In the summer of 1976, a mysterious epidemic of fatal respiratory disease in Philadelphia launched an intensive investigation that resulted in the definition of a new family of pathogenic bacteria, the Legionellaceae. In retrospect, members of the family had been isolated from clinical specimens as early as 1943. Unsolved epidemics of acute respiratory disease dating to the 1950s were subsequently attributed to the newly described pathogens. In the intervening years, the Legionellaceae have been firmly established as important causes of sporadic and epidemic respiratory disease. The sources of

Three "new" pneumonias have recently been recognized which are old diseases whose etiology has only been newly-recognized. Legionella pneumophila (Legionnaires' disease), Pittsburgh Pneumonia Agent (PPA), and Legionella-like Organisms (the Family Legionellaceae) are "rickettsia-like" bacteria which require special media for cultivation. They can each cause nosocomial pneumonia in immunosuppressed patients and all but PPA cause community-acquired pneumonia. The Legionellaceae reside in the environment within water and soil. Diagnosis can be established by morphologic, microbiological, and immun

We report a case of fulminant pneumonia that was due to aspiration of contaminated hot spring water and was not affected by beta-lactam antibiotics. We suspected that the patient had Legionnaires' disease and treated the clinical symptoms with erythromycin. Legionellaceae could not be isolated from sputum or lung biopsy material, but an elevated titer to Legionella pneumophila serogroup 4 was found by indirect immunofluorescence test. We diagnosed the patent as having Legionnaires' disease with improved clinical symptoms. Furthermore, we went to the hot spring that he visited and tried to isol

The purposes of the study were (i) to describe the abundance and epidemiology of Legionellaceae in the man-made environment in a northern Italian area, (ii) to assess the concordance between pulsed-field gel electrophoresis (PFGE) and automated ribotyping (AR) techniques for genotyping L. pneumophila and (iii) to investigate the correlation between serogrouping and genotyping data. Water was sampled from reservoirs in 12 buildings across an area of 80-km radius. Despite the water temperature always being maintained above 55 degrees C, all of the buildings sampled were contaminated with Legione

Bacteria of the genus Legionella persist in a wide range of environmental habitats, including biofilms, protozoa and nematodes. Legionellaceae are 'accidental' human pathogens that upon inhalation cause a severe pneumonia termed 'Legionnaires' disease'. The interactions of L. pneumophila with eukaryotic hosts are governed by the Icm/Dot type IV secretion system (T4SS) and more than 150 'effector proteins', which subvert signal transduction pathways and promote the formation of the replication-permissive 'Legionella-containing vacuole'. The Icm/Dot T4SS is essential to infect free-living protoz

Spinal cord injuries (SCIs) often result in permanent disabilities in humans. One major reason for the lack of recovery is the inability of adult mammalian descending neurons to regenerate their axons after injury. In contrast, several fish species, such as the sea lamprey, exhibit spontaneous axon regeneration and successful functional recovery following a complete SCI. Recent studies have shown that a SCI in rodents and humans induces gut microbiome dysbiosis, which can impair recovery. Therefore, our goal was to examine how the microbiome changes after SCI in a regenerating animal model (th

The Legionellaceae as agents causing pneumonias are widespread distributed and lead to partially very serious diseases, particularly in predisposed persons with impaired resistance. Two of 12 cases observed are reported casuistically. Erythromycin as the drug of choice should be given already at suspicion. Since Legionellosis cannot be differentiated from pneumonias of other etiology on the basis of clinical findings alone, the authors refer to the importance of the relevant microbiologic methods.

Urinary antigen tests are a widely used rapid diagnostic method for Legionella pneumonia. However, conventional urinary antigen tests are unable to detect anything other than Legionella pneumophila serogroup 1. The Ribotest Legionella (Ribotest) can detect all serogroups by using antibodies recognizing L. pneumophila ribosomal protein L7/L12 in addition to the conventional L. pneumophila serogroup 1 lipopolysaccharide. The aim of this study was to evaluate the performance of Ribotest against conventional urinary antigen tests, including the detection of Legionellaceae other than L. pneumophila

There are currently 42 described species of legionellae representing 64 serogroups in the family Legionellaceae and the genus Legionella. The phenotypic characteristics of legionellae are described, including growth requirements, and biochemical characteristics. Identification of legionellae by biochemical tests, fatty acid analysis, ubiquinones, protein profiles, carbohydrate analysis, serology, monoclonal antibodies, and molecular techniques is described. The occurrence and description of Legionella-like amebic pathogens are discussed. The problems of identification to the species level are

A simple absorption step using blocking fluid prepared from a selected campylobacter strain was introduced in parallel with routine legionella serology tests. Over 12 months, 2716 patients were tested for legionella antibodies by the Indirect Fluorescent Antibody Test of whom 58 (2.1%) had a positive titre (> or = 16) in one or more sera. Campylobacter blocking fluid significantly reduced the legionella titres in 17 of these patients (29%) including four patients with diagnostic serology results (two of whom had pneumonia) and 13 patients with non-diagnostic titres. Absorption with campylobact

The genus Legionella, belonging to the family of Legionellaceae, comprises nowadays seven species, among which Legionella pneumophila might be the most important. The identification of the agent is difficult because L. pneumophila is very pretentious requiring peculiar conditions concerning culture medium, temperature, and time. The initial cultivation will succeed the best in an atmosphere enriched with CO2. The demonstration of serum antibodies will succeed by means of indirect immunofluorescence; recently the micro-agglutination test is vastly applied. The clinical picture of Legionellosis

This study provides guidelines on the usefulness of full and 527 bp 16S rRNA gene sequencing and Microseq databases for identifying medically important aerobic Gram-negative bacteria. Overall, full and 527 bp 16S rRNA gene sequencing can identify 26.1 % and 32.6 %, respectively, of medically important aerobic Gram-negative bacteria confidently to the species level, whereas the full-MicroSeq and 500-MicroSeq databases can identify 15.2 % and 26.1 %, respectively, of medically important aerobic Gram-negative bacteria confidently to the species level. A review of the world literature on the "Legionnaires' disease", an infectious disease which was designated in 1979 as "legionellosis" and described in three different clinicoanatomic forms is presented. These forms differ both in manifestations of the disease and in species of the causal agents, legionellae, united in a genus Legionellaceae. Particular attention in the review is paid to morphological lesions observed in legionelloses (macro- and microscopic, and ultrastructural). The necessity of a careful histobacterioscopic examination of tissues in suspected cases of legionellosis and addi

Legionellaceae are composed of a single genus, Legionella, which is currently comprised of over 52 species. L. pneumophila, L. micdadei, L. longbeachae, and L. dumoffi are clinically considered the most important, with L. pneumophila causing >90%1,7 of cases of legionnaires disease (LD). Since LD's recognition in 1976 following an outbreak of pneumonia involving delegates of an American Legion convention in Philadelphia, there has been increased awareness and importance of the isolation, detection, and reporting of these cases. Herein we describe the unfortunate case of a 7 month old boy with

Of the Legionellaceae family, Pittsburgh pneumonia agent (Tatlockia micdadei, Legionella micdadei) is second only to Legionella pneumophila in causing human pneumonia. In nosocomial infection, the patients tend to be immunosuppressed. The clinical presentation is nonspecific, although in immunosuppressed hosts the presentation may mimic that of pulmonary embolus (pleuritic chest pain, nonproductive cough, pleural-based densities on chest rontgenogram). The reservoir for the organism is water, and prevention of nosocomial infections can be accomplished by disinfection of the water supply. Diagn

Legionella gormanii is a fastidious, Gram-negative bacterium known to be the etiological agent of atypical community-acquired pneumonia. The human cathelicidin LL-37 exhibits a dose-dependent bactericidal effect on L. gormanii. The LL-37 peptide at the concentration of 10 µM causes the bacteria to become viable but not cultured. The antibacterial activity of the peptide is attributed to its effective binding to the bacterial membrane, as demonstrated by the fluorescence lifetime imaging microscopy. In this study, to mimic the L. gormanii membranes and their response to the antimicrobial peptid

The article studies the modern problems of legionellosis infection and gives data concerning epidemiological aspects of this disease, especially clinical course of its types (pneumonia, acute respiratory viral infection syndrome, alveolitis, pleuritis), as well as methods of bacteriological and serological diagnostics, and etiotropic treatment. The authors use their own materials to describe the restricted outbreak of legionellosis infection and sporadic cases of this disease. The article makes clinic classification of legionellosis taking into account various publications and the proper exper

From June 24 to July 5, 1996, 3 patients were admitted to the same hospital with atypical pneumonia. One of the patients, a 52-year-old man, demonstrated progressive pulmonary infiltrates and severe hypoxemia, and finally required mechanical ventilation. All 3 patients had elevated antibody titers for Legionella pneumophila serogroup 1, and had visited the same spa prior to the onset of their symptoms. On September 25, 1996 the district health department inspected the spa, and isolated Legionella pneumophila serogroup 1 from the facility's hot water tanks and outlets. Although it has been repo

Legionella pneumophila (L. pneumophila) which is also known as etiologic agent Legionnaires Disease lives in natural water and man made water systems. These bacteria belonging to Legionellaceae family are divided 15 serogroups. Phenotypical methods used for the identification of Legionella isolates are not very discriminatory. In this study we investigated genotypic features of eight L. pneumophila serogroup 1 and 18 L. pneumophila serogroup 2-14 strains isolated from different buildings in Istanbul by randomly amplified polymorphic DNA (RAPD) method. Eight L. pneumophila serogroup 1 strains (
