Targeting immunosenescence for improved tumor immunotherapy.

Tumor immunotherapy has significantly transformed the field of oncology over the past decade. An optimal tumor immunotherapy would ideally elicit robust innate and adaptive immune responses within tumor immune microenvironment (TIME). Unfortunately, immune system experiences functional decline with chronological age, a process termed "immunosenescence," which contributes to impaired immune responses against pathogens, suboptimal vaccination outcomes, and heightened vulnerability to various diseases, including cancer. In this context, we will elucidate hallmarks and molecular mechanisms underly

Microbiome, Immunosenescence, and Chronic Kidney Disease.

The gut microbiome is known as an important predictive tool for perceiving characteristic shifts in disease states. Multiple renal diseases and pathologies seem to be associated with gut dysbiosis which directly affects host homeostasis. The gastrointestinal-kidney dialogue confers interesting information about the pathogenesis of multiple kidney diseases. Moreover, aging is followed by specific shifts in the human microbiome, and gradual elimination of physiological functions predisposes the microbiome to inflammaging, sarcopenia, and disease. Aging is characterized by a microbiota with an ab

Biological age and immunosenescence in Colombian centenarians.

Biological aging and immunosenescence are central to longevity, yet their interplay in centenarians remains unclear. We conducted a cross-sectional study in 160 Colombian centenarians to examine associations between biological age (PhenoAge), immunosenescence and age-related clinical variables. Cytokine profiling (n = 114) and lymphocyte immunophenotyping (n = 42) were assessed. It was observed that better QoL and well-being were significantly associated with lower biological age, while depressive symptoms, prior tobacco use, elevated levels of RANTES and G-CSF as well as a distinct CD8+ T cel

Genetic association of immunosenescence to immune decline.

Immunosenescence is a complex biological process characterized by progressive remodeling of immune function during aging, leading to increased susceptibility to infections, chronic inflammatory diseases, cancer, and reduced vaccine efficacy. While cumulative antigen exposure, infections, lifestyle factors, environmental exposures, and epigenetic influences contribute to immune decline, a growing body of evidence suggests that genetic predisposition also plays an important modulatory role in shaping the molecular trajectory of immune aging. This review discusses how inherited variation in cytok

Sirtuin signaling as therapeutic targets of immunosenescence.

INTRODUCTION: Aging is characterized by immunosenescence, encompassing, immune aging and inflammaging. Because immune competence is a critical determinant of lifespan and healthspan, targeting immunosenescence has become a major objective of geroscience. In this context, sirtuins have emerged as compelling therapeutic candidates. AREAS COVERED: This review examines the role of sirtuin signaling in immunosenescence, focusing on its links with nicotinamide adenine dinucleotide (NAD)+ metabolism, oxidative stress, and inflammatory pathways. Pharmacological strategies aimed at enhancing sirtuin a

Immunosenescence and organoids: pathophysiology and therapeutic opportunities.

Immunosenescence is the gradual deterioration in the functionality of the immune system that has various clinical manifestations, such as a weakened response to vaccination, higher susceptibility to viral/bacterial/fungal infection, and higher incidence rates regarding the ageing-based disorders. Conventionally used models, like animal or 2D models for unraveling the interactions between ageing and immunity are unable to depict the complexity of bodily environments. However, this major limitation can be addressed via using the organoid technology. For clarification, organoids are tissue-resemb

Immunosenescence in Neurological Diseases-Is There Enough Evidence?

The aging of the immune system has recently attracted a lot of attention. Immune senescence describes changes that the immune system undergoes over time. The importance of immune senescence in neurological diseases is increasingly discussed. For this review, we considered studies that investigated cellular changes in the aging immune system and in neurological disease. Twenty-six studies were included in our analysis (for the following diseases: multiple sclerosis, stroke, Parkinson's disease, and dementia). The studies differed considerably in terms of the patient groups included and the cell

B cell immunosenescence in the elderly and in centenarians.

The elderly suffer from an increased susceptibility to infectious disease and cancer. Aging of the immune system contributes to this state of affairs due to immunosenescence. Because repeated intermittent or chronic antigen exposure may lead to lymphocyte clonal exhaustion, chronic antigenic stress plays a part in the compromised immunity of the elderly, who have accumulated a lifetime's exposure to infectious agents, autoantigens, and cancer antigens. Literature on immunosenescence has focused mainly on T cell impairment, but B cell compartment is also affected. The age-dependent B cell chang

Immunosenescence: signaling pathways, diseases and therapeutic targets.

Immunosenescence refers to the abnormal activation or dysfunction of the immune system as people age. Inflammaging is a typical pathological inflammatory state associated with immunosenescence and is characterized by excessive expression of proinflammatory cytokines in aged immune cells. Chronic inflammation contributes to a variety of age-related diseases, such as neurodegenerative disease, cancer, infectious disease, and autoimmune diseases. Although not fully understood, recent studies contribute greatly to uncovering the underlying mechanisms of immunosenescence at the molecular and cellul

Psychological stress on cancer progression and immunosenescence.

Diagnosis and treatment of cancer constitute a deeply stressful experience that involves unique and common problems and generates uncertainty, fear and emotional distress. Furthermore, there are reciprocal interactions between psychological stress and cancer in the clinical settings. Therefore, it is crucial to understand the links of stress with cancer. A growing body of epidemiological and preclinical studies have suggested that stress affects cancer progression, and metastasis and treatment outcomes. Furthermore, stress elicits premature aging and deterioration of the immune system (known a

An immunologist's guide to immunosenescence and its treatment.

INTRODUCTION: Aging causes several changes in the immune system, although immune aging is strongly influenced by individual immunological history, as well as genetic and environmental factors leading to inter-individual variability. AREAS COVERED: We focused on the biological and clinical meaning of immunosenescence. SARS-CoV-2 and Yellow Fever vaccine have demonstrated the clinical relevance of immunosenescence, while inconsistent results, obtained from longitudinal studies aimed at looking for immune risk phenotypes, have revealed that immunosenescence is highly context-dependent. Cellular senescence in lymphoid organs and immunosenescence.

Immunosenescence is a multi-faceted phenomenon at the root of age-associated immune dysfunction. It can lead to an array of pathological conditions, including but not limited to a decreased capability to surveil and clear senescent cells (SnCs) and cancerous cells, an increased autoimmune responses leading to tissue damage, a reduced ability to tackle pathogens, and a decreased competence to illicit a robust response to vaccination. Cellular senescence is a phenomenon by which oncogene-activated, stressed or damaged cells undergo a stable cell cycle arrest. Failure to efficiently clear SnCs re

[Immunosenescence - the sunset over the immune system].

Immunosenescence is characterized by remodeling and dysregulation of immune system due to aging process. These changes affect innate as well as adaptive immunity. Due to the complexity of the physiological processes, in which the mechanisms of the immune system interfere, also other systems of the organism are affected by these changes. Thymus involution as well as chronic antigenic stimulation are the main causes of immunosenescence and lead to a proinflammatory setting of the organism. In addition to impaired immune response against infections or reactivation of latent infections, reduced re

Immunosenescence: a key player in cancer development.

Immunosenescence is a process of immune dysfunction that occurs with age and includes remodeling of lymphoid organs, leading to changes in the immune function of the elderly, which is closely related to the development of infections, autoimmune diseases, and malignant tumors. T cell-output decline is an important feature of immunosenescence as well as the production of senescence-associated secretory phenotype, increased glycolysis, and reactive oxygen species. Senescent T cells exhibit abnormal phenotypes, including downregulation of CD27, CD28, and upregulation of CD57, killer cell lectin-li

Rheumatoid Arthritis, Immunosenescence and the Hallmarks of Aging.

Age is the most important risk factor for the development of infectious diseases, cancer and chronic inflammatory diseases including rheumatoid arthritis (RA). The very act of living causes damage to cells. A network of molecular, cellular and physiological maintenance and repair systems creates a buffering capacity against these damages. Aging leads to progressive shrinkage of the buffering capacity and increases vulnerability. In order to better understand the complex mammalian aging processes, nine hallmarks of aging and their interrelatedness were recently put forward. RA is a chronic auto

Immunosenescence and human healthspan. Lessons from centenarians.

Immunosenescence is a multidimensional remodeling of immunity, characterized by inflammaging, cellular senescence, T-cell exhaustion, and thymic involution, that raises infection and disease risk with age. Emerging evidence, notably from centenarians, shows immune aging follows divergent trajectories: rather than a uniform decline, extreme longevity often reflects adaptive remodeling and a maintained immune equilibrium. Centenarian immune profiles are characterized by selective retention of naïve T cells, expansion of cytotoxic CD4+ and CD8+ subsets, tightly regulated inflammatory signaling, a

Conquering aging-related immunosenescence and tumor immune escape.

With global population aging, senescence has emerged as a key driver of tumorigenesis. Aging-associated molecular changes, including DNA damage, telomere shortening, and epigenetic dysregulation, increase malignancy, while immunosenescence and the senescence-associated secretory phenotype (SASP), reshape the tumor microenvironment to favor immune suppression and tumor escape. Aging also impairs antigen presentation, disrupts ligand-receptor signaling, and compromises tumor suppressive pathways. In the era of immunotherapy, elderly patients face reduced efficacy and increased resistance due to

Immunosenescence: A new direction in anti-aging research.

The immune system is a major regulatory system of the body, that is composed of immune cells, immune organs, and related signaling factors. As an organism ages, observable age-related changes in the function of the immune system accumulate in a process described as 'immune aging. Research has shown that the impact of aging on immunity is detrimental, with various dysregulated responses that affect the function of immune cells at the cellular level. For example, increased aging has been shown to result in the abnormal chemotaxis of neutrophils and decreased phagocytosis of macrophages. Age-rela

Immunosenescence as a driver of the transition from frailty to multimorbidity.

Frailty and multimorbidity are closely intertwined syndromes of later life, yet they are still commonly interpreted through parallel clinical frameworks rather than a shared biological mechanism. In this mini review, we propose that immunosenescence provides a unifying axis linking the transition from frailty to multimorbidity. Rather than representing a simple decline in immune function, immunosenescence is better understood as a maladaptive remodeling process characterized by constrained adaptive immune renewal, repertoire narrowing, chronic low-grade inflammation, impaired immune surveillan
