Maternal-fetal transmission of Toxoplasma gondii tachyzoites acquired during pregnancy has potentially dramatic consequences for the fetus. Current reference-standard treatments are not specific to the parasite and can induce severe side effects. In order to provide treatments with a higher specificity against toxoplasmosis, we developed antibody fragments-single-chain fragment variable (scFv) and scFv fused with mouse immunoglobulin G2a crystallizable fragment (scFv-Fc)-directed against the major surface protein SAG1. After validating their capacity to inhibit T. gondii proliferation in vitro

The self-association of therapeutic antibodies can result in elevated viscosity and create problems in manufacturing and formulation, as well as limit delivery by subcutaneous injection. The high concentration viscosity of some antibodies has been reduced by variable domain mutations or by the addition of formulation excipients. In contrast, the impact of Fc mutations on antibody viscosity has been minimally explored. Here, we studied the effect of a panel of common and clinically validated Fc mutations on the viscosity of two closely related humanized IgG1, κ antibodies, omalizumab (anti-IgE)

The trophoblastic tissue and endothelium of fetal stem vessels in cryostat sections of first-trimester and term human placentae adsorbed erythrocytes sensitized with IgG antibodies of human, rabbit, and guinea pig origin. Native and heat-aggregated human and rabbit IgG, and human IgG subclasses except IgG2 inhibited the hemadsorption. However, the Fc fragment of IgG2 inhibited the hemadsorption similarly to the Fc of pooled IgG. No inhibition was obtained with other immunoglobulin classes or fragments. The inhibitory activity of IgG and Fc fragments was abolished by partial reduction and alkyl

A high-throughput method for domain-specific Fab glycosylation analysis was developed for monoclonal antibodies. Fab glycosylation can affect a product's efficacy and safety. Conventional domain-specific glycan analysis-typically relying on IdeS digestion and affinity chromatography-is labor-intensive. In this study, a mathematical subtraction approach using native Fc and total glycosylation data was initially evaluated but was found to suffer from significant error amplification (up to 8.0%). Accordingly, a practical workflow based on physical separation was established. The method entails ra

The variable domains in Fab regions are well-established contributors to high concentration viscosity of IgG, primarily through charge and hydrophobic interactions. In contrast, the roles of the Fc and the number of self-interacting sites (valency) are less well understood. Here, we investigate the relative contributions of Fab-Fab and Fab-Fc interactions to high concentration viscosity for a diverse panel of 20 IgG1 antibodies, by rheometry, coarse-grained simulations, and molecular surface property analysis. Strikingly, fragmentation of IgG1 into F(ab')2 plus Fc reduced viscosity (-11 to -93

Removal of Fc-fragments from human IgG or Fc-fused proteins. (1) IgG or Fc-fusion proteins are incubated with IdeS-Fc; (2) Fc fragments are retained using protein G agarose beads; and (3) the F(ab')2 fragments or Fc-free proteins are collected by centrifugation in the original buffer without IdeS contamination.

Glycosylation of monoclonal antibodies is a key determinant of their efficacy, stability, and safety, requiring analytical methods capable of resolving site-specific heterogeneity with high sensitivity. In this work, we present a Protein A-assisted, domain-resolved glycan profiling approach for the mass spectrometry-based structural elucidation and quantification of N-glycans linked to Fc and Fab in cetuximab. Antibody domains were selectively extracted using Protein A affinity separation, allowing for independent enzymatic release of N-glycans from Fc and Fab sections, followed by high-resolu

The targeting of a glycosylated antibody Fc fragment to bind to cancer cells by site-selective incorporation of a synthetic ligand is described. Homogeneously glycosylated immunoglobulin G subclass 1 fragment crystallizable (IgG1 Fc) was produced by expression in a glycosylation-deficient yeast strain and subsequent treatment with mannosidase IA. A N-terminal cysteine was generated on the expressed IgG1 Fc by utilizing proteolytic processing enzymes in the yeast secretory pathway. A cyclic RGD peptide thioester 2 was synthesized and then site-selectively attached to the N-terminus of the IgG1

In the last decade, the landscape of treating autoimmune diseases has evolved with the emergence and approval of novel targeted therapies. Several new biological agents offer selective and target-specific immunotherapy and therefore fewer side effects, such as neonatal Fc receptor (FcRn)-targeting therapy. Neonatal Fc receptor-targeted therapies are engineered to selectively target FcRn through various methods, such as Fc fragments or monoclonal anti-FcRn antibodies. These approaches enhance the breakdown of autoantibodies by blocking the immunoglobulin G recycling pathway. This mechanism redu

Excessive influx of immunoglobulin (IgG) into the brain has been reported to induce central nervous system (CNS) dysfunction. Depressed patients may exhibit immune activation manifested by elevated inflammatory markers and pro-inflammatory cytokines. The brain and especially the limbic system contain high concentrations of high affinity Fc receptors. We reviewed the literature on this phenomena and present data on the behavioral effects of pooled normal IgG on the brain. Many disease states are associated with depression and we examined whether this may be linked to high IgG influx. Female Bal

When evaluating the proclivity of a monoclonal antibody (mAb) for non-specific self-assembly, it is common to restrict charge distribution analyses to the variable region of mAbs, leaving out possible contributions from the constant region to the observed sticky behavior. Here, the aim was to study the relationship between charge asymmetry over the entire mAb surface and self-assembly propensity. To do so, we selected three mAbs with decreasing levels of charge asymmetry and evaluated their ability to engage in attractive self-interaction as a function of mAb concentration and ionic strength,

Rituximab is a chimeric monoclonal antibody containing constant kappa-regions of the Fc fragment of IgGI human immunoglobulin and variable fragments of IgG murine immunoglobulin. This modified immunoglobulin structure makes rituximab capable of specifically recognizing CD20 antigen that is mainly expressed on B-lymphocytes. The review considers four main mechanisms of rituximab action leading to reduction of the population of B-lymphocytes in patients with lymphoproliferative processes. All these mechanisms are probably operative under clinical conditions, however, their relative contributions

That in vitro killing by normal and activated macrophages of S. typhimurium and other gram-negative organisms is dependent on the presence of antibody has been confirmed. It has been shown that antibody is required for the binding of S. typhimurium to the surfaces of macrophages. This binding can be inhibited by the Fc portion of immunoglobulin, indicating that the Fc receptors on macrophages are used for binding S. typhimurium. It has also been confirmed that antibody does not appear to be necessary for killing of L. monocytogenes to occur. The organisms bind to the surface of macrophages by

The possible role of specific mechanisms involved in the adherence process of immune aggregates to tissue components of the mouse eye was investigated in an experimental animal model. Passive intravenous administration of immunoglobulin aggregates to mice, resulted in the localisation of these aggregates in various organs, including the eye. In the eye a strong localisation was seen in the episcleral capillary plexus, whereas only a weak deposition was seen in the iris, ciliary body and choroid. No deposits were seen in the retina. To investigate the role of specific receptors for immune compl

Fc domain engineering enables precise modulation of antibody effector functions, serum half-life, conjugation sites, and chain pairing. However, the associated patent landscape is highly complex, shaped by inconsistent mutation numbering, overlapping claims, and variable patent term extensions. This article outlines key technological strategies-ranging from Fcγ receptor and FcRn affinity modulation to antibody-drug conjugate optimization-and illustrates common intellectual property pitfalls with representative patents. Practical approaches to freedom-to-operate assessment are discussed, includ

Delayed in vivo elimination of autologous erythrocytes coated with immunoglobulin G has been reported in autoimmune and inflammatory gastrointestinal diseases. Our aim was to elucidate whether impairment of the macrophages was restricted to the Fc receptors of the reticuloendothelial system or whether complement receptors were also affected. We studied elimination by complement receptors of autologous erythrocytes coated with fragments of C3 and C4 in patients with primary biliary cirrhosis, ulcerative colitis, and alcoholic cirrhosis. Impaired function was seen in all patient groups as compar

This article demonstrates the presence of selective binding activity for the Fc fragment of IgG on the ciliary processes of rabbit eye. Other parts of the eye, including the cornea, iris, choroid, and retina, were negative for such activity. Binding activity was demonstrated in vitro by the specific adherence of IgG-coated sheep red blood cells (IgGEA) to the ciliary processes in frozen sections of whole rabbit eye. IgGEA binding was specifically blocked by IgG but not by albumin or the F(ab')2 fragment of IgG. The data suggest that the ciliary processes, like the choroid plexus and the renal

Conventional production of antibody fragments (e.g. Fab/F(ab')2, VHH/(VHH)2) involves sequential enzymatic cleavage and multi-step purification to remove Fc domains and residual enzymes, a process that is laborious and inefficient for small-scale workflows. To address this, we engineered a bifunctional IdeS-Protein A (IPA) fusion protein, which combines the specific IgG-cleaving activity of IdeS with the Fc-binding capability of Protein A. In solution, this construct enabled simultaneous antibody cleavage and affinity-mediated removal of both the enzyme and Fc fragments in a single step. Furth
