Immunoinformatics for Novel Multi-Epitope Vaccine Development in Canine Parvovirus Infections.

Canine parvovirus (CPV-2) is one of the most important pathogens of dogs of all ages, causing pandemic infections that are characterized by fatal hemorrhagic enteritis. The CPV-2 vaccine is recommended as a core vaccine for pet animals. Despite the intensive practice of active immunization, CPV-2 remains a global threat. In this study, a multi-epitope vaccine against CPV-2 was designed, targeting the highly conserved capsid protein (VP2) via in silico approaches. Several immunoinformatics methods, such as epitope screening, molecular docking, and simulation were used to design a potential vacc

Immunoinformatics approach for multi-epitope vaccine design against P. falciparum malaria.

Plasmodium falciparum (P. falciparum) is a leading causative agent of malaria, an infectious disease that can be fatal. Unfortunately, control measures are becoming less effective over time. A vaccine is needed to effectively control malaria and lead towards the total elimination of the disease. There have been multiple attempts to develop a vaccine, but to date, none have been certified as appropriate for wide-scale use. In this study, an immunoinformatics method is presented to design a multi-epitope vaccine construct predicted to be effective against P. falciparum malaria. This was done thr

Immunoinformatics driven multi-epitope vaccine design targeting clade IIb Mpox variant.

The Monkeypox virus (MPXV), an emerging zoonotic orthopoxvirus, has reemerged as a major global health threat, with the 2022 outbreak predominantly associated with Clade IIb lineages. The surface glycoprotein B21R (OPG210), a major virulence and immune-evasion factor, contains three lineage-defining amino acid substitutions (D209N, P722S, and M1741I) that distinguish Clade IIb from other variants, emphasizing its relevance as a vaccine target. In this study, an integrated immunoinformatics and molecular dynamics-based approach was employed to design a multi-epitope vaccine (MEV) targeting the

Designing a Multi-Epitope Vaccine against Toxoplasma gondii: An Immunoinformatics Approach.

Infection with the intracellular apicomplexan parasite Toxoplasma gondii causes serious clinical outcomes in both human and veterinary settings worldwide. Although approximately one-third of the world's population is infected with T. gondii, an effective human vaccine for this disease remains unavailable. We aimed to design a potential T. gondii vaccine candidate that consisted of the B- and T-lymphocyte epitopes of three parasite immunogenic antigens. Firstly, the immunodominant epitopes expressed within the ROP2, MIC3, and GRA7 proteins of T. gondii were identified. Subsequently, six B-cell

Immunoinformatics based designing a multi-epitope vaccine against pathogenic Chandipura vesiculovirus.

Chandipura vesiculovirus (CHPV) is a rapidly emerging pathogen responsible for causing acute encephalitis. Due to its widespread occurrence in Asian and African countries, this has become a global threat, and there is an urgent need to design an effective and nonallergenic vaccine against this pathogen. The present study aimed to develop a multi-epitope vaccine using an immunoinformatics approach. The conventional method of vaccine design involves large proteins or whole organism which leads to unnecessary antigenic load with increased chances of allergenic reactions. In addition, the process

Multi-epitope based vaccine against yellow fever virus applying immunoinformatics approaches.

Yellow fever disease is considered a re-emerging major health issue which has caused recent outbreaks with a high number of deaths. Tropical countries, mainly African and South American, are the most affected by Yellow fever outbreaks. Despite the availability of an attenuated vaccine, its use is limited for some groups such as pregnant and nursing women, immunocompromised and immunosuppressed patients, elderly people >65 years, infants <6 months and patients with biological disorders like thymus disorders. In order to achieve new preventive measures, we applied immunoinformatics approaches to

Immunoinformatic predictions and characterization of Schistosoma mansoni peptides as candidates for immunodiagnostic.

Schistosoma mansoni represents a significant etiological agent of schistosomiasis, a neglected tropical disease with a global distribution. Although the Kato-Katz technique is an effective diagnostic tool in areas with a high prevalence of the disease, it lacks sensitivity in regions with lower prevalence. The objective of this study was to identify and validate novel immunogenic peptide targets derived from the S. mansoni proteome. The initial set of 14,499 predicted sequences were obtained from the WormBase database, and it was filtered to 8,308 by removing proteins lacking start or stop cod

Design of a Multi-epitope Antigen for Toxoplasmosis Diagnosis: An Immunoinformatics Approach.

Toxoplasma gondii, as an obligatory intracellular protozoan, can infect a diverse array of individuals and warm-blooded creatures. Considering the worldwide public health implications caused by T. gondii infection, it is an important goal to develop effective diagnostic tests or vaccines. The application of natural antigens derived from the parasite in these assessments encounters challenges, including the complexities of culturing the parasites, strain differences, and the high cost of kits produced based on natural antigens. The current bioinformatic study aimed to develop a multi-epitope T.

Design of a cryptococcus neoformans vaccine by subtractive proteomics combined with immunoinformatics.

The emergence of Cryptococcus neoformans has posed an undeniable burden to many regions worldwide, with its strains mainly entering the lungs through the respiratory tract and spreading throughout the body. Limitations of drug regimens, such as high costs and limited options, have directed our attention toward the promising field of vaccine development. In this study, the subtractive proteomics approach was employed to select target proteins from databases that can accurately cover serotypes A and D of the Cryptococcus neoformans. Further, two multi-epitope vaccines consisting of T and B cell
