Research into the phylogenetic relationships of lumpy skin disease virus (LSDV) strains was long overlooked, partially due to its original restricted distribution to sub-Saharan Africa. However, recent incursions into northern latitudes, and a rapid spread causing major economic losses worldwide, have intensified additional research on the disease and the causative virus. This study delineates the phylogeny of LSDV in the context of full genome sequences of strains recovered in the field, as well as strains highly passaged in cell culture. We sequenced the oldest known field strain to date (is

Lumpy skin disease (LSD) is a severe transboundary disease of cattle caused by lumpy skin disease virus (LSDV), which is highly transmissible and responsible for substantial economic losses worldwide. Although cellular immune responses are critical for controlling poxvirus infections, the viral strategies used by LSDV to evade cytotoxic T-cell immunity remain poorly understood. In this study, we demonstrate that LSDV infection markedly downregulates major histocompatibility complex class I (MHC-I) surface expression in host cells. Screening of LSDV-encoded proteins identified LSDV003 as a key

Lumpy Skin Disease Virus (LSDV), a Capripoxvirus of significant veterinary and economic importance, has been reported to manipulate host cellular processes, including autophagy, to enhance its replication and persistence. However, the precise mechanisms by which LSDV interacts with autophagy remain unclear. This study investigates the effect of LSDV infection on host autophagic pathways in MDBK cells, focusing on autophagic flux modulation and its implications for viral replication. Western blot and immunofluorescence analyses demonstrated that LSDV does not robustly induce autophagy but inter

The spread of a severe and often fatal form of lumpy skin disease (LSD) in cattle and water buffaloes has caused widespread mortality and morbidity of these animals in India. To track and understand the genetic changes occurring in the virus and to enable routine surveillance of the virus, multiplexed polymerase chain reaction (PCR) and sequencing methods were developed and validated in this study. Multiplexed nested PCR for LSD virus (LSDV) detection was optimized using skin lesion swabs and nasal samples collected from symptomatic and asymptomatic animals. For genotyping, overlapping PCRs to

Lumpy skin disease virus (LSDV), together with sheeppox virus and goatpox virus, belong to the genus Capripoxvirus within the family Poxviridae. Collectively, they are considered the most serious poxvirus diseases of agricultural livestock. Due to their severe clinical course and consequent loss of production, as well as high mortality of naïve small and large ruminant populations, they are known to have a significant impact on the economy and global trade restrictions of affected countries. Therefore, all capripox diseases are classified as notifiable under the guidelines of the World Organiz

Vaccination against lumpy skin disease (LSD) is crucial for maintaining the health of animals and the economic sustainability of farming. Either homologous vaccines consisting of live attenuated LSD virus (LSDV) or heterologous vaccines consisting of live attenuated sheeppox or goatpox virus (SPPV/GPPV) can be used for control of LSDV. Although SPPV/GTPV-based vaccines exhibit slightly lower efficacy than live attenuated LSDV vaccines, they do not cause vaccine-induced viremia, fever, and clinical symptoms of the disease following vaccination, caused by the replication capacity of live attenua

In this comparative study, we examine the safety of the sheeppox (SPP) and goatpox (GTP) vaccines and the protective response of these vaccines in cattle against a virulent lumpy skin disease (LSD) field strain. The vaccine safety was tested in rabbits, mice and cattle using ten times recommended dose. In the safety trial, none of the vaccinated animals showed any deviation from physiological norms or fever, inappetence or local/ generalized skin reactions. In the challenge trial, both SPP and GTP vaccine groups developed virus-neutralizing antibodies with an average titre of 2.1 log2 at 21 da

Lumpy skin disease (LSD) is caused by Lumpy Skin disease virus (LSDV) belonging to the genus Capripoxvirus (CaPV). The disease is widespread in Africa, the Middle East and Asia and has been present in Egypt since 1988. LSD is mainly transmitted by blood-sucking insects. LSD is clinically distinguished by a high fever, skin nodules, and swollen Lymph nodes. Detecting sub-clinical disease can be challenging however, prompt laboratory investigations are vital. Skin lesions are the main source of infection, although the virus is shed through many excretions and discharges including semen. Disease

Lumpy skin disease (LSD) is a viral disease of cattle and buffalo, caused by a Capripox virus. A field study was performed during an LSD epidemic which occurred in 2012-2013 in Israel, in order to assess the efficacy of two commercial vaccines for protection against LSD. Fifteen dairy herds, vaccinated 2-5 months prior to study onset with a single dose of 10(2.5) TCID50 of RM65 attenuated sheep-pox vaccine, and not affected previously, were enrolled in the study. 4694 cows were randomized to be either vaccinated with a 10(3.5) TCID50/dose of RM65 vaccine (x10RM65) or with a same dose of an att

In this paper, we report the resurgence of lumpy skin disease (LSD) in Kurgan Oblast, Russia, in 2018. The majority of the outbreaks were silent with no mortality and congregated within an area with a radius of about 30 km located 1-50 km away from the national border with Kazakhstan. Following primary molecular diagnosis, LSD virus (LSDV) isolates were analyzed using a panel of PCR assays targeting different genetic loci, namely, LSD008 (vaccine), LSDV126 (field), and GPCR (vaccine and field), for differentiation and genotype assignment. All isolates were positive for the vaccine genotype of

Lumpy skin disease (LSD) has affected many regions of Russia since its first occurrence in 2015. The most devastating year for Russia was 2016, when the virus resurged following a modified stamping-out campaign, causing 313 outbreaks in 16 regions. To avoid unwanted adverse reactions following the use of live attenuated vaccines against LSD virus (LSDV), sheeppox-based vaccines were administered during vaccination campaigns. As a result, LSD was successfully contained in all Russian regions in 2017. In the same year, however, LSD emerged anew in a few regions of the Privolzhsky Federal Distric

The objectives of this work were to determine the site of persistence of lumpy skin disease virus (LSDV) in bulls shedding the virus in semen for a period longer than 28 days, to determine if the virus is present in all fractions of semen and to study lesions that developed in the genital tract. Six serologically negative postpubertal bulls were experimentally infected with a virulent field isolate of LSDV. The polymerase chain reaction (PCR) was performed on sheath washes, vesicular fluid, supernatant and cell-rich fractions of semen from day 10 to day 26 postinfection (p.i.). Bulls that were

Lumpy skin disease (LSD) is an emerging transboundary disease in India. In the recent past, Andhra Pradesh is experiencing outbreaks of LSD in several pockets with a severe economic impact on the farming community. The affected animals showed nodular lesions all over the body in severely affected cases, whereas the lesions were confined to the face, neck, jowl, back, udder, and scrotum in less affected cases. Young ones are highly susceptible to the disease than adults. The mortality was found to be more in young ones than adult cattle which might be due to subsequent secondary bacterial compl

Lumpy skin disease (LSD) has recently expanded its range northwards to include the Balkans, Turkey and Russia. Because there was no solid evidence conclusively verifying the transmission mechanism in the field and LSDV viraemic animals with overt and asymptomatic presentation of disease and their products may represent a risk as an indirect transmission pathway. In this work, we used PCR positivity and infectivity in clinical and subclinical infection to evaluate the safety of meat and offal products from cows infected with the virulent LSDV strain Russia/Dagestan/2015. Lumpy skin disease is an economically important disease of cattle, caused by the lumpy skin disease virus (LSDV; Capripoxvirus). It has a variable clinical appearance but, in severely affected animals, is associated with extensive skin damage, pneumonia and death. The LSDV can be found in the semen of infected bulls for prolonged periods of time, from where it can be transmitted by mating or artificial insemination and cause clinical disease in heifers and cows. In this study, an ejaculate was collected from a LSDV seronegative bull and confirmed free from LSDV DNA by PCR. The ejaculate was sp

Lumpy skin disease virus (LSDV) infection poses a significant threat to the global cattle industry; however, effective therapeutic options remain limited. Cidofovir, a nucleotide analog that inhibits viral DNA polymerase, presents a promising antiviral strategy. This study aimed to elucidate the mechanistic basis of Cidofovir-mediated inhibition of LSDV. The results demonstrated that Cidofovir suppressed LSDV replication and reduced viral DNA polymerase expression in a dose-dependent manner. Transcriptome sequencing (RNA-seq) analysis revealed significant enrichment of apoptotic pathways follo

Lumpy skin disease (LSD) is a transboundary viral disease of cattle caused by lumpy skin disease virus (LSDV), resulting in substantial economic losses and posing a serious threat to the global cattle industry and international trade. Effective serological surveillance is essential for disease control and for evaluating vaccination programs. However, most currently available serological assays rely on a limited number of viral structural proteins as antigens, which restricts antigen diversity and may limit their applicability, particularly in the development of DIVA-compatible diagnostic strat

OBJECTIVE: The resurgence of lumpy skin disease virus isolates of different genotypic natures abolishes the accuracy of assays that target either vaccine or field strain genome. The aim of the present study was to develop a universal real-time PCR assay using TaqMan chemistry to cover field, vaccine, and recombinant strains of lumpy skin disease virus isolates.
RESULTS: The PCR assay was designed based on a LSDV044 target region that offers a unique identification locus to facilitate the sensitive and specific detection of all isolates known to date. The efficiency of amplification, determine

The objective of the present study was to access the titres and duration of maternally derived neutralizing antibodies against lumpy skin disease virus (LSDV) in calves born to immunized dairy cows. The study was conducted in a Greek farm of 200 Holstein cows which were immunized with a homologous Neethling strain-based attenuated vaccine. Composite colostrum samples were obtained from 18 selected cows at the day of calving. Blood samples were obtained from each dam-calf pair prior to the first colostrum feeding and from the calves successively on the third day after calving and on monthly int

Lumpy skin disease virus (LSDV) is being developed as a vector for recombinant vaccines against diseases of veterinary importance. A strategy for generating viral thymidine kinase (TK) gene-disrupted recombinants which are stable and homogeneous using the South African Neethling vaccine strain of LSDV as vector has been developed. To assist with the selection process, the Escherichia coli beta-galactosidase (lacZ) visual marker gene was incorporated into the constructs. However, the use of lacZ has certain limitations. An improved strategy was then devised substituting lacZ with the enhanced g
