Control of Viral Aerosol Dispersion During Simulated Dental Procedures.

AIM: Although air-cleaning systems (ACS) and high-volume evacuation (HVE) have been shown to reduce dental bioaerosols, few works have looked at viruses. This study aimed to assess the effects of HVE and ACS on dispersion of aerosolised particles and airborne viruses in a simulation model of dental bioaerosols using a detailed and comprehensive sampling approach and different tracer molecules. METHODS: MS2 bacteriophage and fluorescein solution were used as viral or dispersion tracers, respectively. These were added independently to the instrument irrigation system and aerosolised during dent

Study on the air gap effect when closing toilet lid on droplet and pathogen escaping from flushing.

BACKGROUND: Closing the toilet lid before flushing is widely recommended to minimize the dispersal of pathogen-laden aerosols. However, an often-overlooked issue is that aerosols can still escape through the air gap between the bowl and seat even when the lid is closed. METHODS: We conducted a pilot investigation on how the gap height and lid influence droplet and bioaerosol escape in a seated toilet. Three microbes were studied, including one bacterium Escherichia coli, and two viruses (MS2 and P22). Bioaerosol escape ratios were calculated as the normalized concentration with the lid closed

MS2 virus-like particles as a versatile platform for multi-disease vaccines: a review.

The coat protein of the MS2 self-assembles into virus-like particles (VLPs) with a diameter of 26 nm. These VLPs are devoid of the phage genome yet are efficiently recognized by the immune system, eliciting robust humoral and cellular immune responses. The structural characteristics of VLPs position them as a promising platform for the development of vaccines and diagnostic tools. Through genetic engineering, antigenic peptides up to 91 amino acids in length can be densely displayed at the N-terminal β-hairpin (AB loop) of the coat protein. Moreover, the fusion of an exogenous sequence with th

Impact of daily disinfection of portable equipment and common areas with a prototype handheld far ultraviolet-C light device on transmission of a DNA marker and bacteriophage MS2 on a long-term care ward.

BACKGROUND: Shared portable medical equipment is a vector for transmission of health care-associated pathogens, but cleaning and disinfection of equipment is often suboptimal. METHODS: We conducted a cross-over trial on a long-term care facility ward to determine the impact of daily low-level disinfection of portable equipment and common areas using a handheld far ultraviolet-C (UV-C) light device versus standard cleaning and disinfection practices. The primary outcome was transmission of a DNA marker and bacteriophage MS2 from inoculated portable vital signs equipment to surfaces on the ward

Preventing microbial contamination during radiological imaging: Experimental evaluation of a multiuse contrast media infusion system incorporating sequential one-way valves within a dual-safety design.

BACKGROUND: Health care-associated infections are a significant cause of patient morbidity, mortality, and health care expenditure. In diagnostic imaging, multiuse contrast media infusion systems are increasingly common; however, their use raises concerns regarding cross-contamination risk. METHODS: A controlled experimental model was used to assess the ability of a multiuse infusion system to prevent microbial contamination during simulated clinical conditions. Escherichia coli and MS2 bacteriophage were selected to model bacterial and viral contamination risks, respectively. Inocula were in

Investigation of enteric adenovirus and poliovirus removal by coagulation processes and suitability of bacteriophages MS2 and φX174 as surrogates for those viruses.

We evaluated the removal of enteric adenovirus (AdV) type 40 and poliovirus (PV) type 1 by coagulation, using water samples from 13 water sources for drinking water treatment plants in Japan. The behaviors of two widely accepted enteric virus surrogates, bacteriophages MS2 and φX174, were compared with the behaviors of AdV and PV. Coagulation with polyaluminum chloride (PACl, basicity 1.5) removed AdV and PV from virus-spiked source waters: the infectious AdV and PV removal ratios evaluated by means of a plaque-forming-unit method were 0.1-1.4-log10 and 0.5-2.4-log10, respectively. Titanium Dioxide Solar Photocatalytic Microbial Inactivation Assessment Utilizing Viability Tests and a Novel Triplex qPCR Assay for Nucleic Acid Degradation Determination.

Photocatalytic oxidation of microorganisms is a powerful alternative to established disinfection approaches, applicable to a variety of water matrices. Bacterial vegetative cells, spores, fungi, and viruses, represent potential biopathogens and photocatalysis targets. Inactivation efficiency is usually evaluated by assessing viability through culture. However, additional inactivation assessment approaches are needed, as some microbes, despite being unculturable, remain metabolically active and pathogenic. Nucleic acid quantification approaches (qPCR) can assess nucleic acid release and degrada

Rapid virucidal activity of an air sanitizer against aerosolized MS2 and Phi6 phage surrogates for non-enveloped and enveloped vertebrate viruses, including SARS-CoV-2.

An air sanitizer was evaluated using an aerobiology protocol, compliant with the U.S. Environmental Protection Agency's Air Sanitizer Guidelines, for virucidal activity against bacteriophages Phi6 and MS2 (used as surrogates for enveloped and non-enveloped human pathogenic viruses). The phages were suspended in a medium containing a tripartite soil load simulating body fluids and aerosolized using a six-jet Collison nebulizer in an enclosed 25 m3 aerobiology chamber at 22 ± 2°C and 50 ± 10% relative humidity. The air sanitizer was sprayed into the chamber for 30 s. Viable phages in the air wer

Conceptual model and experimental framework to determine the contributions of direct and indirect photoreactions to the solar disinfection of MS2, phiX174, and adenovirus.

Sunlight inactivates waterborne viruses via direct (absorption of sunlight by the virus) and indirect processes (adsorption of sunlight by external chromophores, which subsequently generate reactive species). While the mechanisms underlying these processes are understood, their relative importance remains unclear. This study establishes an experimental framework to determine the kinetic parameters associated with a virus' susceptibility to solar disinfection and proposes a model to estimate disinfection rates and to apportion the contributions of different inactivation processes. Analytical Centrifugal Ultrafiltration as a Tool for High Throughput Process Development in Virus Removal Filtration.

Virus removal filtration constitutes an essential component in biopharmaceutical manufacturing processes for both recombinant and plasma-derived protein products. The increasing demand for protein-based therapies necessitates expedited analytical screening methodologies for virus filtration materials and process parameters utilizing high-throughput development (HTPD) platforms. This study examines a novel HTPD methodology, specifically the implementation of analytical centrifugal ultrafiltration, as a scaled-down virus removal filtration tool. Sunlight Inactivation of Influenza A Virus and Bacteriophages on an Acrylic Surface.

Seasonal epidemics and pandemics of influenza cause a large public health burden, with transmission occurring via contact with contaminated fomites. Sunlight has been shown to inactivate viruses in water, but its effect on viruses on surfaces remains poorly understood. This study investigated the inactivation of bacteriophage MS2, bacteriophage Phi6, and influenza A virus H1N1 (IAV) on an acrylic surface exposed to both natural and simulated sunlight. Findings demonstrate that sunlight can rapidly inactivate viruses on an acrylic surface, with the time required for 99% inactivation ranging fro

Short-time thermal inactivation of surrogates of the public transport microbiome with a low-cost thermoresistometer.

In this study, the thermal inactivation of the bacterial genera Staphylococcus, Enterococcus and Burkholderia, which can be found in public transportation environments, as well as the bacteriophage MS2 as a surrogate for potential viral pathogens are investigated. To quantify the thermal inactivation characteristic, an automated and inexpensive thermoresistometer is constructed and set up, which enables the microorganisms to be exposed to short-term thermal shocks. The time dependent temperature curves were measured to account for heat-up and cooling times. Afterwards, the microorganisms were

Refining and in-situ growth of polyaniline endows the cellulose fibers with electrical stimulation sterilization.

Bacteria and virus infections have posed a great threat to public health and personnel safety. For realizing rapid sterilization of the bacteria and virus, electrical stimulation sterilization was adopted to endow cellulose fibers with instantaneous antibacterial and antiviral properties. In the proposed strategy, the fiber is fluffed by mechanical refining, and then by means of the hydrogen bond between hydroxyl and aniline, the polyaniline (PANI) directionally grows vertically along the fine fibers via in-situ oxidative polymerization. Benefiting from the conductive polyaniline nanorod array

Development and application of a quadruple RT-qPCR assay for the simultaneous detection of NoV GI, NoV GII, and HAV in bivalve shellfish.

To achieve rapid and simultaneous detection of NoV GI, NoV GII, and HAV, a quadruple real-time fluorescence quantitative PCR (RT-qPCR) assay was developed using MS2 bacteriophage as a process control virus. The quadruple RT-qPCR assay effectively detected NoV GI, NoV GII, HAV, and MS2 RNA with detection limits of 102 copies/μL, 103 copies/μL, 102 copies/μL, and 103 copies/μL, respectively, within 1 hour 50 minutes. The quadruple RT-qPCR assay could specifically detect NoV GI, NoV GII, HAV, and MS2 without cross-reactions with other common pathogens, demonstrating good reproducibility with intr

Evaluation of hospital-grade disinfectants on viral deposition on surfaces after toilet flushing.

BACKGROUND: Past studies have shown that infectious aerosols created during toilet flushing result in surface contamination of the restroom. The goals of this study were to quantify viral contamination of surfaces in restrooms after flushing and the impact of disinfectants added to the toilet bowl prior to flushing on reducing surface contamination. METHODS: The degree of contamination of surfaces in the restroom was assessed with and without the addition of coliphage MS2 to the toilet bowl before flushing. The bowl water and various surfaces in the restroom were subsequently tested for the p

Survival of enteric bacteria and coliphage MS2 in pure human urine.

AIMS: The survival of Escherichia coli, Salmonella enterica serovar Typhimurium, Enterococcus faecalis and coliphage MS2 was studied in stored, fresh and diluted (1 : 1) human urine at 15 and 30 degrees C. METHODS AND RESULTS: Survival rate was studied by the plate count method. All the organisms showed rapid inactivation in stored urine, but they survived better in diluted and fresh urine. The high pH level and temperature were the major factors found to influence the survival of the micro-organisms with the survival rate being higher at 15 degrees C than at 30 degrees C. CONCLUSIONS: The d

Identification of vaccine candidates by immunization with a complex pool of recombinant VLPs.

Identifying the regions of a protein that can be readily translated into an effective vaccine remains a major challenge in epitope-focused vaccine design, especially when a protein is poorly characterized. Here, we describe an immune-driven strategy for epitope identification which uses the immune system to reveal regions within a candidate protein capable of eliciting targeted antibody responses. Using IL-17A, a pro-inflammatory cytokine, as a model antigen, we engineered a comprehensive library of MS2 bacteriophage virus-like particles (VLPs) displaying peptides representing every possible l

Impact of milk components on recovery of viral RNA from MS2 bacteriophage.

Noroviruses are responsible for approximately 44 % of outbreaks involving dairy products for which causative agents are reported. Recovery of viruses from milk and dairy products is a difficult task. The role of different components of milk in the recovery of viral RNA was evaluated in this study. Four model milk formulations (A-D) were prepared by mixing different combinations of lactose, whey protein, casein, and fat in water. Each model formulation was spiked with five concentrations of bacteriophage MS2. The phenol-guanidine thiocyanate-chloroform protocol was used for extracting viral RNA

Visible light sensitized inactivation of MS-2 bacteriophage by a cationic amine-functionalized C60 derivative.

Recently, we reported the successful synthesis of various hexakis C60 derivatives (i.e., C60 with six functional groups containing NH3+-, CO2H-, or OH-terminals) with enhanced stability in water for aqueous phase application (Lee et al., Environ. Sci. Technol. 2009, 43, pp 6604-6610). Among these newly synthesized C60 derivatives, the cationic hexakis C60 derivative with amine functionality, C60(CR2)6 (R=CO2(CH2)2NH3+CF3CO2-), was found to exhibit remarkable efficiency to inactivate Escherichia coli and MS-2 bacteriophage under UVA irradiation. Herein, we report that this amine-functionalized

Evaluation of electrochemically generated ozone for the disinfection of water and wastewater.

Effective wastewater treatment is critical to public health and well-being. This is especially true in developing countries, where disinfection of wastewater is frequently inadequate. People who live in these areas may benefit from wastewater disinfection using ozone. This study evaluated the ability of a new electrochemical process of ozone generation, which produced ozone continuously at high pressure and concentration by the electrolysis of water, to disinfect tap water and secondarily treated wastewater. Inactivation of Klebsiella terrigena, Escherichia coli, MS2 bacteriophage and poliovir
