TRPA1 activation in non-sensory supporting cells contributes to regulation of cochlear sensitivity after acoustic trauma.

TRPA1 channels are expressed in nociceptive neurons, where they detect noxious stimuli, and in the mammalian cochlea, where their function is unknown. Here we show that TRPA1 activation in the supporting non-sensory Hensen's cells of the mouse cochlea causes prolonged Ca2+ responses, which propagate across the organ of Corti and cause long-lasting contractions of pillar and Deiters' cells. Caged Ca2+ experiments demonstrated that, similar to Deiters' cells, pillar cells also possess Ca2+-dependent contractile machinery. TRPA1 channels are activated by endogenous products of oxidative stress an

The RNA-binding protein Elavl1/HuR is essential for placental branching morphogenesis and embryonic development.

HuR is an RNA-binding protein implicated in a diverse array of pathophysiological processes due to its effects on the posttranscriptional regulation of AU- and U-rich mRNAs. Here we reveal HuR's requirement in embryonic development through its genetic ablation. Obligatory HuR-null embryos exhibited a stage retardation phenotype and failed to survive beyond midgestation. By means of conditional transgenesis, we restricted HuR's mutation in either embryonic or endothelial compartments to demonstrate that embryonic lethality is consequent to defects in extraembryonic placenta. HuR's absence impai

Scar formation in the vestibular sensory epithelium after aminoglycoside toxicity.

Hair cell degeneration and the repair process due to differing types of trauma have been studied extensively in the organ of Corti. It has been determined that, during scar formation, after differing types of trauma to the auditory sensory system, the reticular lamina is maintained with adherens junctions and tight junctions. We investigated the repair process within the vestibular epithelium. Hair cell degeneration was induced by the unilateral application of streptomycin to the inner ears of guinea pigs. Whole mount preparations of all five vestibular organs were processed and examined by fl

Anatomical and molecular insights into avian inner ear sensory hair cell regeneration.

Inner ear sensory hair cells are essential for auditory and vestibular functions. In mammals, loss of these cells leads to permanent hearing loss due to the inability of supporting cells to regenerate hair cells. In contrast, avian species exhibit a remarkable capacity for hair cell regeneration, primarily through the activation and proliferation of supporting cells. This review provides a comprehensive examination of the anatomical and molecular mechanisms underlying sensory hair cell regeneration in two critical avian inner ear structures: the basilar papilla and the utricle. We describe the

Induction of MHC class II antigens on cells of the inner ear.

Growing evidence supports the concept that immune reactions occur in the cochlea, where they can function either in protection or as a source of inflammation. Since immunity is generally initiated by antigen presentation of foreign substances to T cells, antigen-presenting cells expressing major histocompatibility complex (MHC) class II molecules are required. Under resting conditions, cochlear cells usually express no MHC class II. However, we show that exposure to -interferon in vitro induces an increase in MHC class II expression in neonatal cochlear cells of mice. In addition, MHC class II

Neurochemical evidence for afferent GABAergic and efferent cholinergic neurotransmission in the frog vestibule.

Glutamate decarboxylase and choline acetyltransferase activities with magnitudes similar to those of their homologous enzymes in frog nervous tissue were found in homogenates of the frog labyrinth. Transection of the vestibular nerve resulted in a gradual diminution of choline acetyltransferase activity until it reached an 88% decrease 6 weeks after surgery. In contrast, glutamate decarboxylase activity did not suffer any alteration at any time after nerve excision. The presence of their enzymes of synthesis is evidence of the neurotransmitter participation of GABA and acetylcholine in the fro

Transcriptomic change in human utricles after aminoglycoside-induced hair cell ablation: Dynamic alterations to hair cell and supporting cell genes.

Losing sensory hair cells from the mammalian inner ear results in deafness and balance disorders. A major cause of acquired hair cell loss is the use of ototoxic drugs, including aminoglycoside antibiotics. Aminoglycosides are commonly prescribed, and are highly effective and cheap, but they specifically kill hair cells. In mammals, the capacity to regenerate hair cells is extremely limited, with only a small fraction being replaced in the vestibular system. Therefore, the functional deficits arising from their loss are permanent. To date there are no therapeutic strategies to prevent hair cel

Macrophages in the Human Cochlea: Saviors or Predators-A Study Using Super-Resolution Immunohistochemistry.

The human inner ear, which is segregated by a blood/labyrinth barrier, contains resident macrophages [CD163, ionized calcium-binding adaptor molecule 1 (IBA1)-, and CD68-positive cells] within the connective tissue, neurons, and supporting cells. In the lateral wall of the cochlea, these cells frequently lie close to blood vessels as perivascular macrophages. Macrophages are also shown to be recruited from blood-borne monocytes to damaged and dying hair cells induced by noise, ototoxic drugs, aging, and diphtheria toxin-induced hair cell degeneration. Precise monitoring may be crucial to avoid

Immunohistochemical localization of procollagen types I and III during placentation in pregnant rats by type-specific procollagen antibodies.

To examine the sequential localizations of procollagen Types I (Pro I) and III (Pro III) during chorioallantoic placental formation in pregnant rats, we prepared polyclonal anti-rat Pro I- and III-specific antibodies. Biochemical analysis of a fraction containing [14C]-glycine-incorporated collagen from pregnant rat uteri showed that collagen Types I and III were actively synthesized during placental development. We examined 8-, 9.5-, 13-, and 20-day gestation rat uteri immunohistochemically. At Days 8 and 9.5, in the basal decidua facing the fetal cytotrophoblastic giant cell layer and implan

Supporting-cell and extracellular responses to acoustic clicks in the free-standing region of the alligator lizard cochlea.

1. Acoustic clocks were delivered to the tympanic membrane of anesthetized alligator lizards, and electric responses were measured within the free-standing region of the cochlea using glass micropipettes. Responses were recorded intracellularly in supporting cells and extracellularly in the receptor organ and in the scalae. Gross responses were also recorded with wire electrodes in scala tympani. 2. Intra- and extracellular responses contain two components: (1) an early (with latent period less than 0.15 ms after the onset of the click), rate-independent component presumed to originate in the

The combination of scanning and transmission electron microscopy techniques in pathology of the organ of Corti in guinea pigs.

The pathology of the organ of Corti in guinea pigs is studied by means of SEM and TEM. It was the purpose of this study to examine the surface of an experimentally altered organ of Corti by means of SEM screening method). As it was our main interest to find out the localization of acoustic stimulation regarding the tonotopicity on the surface of the organ of Corti, we always examined the whole cochlear by means of SEM first. In acoustic experiments we used long-time pure-tone stimulation beyond the directly damaging sound pressure level, as well as white noise or shots for experimental damage

Sulfamethoxazole increases the risk of preeclampsia through PSMB6: A comprehensive multi-omics and experimental validation study.

BACKGROUND: Environmental and dietary exposure to sulfamethoxazole (SMX) has been detected in pregnant populations, but its impact on preeclampsia (PE)-related placental pathology-particularly through non-therapeutic chronic exposure-remains poorly understood. METHODS: We assessed the effects of maternal SMX exposure on placental structure in pregnant mice. Placental transcriptomic data and Mendelian randomisation (MR) analysis were integrated to identify candidate targets. Molecular docking, molecular dynamics simulations, cellular thermal shift assay (CETSA), and microscale thermophoresis (

LIN28B/let-7 control the ability of neonatal murine auditory supporting cells to generate hair cells through mTOR signaling.

Mechano-sensory hair cells within the inner ear cochlea are essential for the detection of sound. In mammals, cochlear hair cells are only produced during development and their loss, due to disease or trauma, is a leading cause of deafness. In the immature cochlea, prior to the onset of hearing, hair cell loss stimulates neighboring supporting cells to act as hair cell progenitors and produce new hair cells. However, for reasons unknown, such regenerative capacity (plasticity) is lost once supporting cells undergo maturation. Here, we demonstrate that the RNA binding protein LIN28B plays an im

Nodal B-cell lymphomas in Japan--particularly in Tohoku district.

There are different frequencies in the immunological phenotypes of malignant lymphomas in Tohoku and Kyushu districts of Japan. In the Tohoku district, the northern area of Honshu, B-cell lymphomas are more preponderant than T-cell lymphomas. This is just the reverse on the islands of Kyushu and Shikoku. Histologically diffuse lymphoma of large cell type, formerly termed reticulum cell sarcoma or histiocytic lymphoma, was the most frequent (48%) among B-cell lymphomas. It is characteristic of B-cell lymphoma that immunoglobulin is produced in either or both the cellular surface and cytoplasm.

Deletion of the Ebf1, a mouse deafness gene, causes a dramatic increase in hair cells and support cells of the organ of Corti.

Following up on our previous observation that early B cell factor (EBF) sites are enriched in open chromatin of the developing sensory epithelium of the mouse cochlea, we investigated the effect of deletion of Ebf1 on inner ear development. We used a Cre driver to delete Ebf1 at the otocyst stage before development of the cochlea. We examined the cochlea at postnatal day (P) 1 and found that the sensory epithelium had doubled in size but the length of the cochlear duct was unaffected. We also found that deletion of Ebf1 led to ectopic sensory patches in the Kölliker's organ. Innervation of the

Inactivation of maternal Hif-1α at mid-pregnancy causes placental defects and deficits in oxygen delivery to the fetal organs under hypoxic stress.

A critical transition occurs near mid-gestation of mammalian pregnancy. Prior to this transition, low concentrations of oxygen (hypoxia) signaling through Hypoxia Inducible Factor (HIF) functions as a morphogen for the placenta and fetal organs. Subsequently, functional coupling of the placenta and fetal cardiovascular system for oxygen (O2) transport is required to support the continued growth and development of the fetus. Here we tested the hypothesis that Hif-1α is required in maternal cells for placental morphogenesis and function. We used Tamoxifen-inducible Cre-Lox to inactivate Hif-1α i

Conditional Overexpression of Neuritin in Supporting Cell Protects Cochlear Hair Cell and Delays Age-Related Hearing Loss by Enhancing Autophagy.

Age-related hearing loss (ARHL) is a highly prevalent, burdensome sensorineural hearing loss closely associated with impaired autophagic influx. Our previous studies revealed that neuritin, a neurotrophic factor primarily expressed in the central nervous system, could alleviate drug-induced damages in hair cells (HCs) and spiral ganglion neurons. However, its effects on ARHL and whether these effects are closely related to autophagy remain unclear. Using the Nrn1 knock-in mice and cultured cochlear basilar membrane (CBM) of the neonatal mouse, we show that neuritin could restore aging-associat

Pathology of benign paroxysmal positional vertigo revisited.

The pathophysiology of benign paroxysmal positional vertigo (BPPV) is not completely understood. Although the concept of degenerated otoconia transforming the posterior canal (PC) crista into a gravity-sensitive sense organ has gained popular support, several temporal bone (TB) series have revealed similar deposits in normal TBs, suggesting they are a normal change in the aging labyrinth. Furthermore, some TBs from patients with BPPV do not contain particles in the posterior canal. Five TBs from patients with BPPV were studied quantitatively and qualitatively. A small PC cupular deposit was fo

Morphological and functional aspects of the canine placental barrier: evidence supporting a deciduo-chorial model.

ABSTRACT: The canine placenta, characterized by its deciduate and endotheliochorial nature, represents an intermediate placental type between non-invasive and highly invasive placentae. Decidual cells are involved in the prepartum parturition cascade. As they are the only cells expressing the nuclear progesterone receptor (PGR), they play a crucial role in luteal progesterone (P4)-dependent embryo-maternal communication and are essential for maintaining pregnancy. This study focuses on the quantity, morphology, and spatial arrangement of decidual cells. Immunohistochemistry (IHC) was used to l

Selective ablation of pillar and deiters' cells severely affects cochlear postnatal development and hearing in mice.

Mammalian auditory hair cells (HCs) are inserted into a well structured environment of supporting cells (SCs) and acellular matrices. It has been proposed that when HCs are irreversibly damaged by noise or ototoxic drugs, surrounding SCs seal the epithelial surface and likely extend the survival of auditory neurons. Because SCs are more resistant to damage than HCs, the effects of primary SC loss on HC survival and hearing have received little attention. We used the Cre/loxP system in mice to specifically ablate pillar cells (PCs) and Deiters' cells (DCs). In Prox1CreER(T2)+/-;Rosa26(DTA/+) (P
