Isocitrate dehydrogenase 3b is required for spermiogenesis but dispensable for retinal viability.

Isocitrate dehydrogenase 3 (IDH3) is a key enzyme in the mitochondrial tricarboxylic acid (TCA) cycle, which catalyzes the decarboxylation of isocitrate into α-ketoglutarate and concurrently converts NAD+ into NADH. Dysfunction of IDH3B, the β subunit of IDH3, has been previously correlated with retinal degeneration and male infertility in humans, but tissue-specific effects of IDH3 dysfunction are unclear. Here, we generated Idh3b-KO mice and found that IDH3B is essential for IDH3 activity in multiple tissues. We determined that loss of Idh3b in mice causes substantial accumulation of isocitr

Substituted N-phenylitaconamides as inhibitors of mycobacteria and mycobacterial isocitrate lyase.

Novel antimycobacterial drugs are needed, especially those with dual activity against both actively growing and non-replicating subpopulations of mycobacteria. Isocitrate lyase (ICL) is one of proposed targets and this enzyme is inhibited by itaconic acid. That is why we have designed and prepared sixteen amides of itaconic acid and various anilines and amine antimicrobial drugs to evaluate them as potential inhibitors of ICL and antimycobacterial agents. N-Phenylitaconamides were prepared from itaconic anhydride and substituted anilines (yields 57-99%). They were characterized and evaluated a

Adult isocitrate dehydrogenase-mutant brainstem glioma: illustrative case.

BACKGROUND: Adult brainstem gliomas are rare entities that demonstrate heterogeneous biology and appear to be distinct from both their pediatric counterparts and adult supratentorial gliomas. Although the role of histone 3 mutations is being increasingly understood in this disease, the effect of isocitrate dehydrogenase (IDH) mutations remains unclear, largely because of limited data. OBSERVATIONS: The authors present the case of a 29-year-old male with an IDH1-mutant, World Health Organization grade III anaplastic astrocytoma in the dorsal medulla, and they provide a review of the available

Oncogenic isocitrate dehydrogenase mutations: mechanisms, models, and clinical opportunities.

Heterozygous mutations in catalytic arginine residues of isocitrate dehydrogenases 1 and 2 (IDH1 and IDH2) are common in glioma, acute myeloid leukemia, chondrosarcoma, cholangiocarcinoma, and angioimmunoblastic T-cell lymphoma. The mutant enzymes acquire a neomorphic activity that converts α-ketoglutarate (α-KG) to D-2-hydroxyglutarate (D2HG), a rare metabolite. In cells and tissues expressing mutant IDH, D2HG concentrations are highly elevated. D2HG may act as an "oncometabolite" by inhibiting a class of α-KG-dependent enzymes involved in epigenetic regulation, collagen synthesis, and cell s

Isocitrate Dehydrogenase Mutations in Cancer: From Bench to Bedside Applications.

Isocitrate dehydrogenase (IDH) mutations represent pivotal oncogenic drivers across multiple malignancies. Mutant IDH enzymes acquire neomorphic activity that produces the oncometabolite d-2-hydroxyglutarate (D-2HG), which competitively inhibits α-ketoglutarate-dependent dioxygenases and promotes epigenetic reprogramming, differentiation arrest, and malignant transformation. Beyond tumor cell-intrinsic effects, D-2HG profoundly remodels the tumor immune microenvironment by directly suppressing T-cell proliferation and effector functions, silencing natural killer (NK) cell-activating ligands, a

Molecular features of intratumoral hemorrhage in glioblastoma isocitrate dehydrogenase-wildtype.

BACKGROUND: Glioblastoma isocitrate dehydrogenase (IDH)-wildtype (GBM) has been reclassified based on molecular and phenotypic features. Intratumoral hemorrhage is a phenotypic subtype with poorly understood molecular and clinical characteristics. We aimed to characterize the molecular profile and outcomes of hemorrhagic glioblastoma (hGBM) compared with non-hGBM. METHODS: A retrospective analysis of GBMs with pre-operative and post-operative MRI and comprehensive next-generation sequencing of 205 genes was performed. Patients were classified as hGBM or non-hGBM using the Visually Accessible

Isocitrate Dehydrogenase-Mutated Cholangiocarcinoma: Natural History and Clinical Outcomes.

PURPOSE: Clinical-pathologic features and natural history of patients with isocitrate dehydrogenase (IDH)-mutant intrahepatic cholangiocarcinoma (CCA) are not well characterized. Here, we sought to describe the natural history, clinical phenotype, and prognostic impact of advanced, IDH-mutated CCA. METHODS: We conducted a multicentric, retrospective analysis of patients with IDH-mutated (IDH1 or IDH2) CCA between 2010 and 2020. Median overall survival (OS) and progression-free survival (PFS) analyses were performed using the Kaplan-Meier method. Chi-square test was used to analyze disease con

The biology and management of cartilaginous tumors: a role for targeting isocitrate dehydrogenase.

Chondrosarcomas are rare mesenchymal neoplasms defined by the production of abnormal cartilaginous matrix. Conventional chondrosarcoma is the most common histology. The management of primary conventional chondrosarcoma generally is surgical with the possible addition of radiation therapy. Treatment of conventional chondrosarcoma is problematic in unresectable or metastatic disease because the tumors tend to be resistant to standard sarcoma chemotherapy regimens. Previous attempts at targeted therapy, including inhibitors of Hedgehog signaling, the mTOR pathway, and platelet-derived growth fact

Isocitrate dehydrogenase 1 R132H mutation is not detected in angiocentric glioma.

Mutations of isocitrate dehydrogenase-1 gene (IDH1), most commonly resulting in replacement of arginine at position 132 by histidine (R132H), have been described in World Health Organization grade II and III diffuse gliomas and secondary glioblastoma. Immunohistochemistry using a mouse monoclonal antibody has a high specificity and sensitivity for detecting IDH1 R132H mutant protein in sections from formalin-fixed, paraffin-embedded tissue. Angiocentric glioma (AG), a unique neoplasm with mixed phenotypic features of diffuse glioma and ependymoma, has recently been codified as a grade I neopla

Alkylation of isocitrate lyase from Escherichia coli by 3-bromopyruvate.

The inactivation of tetrameric isocitrate lyase from Escherichia coli by 3-bromopyruvate, exhibiting saturation kinetics, is accompanied by the loss of one sulfhydryl per subunit. The substrates glyoxylate and isocitrate protect against inactivation whereas the substrate succinate does not. The modification by 3-bromopyruvate (equimolar to subunits) imparts striking resistance to digestion of isocitrate lyase by trypsin, chymotrypsin, and V8 protease as well as a major decrease in the intensity of tryptophan fluorescence. After alkylation, the sequence Gly-His-Met-Gly-Gly-Lys is found followin

The diagnostic usefulness of isocitrate dehydrogenase (ICDH) in the marmoset (Callithrix jacchus).

Marmosets were given either a hepatotoxin, carbon tetrachloride, orally or an i.m. injection of a mytoxin, chlorpromazine. Although muscle damage alone caused small increases in the plasma levels of lactate dehydrogenase, alanine aminotransferase, aspartate aminotransferase and isocitrate dehydrogenase (ICDH), only the isoenzyme analysis of ICDH can differentiate definitely between liver and muscle damage. Only very severe muscle damage can increase the plasma concentration of this enzyme but, in this case, the elevation of plasma creatinine kinase levels helps differentiation. It is recommend

Computational Investigation of Phytochemicals Targeting Isocitrate Lyase to Inhibit Mycobacterium tuberculosis.

INTRODUCTION: The global burden of tuberculosis (TB) remains a major concern for society that is worsening day by day with the emergence of drug-resistant TB as well as risks associated with latent TB. Isocitrate lyase (ICL) has been shown as a potential target that plays a role in the latent/ dormant stage of M. tuberculosis. Several inhibitors against ICL have been designed and tested, which have various side effects. METHODOLOGY: This study focuses on the phytochemicals from plant extracts, which have anti-tubercular properties. A total of 1413 phytochemicals were virtually screened agains

A Case of Multicentric Isocitrate Dehydrogenase-Wildtype Glioblastoma With Divergent Clonal Evolution.

Here, we present a rare case of multicentric glioblastoma in which two spatially distinct tumors exhibited different molecular profiles. A 56-year-old woman presented with progressive headache and speech difficulties. Magnetic resonance imaging (MRI) of the brain revealed two separate contrast-enhancing lesions, one in the left superior temporal gyrus and the other in the right middle temporal gyrus, with no connecting tract or edema on fluid-attenuated inversion recovery (FLAIR) imaging. Multicentric glioblastoma was suspected, and the patient underwent two-stage surgical resection of both tu

Methylation Profiling Identifies Stability of Isocitrate Dehydrogenase Mutation Over Time.

OBJECTIVE: Isocitrate dehydrogenase (IDH) mutation status is a key diagnostic and prognostic feature of gliomas. It is thought to occur early in glioma tumorigenesis and remain stable over time. However, there are reports documenting a loss of IDH mutation status in a subset of patients with glioma recurrence. Here, we identified patients with a documented loss of IDH mutation status longitudinally and performed multi-platform analysis in order to determine if IDH mutations are stable throughout glioma evolution. METHODS: We retrospectively identified patients from our institution from 2009 t

Interaction of Isocitrate Lyase with Proteins Involved in the Energetic Metabolism in Paracoccidioides lutzii.

BACKGROUND: Systemic mycosis is a cause of death of immunocompromised subjects. The treatment directed to evade fungal pathogens shows severe limitations, such as time of drug exposure and side effects. The paracoccidioidomycosis (PCM) treatment depends on the severity of the infection and may last from months to years. METHODS: To analyze the main interactions of Paracoccidioides lutzii isocitrate lyase (ICL) regarding the energetic metabolism through affinity chromatography, we performed blue native PAGE and co-immunoprecipitation to identify ICL interactions. We also performed in silico an

Structural basis of allosteric activation of Mycobacterium tuberculosis isocitrate lyase 2.

Mycobacterium tuberculosis isocitrate lyase 2 (ICL2) is an allosterically regulated enzyme required for growth on non-glycolytic carbon substrates during infection. Although acetyl-CoA and its analogues are known to activate ICL2, the molecular basis of this regulation has remained unclear. Here, we combine protein NMR, crystallography, molecular dynamics, and mutagenesis to show that two structural features unique to ICL2, the C-terminal domain and a helical substructure in the N-terminal catalytic domain, govern its allostery. Acetyl-CoA binding promotes dimerisation of the C-terminal domain

Noninvasive Identification of Isocitrate Dehydrogenase 1/2 Mutation in Brainstem Gliomas.

PURPOSE: A subset of brainstem gliomas harbor an isocitrate dehydrogenase 1/2 (IDH1/2) mutation, which has important prognostic and treatment implications. We evaluated the radiographic features and the sensitivity of magnetic resonance spectroscopy (MRS) and cerebrospinal fluid cell-free DNA (CSF cfDNA) sequencing at detecting IDH1/2 mutations in a cohort of these tumors. METHODS: We identified IDH-mutant brainstem gliomas by retrospective chart review. IDH1/2 mutation was established by biopsy, CSF cfDNA sequencing, and/or the presence of a 2-hydroxyglutarate (2HG) peak by 3-Tesla MRS. RES

The roles of NADPH and isocitrate dehydrogenase in cochlear mitochondrial antioxidant defense and aging.

Hearing loss is the third most prevalent chronic health condition affecting older adults. Age-related hearing loss affects one in three adults over 65 years of age and is caused by both extrinsic and intrinsic factors, including genetics, aging, and exposure to noise and toxins. All cells possess antioxidant defense systems that play an important role in protecting cells against these factors. Reduced nicotinamide adenine dinucleotide phosphate (NADPH) serves as a co-factor for antioxidant enzymes such as glutathione reductase and thioredoxin reductase and is produced by glucose-6-phosphate de
