Characterization of Highly Branched Dextran VB113 Produced by Leuconostoc mesenteroides VB113 as Traditional Pickle Isolate.

Lactic Acid Bacteria (LAB) from traditional fermented foods serve as a promising source of novel exopolysaccharides (EPSs) with desirable techno-functional properties. In this study, the EPS production ability of LAB strains from traditional pickles was tested and a strong slimy isolate grown in a sucrose-rich environment was further selected. The isolate was identified as Leuconostoc mesenteroides VB113 and its EPS production characteristics were determined. HPLC analysis revealed that glucose was the only monomer in the EPS repeating structure, whereas 1H and 13C NMR spectroscopy analysis de

In vitro probiotic profiling of novel Enterococcus faecium and Leuconostoc mesenteroides from Tunisian freshwater fishes.

Novel lactic acid bacteria isolated from different organs of freshwater fish were examined for their potential application as probiotics in raw and processed foods. Four isolates of Enterococcus faecium and Leuconostoc mesenteroides were identified at the molecular level by 16S rRNA sequencing and random amplification of polymorphic DNA - polymerase chain reaction, and their antimicrobial activity against a panel of pathogens and food-poisoning bacteria was investigated. The whole bacteriocins of the 4 isolates were characterized by enterobacterial repetitive intergenic consensus sequences in

Exopolysaccharide biosynthesis and regulation in Leuconostoc mesenteroides influences water holding capacity of fermented plant-based milks.

Physical instability remains a major challenge in fermented plant-based yoghurts in the absence of added texturizing agents. Cultures can improve water holding capacity (WHC) through production of exopolysaccharide (EPS). This study identified key genes required for sucrose-dependent EPS biosynthesis in Leuconostoc mesenteroides 399 and assessed their contribution to WHC during plant-based milk fermentation. A forward genetic screen for spontaneous mutants with reduced slime formation on sucrose agar identified a loss of function mutation in the predicted dextransucrase gene dsr1. Subsequent s

Novel glutaminase-free l-asparaginase from Leuconostoc mesenteroides BN1: biochemical properties, molecular dynamics, and anticancer activity.

l-Asparaginase is an essential chemotherapeutic enzyme used to treat acute lymphoblastic leukaemia and certain lymphoid malignancies; however, its clinical use can be limited by adverse effects linked to immunogenicity and undesirable l-glutaminase activity. In this study, a novel glutaminase-free l-asparaginase from Leuconostoc mesenteroides BN1 (LmASNase) was cloned, expressed in Escherichia coli, and biochemically characterised. The recombinant enzyme appeared to form a homotetramer and demonstrated specificity towards l-asparagine, with no detectable activity towards l-glutamine. LmASNase

Comparison Between Fluorescent Probe and Ion-Selective Electrode Methods for Intracellular pH Determination in Leuconostoc mesenteroides.

The intracellular pH (pHin) of Leuconostoc mesenteroides subsp. mesenteroides 19D was evaluated by two different methods, fluorescent probe and ion-selective electrode. Two fluorescent probes 5 (and-6)-carboxyfluorescein diacetate succinimidyl ester (cFDASE) and 5 (and-6)-carboxy-2',7'-dichlorofluorescein diacetate succinimidyl ester (cDCFDASE) were tested to evaluate the intracellular pH (pHin) of living cells at a medium pH (pHex) ranged from 5.0 to 6.5. Salicylic acid was used as a probe for the ion-selective electrode method. Cells kept 60-80% of cFDASE probe at all pHex values against 5-1

PHA Production from Cheese Whey and "Scotta": Comparison between a Consortium and a Pure Culture of Leuconostoc mesenteroides.

It is urgent to expand the market of biodegradable alternatives to oil-derived plastics owing to (i) increasingly limited oil availability/accessibility, and (ii) the dramatic impact of traditional plastics on aquatic life, the food chain, all Earth ecosystems, and ultimately, human health. Polyhydroxyalkanoates (PHAs) are promising biodegradable polymers that can be obtained through microbial fermentation of agro-industrial byproducts, e.g., milk and cheese whey. Here, the PHA-accumulating efficiency of a mixed microbial culture (MMC, derived from activated sludges) grown on dairy byproducts

Upregulation of Immune checkpoint PD-L1 in Colon cancer cell lines and activation of T cells by Leuconostoc mesenteroides.

Globally colorectal cancer ranks as the third most widespread disease and the third leading cause of cancer-associated mortality. Immunotherapy treatments like PD-L1 blockade have been used to inhibit the PD-L1 legend, which boosts the activity of cytotoxic T lymphocytes. Recently, studies suggest that some probiotics could potentially enhance the effectiveness of immunotherapy treatments for cancer patients. We found that in Caco-2 and HT-29 cells, the live Leuconostoc mesenteroides treatment resulted an increase in the PD-L1 expression and this treatment stimulated interferon-gamma (IFN-γ) p

The antidiabetic potential of Leuconostoc mesenteroides strain SB1075 fermented soy yoghurt: Insights from mouse intestinal transcriptomics and metabolite analyses.

Fermented foods are recognised as functional due to their health-promoting bioactive metabolites. Our previous study showed enhanced shelf-life with stable functional metabolites of soy yoghurt fermented with Leuconostoc mesenteroides strain SB1075. Building on these findings, we prepared a new batch of SB1075-fermented soy yoghurt for animal experiments and evaluated its antidiabetic potential using transcriptomic and metabolomic approaches. GC-MS profiling confirmed reproducible metabolite patterns in fermented soy yoghurt, with the putative presence of bioactive compounds including D-pinito

Preventive Effect of Probiotic Leuconostoc mesenteroides H40 Against Cognitive Disorder by Anti-Inflammatory, Synaptic Plasticity Regulation, and Antioxidant Effects.

Live Leuconostoc mesenteroides H40 has been reported to have probiotic properties; however, the effect of its live probiotic form on cognitive ability has not been reported. This study investigated modulatory effects of the probiotic L. mesenteroides H40 in an ICR mouse model (male) of cognitive disorders. Cognitive disorders were induced in mice by the addition of scopolamine (1 mg/kg/day) with donepezil (2 mg/kg/day) as a medicinal control. L. mesenteroides H40 significantly attenuated scopolamine-induced cognitive disorder in the novel object recognition and Y-maze tests in a concentration-

Live and heat-killed Leuconostoc mesenteroides counteract the gastrointestinal dysfunction in chronic kidney disease mice through intestinal environment modulation.

Probiotics are well-known therapeutic agents for managing constipation and have been used to improve chronic kidney disease (CKD) progression. However, heat-killed probiotics on CKD remain inadequately explored. This study aimed to evaluate the probiotic potential of lactic acid bacteria derived from natural sources and to investigate the effects of both live and heat-killed Leuconostoc mesenteroides (Ln.m) on renal and gastrointestinal functions in CKD mice. Ln.m was selected from acid and bile salt intolerance tests, non-hemolytic activity, and antibiotic sensitivity. CKD mice demonstrated s

Systems pharmacology and targeted transcriptional profiling suggest the putative neuroprotective role of Leuconostoc mesenteroides in an in vitro Alzheimer's disease model.

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disorder driven by amyloid-beta (Aβ) accumulation, mitochondrial failure, and neuroinflammation. While probiotics show therapeutic potential via the gut brain axis, the molecular mechanisms remain poorly understood. This study investigated the neuroprotective potential of Leuconostoc mesenteroides lysate and its bioactive metabolites in an Aβ-induced SH-SY5Y neuroblastoma model. METHODS: SH-SY5Y cells were challenged with Aβ and treated with L. mesenteroides lysate. Neuroprotective effects were evaluated via ROS accumulat

Leuconostoc mesenteroides fermentation produces butyric acid and mediates Ffar2 to regulate blood glucose and insulin in type 1 diabetic mice.

Type 1 diabetic patients have lower counts of butyric acid-producing bacteria in the dysbiotic gut microbiome. In this study, we demonstrate that a butyric acid-producing Leuconostoc mesenteroides (L. mesenteroides) EH-1 strain isolated from Mongolian curd cheese can reduce blood glucose and IL-6 in the type 1 diabetic mouse model. L. mesenteroides EH-1 fermentation yielded high concentrations of butyric acid both in vitro and in vivo. Butyric acid or L. mesenteroides EH-1 increased the amounts of insulin in Min6 cell culture and streptozotocin (STZ)-induced diabetic mice. Inhibition or siRNA

Enzymatic synthesis of L-DOPA alpha-glycosides by reaction with sucrose catalyzed by four different glucansucrases from four strains of Leuconostoc mesenteroides.

L-DOPA alpha-glycosides were synthesized by reaction of L-DOPA with sucrose, catalyzed by four different glucansucrases from Leuconostoc mesenteroides B-512FMC, B-742CB, B-1299A, and B-1355C. The glucansucrases catalyzed the transfer of d-glucose from sucrose to the phenolic hydroxyl position-3 and -4 of L-DOPA. The glycosides were fractionated and purified by Bio-Gel P-2 column chromatography, and the structures were determined by (1)H NMR spectroscopy. The major glycoside was 4-O-alpha-d-glucopyranosyl L-DOPA, and the minor glycoside was 3-O-alpha-D-glucopyranosyl L-DOPA. The two glycosides
