Thread-Based Bienzymatic Biosensor for Linoleic Acid Detection.

The concentration of nonesterified fatty acids (NEFAs) in biological media is associated with metabolic and cardiovascular disorders (e.g., diabetes, cancer, and cystic fibrosis) and in food products is indicative of their quality. Therefore, the early identification of NEFAs is crucial for both medical diagnosis and food quality assessment. However, the development of a portable and scalable sensor capable of detecting these compounds at a low cost presents challenges due to their considerable chemical and physical stability. This research endeavors to illustrate the viability of detecting li

The Role of Linoleic Acid in Skin and Hair Health: A Review.

Lipids are intimately associated with skin condition. This review aims to discuss the function of linoleic acid (LA, 18:2, ω-6), an essential fatty acid, in skin health and hair growth. In skin, LA can be metabolized into ω-6 unsaturated fatty acid, oxidized derivatives and incorporated into complex lipid molecules, including ω-hydroxy-ceramides. Previous research has revealed that skin diseases including acne, atopic dermatitis and psoriasis are associated with disordered LA metabolism. Studies based on animal or skin cell models suggest that LA or LA-rich vegetable oils, topically applied, e

Linoleic acid as corpse recognition signal in a social aphid.

Social insect colonies constantly produce dead insects, which cause sanitary problems and potentially foster deadly pathogens and parasites. Hence, many social insects have evolved a variety of hygienic behaviors to remove cadavers from the colonies. To that end, they have to discriminate dead insects from live ones, where chemical cues should play important roles. In ants, bees and termites, such corpse recognition signals, also referred to as "death pheromones" or "necromones", have been identified as fatty acids, specifically oleic acid and/or linoleic acid. Meanwhile, there has been no suc

The beneficial and detrimental effects of linoleic acid on autoimmune disorders.

Type 1, or cellular, immune response is characterized by overproduction of IL-1, IL-2, IFN-gamma and TNF-alpha and is the underlying immune mechanism of some autoimmune disorders such as psoriasis, alopecia areata, rheumatoid arthritis, Crohn's disease, multiple sclerosis, insulin-dependent diabetes mellitus and experimental autoimmune uveitis. Type 2 immune response is seen in allergic and antibody-mediated autoimmune diseases and is characterized by IL-4, IL-6 and IL-10 overproduction. Linoleic acid is a precursor of prostaglandin E2 (PGE2) and its intake results in tissue production of PGE2

Toxic response of linoleic acid anilide in female rats.

The toxicity of linoleic acid anilide (LAA) and heated linoleic acid anilide (HLAA) was studied in female rats. Female Sprague-Dawley rats were given 250 mg/kg of LAA or HLAA in mineral oil, by gavage, on alternate days for two weeks. Control rats received an equal volume of mineral oil. The animals were sacrificed at day 1, 7 and 28 following the last dose. Organ-to-body weight ratio was increased for spleen in both LAA and HLAA treated rats at day 1. Lung, kidney and brain showed increases in this ratio at some time point, whereas, thymus in the HLAA group showed a decrease at day 28. Among

Plasma linoleic acid is related to lower hepatic steatosis index and higher linoleate-rich cardiolipin in adults with MAFLD.

Linoleic acid (LA, C18:2n-6) is a required nutrient that supports mitochondrial function as a component of the inner mitochondrial membrane, cardiolipin (CL). Tetralinoleoyl CL (LA4CL) supports optimal fatty acid oxidation, which is impaired in the liver in adults with metabolic dysfunction-associated fatty liver disease (MAFLD). Data were analyzed from 74 adults with MAFLD recruited from the Columbus region, Ohio, United States. Fatty acid content and CL species were measured in blood and peripheral blood mononuclear cells (PBMCs). Liver fat was assessed using magnetic resonance imaging-proto

Human essential fatty acid deficiency: treatment by topical application of linoleic acid.

An essential fatty acid (EFA) deficiency developed in a 19-year-old man who was being maintained on a long-term regimen of fat-free, intravenous hyperalimentation fluids. The EFA deficiency was reversed after 21 days by daily, topical application of linoleic acid to the patient's skin. The ratio of eicosatrienoic acid (20:3, n-9) to eicosatetraenoic acid (20:4, n-6) decreased to normal levels in the skin and serum with clinical improvement of the EFA deficiency syndrome. The cutaneous manifestations (scalp dermatitis, alopecia, and depigmentation of hair) were reversed with continued, topical

Technical note: Improved extraction method with hexane for gas chromatographic analysis of conjugated linoleic acids.

Extraction properties of different solvents (chloroform/methanol, hexane/isopropanol, and hexane) were studied for the gas chromatographic analysis of conjugated linoleic acids (CLA) from probiotic bacteria grown in de Man, Rogosa, and Sharpe medium. As compared with chloroform/methanol and hexane/isopropanol, hexane showed comparable extraction efficiency for CLA from unspent de Man, Rogosa, and Sharpe medium, but showed minimal extraction of oleic acid originated from the emulsifier in broth. The extraction efficiency of CLA by hexane was influenced by the broth pH, showing the optimal pH of

Low cytotoxicity of linoleic acid-derived epoxy-fatty acids in liver and colon cell lines.

Linoleic acid-derived epoxy-fatty acids (EpOMEs) are found in oxidized plant oils and processed foods. Consumption of oxidized oils may lead to a variety of negative health effects. Due to the assumed chemical reactivity of the epoxy-group, toxicity of epoxy-fatty acids, such as 9(10)-EpOME and 12(13)-EpOME, is frequently discussed. Therefore, we investigated the effect of 9(10)-EpOME and 12(13)-EpOME on cell viability by evaluating cell proliferation, LDH leakage, lysosomal integrity and dehydrogenase activity in HepG2 and Caco2 cells. Both tested epoxy-fatty acids show no or low cytotoxicity

Bovine milk fat enriched in conjugated linoleic and vaccenic acids attenuates allergic dermatitis in mice.

BACKGROUND: Orally administered milk fat enriched in conjugated linoleic acid (CLA) and trans-vaccenic acid (VA) ('enriched milk fat'), produced by supplementing the diet of pasture-fed cows with fish and sunflower oil, has been shown previously to suppress the development of allergic airway disease in mice. OBJECTIVE: To investigate whether topical or oral application of enriched milk fat and its two major fatty acids cis-9, trans-11 CLA (c9,t11-CLA) and VA inhibit allergic dermatitis in mice. METHODS: Allergic dermatitis was induced in C57BL/6 mice by epicutaneous sensitization of tape-str

Effect of a High Linoleic Acid Diet on Pregnant Women and Their Offspring.

Nutritional intake during pregnancy can affect gestational length, fetal development, and impact postnatal growth and health in offspring. Perturbations in maternal nutrition with either an excess or deficiency in nutrients during pregnancy may have harmful effects on the offspring's development and increase the risk of developing chronic diseases later in life. In pregnancy, nutrients transfer from the mother to the fetus via the placenta. Essential fatty acids, linoleic acid (LA) and alpha linoleic acid (ALA), can only be obtained in the diet. In Western countries, the ratio of LA and ALA in

Linoleic Acid-Based Transferosomes for Topical Ocular Delivery of Cyclosporine A.

Delivering high-molecular-weight hydrophobic peptides, such as cyclosporine A, across the corneal epithelium remains a challenge that is complicated by other physio-anatomical ocular structures that limit the ocular bioavailability of such peptides. Transferosomes have previously been used to improve transdermal permeability, and have the potential for improving the ocular corneal permeability of applicable drugs. In this study, transferosomes for the potential ocular delivery of cyclosporine A were investigated. Linoleic acid was evaluated for its effect on the stability of the transferosomes

Fulminant hepatitis during self-medication with conjugated linoleic acid.

The present report describes a 63-year-old female who presented with fulminant hepatic failure requiring liver transplantation caused by a weight loss dietary supplement containing conjugated linoleic acid (CLA). Thorough investigation, including liver biopsy, revealed no other cause of hepatotoxicity. In the last few years, a considerable number of reports have been published on toxic hepatitis, associated with non-conventional products, attributed with weight-reducing properties. We emphasize the importance of taking a cautious approach when consuming herbal supplements for the purpose of we

Photochemical production of conjugated linoleic acid from soybean oil.

Conjugated linoleic acid (CLA), an anticarcinogenic compound with numerous other health benefits, is present mainly in dairy and beef lipids. The main CLA isomer present in dairy and beef lipids is cis 9, trans 11 CLA at a 0.5% concentration. The typical minimum human dietary intake of CLA is 10 times less than the 3 g/d suggested requirement that has been extrapolated from animal and cell-line studies. The objectives of this study were to produce CLA isomers from soybean oil by photoisomerization of soybean oil linoleic acid and to study the oxidation status of the oil. Refined, bleached, and

Mechanism of the inhibitory action of linoleic acid on the growth of Staphylococcus aureus.

Linoleic acid, but not stearic acid, inhibited the growth of Staphylococcus aureus NCTC 8325. Growth inhibition was associated with an increase in the permeability of the bacterial membrane. The presence of a plasmid conferring resistance to penicillin (PC plasmid, e.g. pI258blaI-) increased the growth inhibitory and membrane permeability effects of linoleic acid. Under growth inhibitory conditions, linoleic acid was incorporated into the lipid of both PC plasmid-containing and PC plasmid-negative bacteria and there was little difference between these cultures in the uptake or fate of linoleic

Conjugated Linoleic Acid-induced Hepatotoxicity Requiring Liver Transplant.

Acute liver failure (ALF) typically presents with encephalopathy and impairment in the synthetic function of the liver. Weight loss supplements have been associated with ALF, and their use has only been increasing in the United States. We report a case of a 42-year-old woman with a history of Gilbert's syndrome who presented to the hospital with ALF secondary to weight loss supplements, who ultimately required liver transplantation. This is the first known case of conjugated linoleic acid (CLA) toxicity requiring liver transplantation in the United States.

[14C] Linoleic Acid Uptake and Fractionation Assay in Vibrio cholerae.

The gram-negative curved bacillus Vibrio cholerae (V. cholerae) causes the severe diarrheal illness cholera. The work presented here is to assess whether unsaturated fatty acids (UFAs), such as linoleic acid, have the potential to directly affect proteins involved in DNA binding because they are able to enter the cell. In this protocol, we show how to measure linoleic acid entering V. cholerae when added exogenously and determine whether it is able to enter the cytoplasm. This protocol will quantify how much linoleic acid is able to enter the cell and then identify the amount of linoleic acid

Conjugated linoleic acid-rich soy oil triacylglycerol identification.

Conjugated linoleic acid (CLA)-rich soy oil has been produced by soy oil linoleic acid (LA) photoisomerization, but CLA-rich oil triacylglycerol (TAG) characterization was not described. Therefore, the objectives were to identify and quantify new TAG fractions in CLA-rich oil by nonaqueous reversed-phase high-performance liquid chromatography (NARP-HPLC). Analytical NARP-HPLC with an acetonitrile/dichloromethane (ACN/DCM) gradient and an evaporating light scattering detector/ultraviolet (ELSD/UV) detector was used. New TAG peaks from LA-containing TAGs were observed. The LnLL, LLL, LLO, and LL

Isomers of conjugated linoleic acid modulate human preadipocyte differentiation.

Conjugated linoleic acids (CLAs) reduce fat deposition in several mammalian species. Among the proposed mechanisms for this effect are reduced preadipocyte proliferation and differentiation. We measured proliferation and differentiation of cultured human preadipocytes treated with CLAs. Preadipocytes were differentiated with insulin, hydrocortisone, transferrin, and 10% fetal bovine serum, with isobutyl-methylxanthine included for the first 2 d. The differentiation medium contained 200 microM oleic acid (C18:1), 50 microM cis-9,trans-11-CLA (9,11-CLA), or 50 microM trans-10,cis-12-CLA (10,12-C
