Detection of apoptosis in human brainstem by TUNEL assay.

The aim of the present study was to evaluate the distribution of apoptotic neurons in the nuclei of the brainstem. Apoptosis was studied by the terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) assay, in human brainstems taken at autopsy from 15 adults (age range: 25-58 years). The nuclei examined included the hypoglossal nucleus (XII), dorsal motor nucleus of the vagus (DMNV), solitary tract nucleus (STN), vestibular nucleus (Ve), cuneate nucleus (Cu), nucleus of the spinal trigeminal tract (NSTT), principal inferior olivary nucleus (PION), medial inferior olivary

Effects of neonatal conductive hearing loss on brain stem auditory nuclei.

Both postnatal auditory deprivation and experimentally produced conductive hearing losses in mice result in incomplete maturation of most brain stem auditory neurons. The affected groups are: octopus cell, globular cell, small spherical cell, and large spherical cell groups in ventral cochlear nuclei; and the lateral superior olive and medial nucleus of the trapezoid body of the superior olivary complex. When 45 days of auditory deprivation are followed by 45 days of normal acoustic stimulation, there is incomplete maturation of neurons in: multipolar cell, globular cell, small spherical cell,

Medial accessory olivary nucleus asymmetry and its selective degeneration in Parkinson's disease: A human neuropathological study.

OBJECTIVE: The medial accessory olivary nucleus (MAO), a distinct but inconsistently present subnucleus of the human inferior olivary complex, is poorly characterized despite its proposed role in cerebellar motor circuitry. We aimed to investigate its structural asymmetry, functional correlates, and pathological vulnerability in humans. MATERIALS AND METHODS: We performed high-resolution histological analysis (Nissl and immunohistochemistry) in 35 neurologically normal brains to quantify MAO volume and neuronal numerical density and relate these to handedness and lifetime manual dexterity. Noise-induced degeneration in the brain and representation of inner and outer hair cells.

Noise that destroys receptor cells in the chinchilla cochlea also results in degeneration of axonal endings in the brain from the cochlear nerve fibers and the auditory pathways ascending to the superior olivary complex and inferior colliculus. The patterns of degeneration provide experimental evidence for differential representation of inner and outer hair cells and their functions.

[Anatomy of the central auditory pathway. Demonstration with horseradish peroxidase in the guinea pig].

The neurones of the afferent and efferent auditory pathways were labelled by axonally transported horseradish peroxidase. The first neurone of the ascending system is the acoustic nerve with the spiral ganglion. These fibres terminate mainly in the ventral cochlear nucleus (VCN). The neurones of the latter chiefly run to the contralateral superior olivary complex (SOC), whereas the neurones of the dorsal cochlear nucleus (DCN) terminate mainly in the central nucleus of the contralateral inferior colliculus (IC). The nerve cells of the superior olivary complex and nuclei of the lateral lemniscu

Inferior olive lesions in autoimmune glial fibrillary acidic protein astrocytopathy: a report of two cases.

BACKGROUND: Autoimmune glial fibrillary acidic protein (GFAP) astrocytopathy is a newly identified inflammatory nervous system disorder characterized by Immunoglobulin G autoantibodies against GFAP. While several MRI features are considered typical for autoimmune GFAP astrocytopathy, reports of lesions affecting the inferior olivary nucleus (ION) are rare. In this context, we present two cases of autoimmune GFAP astrocytopathy with lesions in the ION. CASE PRESENTATION: Case 1 involved a 64-year-old man with meningoencephalomyelitis, peripheral neuropathy, and autonomic dysfunction. His brain

Problems in interpreting abnormal auditory brainstem responses in comatose patients.

Auditory brainstem responses (ABRs) provide a way to evaluate central nervous system function in young, confused, uncooperative or comatose patients. Auditory brainstem responses represent activity in or around the vestibulocochlear (VIII) cranial nerve, cochlear nucleus, superior olivary complex, lateral lemniscus, and inferior colliculus. Many factors affect recording of the auditory brainstem response. These factors include technical factors, chronic alcoholism, demyelinating diseases, ototoxic drugs, barbiturates, hearing loss, otitis media, and hypothermia.

Malformational complex in an infant with intrauterine influenza viral infection.

An infant with complex malformations of the central nervous system is described. The abnormalities included ependymal damage, aqueduct forking, hydrocephalus, and neuronal heterotopias; agenesis of the cerebellum, pontine, and inferior olivary neutrons; agenesis of the optic and olfactory systems and corpus callosum; and a highly unusual course of the corticospinal fibers in the spinal cord. Immunohistochemical test results showed the presence of influenza virus antigens in the brain. Possible pathogenetic mechanisms are presented.

The olivocerebellar projection to the uvula in the mouse.

The organization of the olivocerebellar projection in the mouse was studied with the use of microinjections of horseradish peroxidase (HRP) or HRP conjugated to wheat germ agglutinin (WGA-HRP). Injections were made in medial, intermediate, and lateral sites along the width of the uvula. The purpose of this investigation was to determine the subnuclear origin of olivary afferents to different mediolateral regions of the uvula. Injections made in or adjacent to the midline of the uvula resulted in the retrograde labeling of cells, bilaterally in the caudal portion of the medial accessory olive (

Compensatory and Dynamic Cerebellar Responses to Striatal Lesions in Experimental Parkinsonism.

Parkinsonian symptoms such as tremors, rigidity, bradykinesia, and postural instability typically arise from basal ganglia dysfunction, but growing evidence suggests cerebellar circuits also play a key role. Here, we investigated multiunit activity (MUA) in the inferior olive (IO), dentate nucleus (DN), and Crus II of the cerebellum in a rat model of tract lesion-induced parkinsonism triggered by ventrolateral striatum (VLS) injury. Thirty-six male Wistar rats were divided into a Lesion group and an Intact group. Monopolar electrodes were implanted to record MUA in IO, DN, or Crus II for four

Inferior olive-selective amplitude suppression and progressive dentato-cortical decoupling during haloperidol-induced parkinsonism: a longitudinal multi-unit activity study.

Drug-induced parkinsonism (DIP) arises from competitive D2 receptor blockade, yet its cerebellar circuit-level dynamics remain poorly characterized. We recorded multi-unit activity simultaneously from four cerebellar structures (dentate nucleus, Crus II, inferior olive, and lobulus Sim B) across three weekly sessions in male Wistar rats receiving chronic haloperidol (1.5 mg/kg/day, subcutaneous) to characterize structure-specific and longitudinally evolving cerebellar population dynamics. Resting-state discharge amplitude was suppressed only in the inferior olive after FDR correction, where al

Dopaminergic projections of the subparafascicular thalamic nucleus to the auditory brainstem.

Neuromodulators can alter the response properties of sensory neurons, including those in the auditory system. Dopamine, which plays a major role in reward and movement, has been shown to alter neural responses in the auditory brainstem and midbrain. Recently we identified the subparafascicular thalamic nucleus (SPF), part of the A11 dopaminergic cell group, as the source of dopamine to the inferior colliculus (IC). The superior olivary complex (SOC) is also a likely target of dopaminergic projections from the SPF because it receives projections from the SPF and contains fibers and terminals im

Central acoustic tract in an echolocating bat: an extralemniscal auditory pathway to the thalamus.

To determine the sources and targets of auditory pathways that bypass the inferior colliculus in the mustache bat, we injected WGA-HRP in the medial geniculate body and related auditory nuclei of the thalamus as well as in the lower brainstem. We used electrophysiological methods to verify that the injection electrode was in an area responsive to sound. The only thalamic injections that produced retrograde transport to cells in auditory nuclei caudal to the inferior colliculus were those that included the suprageniculate nucleus. These injections labeled a group of large multipolar cells lying

Conductive hearing loss results in changes in cytochrome oxidase activity in gerbil central auditory system.

Conductive hearing loss (CHL) restricts auditory input to an intact peripheral auditory system. Effects of deprivation on the central auditory system (CAS) have been debated, although a number of studies support the hypothesis that CHL can cause modification of CAS structure and function. The present study was designed to test the hypothesis that unilateral CHL results in a decrease in cytochrome oxidase (CO) activity in CAS nuclei that receive major afferent input from the affected ear. Gerbils at postnatal day 12 (P21) or 6-8 weeks underwent left unilateral CHL (malleus removal), cochlear ab

[An autopsy case of progressive supranuclear palsy with olivary hypertrophy].

Clinical and pathologic findings of an autopsy case of progressive supranuclear palsy (PSP) with a 7 year clinical course are described. The patient exhibited clinical findings of typical PSP, cerebellar signs and rhythmical myoclonus that was about 2 Hz and synchronous in the eyes, palate, and pharynx, which is so called palatal myoclonus. Pathological findings compatible with those in PSP i.e. loss of nerve cells, neurofibrillary tangles (NFT), and gliosis were found in the globus pallidum, thalamus, subthalamic nucleus, substantia nigra, locus coeruleus, nucleus of Raphe, reticular formatio

Autoradiographic localization of calcitonin gene-related peptide binding sites in human and rat brains.

125I-calcitonin gene-related peptide (CGRP) binding sites were mapped in the human brain and rat brains by in vitro macroautoradiography, and compared to each other. Binding experiments were made to characterize 125I-CGRP binding on the human and rat brains. Scatchard analysis of saturation experiments from slide-mounted sections of the human and rat cerebellum displayed 125I-CGRP binding sites with a dissociation constant (Kd) of 0.17 nM and 0.11 nM, respectively, and a maximal number of binding sites (Bmax) of 96.8 fmol/mg and 23.0 fmol/mg protein. 125I-CGRP binding was time-dependent, rever

Origins of the Auditory Brainstem Response in Mice Using Source Localization of Topographic Multichannel EEG.

The auditory brainstem response (ABR) is a critical tool for assessing auditory brainstem function in biomedical mouse models. Remarkably, despite its importance, the origins of ABR waves specific to mice remain poorly identified. Here, we used EEG source reconstruction to reevaluate the mouse-specific ABR origins. We recorded the topography of ABRs using high-density EEG from the skull of adult mice of either sex combined with parallel multielectrode recordings in the auditory cortex. Individual ABR waves showed a series of distinct spatial topographies across the skull. Wave I' was strongly

MR imaging of the excised human brainstem: a correlative neuroanatomic study.

The purpose of this study was to examine an excised human brainstem with high-resolution MR imaging. The MR images were correlated with myelin-stained histologic specimens and standard anatomic descriptions. Structures visualized included nuclei and/or fibers of all the cranial nerves, with the exception of the trochlear nerve. The topography of the brainstem is discussed. Major tracts such as the corticospinal spinothalamic, spinocerebellar, medial and lateral lemniscus, and other ascending and descending tracts and intrinsic brainstem nuclei such as the olivary complex, pontine nuclei, red n

Far-field acoustic response: origins in the cat.

Short-latency evoked potentials recorded from the vertex of adult cats in response to click stimulation (the far-field acoustic response) were analyzed in a series of lesion experiments to determine the origins of each component. The resultant data indicate that the primary generator of potential is the acoustic nerve; of potential 2, the cochlear nucleus; of potential 3, neurons of the superior olivary complex activated by projections crossing the midline; of potential 4, neurons of the ventral nucleus of the lateral lemniscus and preolivary region activated equally by crossed and uncrossed p
