Kinetics of potassium transport across single distal tubules of rat kidney.

1. The transport of potassium across the distal tubular epithelium was studied in vivo in rats on a normal potassium intake and in rats in which distal tubular potassium secretion was either stimulated by potassium loading or the I.V. administration of a 5% sodium bicarbonate solution or in which potassium secretion was suppressed by dietary deprivation of potassium or sodium.2. (42)K was used to measure unidirectional fluxes across the luminal and peritubular cell membranes and to assess the magnitude of cellular potassium partaking in the transport process. This was accomplished by the simul

Pan AMPK activation protects tubules in rat ischemic acute kidney injury.

Acute kidney injury (AKI) is characterized by an abrupt decline in kidney function and has been associated with excess risks of death, kidney disease progression, and cardiovascular events. The kidney has a high energetic demand with mitochondrial health being essential to renal function, and damaged mitochondria have been reported across AKI subtypes. 5' Adenosine monophosphate-activated protein kinase (AMPK) activation preserves cellular energetics through improvement of mitochondrial function and biogenesis when ATP levels are low, such as under ischemia-induced AKI. We developed a selectiv

Engineered Kidney Tubules for Modeling Patient-Specific Diseases and Drug Discovery.

The lack of engineering systems able to faithfully reproduce complex kidney structures in vitro has made it difficult to efficiently model kidney diseases and development. Using polydimethylsiloxane (PDMS) scaffolds and a kidney-derived cell line we developed a system to rapidly engineer custom-made 3D tubules with typical renal epithelial properties. This system was successfully employed to engineer patient-specific tubules, to model polycystic kidney disease (PKD) and test drug efficacy, and to identify a potential new pharmacological treatment. By optimizing our system we constructed functi

Matrix metalloproteinases and TIMPS in cultured C57BL/6J-cpk kidney tubules.

Restructuring of basement membranes is a hallmark of the pathology of renal cystic disorders. Here, we present findings consistent with the view that basement membrane degradation by matrix metallo-proteinases (MMPs) may contribute to abnormal basement membrane structure in polycystic kidney disease. Cells from cystic kidney tubules embedded in collagen gels appeared to migrate through the gel. This migration through collagen indicated that these cells could degrade the matrix. To examine this activity, we cultured cystic kidney tubules derived from the C57BL/6J cpk/cpk mouse, a hereditary mod

Histochemical studies on peroxisomes in regenerating proximal tubules of the kidney.

Peroxisomes of the regenerating proximal tubules of the rat kidney were investigated after necrosis induced by mercuric chloride. Slices both for light and electron microscopic examinations were incubated in 3,3'-diaminobenzidine (DAB) medium. Peroxisomes were absent in the necrotic epithelium. They appeared on the fourth day of regeneration and later their number increased reaching the normal distribution in the fourth week. They seem to be needed for the functional differentiation of the proximal tubule cells during regeneration.

Biology of the proximal tubule in body homeostasis and kidney disease.

The proximal tubule (PT) is known as the workhorse of the kidney, for both the range and magnitude of the functions that it performs. It is not only responsible for reabsorbing most solutes and proteins filtered by glomeruli, but also for secreting non-filtered substances including drugs and uremic toxins. The PT therefore plays a pivotal role in kidney physiology and body homeostasis. Moreover, it is the major site of damage in acute kidney injury and nephrotoxicity. In this review, we will provide an introduction to the cell biology of the PT and explore how it is adapted to the execution of

[Xanthoid corpuscles in the cytoplasm of rat kidney tubule epithelium].

In older male rats we found fuchsinophilic granula which appeared both singly and in bale-shaped arrangements in the cytoplasm of the epithelias of proximal, subcapsularly located renal tubuli. In its further development it became apparent that they formed small round-shaped clusters which stained with buffered thionine. These clusters finally united to larger ovoid or cone-shaped homogen formations which detached from cytoplasm and appeared unobstructedly in the lumen of canaliculi and in deeper nephron sections, too. The reason why the authors named these formations "xanthoid" is attributabl

Contraluminal bicarbonate transport in the proximal tubule of the rat kidney.

In order to measure the contraluminal bicarbonate flux in situ we applied the stopped flow capillary microperfusion technique and measured the influx of 14C-bicarbonate buffer into cortical tubular cells at pH 8. It was found that the influx in percent of the starting concentration is larger at 20 mmol/l bicarbonate than at 1 mmol/l, indicating a sigmoidal type influx curve. At 20 mmol/l bicarbonate the influx was inhibited by 44%, when Na+ was replaced by choline. Replacement of gluconate by chloride or sulfate did not change H14CO3- influx. At this bicarbonate concentration, influx is inhibi

The selective action of nickel on tubule function in rabbit kidneys.

Two days after intraperitoneal injection of NiCl2 (20 mumol/kg) into rabbits the same apparently uncompetitive inhibition of aspartate reabsorption is seen as was previously observed following acute exposure to other metals. This dose of Ni reduced the calculated maximum tubular transport rate for aspartate (Tm) and the apparent affinity constant (KM) by over 50%, but exerted no effect on either Tm or KM of cycloleucine or glucose reabsorption. The relative selectivity of the nephrotoxic action of Ni and other metals is reviewed; it raises the question whether the acute effects observed are ap

AQP1 expression in the proximal tubule of diabetic rat kidney.

Polyuria is a hallmark symptom and the first clinical manifestation of diabetes mellitus (DM). The glucose that remains in renal tubules was proposed to produce an osmotic effect resulting in polyuria. Although water is reabsorbed in proximal tubules through an aquaporin-1 (AQP1) dependent mechanism, AQP1 role in the genesis of polyuria is unknown. AQP1 expression was studied in a rat model of Type-1 DM at 15-days and 5-months of evolution. A different AQP1 expression pattern was found in both experimental groups, with no changes in AQP1 localization, suggesting that changes in AQP1 may be inv

Kidney Tubule Secretion Can Discriminate the Cause of Acute Kidney Injury in Cirrhosis.

KEY POINTS: In cirrhotic patients with AKI, tubular secretion differentiates those with acute tubular necrosis versus other causes. Different secretory function by cause of AKI may affect treatment approaches and drug dosing in patients with cirrhosis and AKI. BACKGROUND: AKI is a common and severe complication among hospitalized patients with cirrhosis. The most common causes of AKI in cirrhosis are prerenal azotemia (PRA), hepatorenal syndrome (HRS), and acute tubular necrosis (ATN), and treatment depends on its causes. Kidney proximal tubular secretion is an essential mechanism for elimina

Perifusate metabolomics interrogates nutrient exposure and oxidative stress response in kidney tubules.

Understanding metabolism at the organ level is challenging, as tissue metabolite signals are transient and reflect transport, metabolism, and inter-organ communication. Here, we address this challenge by using a perifusion platform that enables time-resolved metabolite measurements in living kidney tissue under controlled input conditions. We apply this approach to native tubule and glomerulus preparations and generate >45,000 longitudinal, time-resolved metabolite measurements using targeted metabolomics. Tubules release tricarboxylic acid cycle and acetylated amino acid intermediates and sho

Recovery of impaired gluconeogenesis in kidney-cortex tubules of gentamicin-treated rabbits.

Rabbits were given gentamicin over a period of 10 days. At 1, 3, 5 and 10 days renal proximal tubules were isolated and glucose synthesis from several substrates was measured. A relationship between the inhibition of renal gluconeogenesis, accompanied by a decline of both pyruvate carboxylase and phosphoenopyruvate carboxykinase (PEPCK) activities, and an increased gentamicin level in kidney-cortex was noticed after 5 days of therapy. Both the rates of glucose formation from various substrates as well as pyruvate carboxylase and the cytosolic PEPCK activity recovered fully within 3 weeks after

Osmotically induced changes in electrical resistance of distal tubules of rat kidney.

Changes in peritubular oncotic pressure do not alter the electrical resistance of distal convoluted tubules. Transverse specific electrical resistance decreases along the distal convoluted tubule in nondiuretic rats. This drop in resistance can be at least in part accounted for by the relative increae in osmolality of late distal fluid. Osmotic gradients influence the magnitude of intercellular rather than transcellular conductance. Hyposmolality of tubular fluid leads to reduced shunting, whereas increased tubular osmolality has the opposite effect. The osmotic effect forms part of an intraep

Molecular patterns of isolated tubulitis differ from tubulitis with interstitial inflammation in early indication biopsies of kidney allografts.

The Banff 2019 kidney allograft pathology update excluded isolated tubulitis without interstitial inflammation (ISO-T) from the category of borderline (suspicious) for acute T cell-mediated rejection due to its proposed benign clinical outcome. In this study, we explored the molecular assessment of ISO-T. ISO-T or interstitial inflammation with tubulitis (I + T) was diagnosed in indication biopsies within the first 14 postoperative days. The molecular phenotype of ISO-T was compared to I + T either by using RNA sequencing (n = 16) or by Molecular Microscope Diagnostic System (MMDx, n = 51). RN

Kidney tubule iron loading in experimental focal segmental glomerulosclerosis.

Kidney iron deposition may play a role in the progression of tubulointerstitial injury during chronic kidney disease. Here, we studied the molecular mechanisms of kidney iron loading in experimental focal segmental glomerulosclerosis (FSGS) and investigated the effect of iron-reducing interventions on disease progression. Thy-1.1 mice were injected with anti-Thy-1.1 monoclonal antibody (mAb) to induce proteinuria. Urine, blood and tissue were collected at day (D)1, D5, D8, D15 and D22 after mAb injection. Thy-1.1 mice were subjected to captopril (CA), iron-deficient (ID) diet or iron chelation
