A Review on the Use of Letrozole in Female and Male Infertility.

Infertility in developing countries is a distinct and complex problem that disproportionately affects women. Though not a physically restraining disease, it causes a huge social burden on the emotional, financial, and psychosocial quotients of those who suffer from it. Assisted reproductive procedures are frequently used to treat infertility. Years ago, the emergence of ovulation induction represented a significant advancement in treating female infertility. Letrozole, an aromatase inhibitor, is a potential therapy for ovulation induction. Numerous clinical conditions, including anovulatory in

Stopping treatment can reverse acquired resistance to letrozole.

Using the intratumoral aromatase xenograft model, we have observed that despite long-lasting growth inhibition, tumors eventually begin to grow during continued letrozole treatment. In cells isolated from these long-term letrozole-treated tumors (LTLT-Ca), estrogen receptor-alpha (ERalpha) levels were decreased, whereas signaling proteins in the mitogen-activated protein kinase cascade were up-regulated along with human epidermal growth factor receptor 2 (Her-2). In the current study, we evaluated the effect of discontinuing letrozole treatment on the growth of letrozole-resistant cells and tu

Can letrozole be repurposed for the treatment of visceral leishmaniasis?

Visceral leishmaniasis, caused by Leishmania infantum in New World countries, is the most serious and potentially fatal form of leishmaniasis, if left untreated. There are currently no effective prophylactic measures, and therapeutic options are limited. Therefore, we investigated whether the aromatase inhibitor letrozole (LET), which is already used to treat breast cancer, has an antileishmanial activity and/or immunomodulatory potential and therefore may be used to treat L. infantum infection. LET was active against L. infantum promastigote and amastigote life cycle stages in an in vitro inf

Nocebo Effect During Letrozole Desensitization: A Case Report.

This report describes the case of a 46-year-old woman with breast cancer and a history of multiple drug hypersensitivity reactions, who developed atypical allergic-like symptoms with three different adjuvant endocrine therapy regimens. As the use of an aromatase inhibitor was considered essential for her treatment, a three-day desensitization protocol to letrozole was implemented. During the protocol, she developed pruritic erythema following placebo administration. Despite this, the protocol was completed, with only mild side effects (oropharyngeal pruritus and headache), which were managed s

The Follicular Output Rate was Improved with 3-Day Letrozole Administration Compared with 5-Day Letrozole Administration Under Progestin-Primed Ovarian Stimulation.

PURPOSE: Progestin-primed ovarian stimulation (PPOS) has been widely employed in in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) cycles. In recent years, letrozole (LE) combined with medroxyprogesterone acetate (MPA) has been used in this protocol to enhance ovarian response. This study compared the effects of a 5-day regimen with those of a 3-day regimen of letrozole within PPOS, focused on the follicular output rate (FORT) and blastocyst formation rates. PATIENTS AND METHODS: From January 2017 to January 2020, 1,754 infertility patients who received PPOS protocol were div

Acantholytic Dyskeratosis Consistent With Grover's Disease After Letrozole Therapy.

We present a rare case of Grover's disease (GD) associated with letrozole therapy in a 66-year-old female with stage IV breast cancer. GD is a dermatological condition characterized by papulovesicular lesions typically found on the chest and trunk. While GD is linked to chemotherapeutic agents, its association with letrozole is not well documented. The patient presented with a pruritic rash on her neck, right arm, and trunk, initially misdiagnosed as contact dermatitis. Despite treatment with triamcinolone acetonide, the rash persisted. A subsequent punch biopsy confirmed acantholytic dyskerat

Roxadustat alleviates metabolic traits in letrozole-induced PCOS mice.

Polycystic ovary syndrome (PCOS) is a highly prevalent disorder in women that is commonly accompanied by metabolic syndrome. Activation of the hypoxia-inducible factor (HIF) pathway is known to alleviate metabolic defects. Hence, this study utilized a preclinical PCOS mouse model to investigate the effects of chemically induced HIF activation on the metabolic traits of PCOS. Prepubertal letrozole treatment was used to generate a PCOS mouse model in the C57Bl6/J strain, and PCOS mice were orally treated with vehicle or roxadustat for six weeks from age 12 weeks onwards to induce HIF activation.

Diacerein ameliorates letrozole-induced polycystic ovarian syndrome in rats.

Polycystic ovary syndrome (PCOS) is the most common gynaecological endocrine disease that causes anovulatory infertility. The current study aimed to explore the possible role of diacerein (DIA), an IL-1β inhibitor, in treating letrozole-induced PCOS in rats that exhibit the metabolic and endocrinal criteria of PCOS patients. PCOS was induced in female Wistar rats by the oral administration of letrozole (1 mg/kg, per orally, p.o.) for 21 days. Rats were then treated with DIA (25 mg/kg/day, p.o.), DIA (50 mg/kg/day, p.o.), or metformin (2 mg/100 g/day, p.o.) for 14 days after the PCOS induction.

Possible interaction between letrozole and long-acting injectable zuclopenthixol.

Mrs A, a 68-year-old woman with paranoid schizophrenia, was on long-term psychiatric treatment with long-acting intramuscular zuclopenthixol, quetiapine and alprazolam when, in April 2012, she was diagnosed with right breast infiltrating ductal carcinoma. After starting treatment with letrozole on 4 July, Mrs A progressively developed extrapyramidal symptoms and these were particularly evident after each zuclopenthixol administration. On 9 January, both quetiapine and alprazolam were stopped due to excessive lethargy. After the administration of the last dose of zuclopenthixol on 26 January, s

Macular oedema due to letrozole: a first case report.

A 72-year-old woman presented with unexplained, progressive, painless visual loss in the right eye during the past six months. At presentation visual acuity (VA) was 3/60 in the right eye and 6/6 in the left eye. Anterior segment examination and intraocular pressures were normal. Dilated fundoscopy revealed significant macular oedema in the right eye and a normal fundus appearance in the left eye. Her medical history was noteworthy for breast ductal carcinoma in situ, for which she had undergone right mastectomy three years earlier. She had not received chemotherapy or radiotherapy but she had

Lichenoid drug eruption induced by letrozole: a case report.

Lichenoid drug eruption (LDE) is an uncommon adverse reaction that can be clinically indistinguishable from idiopathic lichen planus (LP). We report a 63-year-old woman who developed a diffuse pruritic papular eruption with oral mucosal involvement two weeks after initiating letrozole for breast cancer. Histopathology revealed lichenoid interface dermatitis with scattered eosinophils extending into the deep subcutaneous tissue, supporting a drug-induced process. Due to the necessity of endocrine therapy, she was managed with high-potency topical corticosteroids and tacrolimus. Upon histologic

Adropin ameliorates reproductive dysfunctions in letrozole-induced PCOS mouse.

Polycystic ovary syndrome (PCOS) is the most common cause of infertility in reproductive-age women, and its etiology and exact treatment are not yet established. Adropin is a unique hepatokine involved in maintaining energy homeostasis, and its level has been reported to decline in serum and follicular fluid of PCOS women. Thus, present study was designed to investigate the effect of adropin on hormonal and reproductive abnormalities in PCOS mice. PCOS was induced in adult mice by administering letrozole (6 mg/kg body weight) orally for 21 days. PCOS mice were subsequently treated with adropin

Probiotics mitigates ovarian angiogenic disturbance in letrozole-induced PCOS.

BACKGROUND: Among metabolic/endocrine diseases affecting women in the reproductive years, polycystic ovarian syndrome (PCOS) is well documented as the leading cause of female infertility and several complications including dyslipidemia, cardiomorbidities, among others. Vascular endothelial growth factor (VEGF) is a proangiogenic marker which plays crucial roles in endothelial dysfunction, including ovarian dysfunction. Probiotics are gut microbiome that regulate metabolic health via epigenetic modulation of histone. Nevertheless, the present study was designed to investigate the beneficial eff
