Galactosylceramide- and lactosylceramide-loading studies in cultured fibroblasts from normal individuals and patients with globoid cell leukodystrophy (Krabbe's disease) and GM1-gangliosidosis.

The metabolism of galactosylceramide and lactosylceramide in cultured fibroblasts was studied using the lipid-loading test. These compounds were incorporated into the fibroblasts yet only small amounts of the incorporated lipids were hydrolyzed unless additional phospholipid was mixed with the glycolipid before loading. Among phospholipids, phosphatidylserine was the most effective for incorporation and hydrolysis of the glycolipids, while phosphatidylcholine inhibited the incorporation of the glycolipids. Using filtration techniques, light scattering analyses and subcellular fractionation, th

Magnetic Resonance Imaging-negative Encephalomyeloradiculopathy Diagnosed by Anti-lactosylceramide Antibody Positivity and Electrophysiological Findings: A Case Report and Review of the Literature.

A 71-year-old woman developed progressive limb weakness and sensory disturbances below the thoracic level. Although myelopathy was initially considered, spinal magnetic resonance imaging (MRI) showed no abnormalities. Conversely, electrophysiological studies have identified both spinal dysfunction and radiculopathy, suggesting myeloradiculopathy. Initiation of steroid therapy resulted in immediate improvement of clinical symptoms and electrophysiological findings. Finally, anti-lactosylceramide (LacCer) antibodies were detected in the serum and cerebrospinal fluid, and titer changes correlated

Hyperglycemia and alteration in sphingolipids are early mediators of diabetic kidney disease: role of elongase 1 (Elovl1).

Introduction Diabetic kidney disease (DKD) is characterized by impairment of renal glomerular and tubular cells. Low plasma levels of ceramides and lactosylceramides containing very long-chain (VLC) fatty acid were found to be predictive of DKD development. Elongase 1 (Elovl1) is a ubiquitous elongase that elongates C20-C22 fatty acids to generate very long-chain C24 fatty acids. Using a novel transgenic mouse overexpressing Elovl1, we investigated whether modification to sphingolipid fatty acid composition averts DKD development. Methods A transgenic (TG) mouse overexpressing Elovl1 was creat

Specificity of galactosylceramidase activation by phosphatidylserine.

Bovine brain phosphatidylserine effectively activates human brain galactosylceramidase (Hanada, E. and Suzuki, K. (1979) Biochim. Biophys. Acta 575, 410-420). Its effect on the other beta-galactosidase (Gm1-ganglioside beta-galactosidase) in human tissues, genetically distinct from galactosylceramidase, was examined. When partially purified human brain beta-galactosidase preparations, pure with respect to each other, were used as the enzyme source and when lactosylceramide, a common glycosphingolipid substrate for both beta-galactosidases, was used as the substrate, phosphatidylserine activate

In-vitro biosynthesis of sulphatoglycosphingolipids by rat submandibular salivary glands.

A sulphotransferase activity, concentrated mainly in the microsomal fraction, which catalyses the transfer of sulphate group from 3'-phosphoadenosine-5'-phosphosulphate to galactosylceramide and lactosylceramide was demonstrated in rat submandibular and sublingual glands. However, the sulphotransferase activity of this fraction in submandibular glands was about ten times higher than in sublingual glands. Optimum enzyme activity was obtained using the detergent Triton X-100, F-, and Mg2+ at a pH of 6.8. The enzyme did not catalyse the transfer of sulphate to glucosylceramide, trihexosylceramide

B4GALT5 deficiency impairs glycosphingolipid biosynthesis: a new congenital disorder of glycosylation?

Lactosylceramide is a glycosphingolipid precursor synthesized by two dedicated galactosyltransferases, B4GALT5 and B4GALT6. The specific roles of B4GALT5 and B4GALT6 in humans have not yet been clearly defined. Here, we report the first human case with bi-allelic loss-of-function variants in B4GALT5, suggesting that intact B4GALT5 activity is indispensable for normal glycosphingolipid biosynthesis and human development. We identified bi-allelic variants in the B4GALT5 gene in a child presenting with microcephaly, mild cognitive impairment, and bilateral cataracts. B4GALT5/6 double KO cells tra

Abnormalities in cerebral lipids and hepatic cholesterol glucuronide of a patient with GM1-gangliosidosis type 2.

The brain tissue of a patient with GM1-gangliosidosis Type 2 showed an abnormal distribution pattern of lipids, that is, a marked increase in GM1-ganglioside, asialo-GM1 and cholesterol ester, a significant increase of glucosylceramide, lactosylceramide and asialo-GM2, and a decrease in galactocerebroside and sulfatide. On the other hand, the liver tissue showed a normal pattern of glycolipids, that is, the presence of normal amounts of glucosylceramide, lactosylceramide, globotriaosylceramide, globoside, hematoside (GM3) and GD3. However, it was found that the patient's liver contained a larg

Nutrient restriction dynamically alters glucose tolerance and ceramide concentrations in periparturient ewes.

The periparturient period in mammals is characterized by reduced insulin action, which allows nutrient partitioning to support fetal growth and milk synthesis. The sphingolipid ceramide is an established interrogator of insulin signaling in non-ruminants and accumulates during this period in dairy cows. We investigated the effects of short-term nutrient restriction (NR) on glucose tolerance and ceramide metabolism in multiparous ewes (n = 19) during this period. Ewes were fed ad libitum or subjected to two 5-d 50% NR periods in late gestation and early lactation. NR reduced circulating glucose

Circulating Lipoprotein Sphingolipids in Chronic Kidney Disease with and without Diabetes.

Abnormalities of sphingolipid metabolism play an important role in diabetes. We compared sphingolipid levels in plasma and in isolated lipoproteins between healthy control subjects and two groups of patients, one with chronic kidney disease without diabetes (ND-CKD), and the other with type 2 diabetes and macroalbuminuria (D-MA). Ceramides, sphingomyelins, and sphingoid bases and their phosphates in LDL were higher in ND-CKD and in D-MA patients compared to controls. However, ceramides and sphingoid bases in HDL2 and HDL3 were lower in ND-CKD and in D-MA patients than in controls. Sphingomyeli

Sphingolipid composition of human platelets.

Total lipid extracts from washed trypsinized human platelets were fractionated into neutral lipids, glycosphingolipids, and phospholipids by silicic acid chromatography. The concentrations and chemical structures of the neutral and acidic glycosphingolipids were then studied in detail. On the basis of sugar molar ratios, studies of permethylation products, and the action of stereospecific glycosidases on the lipids, identifications were made of four neutral glycosphingolipids. Lactosylceramide was the most abundant type and accounted for 64% of the total neutral glycolipid mixture. The major f

Targeting glycosphingolipid metabolism as a potential therapeutic approach for treating disease in female MRL/lpr lupus mice.

Glycosphingolipids (GSLs) hexosylceramides and lactosylceramides are elevated in lupus mice and human patients with nephritis. Whereas other renal diseases characterized by increased GSL levels are thought to be a result of upregulated GSL synthesis, our results suggest elevated hexosylceramides and lactosylceramides in lupus nephritis is a result of increased catabolism of ganglioside GM3 due to significantly increased neuraminidase (NEU) activity. Thus, we hypothesized GM3 would be decreased in lupus nephritis kidneys and blocking NEU activity would reduce GSLs and improve disease in lupus m

Diabetes and kidney dysfunction markedly alter the content of sphingolipids carried by circulating lipoproteins.

BACKGROUND: We have previously shown that very long ceramides/lactosylceramides predicted the development of macroalbuminuria (MA) in type 1 diabetes and expanded our studies into type 2 diabetes. OBJECTIVE: This study proposes comparing the levels of plasma sphingolipids and their distribution in circulating lipoproteins (VLDL/IDL, LDL, HDL2 and HDL3) between a healthy control group and two groups of subjects with type 2 diabetes, one with and other without MA. METHODS: Plasma and lipoprotein sphingolipids/glycosphingolipids were measured using HPLC-MS/MS in 114 subjects (40 controls; 74 ty

A gut microbiome-lipid axis in early pregnancy is associated with metabolic dysregulation and diabetes risk.

Gestational diabetes mellitus (GDM) reflects metabolic dysregulation that becomes clinically apparent during pregnancy and shares key pathophysiological features with broader forms of diabetes. Gut microbiome-host metabolic interactions may contribute to this process, yet their role in early pregnancy remains incompletely understood. In this prospective nested case-control study within the Tongji-Huaxi-Shuangliu Birth Cohort, 784 pregnant women, including 222 who developed GDM, underwent first-trimester gut metagenomic and plasma lipidomic profiling. Cross-omics analyses were performed to iden

Glycosphingolipids Recognized by Acinetobacter baumannii.

Acinetobacter baumannii is an opportunistic bacterial pathogen associated with hospital-acquired infections, including pneumonia, meningitis, bacteremia, urinary tract infection, and wound infections. Recognition of host cell surface carbohydrates plays a crucial role in adhesion and enables microbes to colonize different host niches. Here the potential glycosphingolipid receptors of A. baumannii were examined by binding of 35S-labeled bacteria to glycosphingolipids on thin-layer chromatograms. Thereby a selective interaction with two non-acid glycosphingolipids of human and rabbit small intes

Urinary lipid profiling for the identification of fabry hemizygotes and heterozygotes.

BACKGROUND: Fabry disease is an X-linked lysosomal storage disorder resulting from a deficiency of the lysosomal hydrolase, alpha-galactosidase, for which enzyme replacement therapy is now available. In this study, we aimed to identify Fabry heterozygotes not only for genetic counseling of families but because it is becoming increasingly obvious that many heterozygous (carrier) females are symptomatic and should be considered for treatment. METHODS: We measured 29 individual lipid species, including ceramide, glucosylceramide, lactosylceramide, and ceramide trihexoside, in urine samples from

Neurodegenerative Lysosomal Storage Disorders: TPC2 Comes to the Rescue!

Lysosomal storage diseases (LSDs) resulting from inherited gene mutations constitute a family of disorders that disturb lysosomal degradative function leading to abnormal storage of macromolecular substrates. In most LSDs, central nervous system (CNS) involvement is common and leads to the progressive appearance of neurodegeneration and early death. A growing amount of evidence suggests that ion channels in the endolysosomal system play a crucial role in the pathology of neurodegenerative LSDs. One of the main basic mechanisms through which the endolysosomal ion channels regulate the function

Comprehensive metabolomics study on the pathogenesis of anaplastic astrocytoma via UPLC-Q/TOF-MS.

Anaplastic astrocytoma (AA) is a malignant carcinoma whose pathogenesis remains to be fully elucidated. System biology techniques have been widely used to clarify the mechanism of diseases from a systematic perspective. The present study aimed to explore the pathogenesis and novel potential biomarkers for the diagnosis of AA according to metabolic differences. Patients with AA (n = 12) and healthy controls (n = 15) were recruited. Serum was assayed with untargeted ultraperformance liquid chromatography-quadrupole/time-of-flight-mass spectrometry (UPLC-Q/TOF-MS) metabolomic techniques. The data
