Leukocyte common antigen--a diagnostic discriminant between hematopoietic and nonhematopoietic neoplasms in paraffin sections using monoclonal antibodies: correlation with immunologic studies and ultrastructural localization.

Immunohistochemical localization of human leukocyte common antigen (LCA), a major membrane glycoprotein restricted to leukocytes, was evaluated in paraffin sections of a wide variety of hematopoietic and nonhematopoietic tissues (294 specimens) with monoclonal antibodies (PD7/26 and 2B11). In nonneoplastic tissues, LCA was identified on B and T lymphocytes, with variable immunoreactivities for plasma cells and histiocytes. By light microscopy and ultrastructurally, LCA was localized predominantly to the cell membrane and was also present focally in the cytoplasm. Myeloid cells at all stages of

Identification of the potential dual inhibitor of protein tyrosine phosphatase sigma and leukocyte common antigen-related phosphatase by virtual screen, molecular dynamic simulations and post-analysis.

Owing to their inhibitory role in regulating oligodendrocyte differentiation and apoptosis, protein tyrosine phosphatase sigma (PTPσ) and leukocyte common antigen-related phosphatase (LAR) play a crucial potential role in treating spinal cord injury (SCI) disease. In this research, the computer aided drug design (CADD) methods were applied to discover the potential dual-target drug involving virtual screen, molecular docking and molecular dynamic simulation. Initially, the top 20 compounds with higher docking score than the positive controls (ZINC13749892, ZINC14516161) were virtually screened

Protein tyrosine phosphatase receptor type C (PTPRC or CD45).

The leucocyte common antigen, protein tyrosine phosphatase receptor type C (PTPRC), also known as CD45, is a transmembrane glycoprotein, expressed on almost all haematopoietic cells except for mature erythrocytes, and is an essential regulator of T and B cell antigen receptor-mediated activation. Disruption of the equilibrium between protein tyrosine kinase and phosphatase activity (from CD45 and others) can result in immunodeficiency, autoimmunity, or malignancy. CD45 is normally present on the cell surface, therefore it works upstream of a large signalling network which differs between cell

A method for the dispersal and characterization of leukocytes from the human female reproductive tract.

PROBLEM: The isolation of human female reproductive tract (RT) cells that maintain viability and are representative of the entire population is essential for a thorough evaluation of mucosal immunity in the reproductive tract mucosa. Here, we describe the isolation of RT cells in high yields and with high viability from the Fallopian tube, uterine endometrium, endocervix, ectocervix and vagina. METHOD OF STUDY: This cell dispersion method uses an enzyme cocktail composed of pancreatin, hyaluronidase, and collagenase (PHC), and employs a 250-microm mesh screen to facilitate cell dispersion. Portal hypertension in common variable immunodeficiency disorders - a single center analysis on clinical and immunological parameter in 196 patients.

BACKGROUND: Liver manifestations and in particular portal hypertension (PH) contribute significantly to morbidity and mortality of patients with common variable immunodeficiency disorders (CVID). Screening strategies and early detection are limited due to the lack of specific diagnostic tools. METHODS: We evaluated clinical, immunological, histological, and imaging parameters in CVID patients with clinical manifestation of portal hypertension (CVID+PH). RESULTS: Portal hypertension was present in 5.6% of CVID patients and was associated with high clinical burden and increased mortality (18%)

Genetic predispositions for the immunological features of chronic active hepatitis.

To assess the frequency and genetic predispositions of concurrent immunological diseases and immunoserological markers in autoimmune hepatitis and chronic viral hepatitis, we assessed 185 patients prospectively, including 122 patients with autoimmune hepatitis and 63 patients with viral disease. Human leukocyte antigens were determined in all patients. Sixty patients (32%) had concurrent immunological diseases, and the majority of the diseases (68%) had known human leukocyte antigen associations. Although patients with autoimmune hepatitis had concurrent immunological diseases more commonly th

Immunohistology of rheumatoid nodules and rheumatoid synovium.

The immunohistological features of rheumatoid nodules and rheumatoid synovium were examined using monoclonal and polyclonal antibodies raised against macrophages, HLA-DR, leucocyte common antigen, and immunoglobulin components. The palisading cells surrounding the necrotic centre of the rheumatoid nodule were shown to be HLA-DR positive leucocytes, mostly histiocytes. The inflammatory infiltrate associated with rheumatoid nodules showed many immunohistochemical similarities to that of rheumatoid synovium, including a preponderance of IgG positive plasma cells, and a similar number and microana

In vitro assessment of parathyroid immunogenicity: the effect of cryopreservation.

Post-parathyroidectomy hypoparathyroidism, although fortunately uncommon, is a disorder of major inconvenience and potential morbidity. Attempts at modifying parathyroid tissue to facilitate allotransplantation without host immunosuppression are warranted. Cryopreservation has been reported to improve survival of canine parathyroid allografts. We employed a modification of the mixed lymphocyte culture to study the effect in vitro of cryopreservation on human parathyroid tissue. Dispersed parathyroid cells from fresh and previously cryopreserved tissue from 10 patients were incubated with unrel

Expression of OKM5 antigen on epidermal cells in mycosis fungoides plaque stage.

To characterize and quantitate potential antigen-presenting cell subsets in the epidermis of patients with cutaneous T-cell lymphoma, epidermal cells in suspension were obtained from involved and uninvolved skin. Involved epidermis contained increased numbers of OKT6+HLA-DR+ Langerhans cells and a variable number of OKM5+ epidermal cells (ECs) in all mycosis fungoides (MF) patients tested (N = 14). The OKM5+ EC population from involved epidermis of MF patients were heterogeneous and comprised both OKM5+HLe1- keratinocytes and OKM5+HLe1+ leukocytes. Uninvolved epidermis, in 6 of 14 patients wit

Role of the CD45 tyrosine phosphatase in signal transduction in the immune system.

CD45, a family of hematopoietic cell specific surface antigens, is the best characterized protein tyrosine phosphatase described to date. Considerable evidence from a number of laboratories suggests that surface expression of this molecule is required for normal signaling events to occur in T lymphocytes, B lymphocytes, and natural killer cells. In this paper, data supporting this contention are reviewed and a model proposing how CD45 may exert its regulatory function on signal transduction via the T cell antigen receptor is presented.

CD45 on CD33br HLA-DR+ immune cells promote cholecystitis via glycine-mediated pathways: A Mendelian randomization study.

Over the past 2 decades, the prevalence of cholecystitis has increased significantly, a trend believed to be influenced by improvements in quality of life and changes in lifestyle habits. However, the precise etiology of cholecystitis remains unclear. Although gallstones are commonly associated with this condition, recent studies suggest that immune cells also play a critical role in their development. This study investigated the relationship between cholecystitis and CD45 on CD33br HLA-DR+ immune cells, focusing on the mediating role of glycine levels, using Mendelian randomization (MR). The

Direct ex vivo flow cytometric analysis of human microglial cell CD4 expression: examination of central nervous system biopsy specimens from HIV-seropositive patients and patients with other neurological disease.

OBJECTIVE: To define a clear ex vivo flow cytometric phenotype for adult human microglia that would distinguish it from all other macrophage lineage cells in the central nervous system (CNS) or blood, and to utilize this phenotype to examine the activation state and CD4 expression of microglia freshly derived from CNS tissue of HIV-positive patients and those with other neurological diseases. DESIGN: Fresh human CNS tissue from both HIV-uninfected and HIV-infected individuals was obtained by biopsy or resection, and cells isolated immediately, labelled for flow cytometry and analysed. miR-155-5p and miR-674-3p presence in peripheral blood and white blood cells as potential early biomarkers of temporal lobe epilepsy.

Epilepsy frequently develops as a result of a brain insult, for example, brain injury or stroke. Currently, no tools are allowing us to predict which trauma patients will eventually develop epilepsy. There is evidence that microRNA levels are altered in the blood, making them attractive candidates for peripheral biomarkers of epilepsy. We analyzed whole blood miRNA levels using a dual-platform approach and selected miR-155-5p and miR-674-3p as candidates for biomarkers of early changes in asymptomatic temporal lobe epilepsy. Additionally, we assessed white blood cell subpopulations containing

The evaluation of leukocytes in response to the in vitro testing of ventricular assist devices.

Infection is a clinically relevant adverse event in patients with ventricular assist device (VAD) support. The risk of infection could be linked to a reduced immune response resulting from damage to leukocytes during VAD support. The purpose of this study was to develop an understanding of leukocyte responses during the in vitro testing of VADs by analyzing the changes to their morphology and biochemistry. The VentrAssist implantable rotary blood pump (IRBP) and RotaFlow centrifugal pump (CP) were tested in vitro under constant hemodynamic conditions. Automated hematology analysis of samples c

Bare lymphocyte syndrome with lack of HLA class I and II antigens. Presentation of two cases.

Bare lymphocyte syndrome (BLS) is a rare disorder characterized by deficient expression of human leukocyte antigens (HLA antigens) and combined immunodeficiency to various degrees. Recurrent severe infections especially due to opportunistic organisms are common. Here, we present two patients with BLS who lack both class I and II antigens (Type III). They had the typical clinical and immunologic findings of BLS. The first patient showed marked improvement in pulmonary symptoms resulting from cytomegalovirus infection by means of gancyclovir treatment. However, intramuscular injections of interf

Enhanced expression of resident leukocyte interferon gamma mRNA in endometriosis.

PROBLEM: Previous studies have shown that the endometrial epithelial/stromal cell proliferative activity of endometriosis is significantly less than that of normal endometrium and that the concentration of resident stromal leukocytes is significantly greater in ectopic than in eutopic endometrium. Other work has shown that interferon gamma (IFN gamma), secreted by resident leukocytes, inhibits endometrial cell proliferation in vitro. Accordingly, we hypothesized that the lower proliferative activity of endometriosis may be related to enhanced resident leukocyte IFN gamma production. This study
