Reconstituted keratin has potential as a raw material for orthopaedic applications. The aim of this study was to investigate the in vivo biocompatibility and osseointegration of keratin materials in an ovine model. Six different modifications of the keratin polymer, based on porous or dense constructs, regenerated by either neutral or acidic treatment, with or without hydroxyapatite, were made as small rods and inserted into drilled round defects in the femur and tibia of sheep. PURPOSE: To understand whether the epithelial phenotype in total sclerocornea is corneal or conjunctival in origin.
METHODS: Four cases of total sclerocornea (male:female = 1:3; mean age = 5.4 ± 4.3; 1-11 years old) who received penetrating keratoplasty (PKP) at our hospital between 2008 and 2011 were included. The DNA sequences encoding beta-keratin have been obtained from Marsh Mugger (Crocodylus palustris) and Orinoco Crocodiles (Crocodylus intermedius). In this investigation the healing pattern of experimental central and marginal pars tensa perforations in cats and rats was studied by using light and scanning electronmicroscopic techniques. The perforations were closed by hyperplastic squamous epithelium exhibiting a marked keratin production. This keratin formed a membranous structure which primarily appeared to close the perforation. No ingrowth of squamous epithelium into the middle ear cavity could be detected. Generally speaking, the perforations had closed within 9-11 days in rats and within 12-14 days in cats.
