Modification of drug-induced tremor by systemic administration of kainic acid and quisqualic acid in mice.

The effects of excitatory amino acids, kainic acid and quisqualic acid, on the tremorine- and harmaline-induced tremor were quantitatively examined in mice using the power spectral analyzing method. The severity of the tremor was determined quantitatively in terms of the cumulative sum of the mean square value of the data. Kainic acid enhanced the tremor induced by tremorine but depressed the tremor induced by harmaline. Quisqualic acid depressed the tremor induced by both tremorine and harmaline in a dose-dependent manner. Kainic acid shifted the frequency of each component of the tremor indu

Cx47 Phosphorylation Exacerbates White Matter Damage and Kainic Acid Induced Epilepsy.

AIMS: Growing evidence implicates dysfunctional myelin in the pathogenesis of temporal lobe epilepsy (TLE). Connexin 47 (Cx47), an oligodendrocytic gap junction protein, maintains myelin integrity. This study investigates the role of Cx47 in myelin impairment and seizure progression in TLE. METHODS: Cx47 and phosphorylated Cx47 (p-Cx47) expression was analyzed in human and mouse TLE brain tissues via Western blot and immunofluorescence. Candidate Cx47 phosphorylation kinases revealed by single-cell RNA sequencing were validated through immunofluorescence, protein docking, and co-immunoprecipi

Kainic acid induction of mossy fiber sprouting: dependence on mouse strain.

After seizures caused by kindling or kainic acid (KA), hippocampal granule-cell axons, the mossy fibers, sprout into the supragranular layer of the rat. The mechanisms underlying this phenomenon remain elusive, but excitotoxic loss of hilar cells, which project to this supragranular layer, is suspected to be a critical determinant. Consistent with this hypothesis, we previously reported that while rats show mossy fiber sprouting after kainate, ICR mice do not. This may be associated with the observation that ICR mice, unlike rats, do not appear to show hilar cell death after KA (McNamara et al

Phenobarbital pre-treatment prevents kainic acid-induced impairments in acquisition learning.

This study examined the protective effect of phenobarbital on kainic acid-induced deficits in acquisition learning. A single kainic acid injection (9 mg/kg i.p.) was administered five days prior to testing using the Morris water maze test. Kainic acid produced deficits in the acquisition of spatial information observed as an increase in latency to a hidden escape platform. Daily phenobarbital treatment (20 mg/kg i.p.) initiated 45 minutes prior to the kainic acid injection blocked the kainic acid-induced deficits in acquisition learning. When daily phenobarbital treatment was initiated 2-3 hou

Decreased severity of ethanol withdrawal behaviors in kainic acid-treated rats.

The involvement of kainate (KA)-sensitive regions in ethanol withdrawal behaviors was investigated in male Wistar rats given three intraperitoneal (IP) injections of KA (12 mg/kg) or saline each followed by recovery at 4 degrees C for 5 h and room temperature for 3 days and a final KA or saline injection at room temperature. Some animals received MK-801 (1 mg/kg, IP) 30 min after each injection and one group received saline only. The saline/saline, saline/MK-801, and KA/MK-801 groups displayed typical ethanol withdrawal behaviors 8-12 h after ethanol withdrawal. These behaviors were attenuated

Increased susceptibility to hippocampal and amygdala kindling following intrahippocampal kainic acid.

The effects of unilateral intrahippocampal injection of kainic acid, a potent neuroexcitant and neurotoxin, on subsequent susceptibility to kindling of the contralateral hippocampus or contralateral amygdala were investigated in albino rats. At the chosen doses (0.20 to 1.25 micrograms dissolved in physiologic saline), the kainic acid-induced lesion was confined to the injected hippocampus and in two cases the ipsilateral entorhinal cortex; never were there contralateral lesions. Approximately 2 to 6 weeks post-injection, each animal received daily afterdischarge-producing electrical stimulati

Intravenous kainic acid induces status epilepticus and late onset seizures in mice.

We set out to establish a novel model of temporal lobe epilepsy (TLE) in a mouse. We sought to induce TLE through the injection of kainic acid (KA) into the tail vein with subsequent development of status epilepticus (SE). Using C57BL/6 mice, we implanted hippocampal EEG recording electrodes before or after injection of KA or phosphate buffered saline (PBS). Video and EEG analysis were conducted to evaluate for SE and development of recurrent seizures, the hallmark of TLE. All mice injected with KA developed SE while those who were injected with PBS did not. Of the animals injected with KA mon

Intracerebroventricular administration of histidine reduces kainic acid-induced convulsive seizures in mice.

Kainic acid (KA)-induced seizures and other experimental models of epilepsy have been proven to be instrumental in identifying novel targets that could be responsible for human icto- and epileptogenesis. We have previously shown that the ablation of pharmacoresistant voltage-gated Ca2+ channels with Cav2.3 as central ion-conducting pore (R-type Ca2+ channel) reduces the sensitivity towards KA-induced epilepsy in mice. In vivo, Cav2.3 channels are thought to be under tight allosteric control by endogenous loosely bound trace metal cations (Zn2+ and Cu2+) that suppress channel gating via a high-

Effect of baicalin on hippocampal damage in kainic acid-induced epileptic mice.

The aim of the present study was to determine the effect of baicalin on the expression of miR-497 and its target B-cell lymphoma-2 (Bcl-2) in the hippocampus of kainic acid (KA)-induced epileptic mice. To establish status epilepticus (SE), 0.1 µg/5 µl KA was injected into the lateral cerebral ventricle in mice, which then received an intraperitoneal injection of baicalin (100 mg/kg) after 1 and 8 h. Hematoxylin and eosin staining was used to observe the pathological changes in morphology and neuronal apoptosis was determined by terminal transferase-mediated dUTP nick end-labeling staining. Wes

High-fat diet-induced obesity exacerbates kainic acid-induced hippocampal cell death.

BACKGROUND: Obesity has deleterious effects on the brain, and metabolic dysfunction may exacerbate the outcomes of seizures and brain injuries. However, it is unclear whether obesity affects excitotoxicity-induced neuronal cell death. The purpose of this study was to investigate the effects of a high-fat diet (HFD) on neuroinflammation and oxidative stress in the hippocampus of kainic acid (KA)-treated mice. RESULTS: Mice were fed with a HFD or normal diet for 8 weeks and then received a systemic injection of KA. HFD-fed mice showed hypercholesterolemia, insulin resistance, and hepatic steato

Acacetin inhibits glutamate release and prevents kainic acid-induced neurotoxicity in rats.

An excessive release of glutamate is considered to be a molecular mechanism associated with several neurological diseases that causes neuronal damage. Therefore, searching for compounds that reduce glutamate neurotoxicity is necessary. In this study, the possibility that the natural flavone acacetin derived from the traditional Chinese medicine Clerodendrum inerme (L.) Gaertn is a neuroprotective agent was investigated. The effect of acacetin on endogenous glutamate release in rat hippocampal nerve terminals (synaptosomes) was also investigated. The results indicated that acacetin inhibited de

Involvement of nitric oxide in kainic acid-induced excitotoxicity in rat brain.

The involvement of nitric oxide (NO) in kainic acid (KA)-induced excitotoxicity was studied in rat brain. With the onset of KA (15 mg kg(-1), s.c.)-induced seizures (convulsions) 30 min after injection, increases in NO, as measured by the formation of citrulline, were seen in cortex (302%), amygdala (171%) and hippocampus (203%). The highest increases were determined 90 min after onset of seizures (120 min after KA injection) with 633%, 314% and 365%, respectively. These changes in NO preceded significant decreases in ATP and phosphocreatine (PCr) ranging from 44 to 53% for ATP and from 40 to

Saccadic eye movements following kainic acid lesions of the pulvinar in monkeys.

Behavioral and anatomical experiments have suggested that the pulvinar might play a role in the generation of saccadic eye movements to visual targets. To test this idea, we trained monkeys to make visually-guided saccades by requiring them to detect the dimming of a small target. We used three different saccade paradigms. On single-step trials, saccades were made from a central fixation point (FP) to a target at 12, 24 or 36 degrees to the left or right. On overlap trials, the FP remained lit during presentation of a target at 12 or 24 degrees. On double-step trials, the target stepped first

Recovery of spatial alternation deficits following selective hippocampal destruction with kainic acid.

Elimination of hippocampal CA3 and CA4 pyramidal cells with kainic acid (KA) induces a performance deficit on a forced-choice alternation task, which recovers over time. In order to determine whether superior cervical ganglion (SCG) fibers sprouting into the hippocampus contribute to behavioral recovery, 31 Sprague-Dawley rats were first trained on a forced-choice task. After reaching criterion performance levels, animals received either KA or saline injections into the hippocampus and were again tested on the forced-choice task. Half of the rats had their SCG removed 35 days after injections,

Neuroprotective Effects of Pinostrobin in a Rat Model of Kainic Acid-Induced Epilepsy.

Epilepsy is a serious neurological disorder characterized by recurrent seizures and neuronal loss. Prolonged seizure activity is associated with changes in apoptosis-related proteins, including Bcl-2-associated X protein (Bax) and caspase-3, as well as alterations in microglial and astroglial responses. This study investigated the neuroprotective effects of pinostrobin, a bioflavonoid, in a kainic acid (KA)-induced epilepsy model and examined its effects on apoptosis-related proteins. Pinostrobin (40 mg/kg) was administered orally once daily for 7 days before and 14 days after intracerebrovent

Sex differences in response to kainic acid and estradiol in the hippocampus of newborn rats.

Premature and full-term human infants are at considerable risk of excitotoxic-mediated brain damage due to hypoxia-ischemia, infection or other trauma. Glutamate receptor activation is a major source of excitoxicity in the adult and developing brain, and the hippocampus is particularly vulnerable to damage. The seven-day-old rat is a widely used model of pediatric brain damage, in large part due to the relative insensitivity of the brain to exogenous glutamate treatment prior to this age. We have reexamined the possible role of glutamate in pediatric brain damage in the newborn rat using kaini

Effects of kainic acid on hypothermia induced by reserpine, oxotremorine and apomorphine in mice.

Antagonists of the norepinephrine reuptake and beta-adrenoreceptor agonists are potent, at once, on the three following tests: antagonism of hypothermia induced by reserpine, oxotremorine and apomorphine. 2-Carboxy-4-isopropenyl-3-pyrrolidine-acetic acid (kainic acid), which is a powerful stimulant of the neurons and a destroyer of the dopaminergic neurons, has been used in these tests to show if it is possible to antagonize hypothermia induced by different substances. The results obtained show that kainic acid is potent on these three tests, thus providing evidence that it is a stimulant of n

Kainic acid seizures and the reversibility of calcium loading in vulnerable neurons in the hippocampus.

The threshold pathological changes in the rat hippocampus following systemic administration of kainic acid (12-15 mg/kg) have been studied in relation to i the duration of EEG seizure activity and ii calcium accumulation in post-synaptic neurons, using the oxalate-pyroantimonate method. The reversibility of the pathological changes and calcium loading has been studied from 40 min to 48 h after the termination of seizure activity. Little or no changes were visible 2-3 h after 12 mg kainic acid per kg, but changes were obvious in most hippocampi directly after 2-3.5 h of seizure activity induced

Kainic acid model of temporal lobe epilepsy: Nanoscale oscillations of hippocampal mitochondria.

Mitochondrial dysfunction is increasingly recognized as a central component of epileptogenesis, yet sensitive biophysical markers capable of capturing early mitochondrial alterations remain limited. Kainic acid (KA) model of temporal lobe epilepsy (TLE) is one of the most reliable tools for understanding the mechanisms of epileptogenesis and induces behavioral, electro-physiological, chemical and histological effects that are similar to those observed in patients with TLE. In the present study, we evaluate the nanoscale oscillatory behavior of mitochondria isolated from primary hippocampal neu

Suppression of AKAP150 palmitoylation alleviates seizures in kainic acid-induced epilepsy mice.

Epilepsy is a chronic and refractory neurological disorder, with drug resistance remaining a major challenge. Identifying new therapeutic targets could facilitate the development of more effective anti-seizure medications. AKAP79/150, a member of A-Kinase Anchoring Proteins (AKAPs) family, plays a critical role in synaptic transmission and plasticity. Although its implication in early epileptogenesis has been reported, its role in epilepsy progression remains unclear. In this study, using a kainic acid (KA)-induced epilepsy mouse model, we found that the expression and palmitoylation of AKAP15
