INTRODUCTION: Citrus huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), is a globally devastating plant disease characterized by severe disruption of host physiology. Although alterations in plant hormone homeostasis, including salicylic acid and abscisic acid (ABA), are hallmarks of HLB, the contribution of plant-associated microbiota to these hormonal changes remains poorly understood. Here, we investigated whether ABA depletion in HLB-affected citrus is associated with microbial metabolism.
METHODS: ABA and its metabolites were quantified in HLB-affected citrus tissue

The phloem-colonizing bacterial pathogen Candidatus Liberibacter asiaticus (CLas) causes the devastating citrus Huanglongbing (HLB). Despite the important roles of roots, how CLas affects roots remains largely unknown. Here we dissected host responses in leaf and root at different time points after CLas inoculation. Our data showed that CLas triggers less reactive oxygen species (ROS) accumulation and phloem callose deposition, which are known to contribute to HLB symptom development in leaves, in roots than leaves. Increased chloroplastic ROS accumulation was detected in CLas-infected leaf co

Ubiquitin-proteasome is a conserved mechanism that regulates cellular responses and disease resistance in plants. However, the regulatory role of ubiquitin-proteasome in the pathogenicity of "Candidatus Liberibacter asiaticus" (CLas), the causal agent of citrus huanglongbing (HLB), one of the most serious citrus diseases, remains poorly defined. In this study, we demonstrated that CLIBASIA_04250 (SDE4250) suppresses 26S proteasome activity in citrus, leading to a reduction in salicylic acid (SA) levels and downregulation of SA-responsive genes. Further investigation revealed that SDE4250 speci

Among citrus diseases, Huanglongbing (HLB) is recognized as the most destructive and economically damaging worldwide. It is mainly associated with 'Candidatus Liberibacter asiaticus' (CLas) being transmitted by Diaphorina citri. There are no curative treatments or commercial citrus varieties resistant to CLas. Wild Aurantioideae species have been widely screened and, recently, Oceanian genotypes graft- and sexually compatible with Citrus were identified as HLB-resistant; however, there is no information regarding their use as interstocks of commercial varieties yet. Under greenhouse conditions

Huanglongbing (HLB, also known as citrus greening) is considered to be the most devastating disease that has significantly damaged the citrus industry globally. HLB is caused by the Candidatus Liberibacter asiaticus (CLas), the fastidious phloem-restricted gram-negative bacterium, vectored by the asian citrus psyllid. To date, there is no effective control available against CLas. To alleviate the effects of HLB on the industry and protect citrus farmers, there is an urgent need to identify or develop inhibitor molecules to suppress or eradicate CLas from infected citrus plant. In this paper, w

'Candidatus Liberibacter solanacearum' (Lso) is a phloem-limited bacterium that severely affects important Solanaceae and Apiaceae crops, including potato, tomato, pepper, tobacco, carrot and celery. This bacterium is transmitted to solanaceous species by potato psyllid, Bactericera cockerelli, and to Apiaceae by carrot psyllids, including Trioza apicalis and Bactericera trigonica. Five haplotypes of Lso have so far been described, two are associated with solanaceous species and potato psyllids, whereas the other three are associated with carrot and celery crops and carrot psyllids. Little is

BACKGROUND: The Toll signaling pathway is a key component of insect innate immunity, yet its role in hemipteran vectors and their interactions with plant pathogens remains poorly understood. The potato psyllid (Bactericera cockerelli), an important agricultural pest in the USA, transmits two haplotypes of 'Candidatus Liberibacter solanacearum' (LsoA and LsoB) that cause severe diseases in solanaceous crops.
METHODS AND RESULTS: We characterized Toll pathway components in the B. cockerelli genome and examined their transcriptional responses during pathogen acquisition. Six Toll receptors and t

Zebra chip disease (ZC) in potato (Solanum tuberosum L. [Polemoniales: Solanaceae]) can produce unmarketable tubers with striped necrotic patterns. ZC is associated with the bacterium "Candidatus Liberibacter solanacearum" (Lso), which is transmitted by the potato psyllid, Bactericera cockerelli (Šulc) (Hemiptera: Triozidae). Potato psyllids are associated with numerous noncrop host plants, especially from the Solanaceae and Convolvulaceae; however, the contribution and importance of these hosts to ZC epidemiology in potato is poorly understood. To clarify seasonal phenologies on two such host

Huanglongbing (HLB) is a devastating citrus disease associated with the gram-negative, phloem-limited, and unculturable bacterium "Candidatus Liberibacter asiaticus (CLas)," which is transmitted by the Asian citrus psyllid Diaphorina citri. Despite extensive research, effective, long-term, and sustainable solutions for managing HLB remain elusive. Oxytetracycline (OTC) is currently used as an emergency measure, but there is an urgent need for alternative compounds to complement or replace OTC. In this study, we identified amicoumacins, a class of antimicrobial compounds produced by the bacteri

Insecticide applications are commonly recommended for managing Diaphorina citri, the vector of huanglongbing (HLB), but their effectiveness in reducing transmission of 'Candidatus Liberibacter asiaticus' (CLas), especially during continuous psyllid influx and shoot growth, remains unclear. This study evaluated the efficacy of foliar application of thiamethoxam and spinetoram in reducing CLas transmission in sweet orange seedlings. Two experiments were conducted up to 13 days after first spray. In experiment 1, CLas-positive psyllids were confined in seedlings every 2 days. Treatments were: con

Plant microbiomes are a rich source of secondary metabolites, often with pathogen- or disease-suppressive traits. Huanglongbing (HLB) is a serious disease of citrus for which there is currently no cure. Here, we tested the hypothesis that bacteria native to the citrus microbiome may possess inhibitory activity against the associated causal agent of HLB, Candidatus Liberibacter asiaticus (CLas). We coupled a robotic microfluidic isolation platform with a high-throughput antimicrobial screening assay using Liberibacter crescens, a culturable surrogate for the unculturable CLas, to identify isola

Citrus Huanglongbing (HLB), caused by "Candidatus Liberibacter asiaticus" (CLas), is a devastating citrus disease worldwide, and its pathogenesis remains poorly understood. Here, we investigated the virulence function of a Sec-dependent effector, metal-dependent hydrolase 1 (MDH1), which is significantly upregulated in citrus during CLas infection compared to in psyllids. Overexpression of MDH1 in citrange reduced its resistance to Xanthomonas citri subsp. citri. RNA-seq and RT-qPCR analyses revealed that MDH1 reprogramed the transcription of genes involved in plant-pathogen interactions and s

Diaphorina citri (Hemiptera: Liviidae) is the main vector for the bacterium 'Candidatus Liberibacter asiaticus', which is associated with citrus greening, also known as Huanglongbing. D. citri transmits 'Ca. L. asiaticus' during its feeding on citrus phloem sap. Transmission occurs in a circulative, propagative, and persistent manner. 'Ca. L. asiaticus' has a small genome (1.2 Mb). Therefore, it acquires most of its nutrients and energetic nucleotides from its hosts. The objective of this study was to assess the effect of 'Ca. L. asiaticus' infection on the level of the free fatty acids in its

Huanglongbing (HLB), associated with "Candidatus Liberibacter asiaticus" (CLas), is a globally devastating plant disease. The highly reduced genome of CLas encodes a number of secretory proteins. The conserved prophage-encoded protein AGH17470 is herein identified as a nonclassical secretory protein. We confirmed that the N-terminal and C-terminal sequences jointly determine the secretion of AGH17470. The transient expression of AGH17470 protein in Nicotiana benthamiana caused hypersensitive response (HR) cell death in infiltrated leaves and systemically infected leaves as well as the dwarfing

Potato psyllid (Bactericera cockerelli Sulc)-transmitted "Candidatus Liberibacter solanacearum" (Lso) has been negatively impacting the potato industry in the United States as well as other potato-producing countries. Lso has been linked to a condition known as zebra chip (ZC) that affects yield and quality of potato tubers. Efforts to find sources of resistance to ZC have primarily focused on greenhouse evaluations based on a single inoculation time prior to harvest. Plant response to infection, however, could be influenced by the developmental stage of the host plant, and ZC may continue to

Huanglongbing, a destructive citrus disease of global importance that is also present in Saudi Arabia, is associated with Candidatus Liberibacter asiaticus (CLas) and remains a major threat to citrus production. Although previous studies have documented sequence variation and prophage polymorphism in CLas, broader comparisons of prophage-associated gene content remain limited. In particular, comparative orthogroup analysis of prophage gene-content conservation across geographically structured CLas populations has rarely been explored. In this study, we analyzed 42 CLas prophage genomes from Sa

INTRODUCTION: Huanglongbing (HLB) is a devastating disease impacting citrus orchards worldwide. Identifying candidate genes and/or genomic regions that significantly influence plants' response to HLB is essential for breeding programs. Nevertheless, this remains a challenge, especially during the early stages of the citrus-pathogen interaction, when phenotypically distinguishing tolerant genotypes is not trivial. This work focused on identifying genomic regions and candidate genes associated with citrus response to Candidatus Liberibacter asiaticus (CLas).
METHODS: Quantitative trait loci (QT

Ponkan mandarin (Citrus reticulata cv. Ponkan) is an important fruit crop planted across different countries. One of its most devastating diseases is citrus huanglongbing (HLB), which is associated with infection by the bacterium Candidatus Liberibacter asiaticus (CLas). Although studies on HLB in Ponkan mandarin have been conducted, symptom heterogeneity and its association with CLas titer under field conditions remain underexplored. We examined the association between symptom variation and CLas infection status in a Ponkan mandarin grove by categorizing symptoms at both the leaf and canopy (

Citrus huanglongbing (HLB) is recognized as the most destructive bacterial disease affecting citrus. Among the causal agents, Candidatus Liberibacter asiaticus (Las) is the predominant species causing HLB epidemics worldwide. Detecting this phloem limited bacterium at early or low titer stages and assessing its cellular activity remain major challenges for HLB research and management. To address these limitations, we developed a one step reverse transcription quantitative PCR (RT qPCR) assay for highly sensitive Las detection using total nucleic acids and the established Li 16S rRNA/rDNA prime
