Circulatory Adipokines and Incretins in Adolescent Idiopathic Scoliosis: A Pilot Study.

Adolescent idiopathic scoliosis (AIS) is a three-dimensional malformation of the spine of unknown cause that develops between 10 and 18 years old and affects 2-3% of adolescents, mostly girls. It has been reported that girls with AIS have a taller stature, lower body mass index (BMI), and bone mineral density (BMD) than their peers, but the causes remain unexplained. Energy metabolism discrepancies, including alterations in adipokine and incretin circulatory levels, could influence these parameters and contribute to disease pathophysiology. This pilot study aims to compare the anthropometry, B

Effects of Different Immunosuppressive Drugs on Incretins in Renal Transplant Patients.

BACKGROUND: Immunosuppressive drugs used in transplantation patients, may contribute to the development of post-transplant diabetes mellitus through their possible adverse effects on incretins. We aimed to compare the effects of different immunosuppressive drugs used in renal transplantation patients on glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) levels. PATIENTS AND METHODS: Forty five subjects were enrolled in the study (cyclosporine-treated 15 and tacrolimus-treated renal transplant patients 15, and healthy volunteers as a control group 15). Oral

The effects of incretins as a double-edged sword on cancer.

PURPOSE: Incretins, such as Glucagon-like peptide-1 (GLP-1) and Glucose-dependent insulinotropic peptide (GIP), are hormones known to stimulate insulin secretion and are widely used in the management of type 2 diabetes mellitus (T2DM). Recent evidence suggests that these hormones, and incretin-based therapies like DPP-4 inhibitors and GLP-1 receptor agonists, may have proliferative effects on pancreatic β-cells, raising concerns about a potential link to certain cancers. This review aims to evaluate both the beneficial and detrimental effects of incretins in cancer development and therapy. ME

Incretins and microvascular complications of diabetes: neuropathy, nephropathy, retinopathy and microangiopathy.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs, incretin mimetics) and dipeptidyl peptidase-4 inhibitors (DPP-4is, incretin enhancers) are glucose-lowering therapies with proven cardiovascular safety, but their effect on microvascular disease is not fully understood. Both therapies increase GLP-1 receptor agonism, which is associated with attenuation of numerous pathological processes that may lead to microvascular benefits, including decreased reactive oxygen species (ROS) production, decreased inflammation and improved vascular function. DPP-4is also increase stromal cell-derived factor

Pleiotropic actions of the incretin hormones.

The insulin secretory response to a meal results largely from glucose stimulation of the pancreatic islets and both direct and indirect (autonomic) glucose-dependent stimulation by incretin hormones released from the gastrointestinal tract. Two incretins, Glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1), have so far been identified. Localization of the cognate G protein-coupled receptors for GIP and GLP-1 revealed that they are present in numerous tissues in addition to the endocrine pancreas, including the gastrointestinal, cardiovascular, central nervous

Incretin-Based Therapies: A Testable Hypothesis Linking Incretin Signaling, Mitochondrial Redox, and OXPHOS Efficiency.

OBJECTIVE: Incretin-based obesity therapies (IBTs), especially GLP-1 receptor agonists (GLP-1 RAs), effectively treat obesity and improve comorbidities. However, their impact on energy metabolism is unclear. A recent case of acute generalized muscle weakness in a patient with mitochondrial myopathy after tirzepatide exposure raises concerns about IBTs' potential to uncouple oxidative phosphorylation (OXPHOS). Given the widespread use of IBTs, this inquiry is crucial due to potential adverse effects. METHODS: A narrative literature review of evidence linking incretins and IBTs to mitochondrial

Role of incretins in Alzheimer's disease and their impact on behavioral aspects.

Recent studies highlight the therapeutic potential of glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic peptide (GIP), two key incretin components, in neurodegenerative diseases, particularly Alzheimer's disease (AD). Although the incretin pathway may also affect other neurodegenerative conditions, this discussion focuses on AD. The therapeutic benefits are largely attributed to their role in enhancing energy metabolism. In vivo and in vitro studies have demonstrated that GLP-1 and GIP exert neuroprotective effects through the inhibition of oxidative stress, neuroinflammatio

Incretins for obesity: closer to a nicotine patch than a silver bullet.

INTRODUCTION: Obesity is a pandemic and a key driver of type 2 diabetes mellitus, heart disease, and healthcare spending. Incretin drug therapy prescribing for obesity has dramatically increased in recent years, necessitating researchers and clinicians to rapidly update their understanding of and approach to obesity management. AREAS COVERED: We assembled a multi-disciplinary team of experts to identify and complete a narrative review of the relevant literature regarding behavioral interventions, nutrition, physical activity, incretin drug therapy for weight loss, and the management of incret

Renin-Angiotensin System in Liver Metabolism: Gender Differences and Role of Incretins.

The impaired hepatic lipids and carbohydrates metabolism result in various metabolic disorders, including obesity, diabetes, insulin resistance, hyperlipidemia and metabolic syndrome. The renin-angiotensin system (RAS) has been identified in the liver and it is now recognized as an important modulator of body metabolic processes. This review is intended to provide an update of the impact of the renin-angiotensin system on lipid and carbohydrate metabolism, regarding gender difference and prenatal undernutrition, specifically focused on the role of the liver. The discovery of angiotensin-conver

Incretins Enhance PGF2α-Induced Synthesis of IL-6 and Osteoprotegerin in Osteoblasts.

Incretins including glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1), which are secreted from the small intestine after oral food ingestion, are currently well-known to stimulate insulin secretion from pancreatic β-cells and used for the treatment of type 2 diabetes mellitus. We have previously reported that prostaglandin F2α (PGF2α) stimulates the synthesis of interleukin-6 (IL-6) and osteoprotegerin in osteoblast-like MC3T3-E1 cells, and that IL-6 and osteoprotegerin release are mediated through the p44/p42 mitogen-activated protein (MAP) kinase, p38 MAP

Effects of meal and incretins in the regulation of splanchnic blood flow.

OBJECTIVE: Meal ingestion is followed by a redistribution of blood flow (BF) within the splanchnic region contributing to nutrient absorption, insulin secretion and glucose disposal, but factors regulating this phenomenon in humans are poorly known. The aim of the present study was to evaluate the organ-specific changes in BF during a mixed-meal and incretin infusions. DESIGN: A non-randomized intervention study of 10 healthy adults to study splanchnic BF regulation was performed. METHODS: Effects of glucose-dependent insulinotrophic polypeptide (GIP) and glucagon-like peptide 1 (GLP-1) infu

WNT/beta-catenin increases the production of incretins by entero-endocrine cells.

AIMS/HYPOTHESIS: Glucose-dependent insulinotropic peptide (GIP) plays a pivotal role in the regulation of glucose homeostasis. Rates of diet-induced obesity, insulin resistance and type 2 diabetes are decreased when GIP signalling is disturbed in mice, suggesting that GIP plays a role in the onset of type 2 diabetes. WNT signalling is linked to type 2 diabetes and induces synthesis of the other incretin, glucagon-like peptide 1 (GLP-1). GLP-1 analogues improve treatment of type 2 diabetes patients in whom GLP-1 signalling is intact and have captured clinical attention. GIP levels are altered a

Impaired beta cell sensitivity to incretins in type 2 diabetes is insufficiently compensated by higher incretin response.

BACKGROUND AND AIMS: The incretin effect is impaired in type 2 diabetes (T2D), but the underlying mechanisms are only partially understood. We investigated the relationships between the time course of the incretin effect and that of glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) during oral glucose tolerance tests (OGTTs), thereby estimating incretin sensitivity of the beta cell, and its associated factors. METHODS AND RESULTS: Eight patients with T2D and eight matched subjects with normal glucose tolerance (NGT) received 25, 75, and 125 g OGTTs and cor

Recent advances in incretin-based therapy for MASLD: from single to dual or triple incretin receptor agonists.

Clinically effective pharmacological treatment(s) for metabolic dysfunction-associated steatotic liver disease (MASLD) and its progressive form metabolic dysfunction-associated steatohepatitis (MASH) represent a largely unmet need in medicine. Since glucagon-like peptide-1 receptor agonists (GLP-1RAs) have been licensed for the treatment of type 2 diabetes mellitus and obesity, they were one of the first drug classes to be examined in individuals with MASLD/MASH. Successful phase 2 randomised clinical trials with these agents have resulted in progression to phase 3 clinical trials (principally

New Horizons for Incretin-Based Therapeutics.

Glucagon-like peptide-1 (GLP-1)-based therapies marked a major breakthrough in obesity pharmacotherapy, while subsequent advancements targeting multiple entero-pancreatic hormones now deliver weight loss previously achievable only with bariatric surgery, alongside improvements in obesity-related complications. Despite this progress, significant challenges persist, including reduced efficacy in diabetic populations, gastrointestinal adverse effects, and high costs that drive a real-world adherence gap. Consequently, next-generation therapies, including receptor polyagonists, oral formulations,
