant appearances. Sutures and fontanelles, synostosis, abnormalities of head shape without craniosynostosis, congenital calvarial defects, cranial fractures, bone tumors, and postoperative cranial vault can be assessed easily with 3D CT imaging. We aimed to discuss the common normal, variant, and pathological findings that 3D CT imaging can aid to diagnose as well as explain the usefulness of 3D CT

Three-dimensional US of the fetus: volume imaging.

PURPOSE: To retrospectively compare the rapidity, efficiency, and accuracy of three-dimensional (3D) and two-dimensional (2D) ultrasonography (US) for complete anatomic survey in fetuses at 17-21 weeks of gestation. MATERIALS AND METHODS: Institutional review board approval was obtained, informed consent was waived, and the study was HIPAA compliant. Fifty consecutive women undergoing fetal anatomic survey at 17-21 weeks of gestation formed the study cohort. After standard 2D US was performed by one of eight sonographers, the same sonographer also obtained five 3D volumes to encompass the ent

Three-dimensional imaging diagnosis for cranial base lesions.

Diagnosis of neurological disease by imaging has greatly improved in recent years. Precise morphological information can be obtained by computed tomography and magnetic resonance imaging, and biochemical or electrophysiological dynamic changes can be assessed by positron emission tomography or magnetoencephalography. The transcription of such biochemical or electrophysiological data on computed tomographic scan or magnetic resonance imaging can precisely demonstrate the local dynamic information in the brain. Three-dimensional imaging also provides accurate intelligence on the morphological st

Three-dimensional CT-imaging of the ossicular chain.

In eight patients, with a known air-conduction hearing loss, three-dimensional images of the ossicular chain were constructed based on heavily overlapping high resolution CT-slices, obtained from a spiral CT data set. High quality three-dimensional images could be obtained in all eight patients. Supplementary information was derived from these three-dimensional images in three patients; in one, this supplementary information had therapeutic importance. The slight image quality loss in spiral-CT of the temporal bone, compared with conventional CT, is in some cases largely compensated by the pos

Evaluation of craniosynostosis with three-dimensional CT imaging.

Technological advances in computer image analysis have made possible three-dimensional (3D) surface reformations of anatomic structures from contiguous axial CT slices. We have recently utilized this new imaging technique in six patients with primary, isolated craniosynostosis. In each case 3D CT demonstrated the exact extent of synostosis, and in five cases 3D CT images facilitated surgical planning by precisely localizing the proper site for craniectomy. Three-dimensional CT may be a useful adjunct to imaging and surgical planning in this condition.

Three-dimensional imaging of a juvenile Tillaux fracture.

The juvenile Tillaux fracture is an injury involving the articular surface of the ankle joint. The fracture sometimes requires open reduction and internal fixation. The decision to adopt an operative or nonoperative policy in each case can only be made with precise imaging of the anatomy which can be provided by three-dimensional reformation of computed tomography (CT) scanning.

Three-Dimensional Optical Imaging for Pectus Excavatum Assessment.

BACKGROUND: Corrective surgery for pectus excavatum often relies on the Haller index (HI), derived from chest roentgenograms or computed tomography; however, this exposes children to potentially unnecessary radiation. Our aim was to develop a novel three-dimensional (3D) optical imaging technique to accurately measure chest wall dimensions in a clinically relevant manner. METHODS: Patients with pectus excavatum were imaged using a 3D structured light scanner. Patient characteristics, including height, weight, body mass index, and radiographic HIs (rHI) were recorded. We defined the optical in

Perspective: three-dimensional imaging of the musculoskeletal system.

An evaluation of musculoskeletal disorders in 202 patients using three-dimensional CT displays has revealed the usefulness of the technique, especially in patients with fractures of skeletal areas with complex anatomy, articular disorders of the hip, and spinal stenosis. In some cases, plastic models of diseased areas have been created from the CT data and are reasonably accurate, providing a graphic representation of the disease and the ability to perform rehearsal surgery. A preliminary investigation of three-dimensional displays of MR images indicates that the technique is feasible, althoug

Three-dimensional ultrasonic images of normal fetus.

BACKGROUND: The purpose of this study was to assess the usefulness and feasibility of 3-dimensional (3D) ultrasound for routine fetal observation. METHODS: Eighty-five normal pregnancies with gestational ages of 20-36 weeks were studied. A conventional ultrasound scanner with a specially designed operation unit, frame memory, and 3D probe was the apparatus used in this study. The unit has a circuit board consisting of digital integrated circuit chips designed to make calculations of volume ray tracings for volume rendering in real time. For each fetus, scanning was performed with different le

Three dimensional image alignment, registration and fusion.

Combined assessment of three dimensional anatomical and functional images (SPECT, PET, MRI, CT) is useful to determine the nature and extent of lesions in many parts of the body. Physicians principally rely on their spatial sense to mentally re-orient and overlap images obtained with different imaging modalities. Objective methods that enable easy and intuitive image registration can help the physician arrive at more optimal diagnoses and better treatment decisions. This review describes a simple, intuitive and robust image registration approach developed in our laboratory. It differs from mos

[Three-dimensional ultrasound image of the infant hip].

Ultrasound imaging of the infant hip has gained wide acceptance, especially in diagnosis and therapy of infant hip dysplasias. Problems are the prognosis with regard to normal and pathological alpha-angle and treatment using cast, harness, or pillow. The well-known two-dimensional ultrasound techniques are not able to reproduce the correct relationship between femoral head and acetabulum. The use of the new three-dimensional technique is independent of the position of the infant hip. Although a special view into the acetabulum is possible, at present this method is not suitable for screening p

Three-dimensional and four-dimensional HDlive-rendered images of thanatophoric dysplasia.

We present herein our first experience with a case of thanatophoric dysplasia (type I) that was diagnosed during the second trimester using three- and four-dimensional HDlive ultrasonography. The HDlive rendering mode clearly showed the anatomical features of thanatophoric dysplasia: external malformations and skeletal abnormalities, including extremely short limbs, flattened vertebral bodies, and short horizontal ribs, among others. HDlive can provide valuable, highly realistic images for the differential diagnosis of skeletal dysplasia. It may also play an important complementary role when c
