Accurate intraoperative assessment of parathyroid blood flow is crucial to preserve function postoperatively. Indocyanine green (ICG) angiography has been successfully employed, however its conventional application has limitations. A label-free method overcomes these limitations, and laser speckle contrast imaging (LSCI) is one such method that can accurately detect and quantify differences in parathyroid perfusion. In this study, twenty-one patients undergoing thyroidectomy or parathyroidectomy were recruited to compare LSCI and ICG fluorescence intraoperatively. An experimental imaging devic

BACKGROUND: Handheld laser speckle contrast imaging (LSCI) is crucial in clinical settings, but motion artifacts (MA) can compromise perfusion image reliability. Current prevention and suppression methods are often impractical or complex. Machine vision techniques, promising in medical imaging, could improve signal quality, but their use in suppressing MA is still unexplored.
OBJECTIVE: We propose an innovative method based on linear regression for MA correction (MAC) in LSCI and validate it in vivo.
METHODS: We performed paired handheld and mounted LSCI measurements on 14 subjects with psor

OBJECTIVE: Facial nerve palsy (FNP) is a common disease in the otorhinolaryngological department. Besides the main symptom of motionlessness occurring in the ipsilateral facial muscles in FNP, there are other common complaints of numbness, stiffness and tightness in ipsilateral face described by the patients. Based on our pilot study, we further investigated the relevance between these complaints and facial microcirculation.
METHOD: Function of facial microcirculation was evaluated by laser speckle contrast imaging (LCSI). Facial perfusion was measured in 143 patients with facial nerve palsy

BACKGROUND: Ischemia at the site of an intestinal anastomosis is one of the most important risk factors for anastomotic leakage (AL). Consequently, adequate intestinal microperfusion is essential for optimal tissue oxygenation and anastomotic healing. As visual inspection of tissue viability does not guarantee an adequate objective evaluation of intestinal microperfusion, surgeons are in dire need of supportive tools to decrease anastomotic leakage after colorectal surgery.
METHODS: In this feasibility study, laparoscopic laser speckle contrast imaging (LSCI) was used to evaluate intestinal m

Laser speckle contrast imaging (LSCI) is a non-invasive technique for measuring superficial blood perfusion over large areas. With its high spatiotemporal resolution, LSCI semi-quantifies heterogeneous blood flow changes across different regions within the imaged area. Here, we demonstrate the technique of LSCI to record blood flow during unilateral renal ischemia-reperfusion injury (IRI) in mice. Following right nephrectomy, the position of the mouse and the focal point of the objective of the LSCI system were adjusted to ensure the left kidney was in the field of view. LSCI scans were perfor

SIGNIFICANCE: Quantitatively mapping both cerebral blood flow and tissue dynamics from laser speckle contrast imaging (LSCI) is powerful for studying cerebral blood flow and neurovascular coupling, particularly in the context of stroke. Conventional multi-exposure fitting is slow and difficult to scale. Efficient, physically grounded methods are needed to extract both vascular and tissue dynamic biomarkers from LSCI data.
AIM: We aim to develop and validate a physics-informed neural network (PINN) that quantitatively estimates fast (vascular) and slow (tissue-related) speckle decorrelation pa

BACKGROUND: Intestinal resection causes inevitable vascular damage, which cannot always be seen during an intraoperative clinical assessment of local intestinal perfusion. If left unaltered, impaired perfusion can lead to complications, such as anastomotic leakage (AL). Therefore, we demonstrate the use of a novel laparoscopic laser speckle contrast imaging (LSCI)-based approach in order to assess local intestinal perfusion during the construction of intestinal anastomoses.
METHODS: Three segments were isolated from the small intestine of a pig, while the perfusion of each was compromised by

INTRODUCTION: Dysfunctional microcirculation is associated with cardiovascular risk factors, chronic disease such as diabetes and acute conditions like septic shock. The non-invasive optical techniques laser Doppler flowmetry (LDF) and diffuse reflectance spectroscopy (DRS) are often used to measure perfusion and oxygen saturation, but are limited to single-point measurements making them sensitive to spatial variations. The imaging modalities multi-exposure laser speckle contrast imaging (MELSCI) and multi-spectral imaging (MSI) overcome this limitation by capturing the parameters in a larger

The dual-display laparoscopic imaging system (DLIS), which integrates laser speckle contrast imaging (LSCI) with white-light RGB mode, enhances surgical decision-making during minimally invasive procedures. However, motion artifacts and dynamic perfusion necessitate reduced statistical frames, which increases noise and degrades imaging quality. Although the structure-guided anisotropic approach optimizes local contrast calculation, it fails to suppress global statistical noise. To address this, we propose a green (G) channel-informed collaborative filtering framework, which leverages the hemog

Laser speckle contrast imaging (LSCI) is an important non-invasive capability for real-time imaging for tissue-perfusion assessment. Yet, the size and weight of current clinical standard LSCI instrumentation restricts usage to mainly peripheral skin perfusion. Miniaturization of LSCI could enable hand-held instrumentation to image internal organ/tissue to produce accurate speckle-perfusion maps. We characterized a 1mm2 chip-on-tip camera for LSCI of blood perfusion in vivo and with a flow model. A dedicated optical setup was built to compare chip-on-tip camera to a high specification reference

Laser speckle contrast imaging (LSCI) is an under-utilized retinal imaging modality capable of non-invasively generating contrast-free, widefield maps of retinal blood flow from which multiple metrics of retinal hemodynamic function can be derived using relatively simple flood illumination instrumentation. Technical and computational advancements have improved spatiotemporal resolution and data extraction to the extent that three-peak blood flow waveforms, currently unaccounted for in LSCI nomenclature, are consistently revealed in the murine retina. Herein, we utilize a custom LSCI instrument

Accurate intraoperative identification of necrotic intestinal segments remains challenging owing to the limitations of invasive methods or single-parameter techniques. A multimodal system integrating laser speckle contrast and multispectral imaging was developed. The noninvasive system simultaneously acquires images of blood flow index (BFI) and tissue oxygen saturation (StO2), which enables the assessment of tissue metabolic rate of oxygen (tMRO2). For in vivo calculation of StO2 in the small intestine, a linear transformation algorithm based on three hemoglobin isosbestic points was employed

BACKGROUND: In reconstructive surgery, adequate perfusion of flaps is essential for successful outcomes. Assessing normal flap perfusion and differentiating it from problematic perfusion is important and time-consuming. Laser speckle contrast imaging (LSCI), an optical technique for quantitative microcirculation assessment, can be used to non-invasively monitor flap perfusion. We designed a wireless handheld LSCI device and investigated its clinical feasibility in Deep Inferior Epigastric Perforator (DIEP) flap reconstruction surgery.
METHODS: In a case series study with 15 patients and 20 DI
