Causes of fatigue in slow-twitch rat skeletal muscle during dynamic activity.

Skeletal muscle fatigue is most often studied in vitro at room temperature and is classically defined as a decline in maximum force production or power output, exclusively linked to repeated isometric contractions. However, most muscles shorten during normal use, and we propose that both the functional correlate of fatigue, as well as the fatigue mechanism, will be different during dynamic contractions compared with static contractions. Under isoflurane anesthesia, fatigue was induced in rat soleus muscles in situ by isotonic shortening contractions at 37 degrees C. Muscles were stimulated rep

Calcaneal tendon stiffness is not associated with dynamic time-dependent contractile output.

The ability to rapidly generate muscular torque and velocity is important in specialized activities and daily tasks of living. Tendon stiffness is one factor in the neuromuscular system that influences musculoskeletal torque transmission. Previous studies have reported weak-to-moderate correlations between tendon stiffness and rate of torque development (RTD). However, these correlations have been reported only for isometric contractions, which may not be relevant to contractions involving joint rotation (i.e., dynamic). The purpose was to investigate the effect of calcaneal tendon stiffness o

Passive thigh heating improves isokinetic but not isotonic muscle function.

PURPOSE: Optimal skeletal muscle function occurs when tissue temperature is elevated above resting. This study examined muscle torque production and maximal velocity responses to passive thigh heating on two occasions to determine the efficacy and reliability of the intervention. METHODS: Twenty active, young participants (10 female) completed two identical visits whereby one thigh was wrapped in a water perfused garment circulating 50 °C water for 90 min; with the contralateral limb remaining unheated. Four maximal isokinetic repetitions were conducted at three speeds (slow, 60°/s; moderate,

Movement Evoked Pain and Mechanical Hyperalgesia after Intramuscular Injection of Nerve Growth Factor: A Model of Sustained Elbow Pain.

OBJECTIVE: Lateral epicondylalgia presents as lateral elbow pain provoked by upper limb tasks. An experimental model of elbow pain provoked by movement/muscle contraction and maintained over several days is required to better understand the mechanisms underlying sustained elbow pain. This study investigated the time course and pain location induced by nerve growth factor (NGF) injection into a wrist extensor muscle, and whether movement and muscle contraction/stretch provoked pain. METHODS: On Day 0 twenty-six painfree volunteers were injected with NGF (N = 13) or isotonic saline (randomized)

Sustained skeletal muscle power for cardiac assist devices: implications of metabolic constraints.

A device to harness power from skeletal muscle contracting in a linear configuration is under development. This application requires a sustained level of power that is dependent upon muscle mechanics and metabolic properties. A biomechanical muscle model and a metabolic model constructed from experimental data were used to predict maximum power available in a sustainable region of loading and stimulation conditions. Latissimus dorsi (LD) of four goats were evaluated in vivo after a 10 week in situ conditioning protocol with an implanted Telectronics myostimulator. The LD insertion was reconnec

Post-transcriptional regulation of alpha-smooth muscle actin determines the contractile phenotype of Dupuytren's nodular cells.

The objective was to study Dupuytren's myofibroblast cells in constrained collagen matrices in order to more closely emulate their in vivo environment and, to correlate their contractility with alpha-smooth muscle actin (alpha-SMA) expression and determine if dermal fibroblasts regulate Dupuytren's myofibroblast phenotype. Isotonic and isometric force contraction by cells isolated from Dupuytren's nodules, palmar and non-palmar skin fibroblasts was measured in collagen matrices. The effect of co-culturing nodule cells with dermal fibroblasts on isometric contraction was examined. Isometric con

Evaluation of the torque-velocity test of the BTE-Primus as a measure of sincerity of effort of grip strength.

An inverse linear relationship exists between torque and velocity in the mid-ranges of an isotonic maximal contraction in a single joint movement (such as the elbow and knee). We hypothesized that submaximal effort does not produce a linear torque-velocity relationship because replicating a submaximal isotonic contraction requires an enormous amount of proprioceptive feedback and the nervous system may not be able to accurately replicate both force and speed of contraction. If this hypothesis is true, the torque-velocity test of the BTE-Primus would be an effective method for assessing sinceri

Performance fatigability on a constant-load dynamic task is greater in females with moderate-severity Parkinson's disease than healthy-aging controls.

Parkinson's disease (PD) is a neurodegenerative disorder associated with self-reported fatigue. While fatigue is a disabling symptom, performance fatigability, a decline in strength or power during sustained tasks, remains poorly understood, particularly during isotonic (constant-load) contractions, which are encountered in everyday activities. We assessed performance fatigability and recovery in people with PD compared to healthy-aging controls. Fifteen females with PD and 14 healthy-aging controls underwent neuromuscular testing of knee extensors using dynamometry. Participants then performe

Three-dimensional imaging of the stomach: role of pylorus in the emptying of liquids.

Using the dynamic spatial reconstructor and manometry, we measured gastric emptying, pyloric size, and antral contractile activity in three anesthetized dogs in the prone position. Our objective is to evaluate the effect of intestinal perfusion of equicaloric, neutral, and isosmolar nutrients (Maltose 32.5 mg/ml, casein hydrolysate 32.5 mg/ml, and oleic acid 15.5 mg/ml) at a constant rate of 10 ml/min in the proximal jejunum on the dynamics of the antropyloric region. Isotonic saline perfusion was used as a control solution. Test meal in the stomach was 600 ml of isotonic Gastrograffin. Antral

[The level of cyclic nucleotides during spontaneous smooth-muscle contractions in the guinea pig].

The relationships between isotonic tension and changes in cAMP and cGMP levels was studied in isolated preparations of the guinea-pig taenia coli and myometrium during spontaneous contractions. No significant changes in cyclic nucleotide levels were detected at any stage of the spontaneous contraction-relaxation cycle. The finding suggests that these changes play no significant role in the regulation of spontaneous contractions of the taenia coli and myometrium. However, other data obtained suggests the possibility of combined actions of the cGMP and phosphatidylinositols in control of smooth

[Immediate effects of therapeutic interventions on the central motor system and their practical consequences].

Physiotherapy and occupational therapy have changed over recent years influenced by our growing neurophysiological knowledge especially about motor learning. Understanding the direct effects of therapeutic interventions on the central motor system is crucial for further development of therapeutic approaches. Recent studies are discussed dealing with the facilitatory effects of different therapeutic interventions and the question whether or not the activation of the healthy upper limb has a facilitatory or inhibitory net-effect on the damaged central motor system. [Temperature dependence of bronchial and tracheal reactivity of isolated respiratory tract preparations of the guinea pig].

The intrapulmonary loss of heat due to hyperventilation may be considered as essential pathogenetic factor in producing exercise induced asthmatic response. The temperature dependence of smooth muscle contraction was investigated using isolated trachea and isolated lung strips of guinea pigs. The measurements of the contraction responses to cumulative dose administrations of acetylcholine (10(-9) to 10(-3) M) were made in thermostated organ baths (isotonic contraction, modified Krebs-Henseleit solution, pH 7.4, air gassing). The standardized measurement data were adjusted to a non-linear funct

Sensitivity of the airway smooth muscle in terms of force, shortening and stiffness.

Eight pig tracheal strips were stimulated to contract with log increments of methacholine from 10-8 to 10-5 M. For each strip, the concentration-response was repeated four times in a randomized order to measure isometric force, isotonic shortening against a load corresponding to either 5 or 10 % of a reference force, and average force, stiffness, elastance and resistance over one cycle while the strip length was oscillating sinusoidally by 5 % at 0.2 Hz. For each readout, the logEC50 was calculated and compared. Isotonic shortening with a 5 % load had the lowest logEC50 (-7.13), yielding a gre

Inotropic and lusitropic effects of chlorpromazine on rat left ventricular papillary muscle.

The in vitro effects of chlorpromazine on rat cardiac papillary muscle were tested at 10(-6), 10(-5) and 10(-4) M. Mechanical parameters were determined from the contraction and relaxation phases under isotonic and isometric conditions in order to assess contraction, relaxation, contraction-relaxation coupling and load sensitivity of relaxation. The peak power output Emax was determined from the force-velocity relationship. At 10(-6) M, a slight positive inotropic effect was observed, probably related to modifications in cross-bridges kinetics. Negative inotropic effects were observed with 10(

Low potency of Ca antagonists in smooth muscle from different levels of the respiratory tract.

Respiratory smooth muscle at different levels of the respiratory tract was isolated from rabbit, rat and guinea-pig and incubated with the Ca antagonists, verapamil, D-600, felodipine, and nifedipine. There was no effect of the Ca antagonists when added on top of an already developed contraction. When added before the contractile agent at high concentrations (10-100 microM) the Ca antagonists reduced the contractions. Lanthanum (2 mM) was considerably more effective to prevent contractions than the Ca antagonists. Preparations from the trachea and bronchus responded in all animals, whereas the

Aminophylline increases submaximum power but not intrinsic velocity of shortening of frog muscle.

We hypothesized that methylxanthines, such as aminophylline, increase the power developed by submaximally activated frog skeletal muscles by increasing the force developed at any given velocity of shortening. Frog semitendinosus muscles were excised and tested at 20 degrees C in oxygenated control and aminophylline Ringer solutions. Force-velocity relationships were determined and power was calculated from muscles stimulated at frequencies of 80 and 300 Hz. The 300-Hz frequency of stimulation produced a maximum rate of force development. In 50 and 500 microM aminophylline, twitch force increas

Magnetic Resonance Compatible Knee Extension Ergometer.

A magnetic resonance (MR) compatible ergometer has been developed to study contracting lower limb muscles during acquisition of MR spectroscopy data, a technique to noninvasively measure metabolic energy in muscle tissue. Current active and passive MR-compatible ergometer designs lack torque or velocity control to allow precise mechanical measurements during isotonic and isokinetic contractions; incorporating load and velocity controllers while maintaining MR-compatibility is the main challenge. Presented in this paper is the design and evaluation of an MR-compatible ergometer designed to cont
